Abstract
本发明公开一种用于检测糖胺聚糖的碳化聚合物点单组件荧光阵列传感器的制备方法,以聚乙烯亚胺和小分子卟啉为前驱体,通过缩合聚合反应一步水热法合成碳化聚合物点,再通过与铜离子溶液共混,制得CPDs‑Cu2+荧光阵列传感器。利用本发明制得的CPDs‑Cu2+荧光阵列传感器能够摆脱高特异性元件与分析物结合传感模式,用于识别和检测生物样本中多种糖胺聚糖分。

本发明属于生物传感器技术领域,具体涉及一种用于检测糖胺聚糖的碳化聚合物点单组件荧光阵列传感器及其制备方法,可实现多靶标检测。
背景技术
糖胺聚糖(Glycosaminoglycans,GAGs)是一类普遍存在于各种组织中的无支链线性多糖,因其亲水性和高负电荷密度在各种病理和生理过程中发挥着关键作用。然而,每种GAGs表现出不同的生物功能和临床应用。例如,肝素(Heparin,Hep)作为负电荷密度最高的,在临床上被广泛用作抗凝血药物,在包括抗病毒、抗癌、心脏病发作及神经系统疾病中具有关键性的作用;硫酸软骨素(Chondroitin sulfate,Chs)通过与各种分子发生特异性相互作用改变细胞间的信号传递,可用于治疗炎症,临床上也可以有效治疗关节炎、风湿病、肩周炎等疾病;透明质酸(Hyaluronic acid,HA)可以与细胞表面受体连接达到激活信号通路的效果,调控细胞功能、组织发育、伤口愈合和肿瘤进展转移,常用于眼科手术和皮肤护理产品;硫酸葡聚糖(Dextran sulfate,DS)被广泛用作降血脂药等;上述这些GAGs的化学结构中有类似的二糖重复单元,因此区分GAGs是非常有难度的。但是,区分不同的GAGs是能够避免严重的安全问题,对于临床应用有巨大意义的。当前GAGs的分析主要是结合核磁共振波谱(NMR)、质谱(MS)、高效液相色谱(HPLC)及电泳法等分析技术进行。但这些方法所用到的设备昂贵,且数据分析需要有经验的技术人员,因此开发简便可靠的GAG质量监测方法具有重要意义。
近年来,荧光传感器以其反应快速、检测限低、技术简单和无创的优点获得了众多科学研究者极大的关注。目前,市面上已经开发了一系列荧光传感器实现离子、小分子、大分子等的检测。然而,传统的荧光传感器需要一个对分析物具有高特异性的受体,即“锁和钥匙”传感模式,使得传统的荧光传感器几乎不可能同时识别不同的分析物。
作为一种性能优异的荧光纳米材料,近年来碳化聚合物点在生物成像、生物传感、光催化、药物传递等领域应用广泛。碳化聚合物点(Carbonized polymer dots,CPDs)是一种由有机聚合物链和高度交联的碳核组成的新型荧光纳米材料。与传统碳点(CDs)不同的是,CPDs通常由碳杂化结构/聚合物组成,而不是碳主体结构,这种独特结构特性使CPDs既具备聚合物的性能,又保留了传统CDs的优异光学性能。此外,CPDs的聚合物链结构使得其很容易与无机材料、聚合物和功能分子共价结合实现功能化。公开号为CN113030042A的专利“一种检测乙醛的荧光传感器及其合成方法与应用”,具体公开了采用水热反应合成碳化聚合物点,并由碳化聚合物点与稀土离子复合而成的传感器,用于在光化学分析、食品化学领域中的乙醛检测。
综上所述,制备能够实现多个目标的高通量测定和模式识别的荧光传感器,将其用于识别生物液体中的GAGs具有现实意义。
发明内容
