Abstract
The invention provides a composition with an elastase inhibition effect and an eye and face tightening essence. The composition is prepared by carefully selecting special plants such as peach gum, monkey bread tree pulp, saffron pistil and the like, reasonably mixing, and extracting by a modern biological fermentation technology. Experiments prove that the skin-care gel has good elastase activity inhibition capability, and can avoid elastin loss when being applied to cosmetics, so that the skin reappears Q, is compact, tender and full, and can be used for rejuvenating and brightening.

Background
Skin is a very complex body organ whose biomechanical properties are the result of complex interactions between different layers of skin (e.g. epidermis, dermis and subcutaneous tissue). Reflected in the combination of elastic, viscous and plastic properties of the skin biomechanics.
Skin elasticity is the ability of the skin to stretch and recover its original shape. Elastin is a type of fibrin that is present in the extracellular matrix and is critical to the elasticity of the lungs, arteries, ligaments and skin. During maturation and aging of the human body, the dissolution of elastin accelerates the development of aging diseases. When elastin is excessively lost, skin elasticity is gradually lost, and when severe, skin sags, and the appearance of aging is presented. One of the focus of research in the cosmetic field is how to keep skin well elastic.
For the above-mentioned cosmetics for improving skin elasticity, plant-derived materials have been more advantageous in skin irritation or safety than chemically synthesized materials, and are also more acceptable to consumers. In recent years, researches of a plurality of scholars find that some plants or Chinese herbal medicines have the effect of inhibiting elastase, and a plurality of manufacturers and brands of cosmetics containing plant components are introduced, so that the realization of skin elasticity maintenance through plant-derived raw materials has become an industry hotspot.
Peach gum is a nutrient secreted by peach trees after injury for healing wounds, and is collected in summer and autumn, and the components of peach gum mainly comprise galactose, rhamnose, alpha-glucuronic acid, carbohydrate, fat, protein and the like. Plum fruit in the "Ben Cao gang mu" says that peach gum is: when the peaches are luxuriant, cutting bark by a knife, overflowing glue for a long time, harvesting, soaking by Sang Huishang, and exposing to dry. The peach gum is shiny and amber, is semitransparent, elastic, is like jelly, is also called peach oil, peach fat and peach tree gum, has flat property, sweet and bitter taste, is nontoxic, and has the effects of harmonizing blood and tonifying qi, promoting the production of body fluid to quench thirst, relaxing bowel, nourishing skin and tendering skin and the like. Peach gum has rich resources in hilly and mountainous areas in China, but the original peach gum has the problems of difficult dissolution, poor chemical stability, poor thermal stability and the like, and restricts the application of the peach gum in industry.

In some embodiments, the cosmetic composition may further comprise plant materials extracted from the following parts by weight: 10 parts of red sage root, 3 parts of baikal skullcap root and 5 parts of tree peony root bark.
The peach gum is a reddish brown or yellowish brown gum-like substance secreted from the bark of peach trees. Has mild nature, sweet and bitter taste, no toxicity, and has the effects of regulating blood, invigorating qi, promoting salivation, quenching thirst, loosening bowel to relieve constipation, caring skin, and caring skin.
The monkey-bread tree pulp is mature fruit of monkey-bread tree of kapok family. Monkey bread is the whole body drug. Fruit, leaf and bark can be used as medicine for relieving inflammation, defervescing and treating malaria. The invention relates to red sage root, which is the dried root and rhizome of red sage root of Labiatae. Has effects of promoting blood circulation, removing blood stasis, dredging channels, relieving pain, clearing heart fire, relieving restlessness, cooling blood, and resolving carbuncle.
The radix scutellariae related by the invention is the dried root of the perennial herb radix scutellariae of the genus radix scutellariae of the family Labiatae. Has effects of clearing heat, eliminating dampness, purging pathogenic fire, removing toxic substances, stopping bleeding, and preventing miscarriage.
The tree peony root bark related by the invention is a dried tree peony root bark of the Ranunculaceae plant. Bitter and pungent, slightly cold, and good effects in clearing heat and cooling blood, promoting blood circulation and removing blood stasis, and removing deficiency heat.
In some embodiments of the present invention, there is also provided a method for preparing the cosmetic composition, comprising the steps of: (1) Cleaning peach gum, soaking in clear water, and then treating at high temperature and high pressure to obtain peach collagen liquid; (2) Crushing monkey bread pulp and crocus sativus pistil, adding water, peach collagen liquid and honey, mixing uniformly, and sterilizing under high pressure to obtain a culture medium; (3) inoculating bifidobacterium seed liquid and fermenting; (4) And after fermentation, adding a butanediol aqueous solution, stirring and extracting, press-filtering to obtain a filtrate, and centrifuging at a high speed to obtain a supernatant.

