技术领域
本发明涉及一种抗炎、抑菌功能肽,尤其是涉及一种鸭肝蛋白源抗炎和抗菌功能肽及其制备方法和应用。
背景技术
鸭肝的营养价值较高,但由于其强烈的泥土或金属的特殊气味,除了极少量的直接食用鸭肝和加工食品的辅料外,大部分鸭肝都被加工成肥料或饲料,利用附加值低。生物活性肽是由共价键连接具有特定顺序的天然氨基酸形成线形、环形结构,并通过调控生物体的代谢功能发挥一定的生物学或生理作用。根据最新的Biopep数据库的统计数据,已收录的4,442种生物活性肽,包括768种抗氧化肽,1,077种抗高血压肽,512种抗菌肽,但抗炎肽仅有41种(2022年4月更新)。因此,我们将目光转移至以鸭肝为蛋白来源开发含有天然抗炎和抗菌能力的生物活性肽。目前,国内外还没有公开关于鸭肝蛋白源抗炎抑菌功能肽及其制备方法的相关研究报道。
发明内容
本发明所要解决的技术问题是提供一种活性高、特异性强的鸭肝蛋白源抗炎和抗菌功能肽及其制备方法和应用。
本发明解决上述技术问题所采用的技术方案为:一种鸭肝蛋白源抗炎和抗菌功能肽,包括DLTGIPPAP(P1),ELKPTPEGDL(P2),IDVSPDSPDHY(P3),IYVDAVINH(P4),LDSNLDLKF(P5),LGEHNIDV(P6),LVYPFPGPI(P7),QTNLVPYPR(P8),SLVYPFPGPIPN(P9)和VIESPPEI(P10)中的至少一种肽。
上述鸭肝蛋白源抗炎和抗菌功能肽的制备方法,包括以下步骤:
(1)提取鸭肝蛋白;
(2)将鸭肝蛋白酶解制备得到含有抗炎和抗菌肽的酶解粗提物;
(3)将酶解粗提物通过Sephadex G-15分离得到抗炎和抗菌肽分离物;
(4)将抗炎和抗菌肽分离物通过RP-HPLC纯化得到鸭肝蛋白源抗炎和抗菌功能肽,经LC-MS/MS鉴定分别为DLTGIPPAP,ELKPTPEGDL,IDVSPDSPDHY,IYVDAVINH,LDSNLDLKF,LGEHNIDV,LVYPFPGPI,QTNLVPYPR,SLVYPFPGPIPN和VIESPPEI。
进一步,步骤(1)具体为:取鸭肝,去筋,切碎后加水按质量体积比1 g:6 mL的比例于匀浆机上充分匀浆,用1 mol/L氢氧化钠调pH至12,不断搅拌,于8000 rpm,4 ℃离心10min,取中间层,继续用盐酸调pH至5.5,不断搅拌,于8000 rpm,4 ℃离心10 min,取沉淀得到鸭肝蛋白,冻干后冷冻保存备用。
进一步,步骤(2)具体为:在底物质量分数5%的鸭肝蛋白悬液中按添加量5000 U/g加混合酶,于pH 7.5,温度50℃,磁力水浴锅中酶解4 h后,95℃加热15 min灭酶,终止水解,将pH调至4.5,然后于8000 rpm,4 ℃,离心10min,以去除未酶解的蛋白质,得到含有抗炎和抗菌肽的酶解粗提物,然后冷冻干燥,保存在-40 ℃冰箱。
进一步,所述的混合酶为碱性蛋白酶、风味蛋白酶、木瓜蛋白酶和中性蛋白酶按质量比2:1:2:2的比例混合而成。
进一步,步骤(3)具体为:将0.1g酶解粗提物溶于10mL蒸馏水中,然后用0.45 μm水相过滤膜过滤后,采用Sephadex G-15葡聚糖凝胶柱分离,上样量为1.5 mL,超纯水以1 mL/min的流速洗脱,检测波长为220 nm,共得到4个吸收峰,收集第1个吸收峰得到抗炎和抗菌肽分离物。