为了解决现有技术中的问题,本发明考虑引入“化学鼻/舌”策略的传感阵列,以实现对多个目标的高通量测定和模式识别;对于这种策略,不同的分析物会产生差异化的响应,这些独特的响应可以很容易地被收集并用于区分分析物,基于此,本发明提供一种用于检测糖胺聚糖的碳化聚合物点单组件荧光阵列传感器,摆脱了高特异性元件与分析物结合传感模式,可用于检测生物样本中多种糖胺聚糖分。
本发明的技术方案如下:
本发明提供一种用于检测糖胺聚糖的碳化聚合物点单组件荧光阵列传感器的制备方法,以聚乙烯亚胺和小分子卟啉为前驱体,通过缩合聚合反应一步水热法合成碳化聚合物点,再通过与铜离子溶液共混,制得CPDs-Cu2+荧光阵列传感器。
进一步的,所述用于检测糖胺聚糖的碳化聚合物点单组件荧光阵列传感器的制备方法,包括以下步骤:
S1、将聚乙烯亚胺和小分子卟啉分别配制成浓度为50mg/mL和1mg/mL的聚乙烯亚胺水溶液和小分子卟啉水溶液;
S2、将聚乙烯亚胺水溶液和小分子卟啉水溶液加入到去离子水中,在170~190℃水热条件下反应10~12h,当反应冷却到室温时,初步过滤纯化去除聚合颗粒,然后通过超滤离心管进一步纯化,最后冷冻干燥得到碳化聚合物点CPDs;
S3、在磷酸盐缓冲液PBS中配置Cu2+溶液,与碳化聚合物点CPDs均匀混合,在室温下孵育15~20min制备CPDs-Cu2+荧光阵列传感器。
进一步的,所述S1中小分子卟啉为5,10,15,20-四(4-羧基苯基)卟啉。
进一步的,所述S2中聚乙烯亚胺水溶液、小分子卟啉水溶液和去离子水的体积比为1:1.2:12。
进一步的,所述S2中初步过滤纯化采用0.22μm的水相滤膜;所述超滤离心管的分子量分别为10000和30000,进行两次超滤离心纯化。
进一步的,所述S3中Cu2+溶液浓度为10~40μM;所述碳化聚合物点CPDs浓度为0.1mg/mL。
本发明还提供一种用于检测糖胺聚糖的碳化聚合物点单组件荧光阵列传感器,采用用于检测糖胺聚糖的碳化聚合物点单组件荧光阵列传感器的制备方法制备而成。
本发明进一步提供一种碳化聚合物点单组件荧光阵列传感器在生物样本中同时识别检测多种糖胺聚糖分子的应用。
进一步的,将制得的碳化聚合物点单组件荧光阵列传感器反应体系与生物样本振荡混匀后,用荧光酶标仪360nm激发后检测667nm和469nm发射荧光值。
相较于现有技术,本发明的有益效果在于:
1、与现有的荧光传感器相比,本发明制备的聚合碳点荧光传感器摆脱了必须借助高特异性元件才能与单一分析物结合的传感模式,通过CPDs-Cu2+荧光阵列传感器与分析物结合形成的荧光指纹图谱形成传感阵列,从而实现多元分析物高通量检测,在本发明中可检测生物样本中多种糖胺聚糖分子;对于不同的GAGs具有不同数量的磺酸基团,与金属离子具有不同的结合位点,由此产生了不同的相互作用力,所以GAGs能够以不同的力度与表面的Cu2+结合,通过相互竞争原理使CPDs从CPDs-Cu2+中脱离,随着CPDs-Cu2+传感器中GAGs的引入,由于Cu2+淬灭CPDs不同荧光发射峰的程度有差异,GAGs对传感器的不同作用程度可以转化为差异化的光学信号模式,再将收集到的数据矩阵通过主成分分析(PCA)方法进行分析识别,从而实现传感元件对生物样本中多种GAGs的检测;此外,还可以基于荧光指纹图谱开发更多类似结构的生物标志物;