The cosmetic composition prepared by the embodiment of the invention can be used as an additive in cosmetics, and the cosmetics can be cream, emulsion, essence, facial mask and toner according to dosage form.
The embodiment of the invention also provides an eye and face tightening essence, which comprises the following components in percentage by mass: 1-5% of 1, 3-propanediol, 1-5% of corn gluten amino acid, 1-5% of saccharide isomer, 0.1-1% of glycerolyether-26, 0.1-1% of glyceroglycoside, 0.1-1% of hydroxypropyl cyclodextrin, 0.1-0.5% of acrylamide dimethyl taurate/VP copolymer, 0.4-0.6% of 1, 2-hexanediol, 0.4-0.6% of p-hydroxyacetophenone, 1-10% of the cosmetic composition and the balance of water.

(1) Cleaning peach gum, and soaking in clear water, wherein the weight ratio of the peach gum to the clear water is 1:10; then treating for 2 hours at 130 ℃ and 0.5MPa to obtain peach collagen liquid;
(2) Crushing the monkey bread pulp and the crocus sativus pistil, and then adding water, peach collagen liquid and honey, wherein the weight ratio of the monkey bread pulp to the crocus sativus pistil to the water to the peach collagen liquid to the honey is 1:1:50:50:1; uniformly mixing, and sterilizing at 120 ℃ and 5MPa for 1 hour to obtain a culture medium;

