2. Experimental methods
Grouping, modeling and dosing:
60 mice were selected and randomized after 7d acclimation into Normal Control (NC) and model groups (n = 50). The mice in the NC group are fed with common feed, the mice in the model group are fed with high-fat high-sugar feed, after the mice in the model group are fed with diet-induced insulin resistance for 8 weeks, the mice in the model group are injected with STZ (75 mg/kg) in the abdominal cavity, and the mice with hyperglycemia after successful modeling are divided into a diabetes model group (MC), a positive control group (PC), a low-dose group (L) of a traditional Chinese medicine composition, a medium-dose group (M) of the traditional Chinese medicine composition and a high-dose group (H) of the traditional Chinese medicine composition; wherein, the L group, the M group and the H group are respectively irrigated with low, medium and high doses of the traditional Chinese medicine composition prepared in the example 1, 200mg/kg of metformin is administered to the PC group, and physiological saline with the same volume is administered to the MC group and the NC group for continuous irrigation for 4 weeks.
And (4) observation indexes are as follows:
mental status, body weight, blood glucose, serum insulin levels, oral glucose tolerance, total cholesterol, triglycerides, low density lipoprotein cholesterol and high density lipoprotein cholesterol were measured in 6 groups of mice, respectively.
The experimental indexes are as follows:
(1) Before molding, all groups of mice have good mental state, white and bright hair, and normal drinking water, food intake and defecation. After the model building is successful, except for the normal group, the mice in each group show different degrees of lassitude, withered hair color, polydipsia, diuresis, polyphagia, reduction of activity and the like, and the symptoms of 'three high and one few' appear. After the results of the dose groups of the traditional Chinese medicine composition prepared by the metformin and the example 1 are dry, the symptoms of mice in each group are relieved, the activity is enhanced, the spirit is improved, and the hair color glossiness is improved compared with that of an MC group, which shows that the traditional Chinese medicine composition prepared by the invention is beneficial to maintaining the blood sugar health level.
(2) The effect on mouse body mass is shown in FIG. 1.
As can be seen from fig. 1, the MC group showed a downward trend in body weight (P < 0.01) compared to the normal group (NC), which is probably due to abnormal metabolism of structural proteins of diabetic mice, resulting in a decrease in food intake; the weight of the PC group gradually increased compared with that of the MC group; after 1 month of intervention, the traditional Chinese medicine composition prepared in example 1 has different weight reducing effects in each administration group compared with the MC group, especially in the H group (P < 0.01).
(3) The results of the effect on fasting plasma glucose concentration in mice are shown in FIG. 2.
As can be seen from FIG. 2, the FBG of the NC group mice is substantially unchanged and is between 5 and 7 mmol/L. Compared with the NC group, the MC group had a significant increase in FBG (P < 0.01), and was maintained in a hyperglycemic state, i.e., the model was considered successful and the stability of the diabetes model was good. After the intervention of each dosage of the traditional Chinese medicine composition prepared in the embodiment 1 for 1 month, compared with the MC group, the FBGs of the mice in the PC group and the L, M and H groups of the recipe are all reduced, and the difference is of great statistical significance (P < 0.01). The dosage groups of the traditional Chinese medicine composition prepared by the invention can better regulate and control the fasting blood glucose concentration level of T2DM mice, and present a certain dose-effect relationship, and the ability of maintaining the healthy blood glucose level is reduced along with the reduction of the dosage.
(4) The effect on mouse oral glucose tolerance and area under the curve results are shown in figure 3.
As can be seen from fig. 3, within 120min of the test, the blood glucose concentration of each group of mice tended to increase first and then decrease, and the blood glucose of the MC group was significantly increased (P < 0.01) compared to the NC group, indicating that T2DM mice had impaired glucose tolerance. Blood glucose values of each group began to fall back to the vicinity of the initial value at 120min, but blood glucose levels of the MC group were high, and blood glucose of mice of each administration group tended to fall back to the initial value, wherein blood glucose of mice of the H group was significantly reduced.
(5) The effect on blood lipid metabolism in mice is shown in table 1.
TABLE 1 Effect on blood lipid metabolism in diabetic mice
Figure BDA0004032537040000091
As can be seen from Table 1, the MC group showed significantly higher TC, TG and LDL-C contents (P < 0.01) than the NC group, but the HDL-C was significantly lower than the control group, indicating that the lipid metabolism of the mice under hyperglycemia was abnormal. After the administration, the serum TC, TG and LDL-C of the PC group and the mice of the L, M and H groups are reduced to different degrees (P < 0.05) compared with the MC group, wherein the concentration reduction of the TC and the LDL-C of the L group is not obvious (P > 0.05). The HDL-C content of each group is obviously increased compared with that of the MC group, and the function of accelerating the metabolism of TC is achieved. Therefore, each dose group can regulate the blood lipid level of the mouse to a certain extent, the low dose group has weaker capability, the M group and the PC group have similar blood lipid regulation capability, and the H group has more obvious effect on improving the blood lipid level.
(6) The effects on mouse insulin, HOMA-IR are shown in FIGS. 4-5.
As can be seen from FIGS. 4 to 5, the serum insulin levels were decreased and the insulin resistance was significantly increased in each group as compared with the NC group. Wherein, the serum insulin level of the MC group is remarkably reduced compared with that of the NC group (P < 0.01), but the insulin resistance is remarkably enhanced (P < 0.01); the difference in serum insulin levels between the dose-administered groups was not statistically significant (P > 0.05) compared to the MC groups, but insulin resistance was significantly lower than the MC groups. The effect of the traditional Chinese medicine composition for maintaining the healthy blood sugar level is not realized by promoting insulin secretion, but is realized by reducing the insulin sensitivity index and improving the insulin resistance level of a T2DM mouse so as to achieve the purpose of maintaining the healthy blood sugar level.
The above description is only for the purpose of illustrating the preferred embodiments of the present invention and is not to be construed as limiting the invention, and any modifications, equivalents, improvements and the like that fall within the spirit and principle of the present invention are intended to be included therein.