2、与其他碳点纳米生物传感器相比,本发明制得的聚合碳点元件材料在保留碳点材料优异的光学性能的同时具有聚合物的优势,碳化聚合物点CPDs同时具备传统碳点CDs和小分子卟啉TCPP的光学特性,增加了传感器的光学可调性、功能多元性和生物兼容性;
3、本发明提供水热法制备碳化聚合物点CPDs,水热反应进行的聚合和碳化过程中,带有氨基的PEI被碳化到更大程度;而带有羧基的TCPP单元由于被PEI包裹着,则部分碳化,小分子TCPP不仅贡献了一个强的荧光发射峰,而且起到了交联剂的作用,与PEI通过氨基和羧基之间的反应联结形成更大的聚合物,用聚合物和小分子作为共聚物通过水热法获得的集成式CPDs,不仅可以保持TCPP原有的性能,而且还提高了TCPP的水溶性和光稳定性;
4、与传统成熟应用的质谱、核磁共振等检测技术相比,本发明利用碳化聚合物点单组件荧光阵列传感器进行糖胺聚糖检测,方法具有成本低廉、操作简单、不依赖大型设备、特异性高、辨识度高等优势。
5、本发明的检测传感器平台在检测应用中,试剂成本接近生化反应平台,操作可适配疫情以来各医疗机构普遍配置的荧光定量PCR仪,无需额外购置检测设备,检测实验操作简单,拥有荧光传感检测的灵敏度和特异性,同时具备更多生物标记物开发的基础。

具体实施方式
为了使本发明要解决的技术问题、技术方案及有益效果更加清楚明白,以下结合实施例,对本发明做进一步的详细说明。应当理解,此处所描述的具体实施例仅仅用以解释本发明,并不用于限定本发明。
实施例1
如图1所示,本实施例提供一种用于检测糖胺聚糖的碳化聚合物点单组件荧光阵列传感器的制备方法,以聚乙烯亚胺和小分子卟啉为前驱体,通过缩合聚合反应一步水热法合成碳化聚合物点,再通过与铜离子溶液共混,制得CPDs-Cu2+荧光阵列传感器,具体包括以下步骤:
S1、将聚乙烯亚胺PEI和小分子卟啉TCPP分别配制成浓度为50mg/mL和1mg/mL的聚乙烯亚胺水溶液和小分子卟啉水溶液;在本实施例中小分子卟啉为5,10,15,20-四(4-羧基苯基)卟啉;
S2、将聚乙烯亚胺水溶液和小分子卟啉水溶液加入到去离子水中,聚乙烯亚胺水溶液、小分子卟啉水溶液和去离子水的体积比为1:1.2:12;在本实施例中,将1mL聚乙烯亚胺PEI水溶液和1.2mL小分子卟啉水溶液加入量到12mL去离子水中;在170℃水热条件下反应12h,当反应冷却到室温时,采用0.22μm的水相滤膜进行初步过滤纯化去除聚合颗粒,然后分别通过分子量为10000和30000的超滤离心管进一步纯化,最后冷冻干燥得到碳化聚合物点CPDs;
S3、在磷酸盐缓冲液PBS(1mM,pH=7.5)中配置Cu2+溶液,其中,Cu2+溶液浓度为20μM,与经过S2制得的碳化聚合物点CPDs均匀混合,碳化聚合物点CPDs浓度为0.1mg/mL,在室温下孵育15min制备CPDs-Cu2+荧光阵列传感器。
实施例2
本实施例提供一种用于检测糖胺聚糖的碳化聚合物点单组件荧光阵列传感器的制备方法,包括以下步骤:
S1、将聚乙烯亚胺PEI和小分子卟啉TCPP分别配制成浓度为50mg/mL和1mg/mL的聚乙烯亚胺水溶液和小分子卟啉水溶液;除实施例1中小分子卟啉为5,10,15,20-四(4-羧基苯基)卟啉外,在本实施例中小分子卟啉还可以为其它四羟基卟啉;
S2、将聚乙烯亚胺水溶液和小分子卟啉水溶液加入到去离子水中,在本实施例中,将5mL聚乙烯亚胺PEI水溶液和6mL小分子卟啉水溶液加入到60mL去离子水中;在190℃水热条件下反应10h,当反应冷却到室温时,采用0.22μm的水相滤膜进行初步过滤纯化去除聚合颗粒,然后分别通过分子量为10000和30000的超滤离心管进一步纯化,最后冷冻干燥得到碳化聚合物点CPDs;
