Abstract
本发明涉及微生物和食品技术领域,具体涉及到一种微小根毛霉及其在生产替代蛋白中的应用。该菌株的保藏编号为CGMCC NO.40441,可利用简单的底物快速生长,发酵获得蛋白含量高于60%的菌丝体,氨基酸种类丰富,同时富含膳食纤维、不饱和脂肪酸、以及多种矿物质。本发明的微小根毛霉菌株在替代蛋白制品及其它供人类食用的食品中有广泛的应用价值。

技术领域
本发明涉及食品技术领域,具体涉及到一株产菌丝蛋白的微小根毛霉及其在生产替代蛋白中的应用。
背景技术
动物肉是人类饮食中重要的一部分,不仅因为它富含蛋白质、维生素等营养物质,还因为它独特的质构和口感。然而全球人口增长导致对蛋白质的需求飙升,传统的畜牧业将不足以提供足够的蛋白质供人类消费。另一方面,基于环保健康等理念越来越多的人转向弹性饮食。非动物源的可食用产品可在成分和质地上模仿肉类,被认为是动物蛋白产品的理想替代品。
随着科技的进步,近年来出现了许多新兴的肉类替代品。其中以植物蛋白为原料,通过挤压等技术模仿动物肉质构的产品已经在在市场上销售,然而在营养均衡和感官特性方面仍存在不足。另一种解决方案是菌丝蛋白,即从丝状真菌中提取蛋白质作为食品加工原料。菌丝蛋白具有诸多优点,富含高比例蛋白质、必需氨基酸、膳食纤维、维生素、矿物质、不饱和脂肪酸等营养成分。菌丝蛋白原料与肉类纹理高度相似,具有令人愉悦的稠度和味道,非常适合用于食品生产。同时,丝状真菌的生长速度快,可在连续发酵中培养,单位面积产率高,不受环境和气候等因素的影响,保证生产既快速又高效。目前已有多个菌丝蛋白产品已经通过GRAS认证,被用作人类饮食中蛋白质和微量营养素补充物。如英国Quorn公司的Mycoprotein、美国Nature’s Fynd公司的Fy protein、以及瑞典Mycorena公司的Promyc,均已用于食品开发。
事实上,丝状真菌在人类食品中有很长的使用历史,特别是在发酵食品过程中,不仅可以提高原料的营养价值,还可以抑制其他致病微生物带来的潜在风险。普洱茶作为我国特有的发酵黑茶产品,是获取高价值真菌的资源宝库。许多研究表明,普洱茶生产的各个工艺中都有丝状真菌的参与,比如黑曲霉、米根霉、微小根毛霉等。当前,微小根毛霉常用于生产食品工业相关的酶(脂肪酶、淀粉酶、半乳糖醛酸酶)。最近,荷兰The protein brewery公司应用微小根毛霉开发真菌蛋白产品Fermotein,其在过敏性、毒素、次级代谢产物等方面的评估均表明该真菌蛋白的食用安全性。可以预见,未来利用微小根毛霉发酵产生的真菌生物质制备替代蛋白原料,具有巨大的发掘空间。
发明内容
针对现实的需求,本发明人经过分离筛选获得一株高效生产菌丝蛋白的微小根毛霉菌株,其能够利用简单的底物,转化为蛋白含量的菌丝体产物。
因此,本发明首先提供一株微小根毛霉FF23,于2022年12月14日保藏于中国微生物菌种保藏管理委员会普通微生物中心(简称CGMCC),保藏单位地址为在北京市朝阳区北辰西路1号院3号,保藏编号为CGMCC NO. 40441。所述微小根毛霉FF23是从普洱茶中分离获得,经鉴定是微小根毛霉Rhizomucorpusillus。
本发明由此提供所述的微小根毛霉在生产替代蛋白的应用。
进一步地,本发明提供利用所述的微小根毛霉生产菌丝蛋白的方法,其特征在于,通过所述微小根毛霉的发酵以产生菌丝蛋白,其中所述微小根毛霉优选的在化学成分确定的培养基中生长或培养。
在一些具体的实施方式中,所述培养基中无机氮源为磷酸铵、硫酸铵或酒石酸铵。更优选的,所述无机氮源为磷酸二氢铵。进一步优选的,磷酸二氢铵在发酵中提供1.5-3.5g/L的氮源,更优选的添加量为2.5 g/L。
在一些具体的实施方式中,所述碳源为葡萄糖,进一步优选的,所述微小根毛霉能够耐受10-40 g/L的碳源。更优选的,碳源添加量在10 g/L。
在另一些具体的实施方式中,所述发酵培养基pH范围广,进一步优选的发酵培养基pH 范围在3.5-5.5,更优选的pH为3.5。