进一步,步骤(4)具体将抗炎和抗菌肽分离物用蒸馏水配置成5 mg/mL的溶液,用0.22 μm超滤膜过滤并超声脱气后,使用RP-HPLC进行纯化,流动相A为含0.1wt%三氟乙酸的水溶液,流动相B为含0.1wt%三氟乙酸的乙腈溶液,以线性梯度方式洗脱0~2 min,0~5%乙腈;2~30 mim,5%~25%乙腈;30~35 min,25%~40%乙腈;35~60 min,5%乙腈,上样量为100 μL,检测波长为214 nm,流速为1 mL/min,分离得到6个峰,收集第1个峰,冷冻干燥,经LC-MS/MS鉴定为DLTGIPPAP(P1),ELKPTPEGDL(P2),IDVSPDSPDHY(P3),IYVDAVINH(P4),LDSNLDLKF(P5),LGEHNIDV(P6),LVYPFPGPI(P7),QTNLVPYPR(P8),SLVYPFPGPIPN(P9)和VIESPPEI(P10)。
上述鸭肝蛋白源抗炎和抗菌功能肽在制备炎症因子GAPDH、TNF-α、IL-6、COX-2和/或NF-κB抑制剂方面的用途。
上述肽VIESPPEI在制备结肠炎小鼠肠道菌群调节药物或者结肠炎治疗药物方面或者小鼠结肠内梭状芽孢杆菌抑制剂的用途。
与现有技术相比,本发明的优点在于:本发明一种鸭肝蛋白源抗炎和抗菌功能肽及其制备方法和应用,利用多种酶水解鸭肝蛋白获得小分子活性肽,进一步纯化并鉴定鸭肝肽,得到了10种新的抗炎肽,通过体外合成、计算机模拟、分子对接确认肽的生物学功能,在RAW 264.7细胞和C57BL/6J小鼠肠道内中探讨抗炎、抗菌肽的生物活性,并筛选出1种新的强效抗炎、抗菌肽,提供了其可替代非甾体抗炎药和糖皮质激素类药物的应用价值。不仅可以解决鸭肝副产物的高值化利用,增加经济效益,还解决了工业生产肽的得率低、特异性不强、质量不稳定等问题,提供了具有抗炎、抗菌功能的肽高效生产方法。

本发明中的以下缩略语具体定义为:LC-MS/MS,液相二级质谱;RP-HPLC,反相高效液相色谱;LPS,脂多糖;PCR,聚合酶链式反应;qPCR,实时荧光定量核酸扩增检测系统;ELISA,酶联接免疫吸附剂测定;DCFH-DA,活性氧ROS荧光探针;SOD,超氧化物歧化酶;DSS,葡聚糖硫酸钠盐。
一、具体实施例
一种鸭肝蛋白源抗炎和抗菌功能肽的制备方法,包括以下步骤:
1、提取鸭肝蛋白
首先,取鸭肝适量,去筋,切碎后加水1:6(m/v,g/mL)于匀浆机上充分匀浆,1 mol/L氢氧化钠调至pH 12,不断搅拌,8500 rpm,4 ℃离心10 min,取中间层,继续用盐酸调至pH5.5,不断搅拌,8500 rpm,4 ℃离心10 min取沉淀,冻干后冷冻保存备用,凯氏定氮法测定鸭肝中粗蛋白质含量为79.58%。
2、制备抗炎和抗菌肽粗提物
首先,向底物质量分数5%的鸭肝蛋白悬液加混合酶5000 U/g,pH 7.5;温度为50℃;碱性蛋白酶:风味蛋白酶:木瓜蛋白酶:中性蛋白酶在2:1:2:2的条件下,磁力水浴锅中酶解4 h后,95 ℃加热15 min灭酶,终止水解,将pH调至4.5,然后离心(8000 rpm, 4 ℃,10min),以去除未酶解的蛋白质,得到含有抗炎和抗菌肽的酶解粗提物,然后冷冻干燥,保存在-40 ℃冰箱。
3、Sephadex G-15分离小分子肽
首先,将0.1g酶解粗提物溶于10mL蒸馏水中,然后用0.45 μm水相过滤膜过滤后,采用Sephadex G-15葡聚糖凝胶柱分离,上样量为1.5 mL,超纯水以1 mL/min的流速洗脱,检测波长为220 nm,共得到4个吸收峰,收集第1个吸收峰得到抗炎和抗菌肽分离物F1。
4、RP-HPLC纯化