Test 1 elastase Activity inhibition test
Elastase, which is a proteolytic enzyme with extremely high selectivity and specificity, has a catalytic hydrolysis effect on carboxyl-containing polypeptide bonds of many amino acids such as glycine, leucine, alanine, valine, etc., and can decompose elastin in connective tissue proteins. The elastase inhibitor can effectively inhibit the activity of elastase and slow down the degradation speed of elastin.
According to the reference: li Xiaojing, sun Peidong et al, inhibition of elastase by Polygonum cuspidatum extract [ J ]. Natural product research and development, 2012,24 (03): 378-380. The cosmetic compositions prepared in examples 1-3 and comparative examples 1-3 were subjected to an elastase activity inhibition test by the method described.
The specific test steps are as follows:
taking a plurality of conical flasks, sequentially adding Congo red-elastin and borate buffer solution, oscillating for 10min at constant temperature in a water bath at 37 ℃, respectively adding elastase solution and different test objects, and measuring the absorbance of each tube at 495 nm.
The inhibition rate of the test substance on the enzyme activity is calculated as follows:
inhibition (%) = (1-An/A0) ×100%;
wherein A0 is the initial absorbance value without the addition of the test substance.
FIG. 1 is a bar graph showing the inhibition of elastase by the test substances (1% aqueous solution of the cosmetic compositions prepared in examples 1 to 3 and comparative examples 1 to 3, 1mg/mL of polydatin standard as a positive control).
As can be seen from FIG. 1, the cosmetic compositions prepared in examples 1 to 3 of the present invention have good elastase inhibitory activity.
Example 5
An eye and face tightening essence comprises the following components in percentage by mass: 1, 3-propanediol 1%, corn gluten amino acid 5%, saccharide isomer 1%, glycerolyether-26%, glyceroglycoside 0.1%, hydroxypropyl cyclodextrin 1%, ammonium acryloyldimethyl taurate/VP copolymer 0.1%, 1, 2-hexanediol 0.6%, p-hydroxyacetophenone 0.4%, 10% of the cosmetic composition prepared in example 3, and the balance of water.
The preparation process comprises the following steps: adding water, hydroxypropyl cyclodextrin, 1, 2-hexanediol and p-hydroxyacetophenone into an emulsifying pot, then adding 1, 3-propanediol, glycereth-26 and acrylamide-dimethyl taurate/VP copolymer which are uniformly mixed in advance, heating to 85 ℃, and preserving heat and stirring for 30 minutes; cooling to 48 deg.C, sequentially adding corn gluten amino acids, saccharide isomer, glyceroglycosides, and cosmetic composition, and stirring; and discharging after the inspection is qualified.
Example 6
An eye and face tightening essence comprises the following components in percentage by mass: 5% of 1, 3-propanediol, 1% of corn gluten amino acids, 5% of saccharide isomer, 0.1% of glycerolyether-26, 1% of glyceroglycosides, 0.1% of hydroxypropyl cyclodextrin, 0.5% of ammonium acryloyldimethyl taurate/VP copolymer, 0.4% of 1, 2-hexanediol, 0.6% of p-hydroxyacetophenone, 1% of the cosmetic composition prepared in example 4 and the balance of water. The preparation process is the same as in example 5.
Trial 2 crowd trial test
30 female volunteers 35-60 years old were summoned, and the eye-surface tightening essence prepared in example 5 was subjected to a crowd trial test. Firstly, a subject is required to be well matched with a test, and a living rule is kept during the trial of a product; all contents of the informed consent form can be read and understood and signed voluntarily; the use of skin care products was discontinued during the trial period, no further clinical trials at any other study center were taken and no cosmetic, pharmaceutical and health care products having any effect on the results were agreed to be used.
2.1 Patch test
The skin closed patch test was performed on volunteers according to the method described in the cosmetic safety Specification (2015 edition) for human skin patch test.
The test object is placed in the plaque laboratory in an amount of about 0.020g to about 0.025g. The test substance is cosmetic product original substance, and the control hole is blank (without any substance). The patch test with the test object is applied to the back or forearm of the subject by hypoallergenic tape, and is applied to the skin with palm light pressure for 24 hr. Skin reactions were observed according to the criteria of Table 1 for 30min (after disappearance of the indentations), 24h and 48h, respectively, after removal of the subject plaque tester.

The test results of the skin-sealing patch show that no skin reaction of more than grade 2 occurs in 30 persons after 30 minutes (after the indentation disappears), 24 hours and 48 hours. Therefore, the eye and face tightening essence prepared by the embodiment of the invention does not show potential adverse reaction to human skin.
2.2 efficacy assessment
There are various methods for measuring skin elasticity, and more commonly used in the art is the skin elasticity meter (Cutometer MPA 580) which uses suction force (vacuum) perpendicular to the skin surface as stress.
Test arrangement: after skin cleaning, a proper amount of samples are taken and rubbed on the facial skin of a volunteer, and the samples are taken once in the morning and evening. Before use, after use for 4 weeks and after use for 8 weeks, skin elasticity is measured by a skin elastometer, and the probe repeatedly measures 3 times in the same measured area during measurement, and an average value is obtained. The specific test results are shown in table 2. Wherein the output data R7 of the skin elastometer represents the ratio of the timely rebound quantity Ur of the skin without negative pressure to the maximum stretching quantity Uf of the skin with negative pressure, and represents the compaction degree of the skin, and the numerical value is as good as the closer to 1.