在优选的实施方案中,在本发明的方法中生长的嗜热真菌是不需要在化学成分确定的培养基中存在任何维生素的菌株。
在一些具体实施方式中,所述微小根毛霉的蛋白质含量高于60%,氨基酸组成丰富,富含人体所需的9种必须氨基酸,且必须氨基酸占比45.27%。
在另一些实施方式中,所述微小根毛霉产物在替代蛋白产品中作为优质脂肪的来源,其中总脂肪在干重中占比9.46%,进一步地,不饱和脂肪酸在脂肪中占比65.01%。
在另一些实施方式中,所述微小根毛霉产物在替代蛋白产品中作为矿物质元素钙、镁、钾、钠、磷来源。
在另一些实施方式中,所述微小根毛霉产物含有丰富的膳食纤维。
本发明的微小根毛霉来源于我国传统发酵食品普洱茶。其能够耐受37℃以上高温、适应低PH环境,利用简单的碳源、氮源快速生长,生物质转化率高,获得的菌丝体中菌丝蛋白的含量高于60%,氨基酸种类丰富,具有良好的食品应用基础。该微小根毛霉产物种类丰富,可发酵获得包括蛋白质、脂质、膳食纤维、矿物质等多种营养成分,具有广泛的应用价值。本发明的微小根毛霉真菌生物质可用于肉类替代品、肉类增量剂、富含蛋白质的食品、富含纤维的食品或新型含真菌蛋白的食品。

具体实施方式
下面通过具体实施例对本发明的技术方案进行清楚、完整的描述。所描述的实施例仅是本发明一部分实施例,而不是全部实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
下述实施例中所使用的实验方法如无特殊说明,均为常规方法。实施例中所用的材料、试剂等,如无特殊说明,均可从商业途径得到。
实施例1:普洱茶中耐热真菌的筛选
普洱茶样品于2022年06月收集自云南西双版纳地区产茶基地。将样品在均质机中打碎,使用倾注法分离样品中的微生物菌株。
倾注法具体是指称取0.1 g粉碎后的茶叶样品,加入40 mL 0.1%蛋白胨水溶液中,28℃ 150r/min 振荡1h,倒入冷却至45℃的40 mL双料DRBC培养基混匀,每80 mL培养基倒2个方形平板。将处理后的平板置于37℃培养5-10 d。根据菌落表型特征(包括大小、形状、颜色、质地、有无渗透液、有无可溶性色素等)挑取菌落于PDA平板上进行分离纯化。分离纯化次数大于等于3次,直至获得纯菌落。从样品中获得不同表型的耐热真菌144株。
实施例2:真菌基因组的提取及鉴定
收集PDA平板上生长的菌丝体,通过真菌基因组提取试剂盒提取总DNA,然后采用真菌通用引物扩增真菌ITS区域。扩增产物经1%琼脂糖凝胶电泳分离后,采用胶回收试剂盒回收后测序。采用BLAST方式将其与GenBank数据库中的序列进行比对。BLAST比对将144株真菌鉴定为Rhizomucor、Aspergillus、Lichtheimia、Schizophyllum、Irpex、Trametes、Penicillium、Alternaria、Coprinellus、Cubamyces等10个属的真菌。鉴定后的菌株保存在10%甘油中,于-80℃保藏。
进一步的,其中18株分离菌株被鉴定为微小根毛霉,命名为RP-1~RP-18。
实施例3:微小根毛霉菌株生长能力筛选
在包含葡萄糖40 g/L、酒石酸铵10 g/L、KH2PO47 g/L、MgSO4·7H2O 1.5 g/L、CaCl2·2H2O 0.1 g/L、Na2HPO42 g/L、ZnSO4·7H2O 1 mg/L、FeCl3·6H2O 8 mg/L、Co(NO3)2·6H2O 0.1 mg/L、MnSO4·5H2O 0.1 mg/L、CuSO4·5H2O 0.1 mg/L,琼脂20 g/L,pH 6.0的培养基平板上评估微小根毛霉的生长能力。
孢子悬浮液以1×105个点样到培养皿中央,置于35 ℃静置培养。在1-5 d内测量菌落直径以评价菌株生长能力。