首先,抗炎和抗菌肽分离物F用蒸馏水配置成5 mg/mL的溶液,并超声脱气;然后使用RP-HPLC纯化鸭肝蛋白抗炎、抗菌肽分离物F1;流动相A为含体积分数0.1%三氟乙酸的水,流动相B为含体积分数0.1%三氟乙酸的乙腈,以线性梯度方式洗脱0~2 min,0~5%乙腈;2~30 mim,5%~25%乙腈;30~35 min,25%~40%乙腈;35~60 min,5%乙腈,上样量为100 μL,检测波长为214 nm,流速为1 mL/min,分离得到6个峰,收集第1个峰(F1-1),冷冻干燥。其中含0.1wt%三氟乙酸的水溶液的配制方法如下:称取1克三氟乙酸,溶于999克水中即可得到含0.1wt%的三氟乙酸的水溶液;含0.1wt%三氟乙酸的乙腈溶液的配制方法如下:称取1克三氟乙酸,溶于999克水中乙腈即可得到含0.1wt%的三氟乙酸的乙腈溶液。
5、LC-MS/MS鉴定活性组分
首先,使用LC-MS/MS鉴定,根据MS/MS光谱、查阅鸭肝蛋白库以及通过比对Biopep数据库并用蛋白质组学软件MaxQuant(1.6.2.10版)分析MS/MS数据。序列在Anasplatyrhynchos的蛋白质组参考数据库中检索,以匹配亲本蛋白。筛选肽段序列的标准是根据质谱的可信度与峰强度。经鉴定F1-1为10条首次发现的抗炎、抗菌肽,分别是DLTGIPPAP(P1),ELKPTPEGDL(P2),IDVSPDSPDHY(P3),IYVDAVINH(P4),LDSNLDLKF(P5),LGEHNIDV(P6),LVYPFPGPI(P7),QTNLVPYPR(P8),SLVYPFPGPIPN(P9),VIESPPEI(P10)。
二、实验结果分析
1、四种酶水解鸭肝蛋白制备抗炎和抗菌肽
水解度是评价酶解效果最直观的指标。而自由基清除率代表了抗氧化活性的大小,然而抗氧化与抗炎有密切的关系。故选择水解度为主指标,OH与DPPH自由基清除率为参考指标,综合评价酶解过程。由图1可知,在五种蛋白酶中,复合蛋白酶水解后DPPH自由基清除率最弱,碱性蛋白酶的自由基清除率最强,但水解度比风味蛋白酶小,故删除复合蛋白酶。

如表1所示,加酶量5000 U/g,pH 7.5;温度为50 ℃;碱性蛋白酶:风味蛋白酶:木瓜蛋白酶:中性蛋白酶在2:1:2:2的条件下,底物水解度较高,且·OH与DPPH自由基清除率均较强,采用优化后的酶解条件为:底物质量分数5%的鸭肝蛋白悬液在最佳酶解条件下,磁力水浴锅中酶解4 h后,95 ℃加热15 min灭酶,终止水解。将pH调至4.5,然后离心(8000rpm,10 min,4℃),以去除未消化的蛋白质,旋蒸后冻干,保存在-40℃冰箱。
四种酶水解产物的分子量分布如图2所示,鸭肝蛋白经四种酶协同水解后,获得了分子量较小的多肽,其中小分子肽(<1500 Da)达到70.25%,而>3000 Da的肽百分比含量仅为1.43%。已有研究表明,小分子量的肽吸收耗能低,载体不易饱和,并且吸收速度更快,尤其是二肽和三肽,可完整地被肠上皮细胞吸收,生物活性高等优点,并且很多被证实具有抗炎活性的肽分子量均在1500 Da以下。
2、水解物的分离、纯化、鉴定
图3是对鸭肝水解物的分离。鸭肝蛋白酶解产物经Sephadex G-15初步分离后,共得到4个吸收峰。之后在RAW 264.7细胞中验证鸭肝蛋白酶解粗提物对于炎症因子释放的抑制效果(图4)。一氧化氮(NO)作为一种自由基,不仅是对抗侵袭性病原体(细菌、病毒等)的强毒分子,也是炎症细胞信号通路中强有力的血管扩张剂。如图4-(1)所示,层析四个组分以100 µg/mL作用于RAW 264.7细胞后,与使用LPS刺激的细胞中NO的释放量比未刺激组增加了9.54倍(P<0.05)。相对于LPS诱导组,F1、F2、F3和F4均可以降低炎症细胞中NO的释放量,但各组分之间差异不显著。TNF-α和IL-6作为促炎或抗炎细胞因子主导炎症反应以及终止,在炎症中发挥关键作用,故经常被用来指示炎症状态。如图4-(2)和(3),与未刺激组相比,LPS刺激后,TNF-α和IL-6的释放量显著增加,说明LPS诱导后引发细胞炎症,释放大量炎症因子(P<0.05),F1和F4均可以显著降低炎症细胞中TNF-α的释放量。而只有F1对IL-6产生有较强的抑制作用,抑制率为42.69%,其余F2,F3和F4与LPS损伤组无显著差异,故选取组分F1作为抗炎、抗菌肽粗品。