S3、在磷酸盐缓冲液PBS(1mM,pH=7.5)中配置Cu2+溶液,其中,Cu2+溶液浓度为40μM,与经过S2制得的碳化聚合物点CPDs均匀混合,碳化聚合物点CPDs浓度为0.1mg/mL,在室温下孵育20min制备CPDs-Cu2+荧光阵列传感器。
实施例3
本实施例提供一种用于检测糖胺聚糖的碳化聚合物点单组件荧光阵列传感器的制备方法,包括以下步骤:
S1、将聚乙烯亚胺PEI和小分子卟啉TCPP分别配制成浓度为50mg/mL和1mg/mL的聚乙烯亚胺水溶液和小分子卟啉水溶液;
S2、将聚乙烯亚胺水溶液和小分子卟啉水溶液加入到去离子水中,在本实施例中,将10mL聚乙烯亚胺PEI水溶液和12mL小分子卟啉水溶液加入到120mL去离子水中;在180℃水热条件下反应11h,当反应冷却到室温时,采用0.22μm的水相滤膜进行初步过滤纯化去除聚合颗粒,然后分别通过分子量为10000和30000的超滤离心管进一步纯化,最后冷冻干燥得到碳化聚合物点CPDs;
S3、在磷酸盐缓冲液PBS(1mM,pH=7.5)中配置Cu2+溶液,其中,Cu2+溶液浓度为10μM,与碳化聚合物点CPDs均匀混合,碳化聚合物点CPDs浓度为0.1mg/mL,在室温下孵育18min制备CPDs-Cu2+荧光阵列传感器。
实施例4
本实施例提供一种用于检测糖胺聚糖的碳化聚合物点单组件荧光阵列传感器,根据上述实施例1的制备方法制得;制得的碳化聚合物点单组件荧光阵列传感器形态和尺寸通过透射电子显微镜(TEM)进行表征,如图2A和2B可看出,在水热条件下所合成的碳化聚合物点CPDs显示出分散均匀的球形结构,平均直径为25.76nm;使用FT-IR和XPS分析CPDs的表面化学组成,如图2C所示,主要有3440和1650cm-1两个明显的吸收峰,对应着O-H/N-H和C=O基团的拉伸振动峰,表明CPDs中存在氨基、羧基和羟基;图3A的XPS图谱显示,在284.80、398.30和530.43eV处有三个明显的峰,分别是C1s、N 1s和O1s,其原子占有率分别为72.02%、20.13%和7.85%;C 1s的高分辨率能谱在287.08、284.79、283.91eV处表现出三个峰,对应于C=O、C-N/C-O和C=C/C的结合能(如图3B);此外,高分辨XPS能谱(N 1s和O1s)显示,CPDs中存在C-N,C-O,C=O和N-H键等(如图3C和3D);上述结果表明,水热法成功制备CPDs,水热反应进行的聚合和碳化过程中,带有氨基的PEI被碳化到更大程度;而带有羧基的TCPP单元由于被PEI包裹着,则部分碳化,具体而言,小分子TCPP不仅贡献了一个强的荧光发射峰,而且起到了交联剂的作用,与PEI通过氨基和羧基之间的反应联结形成更大的聚合物,用聚合物和小分子作为共聚物通过水热法获得的集成式CPDs,不仅可以保持TCPP原有的性能,而且还提高了TCPP的水溶性和光稳定性;