18株菌在平板上培养5 d后,有7株菌生长速度较慢,菌落直径较小,未用作进一步筛选。其余11株菌中,RP-3、RP-4、RP-5、RP-16、RP-17培养4d时,生长直径达到85 mm,RP-1、RP-2、RP-7、RP-10、RP-13、RP-14培养5d时,生长直径达到85 mm。这11株菌有较快的生长速度,用作下一步筛选。
实施例4:微小根毛霉菌株蛋白生产能力筛选
将11株微小根毛霉以1×106CFU/mL的孢子接种到100 mL液体培养基发酵,以评估其蛋白生产能力(表1)。在35℃下,220 rpm振荡培养48h。
所述液体发酵培养基组成包括:葡萄糖10 g/L、磷酸二氢铵20.54 g/L、KH2PO47 g/L、MgSO4·7H2O 1.5 g/L、CaCl2·2H2O 0.1 g/L、Na2HPO4 2 g/L、ZnSO4·7H2O 1 mg/L、FeCl3·6H2O 8 mg/L、Co(NO3)2·6H2O 0.1 mg/L、MnSO4·5H2O 0.1 mg/L、CuSO4·5H2O 0.1mg/L,pH 4.5。
发酵结束,过滤收集微小根毛霉菌丝体,于70℃下干燥24h至恒重,测定真菌生物质含量和蛋白浓度。

本发明筛选到的微小根毛霉具有很高的底物转化能力,48h发酵后真菌生物量大于等于3.98g/L。其中有6株微小根毛霉菌株发酵后生物质蛋白浓度高于50%,总蛋白含量高于2g/L。其中微小根毛霉RP-2(RhizomucorpusillusFF23),发酵后生物质产量最高达4.37g/L,蛋白浓度达到52.16%,总蛋白含量最高达到2.28g/L。
实施例5:R.pusillusFF23的形态分析
Schipper在1978年的相关研究对毛霉属菌株及其相关属菌株进行全面分类学修订,将毛霉属和根毛霉属的分类学地位得以分开。根毛霉属菌株与毛霉属菌株的差异在于,根毛霉属菌株具有嗜热特性,同时在形态上有匍匐茎和根状茎的存在。使用R.pusillusFF23的ITS序列进行Blast比对,经MrBayes获得贝叶斯分析法进化树,本发明筛选到的菌株R. pusillusFF23被分类为R. pusillus(图2)。
R. pusillusFF23在PDA平板上生长四天的菌落形态如图3所示。PDA上的菌落在35℃下快速生长,呈丝绒状至絮状,最初为黄褐色,然后变为灰褐色。40℃和45℃生长更快,但在25℃时生长受到抑制。平板上菌丝宽度为11-14µm,无分隔并形成孢子囊,偶尔具有不发育的黄褐色根茎。孢子管载体透明至浅棕色,宽8-18µm,分枝,有时在孢子囊下面稀疏间隔。孢子囊为黄褐色和近球形,壁光滑,直径可达3.8-4.2µm,具有倒卵形至近球形的小柱,平均长20-30 µm。
R. pusillusFF23在显微镜下的的菌丝体形态结构如图3所示。
实施例6:R. pusillusFF23蛋白发酵条件优化
比较不同的氮源种类、碳氮比、PH、葡萄糖含量、发酵时间等因素对R. pusillusFF23发酵生物质蛋白含量的影响。除碳源、无机氮源、pH外,培养基其它组成为KH2PO45-10 g/L、MgSO4·7H2O 1-5 g/L、CaCl2·2H2O 0.1-1 g/L、Na2HPO41-5 g/L、ZnSO4·7H2O 0.1-1mg/L、FeCl3·6H2O 1-10 mg/L、Co(NO3)2·6H2O 0.1-1 mg/L、MnSO4·5H2O 0.1-1mg/L、CuSO4·5H2O 0.1-1 mg/L。
R. pusillusFF23以1×106CFU/mL的孢子数接种到100 mL下述液体培养基发酵,35℃、220rpm发酵。
培养基1:葡萄糖10 g/L、以磷酸氢二铵为氮源、C/N比2.4,pH2.5,发酵36 h,生物质含量0.049 g/100ml,蛋白浓度77.56%,总蛋白含量0.04 g/100ml。
培养基2:葡萄糖20 g/L、以硫酸铵为氮源、C/N比1.6,pH2.5,发酵60h,生物质含量0.021g/100ml,蛋白浓度83.99%,总蛋白含量0.02g/100ml。