使用RP-HPLC对层析获得的F1组分进一步纯化,如图5所示得到6个组分,分别为F1-1、F1-2、F1-3、F1-4、F1-5、F1-6。之后在RAW 264.7细胞中验证鸭肝6组分对于炎症因子释放的抑制效果(图6)。如图6-(1)所示,6个组分均能降低细胞中NO的释放量,但抑制能力具有显著差异(P<0.05),其中F1-1、F1-3和F1-4组中NO含量较低,与LPS诱导组相比,NO释放的抑制率分别为55.46%、45.30%和37.49%。然而,如图6-(2)所示,F1-1、F1-3和F1-4这三个组分在抑制TNF-α释放的方面有显著差异(P<0.05)。其中,F1-1组分能有效抑制的肿瘤坏死因子TNF-α的释放,抑制率为61.02%。如图6-(3)所示,F1-1、F1-3和F1-4这三个组分在IL-6的释放方面,无明显差异。因此,确定选取组分F1-1。
使用LC-MS/MS鉴定了抗炎活性较强的馏分F1-1,根据MS/MS光谱、查阅鸭肝蛋白库以及通过比对Biopep数据库,发现了10条肽段如图7与表2,分别是DLTGIPPAP(P1),ELKPTPEGDL(P2),IDVSPDSPDHY(P3),IYVDAVINH(P4),LDSNLDLKF(P5),LGEHNIDV(P6),LVYPFPGPI(P7),QTNLVPYPR(P8),SLVYPFPGPIPN(P9),VIESPPEI(P10)。与Biopep数据库比对,发现这10条抗炎、抗菌肽均是新的未被发现的。

3、肽的理化性质和生物活性
如表3和4所示,10条抗炎、抗菌肽的疏水性为+4.22~+20.78 kcal/mol,P2、P3、P5、P7、P8的亲水性为-1.11~-0.11 kcal/mol。结果表明,所获产物中有5种多肽具有两亲性,可以增大其溶解度。脂肪族氨基酸指数为75.56~162.22,说明这10条肽热稳定性良好,肽的净电荷为-3至+1,pI的范围是2.93到9.60。鉴定得到的鸭肝蛋白肽均显示出良好的ACE(血管紧张素转换酶),其中90%的肽序列显示出能抑制二肽基肽酶III或二肽基肽酶IV的能力,具有抗菌与抗炎的活性。

4、10条抗炎、抗菌肽在细胞中抗炎活性评价及检验NF-κB信号通路
通过使用LPS作为诱导剂,构建RAW 264.7细胞模型,确定10条鸭肝抗炎、抗菌肽在RAW 264.7细胞中的抗炎能力。利用qPCR检测抗炎活性,以GAPDH的表达作为内参基因,设计用于基因扩增的引物表5。最后,通过Western Blot检验NF-κB信号通路。

10条抗炎、抗菌肽在细胞中抗炎活性评价结果如图8所示。图8-(1)为用CCK-8法检测合成肽对细胞的毒性,除肽ELKPTPEGDL和肽LGEHNIDV在200 μg/mL时,细胞存活率降至90%以下,其余8条肽段在25-200 μg/mL浓度范围内,细胞存活率在90%以上。并且肽P1,P3,P4,P8在该浓度范围内,细胞存活率均>100%,说明这四条肽序列能够显著促进细胞增殖,这可能是由于肽作为一种氮源,被加入到DMEM基本培养基后促进RAW 264.7细胞的生长。考虑到P2和P6在200 μg/mL时对细胞有一定的毒性,因此,在接下来的研究中,选择25、50和100μg/mL来评估合成肽的抗炎潜力。