本实施例中,在温和条件下制备的传感元件碳化聚合物点CPDs具有双发射峰的光学性能,可开发为比率荧光探针传感器;在pH=7.5的反应体系中,激发波长为330nm到400nm,CPDs具有469nm和667nm处明显的荧光发射峰,360nm激发波长为最大激发波长(如图4A);激发光强度受反应体系pH影响较小,pH在4.0-8.5区间内均可进行反应,其中CPDs的荧光强度及双发射波长比率(F667/F469)在pH7.5时呈现最大值;随着反应体系中Cu2+的加入,会产生Cu2+对传感元件CPDs的荧光淬灭作用,如图4B所示,固定CPDs的浓度(0.1mg/mL),将不同浓度的Cu2+与之均匀混合,Cu2+的浓度从0到60μM变化的同时,667nm处的荧光强度明显逐步减弱,而469nm处的荧光强度缓慢减弱,这直接导致F667/F469值的相应减小。
实施例5
CPDs-Cu2+带有正电荷,而不同种类的糖胺聚糖GAGs具有不同程度的负电荷,且具有不同位置的亲水基团,与带正电荷的CPDs-Cu2+会有不同的附着位点,具有不同的亲和力,进而产生差异化的光学信号;在本实施例中将四种糖胺聚糖GAGs肝素(Hep)、硫酸软骨素(Chs)、透明质酸(HA)、硫酸葡聚糖(DS)各自50μg/mL的浓度分别加入到依据本发明制得的CPDs-Cu2+传感器阵列溶液中,测试传感器对不同GAGs的荧光信号(即F469、F667和F667/F469),将收集到的荧光信号转化为数据柱状图(1-F/F0,F0为添加GAGs前的反应液初始荧光值,F为添加GAGs后的检测荧光值)和相应的热图,图5A和5B显示了CPDs-Cu2+传感器对4种GAGs的不同响应程度;使用PCA法通过降低数据的维度将原始数据集变换为三个主要组成部分,选用其中两个主成分来绘制二维PCA图,其中一个点代表特定CPDs-Cu2+传感器阵列对GAGs的一次平行信号,由于GAGs与Cu2+存在配位作用,不同的GAGs可在不同程度上影响CPDs-Cu2+传感器的荧光发射强度,产生不同的信号,因此,图5C中显示了4种GAGs划分的4个明显离散的集群,确定了依据本发明制得的CPDs-Cu2+传感器的识别能力;
CPDs-Cu2+传感器还可使用PCA法对共存GAGs进行检测识别,以DS/Chs和HA/Hep=50/0、40/10、30/20、25/25、20/30、10/40和0/50(μg/mL)的混合物作为分析物(如图6);对于不同摩尔比的DS/Chs混合物,荧光信号是不同的,荧光响应模式在PCA结果中是明显离散的(如图6A和6B);同样的结论也可以在HA/Hep的混合样品中得到,每个摩尔比均显示出独特的荧光信号,这使得它们能够在PCA图中被准确地分辨出来(图6C和6D)
实施例6
依据本发明提供的方法制得的CPDs-Cu2+传感器可使用PCA法对不同浓度的某一种GAGs实现半定量识别;以Hep检测为例,如图7A所示,随着Hep浓度的增加,F469、F667和F667/F469的淬灭率是呈一定梯度变化的,传感器对分析物Hep的信号差异可以直观地表示为热图(如图7B),它们呈现出渐变色的趋势,同时PCA图也遵循一定的规律,即相同浓度的数据点聚集成簇,不同浓度数据点之间明显区分开无重叠(如图7C),拟合Hep浓度和PC1值的相关线性曲线,相对系数(R2)可达到0.9643(如图7D)。
实施例7
本实施例提供一种碳化聚合物点单组件荧光阵列传感器在生物样本中同时识别检测多种糖胺聚糖分子的应用,具体步骤如下:
(1)将依据实施例1制备好的CPDs-Cu2+单组件荧光阵列传感器反应体系加入黑色样孔板中(80μL),荧光酶标仪360nm激发后检测背景667nm和469nm发射荧光值;