培养基3:葡萄糖10 g/L、以磷酸二氢铵为氮源、C/N比1.6,pH3.5,发酵48h,生物质含量0.401g/100ml,蛋白浓度64.6%,总蛋白含量0.259g/100ml。
培养基4:葡萄糖15 g/L、以硫酸铵为氮源、C/N比2.8,pH3.5,发酵36 h,生物质含量0.275g/100ml,蛋白浓度73.00%,总蛋白含量0.20g/100ml。
培养基5:葡萄糖5 g/L、以硝酸钾为氮源、C/N比1.6,pH4.5,发酵36 h,生物质含量0.192g/100ml,蛋白浓度38.95%,总蛋白含量0.07g/100ml。
培养基6:葡萄糖20g/L、以磷酸氢二铵为氮源、C/N比2.8,pH4.5,发酵48h,生物质含量0.294g/100ml,蛋白浓度64.65%,总蛋白含量0.19g/100ml。
培养基7:葡萄糖5 g/L、以硫酸铵为氮源、C/N比2.4,pH5.5,发酵48h,生物质含量0.245g/100ml,蛋白浓度58.66%,总蛋白含量0.14g/100ml。
培养基8:葡萄糖20 g/L、以磷酸二氢铵为氮源、C/N比2,pH5.5,发酵36 h,生物质含量0.859g/100ml,蛋白浓度49.66%,总蛋白含量0.43g/100ml。
上述实验结果表明,PH对生物质和蛋白含量产生最大影响,随着PH的降低,蛋白含量升高,但是PH降至2.5时,严重抑制了R. pusillusFF23的生长;其次是氮源,硝酸钾作为氮源产生最低的生物质和蛋白含量,硫酸铵促进了菌丝蛋白的蛋白浓度,而磷酸二氢铵作为氮源有更高的生物质含量。在实际生产中,生物质含量和蛋白浓度都是重要的参考指标。综合上述发酵条件,以培养基3为最优发酵培养基,发酵后达到0.401g/100ml的生物质含量及64.6%的蛋白浓度。
实施例7:R. pusillusFF23单细胞蛋白的营养价值
加入5-10 mL无菌水至菌株培养平板,并用一次性无菌涂布棒从R. pusillusFF23平板表面刮下孢子,制备孢子悬浮液。在光学显微镜下计数孢子,用作液体发酵的接种物。
将R. pusillusFF23以1×106CFU/mL接种到pH3.5的800mL发酵培养基中。220rpm、35℃培养48h。使用真空抽滤泵过滤,同时使用3倍体积的无菌水清洗。将收集到的生物质滤饼进一步冷冻干燥并研磨,得到干物质含量约3.2g的粉末状微小根毛霉生物质。冻干后的R. pusillusFF23生物质水分含量2.88%。
所述发酵培养基组成为:葡萄糖10 g/L、磷酸二氢铵 20.54 g/L、KH2PO47 g/L、MgSO4·7H2O 1.5 g/L、CaCl2·2H2O 0.1 g/L、Na2HPO4 2 g/L、ZnSO4·7H2O 1 mg/L、FeCl3·6H2O 8 mg/L、Co(NO3)2·6H2O 0.1 mg/L、MnSO4·5H2O 0.1 mg/L、CuSO4·5H2O 0.1 mg/L,pH3.5。
、R. pusillusFF23生物质基本营养成分
冻干后的R. pusillusFF23生物质的营养组成按照表2所述国家准方法进行检测并获得相应的结果。

如表2所示,本发明微小根毛霉生物质的营养价值主要是由于其高蛋白和膳食纤维含量,其干物质中含有64.6%的蛋白以及16.68%的总膳食纤维。R. pusillusFF23生物质含有9.46%的脂肪,其中3.21%为饱和脂肪,3.74%为单不饱和脂肪,2.41%为多不饱和脂肪。因此,R. pusillusFF23的生物质不仅可作为替代蛋白原料,同时也是不饱和脂肪酸的来源。
人类饮食中蛋白质主要来源于动物制品和植物源食品等。这些主要食物的蛋白质含量如表3所示。动物源制品中蛋白质含量在基于干重的30.98%-43.44%不等,植物源制品中蛋白质含量在基于干重的40%-55.72%。本发明所提供的R. pusillusFF23生物质在优化的工艺下达到64.60%的蛋白质含量,远高于其它食品。

2、R. pusillusFF23的氨基酸谱