图8-(2)是鸭肝抗炎、抗菌肽对炎症细胞释放NO的影响,LPS诱导的RAW 264.7细胞NO浓度是未刺激组的10.51倍,而在25-100 μg/mL的IDVSPDSPDHY、IYVDAVINH、LVYPFPGPI和VIESPPEI预处理下,NO的产生量明显减少(P<0.05)。其中LVYPFPGPI和VIESPPEI以剂量的方式抑制炎症细胞中NO的释放,在浓度为100 μg/mL,抑制率分别为47.24%和56.32%。故选择IDVSPDSPDHY、IYVDAVINH、LVYPFPGPI和VIESPPEI为研究目标,ELISA测定其对于细胞中炎症因子的释放量的影响。
鸭肝抗炎、抗菌肽对炎症细胞释放TNF-α的影响如图8-(3)所示,RAW 264.7单独用LPS处理6 h后,TNF-α的释放量对比空白组有显著增加(P<0.05),而4种鸭肝抗炎、抗菌肽预处理后,细胞中TNF-α释放量均有不同程度的下降。并且,VIESPPEI以剂量依赖的方式降低炎症细胞中的TNF-α的含量,在浓度为100 μg/mL时,对TNF-α释放的抑制率达42.48%。如图8-(4)所示,IDVSPDSPDHY、LVYPFPGPI和VIESPPEI在不同浓度下显示不同的IL-6抑制活性。IDVSPDSPDHY在低剂量时对IL-6有最强的抑制作用(27.04%),但当剂量超过25 μg/mL时抑制作用减弱。这可能与细胞耐受性有关。因为,肽直接与靶细胞表面的特定受体结合,来实现其免疫调节作用。正因为它不与病原体直接相互作用,复杂的机制导致靶点上的肽剂量不同。肽与受体结合受限,致使调节作用减弱。炎症反应与氧化应激有密切关联,TNF-α可直接诱导ROS的产生,而机体中ROS的积累造成细胞氧化损伤进而加剧炎症反应。结果如图8-(5)所示,LPS诱导组的RAW 264.7细胞,荧光强度明显高于空白组和给药组,说明在此条件下细胞内积累大量了ROS。然而,使用四种肽IDVSPDSPDHY(P3)、IYVDAVINH(P4)、LVYPFPGPI(P7)和VIESPPEI(P10),100 μg/mL预孵育18 h预处理后,细胞ROS荧光强度相比LPS诱导组有所减弱,表明细胞中的ROS含量减少,减轻了LPS引起的细胞氧化应激反应。说明四种肽作用于RAW 264.7细胞后,能减轻LPS刺激导致的细胞氧化应激。
结果如图9所示,LPS刺激细胞后,会促进炎症介质如TNF-α,IL-6,IL-1β,COX-2和一氧化氮合酶(iNOS)等的释放,导致炎症浸润从而促进并扩大炎症。为了进一步阐明合成肽抑制LPS诱导的促炎因子表达的机制,用RT-qPCR方法检测了25、50和100 μg/mL的RAW264.7细胞中TNF-α,IL-6,COX-2和NF-κB mRNA的表达量。如图9-(1)所示,为根据NCBI上基因信息,利用Primer Primier 5软件设计的引物,经过PCR验证后,每对引物特异性良好。如图9-(2)所示,与LPS损伤组相比,肽IDVSPDSPDHY(P3)、LVYPFPGP(P7)和VIESPPEI(P10)对TNF-α的mRNA表达量有明显的抑制作用(P<0.05)。当三种肽浓度均为100 μg/mL作用于炎症细胞时,抑制率分别为50.50%,59.28%和44.60%。如图9-(3)所示,对IL-6 mRNA具有明显抑制效果的是P3,在其浓度为25 μg/mL,抑制率达到16.37%。COX-2衍生的前列腺素是重要的炎症调节因子。