(2)反应体系中加入血清样本(黄帽采血管离心后分离得到)20μL,振荡混匀后用荧光酶标仪360nm激发后检测667nm和469nm发射荧光值;
(3)计算统计F667、F469和F667/F469的值,进而进行PCA法得到结果。
在本实施例中使用依据本发明制得的传感器CPDs-Cu2+检测胎牛血清中人工混合的GAGs标准品,如图8所示,传感器对不同组合类型的GAGs(DS/Chs和HA/Hep按照摩尔比50/0、40/10、30/20、25/25、20/30、10/40和0/50)有不同的荧光信号,对不同的GAGs,热图呈现着交叉式的彩色差异,通过降维处理,在PCA图中表现为明显分离的簇,确定传感器对血清中的GAGs具有良好的分辨能力。

Claims (9)
1.一种用于检测糖胺聚糖的碳化聚合物点单组件荧光阵列传感器的制备方法,其特征在于:以聚乙烯亚胺和小分子卟啉为前驱体,通过缩合聚合反应一步水热法合成碳化聚合物点,再通过与铜离子溶液共混,制得CPDs-Cu2+荧光阵列传感器。
2.如权利要求1所述的一种用于检测糖胺聚糖的碳化聚合物点单组件荧光阵列传感器的制备方法,其特征在于,包括以下步骤:
S1、将聚乙烯亚胺和小分子卟啉分别配制成浓度为50mg/mL和1mg/mL的聚乙烯亚胺水溶液和小分子卟啉水溶液;
S2、将聚乙烯亚胺水溶液和小分子卟啉水溶液加入到去离子水中,在170~190℃水热条件下反应10~12h,当反应冷却到室温时,初步过滤纯化去除聚合颗粒,然后通过超滤离心管进一步纯化,最后冷冻干燥得到碳化聚合物点CPDs;
S3、在磷酸盐缓冲液PBS中配置Cu2+溶液,与碳化聚合物点CPDs均匀混合,在室温下孵育15~20min制备CPDs-Cu2+荧光阵列传感器。
3.如权利要求2所述的一种用于检测糖胺聚糖的碳化聚合物点单组件荧光阵列传感器的制备方法,其特征在于,所述S1中小分子卟啉为5,10,15,20-四(4-羧基苯基)卟啉。
4.如权利要求2所述的一种用于检测糖胺聚糖的碳化聚合物点单组件荧光阵列传感器的制备方法,其特征在于,所述S2中聚乙烯亚胺水溶液、小分子卟啉水溶液和去离子水的体积比为1:1.2:12。
5.如权利要求2所述的一种用于检测糖胺聚糖的碳化聚合物点单组件荧光阵列传感器的制备方法,其特征在于,所述S2中初步过滤纯化采用0.22μm的水相滤膜;所述超滤离心管的分子量分别为10000和30000,进行两次超滤离心纯化。
6.如权利要求2所述的一种用于检测糖胺聚糖的碳化聚合物点单组件荧光阵列传感器的制备方法,其特征在于,所述S3中Cu2+溶液浓度为10~40μM;所述碳化聚合物点CPDs浓度为0.1mg/mL。
7.一种用于检测糖胺聚糖的碳化聚合物点单组件荧光阵列传感器,其特征在于,采用如权利要求1-6中任一所述的用于检测糖胺聚糖的碳化聚合物点单组件荧光阵列传感器的制备方法制备而成。
8.一种如权利要求1-6任一所述的制备方法制得的碳化聚合物点单组件荧光阵列传感器或如权利要求7所述的碳化聚合物点单组件荧光阵列传感器在生物样本中同时识别检测多种糖胺聚糖分子的应用。
9.如权利要求8所述的应用,其特征在于,将制得的碳化聚合物点单组件荧光阵列传感器反应体系与生物样本振荡混匀后,用荧光酶标仪360nm激发后检测667nm和469nm发射荧光值。