使用高效液相色谱的方法检测R. pusillusFF23生物质的氨基酸组成,如表4所示。必需氨基酸指苏氨酸、缬氨酸、异亮氨酸、亮氨酸、苯丙氨酸、赖氨酸、蛋氨酸、色氨酸这9种氨基酸。R. pusillusFF23包含这9种必须氨基酸且必须氨基酸占比达到45.27%。该结果表明R. pusillusFF23的必须氨基酸比例与鸡肉(45%)相当,且高于大豆(36%)、牛奶(41%)、以及乳清蛋白(43%)。

3、R. pusillusFF23的矿物质含量
经检测,上述得到的R. pusillusFF23生物质含有多种矿物质,如表5所示。钙、磷、镁对骨骼健康均非常重要。

所有数据均基于干重。
本发明生物质R. pusillusFF23中矿物质含量与其它菌丝蛋白产品相比有更高的P和Fe含量,与生肉(猪肉、牛肉)相比,有更高的Mg、P、Fe、Cu和Mn。磷和镁在人体的组织细胞以及人体的骨骼构成中起着重要的作用,铁参与氧的运输与储存,维持正常造血功能、增强免疫。锰元素是人体内多种生物酶的组成成分,对生长、发育、繁殖和内分泌都有影响。锰与铜一样,参与造血过程,改善人体对铜的利用功能和肌体的造血功能。
应用例1:发酵后生物质的处理
按照实施例7中描述的方法发酵获得生物质。真菌生物质发酵完成后在65℃下热处理30min以去除细胞内RNA。之后将发酵后生物质进一步加热至90℃保持30min,以杀死真菌细胞。之后通过100目的纱布过滤,使用5倍体积的无菌水洗涤以完全除去培养基。获得水分含量85%的生物质用于替代蛋白产品开发。
应用例2:含有R. pusillusFF23生物质的块状产品的生产
按照应用例1所述获得含水量85%的生物质,将其与2%的甲基纤维素和2%的菜籽油混合均匀。通过压饼机压制为厚度为1cm饼状生物质,置于-20℃冷冻24 h后取出放至室温。置于蒸锅中加热30 min,获得块状生物质饼。
将块状生物质饼与橄榄油、蚝油、鸡蛋清、红酒、淀粉等风味物质混合腌制后,可用于汉堡类产品的制作。
应用例3:含有R. pusillusFF23生物质的肉丸的生产
按照应用例2所述的方法获得块状生物质饼,使用绞肉机将所得生物质饼打碎至肉末状。向其中加入水,鸡蛋、少许鸡精,淀粉、蚝油、生抽、料酒、盐等风味物质,混合均匀。将混合物制备为球状肉丸。将球状肉丸在140°C的烤箱中热处理40分钟,并在180°C下油炸2分钟获得含有R. pusillusFF23生物质的肉丸产品。
Claims (6)
1.一种微小根毛霉(Rhizomucorpusillus),其特征在于,于2022年12月14日保藏于中国微生物菌种保藏管理委员会普通微生物中心,其保藏编号为CGMCC NO. 40441。
2.如权利要求1所述的微小根毛霉在生产替代蛋白中的应用。
3.一种利用权利要求1所述的微小根毛霉生产替代蛋白的方法,其特征在于,通过所述微小根毛霉的发酵,从发酵物中获得替代蛋白。
4.如权利要求3所述的微小根毛霉生产替代蛋白的方法,其特征在于,所述发酵的培养基中无机氮源为磷酸二氢铵、硫酸铵或酒石酸铵中的一种或多种。
5. 如权利要求4所述的微小根毛霉生产替代蛋白的方法,其特征在于,所述发酵的培养基中除碳源、无机氮源,培养基的其它组成为:KH2PO4 5-10 g/L、MgSO4·7H2O 1-5 g/L、CaCl2·2H2O 0.1-1 g/L、Na2HPO4 1-5 g/L、ZnSO4·7H2O 0.1-1mg/L、FeCl3·6H2O 1-10 mg/L、Co(NO3)2·6H2O 0.1-1 mg/L、MnSO4·5H2O 0.1-1 mg/L、CuSO4·5H2O 0.1-1 mg/L。
6.如权利要求3至5任一项所述的微小根毛霉生产替代蛋白的方法,其特征在于,所述发酵的培养基的pH在3.5-7.0范围内;在所述发酵完成后,使用真空抽滤泵过滤,使用无菌水清洗,将收集到的生物质滤饼进一步冷冻干燥,得到滤饼生物质或进一步通过研磨,得到粉末状生物质。