如图9-(4)所示,4种鸭肝抗炎、抗菌肽作用于细胞后,均能显著降低COX-2mRNA表达量(P<0.05),其中,P3和P7均在浓度为50 μg/mL时,对COX-2 mRNA的抑制作用最强,抑制率分别达到79.41%,84.03%,说明,P3和P7对COX-2 mRNA具有较好的抑制活性。NF-κB是脂多糖刺激后重要的上游转录因子。如图9-(5)所示,四种肽作用于RAW 264.7细胞后,NF-κB的mRNA表达量均比LPS损伤组低。其中,P3和P10均以剂量依赖性降低NF-κB的mRNA表达量,在100 μg/mL的浓度时,抑制率达到47.64%和30.93%。综上说明,P3,P7和P10在4种炎症介质的基因表达水平上,抑制性较强,表现出较好的抗炎活性。
如图9-(6)所示,为进一步研究肽(P3、P7、P10)对NF-κB上游相关调控因子的调节作用,采用Western blotting检测NF-κB p65和IκBα的蛋白表达和磷酸化。如图9-(7)所示,当浓度为50 μg/mL P3和P10预处理细胞后,与LPS损伤组相比,可显著抑制细胞质内IκBα的磷酸化和降解(P<0.01),抑制率分别为52.33%和53.68%。而经P7治疗后,IκBα磷酸化水平仅降低了29.53%。如图9-(8)所示,LPS使NF-κB p65在细胞核中的表达增加了4.44倍,而三种肽预处理后NF-κB p65的表达显著降低,抑制率分别为79.94%、54.36%和56.67%。结果表明,三种鸭肝抗炎、抗氧化肽的抗炎作用机制归因于抑制IκBα蛋白磷酸化和NF-κB p65核移位,来调控NF-κB信号通路。其中P10的对抑制NF-κB信号通路激活的效果最明显,故选择此肽段进一步研究在小鼠体内的抗炎活性。
三、应用实施例
肽VIESPPEI对小鼠体内抗炎活性与肠道菌群的影响
首先,采用3.5% DSS诱导结肠炎模型。SPF级C57BL/6J小鼠(20±2 g,雌雄各20只,共40只,6-8周龄),在SPF级实验室中恒温(22-24 °C)和12/12 h光−暗周期条件下饲养,测定结肠MPO,SOD和CAT的含量。采用DNA抽提试剂盒取小鼠粪便微生物群的总DNA,通过核酸电泳并检测DNA浓度。以基因组DNA为模板,PCR扩增16S rRNA基因V3-V4区。使用电泳检测PCR产物,然后使用磁珠纯化,并进行二轮PCR扩增。重复上一步骤后,对PCR产物进行Qubit定量。最后根据PCR产物浓度进行等量混样,并上机测序。
结果如图10-(1)所示,空白组小鼠在试验周期内,小鼠体重整体呈现增长的趋势。第五天时,经DSS处理组的体重降至最低,为原始体重的85.60%,明显低于空白组的106.11%。停止饮用DSS溶液后,小鼠的体重略微升高,但第八天小鼠体重与空白组仍有较大差异,说明DSS对小鼠的损伤是不可逆的。而服用了不同剂量肽的组,小鼠体重降低变缓。如图10-(2)所示,从饮用DSS的第3天起,除空白组,其余组DAI评分持续增加。至第5天,DSS诱导组的DAI评分到达峰值,大便呈膏状液体,普遍出现直肠出血情况。然而,不同剂量的肽对DAI评分有不同程度的降低。观察显示,即使停止饮用DSS,DSS组的DAI评分,虽略微下降,但仍然高于中剂量组和高剂量组。证明肽VIESPPEI在UC小鼠结肠炎恢复过程中的有益作用。
如图10-(3)所示,对照组小鼠结肠长度平均为5.075±0.46 cm,明显短于空白组(P<0.01),而治疗组小鼠结肠长度显著增加。并且,结肠长度与小鼠体重以及DAI评分的变化趋势相似。高剂量的肽的干预虽然比空白组仍要短9.06%,但比对照组显著增加了26.10%(P<0.01),说明给予高剂量的肽能明显改善小鼠结肠萎缩变短的症状.
根据图10-(4)的HE染色,由DSS诱导后,结肠组织出现不同程度的上皮结构破坏、炎症细胞浸润等病理变化。空白组的结肠组织具有清晰完整的黏膜层、正常的隐窝、丰富的杯状细胞,且并无炎症细胞浸润。与空白组比,DSS刺激后的组织明显受损,表现为杯状细胞减少、隐窝间距增大且表面不规则和大量炎症细胞浸润等形态。结肠中的杯状细胞分泌的粘液层可以限制细菌的相互作用。若杯状细胞功能受损或减少与结肠炎的发病有关。并且结肠炎症细胞浸润的增加可能导致结肠炎过程中细胞因子的过度产生。低剂量和中剂量组的隐窝间距仍较大,仍有部分隐窝表面不规则,肠粘膜被侵蚀;然而,高剂量组比空白组比,杯状细胞仍减少;但相对于DSS组,仅有少量炎症细胞浸润到黏膜和黏膜下层,黏膜肌层比较清晰,U型隐窝数量增加。以上结果表明肽VIESPPEI能减轻DSS诱导的结肠炎小鼠的结肠组织形态学变化,其中高剂量组(100 mg/kg)的效果好于低剂量组(10 mg/kg)。
炎症导致受损组织中性白细胞的聚积,在结肠炎缓解过程中,细胞数量会减少,中性白细胞中含有一定量的MPO。MPO是一种血红素蛋白,在机体产生和调节炎症反应等多方面发挥作用。由表6所示,对照组小鼠结肠组织中MPO的酶活明显增高,是空白组的的2.88倍。给小鼠灌胃不同剂量的肽后,其结肠组织中MPO酶活较对照组而言,显著下降(P<0.05)。其中,高剂量组MPO酶活比对照组降低了51.38%。在免疫调节因素中,氧化应激是导致疾病持续发展的主要诱因之一。模型组由于ROS释放,抗氧化系统受损,从而损害肠粘膜,最终导致肠道炎症。此外,结肠炎可以通过抑制氧化应激的过程来预防。SOD和CAT作为细胞中重要保护酶,它们通过协同工作能够有效防止细胞内ROS水平过高,抑制氧化损伤。对照组的SOD和CAT活力显著下降,分别为61.86±5.61 U/mg和19.11±1.17 U/mg,与MPO含量成反比。用高剂量的肽进行的预处理的治疗组SOD和CAT的活力分别显著提高至105.47±4.65、33.62±1.25 U/mg(P<0.05)。

DSS诱导和给药会使小鼠肠道粪便菌群的菌属相对丰度出现差异。DSS刺激后,与空白组相比,会显著降低
Muribaculaceae菌属和毛螺旋菌属的比例。
Muribaculaceae菌属和毛螺旋菌属均属于小鼠肠道内的有益菌属。其中,
Muribaculaceae可以促进葡萄糖代谢,减少促炎状态。毛螺旋菌属是SCFA生产者,SCFA不仅可作为能量来源,还具有免疫调节和抗炎的作用。空白组和对照组的
Muribaculaceae菌属和毛螺菌属总含量分别为60.46%、38.86%。而给予高剂量的肽后,两种有益菌的含量为50.50%,比对照组显著提高了11.64%,减小了与空白组的差异。
如图10-(5)所示,各组拟杆菌的比例分别为1.40%、12.41%,12.40%、12.79%、17.53%,可见经DSS诱导后小鼠结肠内的拟杆菌属较空白组而言,均显著上升。而对照组的梭状芽孢杆菌总含量为6.99%是空白组的2.6倍,在高剂量给药组降低至3.74%。研究表明梭状芽孢杆菌属于有害菌属,其丰度的增加会导致代谢紊乱,引起局部或全身性炎症。而摄入肽VIESPPEI能显著降低其丰度,表明VIESPPEI具有显著抗菌效果。
如图10-(6)所示,DSS刺激组与空白组分离明显,重叠区域较小,说明DSS刺激后会大幅度改变肠道菌群群落结构,而给药组样本基本重叠,表明其肠道菌群丰度和多样性基本相似,但与空白组间出轻度分离,重叠区偏小,表明其与正常小鼠的肠道菌群丰度和多样性有差异。结果表明:DSS刺激后,肠道菌群群落结构改变,但100 mg/kg VIESPPEI肽可逆转结肠炎小鼠肠道菌群的失调,缓解和恢复结肠炎症。表明其优异的抗炎、抗菌能力。
综上所述,本发明一种鸭肝蛋白源抗炎、抗菌功能肽,提供了可替代非甾体抗炎药和糖皮质激素类药物的生物活性肽,解决了工业生产肽的得率低、特异性不强、质量不稳定等问题,增加了数据库中抗炎、抗菌肽种类。通过提取鸭肝蛋白、确认氨基酸类型和复合酶解鸭肝蛋白解决了鸭肝作为加工生产的副产物,因其难闻的气味,食用率低,产品附加值不高的问题。
上述说明并非对本发明的限制,本发明也并不限于上述举例。本技术领域的普通技术人员在本发明的实质范围内,做出的变化、改型、添加或替换,也应属于本发明的保护范围。
Claims (9)
1.一种鸭肝蛋白源抗炎和抗菌功能肽,其特征在于包括DLTGIPPAP,ELKPTPEGDL,IDVSPDSPDHY,IYVDAVINH,LDSNLDLKF,LGEHNIDV,LVYPFPGPI,QTNLVPYPR,SLVYPFPGPIPN和VIESPPEI中的至少一种肽。
2.一种权利要求1所述的鸭肝蛋白源抗炎和抗菌功能肽的制备方法,其特征在于包括以下步骤:
(1)提取鸭肝蛋白;
(2)将鸭肝蛋白酶解制备得到含有抗炎和抗菌肽的酶解粗提物;
(3)将酶解粗提物通过Sephadex G-15分离得到抗炎和抗菌肽分离物;
(4)将抗炎和抗菌肽分离物通过RP-HPLC纯化得到鸭肝蛋白源抗炎和抗菌功能肽,经LC-MS/MS鉴定分别为DLTGIPPAP,ELKPTPEGDL,IDVSPDSPDHY,IYVDAVINH,LDSNLDLKF,LGEHNIDV,LVYPFPGPI,QTNLVPYPR,SLVYPFPGPIPN和VIESPPEI。
3.根据权利要求2所述的一种鸭肝蛋白源抗炎、抗菌功能肽的制备方法,其特征在于步骤(1)具体为:取鸭肝,去筋,切碎后加水按质量体积比1 g:6 mL的比例于匀浆机上充分匀浆,用1 mol/L氢氧化钠调pH至12,不断搅拌,于8000 rpm,4 ℃离心10 min,取中间层,继续用盐酸调pH至5.5,不断搅拌,于8000 rpm,4 ℃离心10 min,取沉淀得到鸭肝蛋白,冻干后冷冻保存备用。
4.根据权利要求2所述的一种鸭肝蛋白源抗炎、抗菌功能肽的制备方法,其特征在于步骤(2)具体为:在底物质量分数5%的鸭肝蛋白悬液中按添加量5000 U/g加混合酶,于pH7.5,温度50℃,磁力水浴锅中酶解4 h后,95℃加热15 min灭酶,终止水解,将pH调至4.5,然后于8000 rpm,4 ℃,离心10min,得到含有抗炎和抗菌肽的酶解粗提物,然后冷冻干燥,保存在-40 ℃冰箱。
5.根据权利要求4所述的一种鸭肝蛋白源抗炎和抗菌功能肽的制备方法,其特征在于所述的混合酶为碱性蛋白酶、风味蛋白酶、木瓜蛋白酶和中性蛋白酶按质量比2:1:2:2的比例混合而成。
6.根据权利要求4所述的一种鸭肝蛋白源抗炎和抗菌功能肽的制备方法,其特征在于步骤(3)具体为:将0.1g酶解粗提物溶于10mL蒸馏水中,然后用0.45 μm水相过滤膜过滤后,采用Sephadex G-15葡聚糖凝胶柱分离,上样量为1.5 mL,超纯水以1 mL/min的流速洗脱,检测波长为220 nm,共得到4个吸收峰,收集第1个吸收峰得到抗炎和抗菌肽分离物。
7.根据权利要求5所述的一种鸭肝蛋白源抗炎和抗菌功能肽的制备方法,其特征在于步骤(4)具体为将抗炎和抗菌肽分离物用蒸馏水配置成5 mg/mL的溶液,用0.22 μm超滤膜过滤并超声脱气后,使用RP-HPLC进行纯化,流动相A为含0.1wt%三氟乙酸的水溶液,流动相B为含0.1wt%三氟乙酸的乙腈溶液,以线性梯度方式洗脱0~2 min,0~5%乙腈;2~30 mim,5%~25%乙腈;30~35 min,25%~40%乙腈;35~60 min,5%乙腈,上样量为100 μL,检测波长为214 nm,流速为1 mL/min,分离得到6个峰,收集第1个峰,冷冻干燥,经LC-MS/MS鉴定分别为DLTGIPPAP,ELKPTPEGDL,IDVSPDSPDHY,IYVDAVINH,LDSNLDLKF,LGEHNIDV,LVYPFPGPI,QTNLVPYPR,SLVYPFPGPIPN和VIESPPEI。
8.一种权利要求1中所述的鸭肝蛋白源抗炎和抗菌功能肽在制备炎症因子GAPDH、TNF-α、IL-6、COX-2 和/或NF-κB抑制剂方面的用途。
9.一种权利要求1中所述的肽VIESPPEI在制备结肠炎小鼠肠道菌群调节药物或者结肠炎治疗药物方面或者小鼠结肠内梭状芽孢杆菌抑制剂的用途。
