Abstract
The invention relates to the technical field of feed additives, in particular to a preparation method of a paulownia flower extract for a feed anti-microbial additive and a leacher. The preparation method of the paulownia flower extract for the feed anti-bacterial additive comprises the following steps: adding dried crushed paulownia flower into an organic solvent to obtain a mixture; adding the mixture into the interior of a leaching device, and leaching at a preset temperature; filtering and discharging the hot extract after extraction is finished, and obtaining filtrate; washing the residue with the organic solvent to obtain a washing solution; and cooling the filtrate and the washing liquid, mixing, and evaporating under reduced pressure to remove the solvent to obtain light red viscous liquid, namely the paulownia flower extract. The paulownia flower extract has natural fragrance, and can reduce the original pungent smell of the feed, cover the unpleasant smell of the feed and improve the disease resistance; the preparation method is simple, low in cost, low in energy consumption, stable in property, free of toxic and side effects, and capable of reducing irritation of the feed, improving sweet taste, improving aftertaste and increasing feed intake of animals.

Technical Field
The invention relates to the technical field of feed additives, in particular to a preparation method of a paulownia flower extract for a feed anti-microbial additive and a leacher.
Background
The flos Paulowniae is dried flower of Paulownia of Scrophulariaceae, has wide distribution, is mainly produced in yellow river basin, has large quantity and has wide pharmacological activity. It is rich in organic substances such as protein, amino acids, polysaccharides, glucoside, quercetin, kaempferol, homoeriodictyol, 4,5, 7-trihydroxy-3-methoxyflavone, 5-hydroxy-3, 4, 7-trimethoxybiflavone, luteolin, apigenin, benzaldehyde, vanillin, nonanal, indole, ursolic acid, palmitic acid, 4-methylphenol, 2-methoxyphenol, 3, 4-dimethoxyphenol, 3, 5-dimethoxyphenol, benzyl alcohol, 4-methoxybenzyl alcohol, 3-methoxyphenetol, phenylpropanol, α -terpineol, β -gu-catanol, 1, 4-dimethoxybenzene, daucosterol, verbascoside, 1-acetyl-2- (3 '-hydroxy) octadecanoic acid ester, 2, 4-decadiene, anisene, pinene, romonene, octone, p-methoxyanisole, methyl anisate, dipalmitone, 3' -O-metalacerate, minone, etc., most of the components are fragrant substances, and have the effects of promoting the circulation of qi, inducing resuscitation, relieving exterior syndrome, eliminating dampness, relieving pain, dispersing lung qi and the like, and due to the existence of the organic compounds, the paulownia flower has the effects of resisting oxidation, resisting bacteria, diminishing inflammation, diminishing swelling, growing hair, killing insects, stopping bleeding, relieving cough, relieving asthma, eliminating phlegm, treating tracheitis and the like.
The pig has very obvious preference on sweet food, and experiments prove that in the growth stage (1-28 days) of piglets, the addition of 150mg/kg of additive into the daily ration can achieve larger daily feed intake and can also obtain larger daily gain in the stage. In addition, experiments prove that the additive has an antioxidant function, and can effectively relieve the oxidation degree of the feed along with the prolonging of the storage time of the feed. The essence is the core of the feed additive, and can stimulate the smell of animals to generate a food calling effect. The fragrance composition of the additive is divided into three parts of top fragrance, body fragrance and bottom fragrance according to different volatility. Top notes are the first smells of fragrance to be smelled, which impart diffusion to the additive. Body notes are the smells that can be heard after the top note, and are usually the most important components of additives that impart characteristics to the fragrance. Together with the body notes, the base notes determine the tolerance of the additive to time and temperature, and are important when the feed needs to be expanded and pelleted, and the base notes impart the flavor retention properties of the additive. The proportion and the content of the three components are different, so that different types of odor types can be formed, and the application characteristics of the additive are determined by the different types. Therefore, the traditional additive also has the defects of non-lasting fragrance, over-concentrated fragrance, unstable fragrance and the like.

In order to solve the technical problems, the invention provides a preparation method of a paulownia flower extract for a feed anti-additive, which comprises the following steps: adding dried crushed paulownia flower into an organic solvent to obtain a mixture; adding the mixture to the interior of a leaching vessel and leaching at a predetermined temperature; filtering and discharging the hot extract after extraction is finished, and obtaining filtrate; washing the residue with the organic solvent to obtain a washing solution; and cooling the filtrate and the washing solution, mixing, and evaporating under reduced pressure to remove the solvent to obtain light red viscous liquid, namely the paulownia flower extract.
Preferably: the proportion of the paulownia flower extract in the feed substitute antibiotic additive is 15-20%;
the feed substitute antibiotic additive accounts for 2-5% of the feed.
Preferably: the feed anti-bacterial additive comprises the following components in parts by weight:
the proportion of the paulownia flower extract, the apple aroma compound and the carrier is 20-30 parts: 10-20 parts of: 60-70 parts.
Preferably: the apple milk flavor compound is a compound of 2-methyl ethyl butyrate, hexyl acetate, phenethyl isobutyrate, cinnamyl isovalerate, 5- (6) -decenoic acid and a stabilizer; the weight proportion of the added components is 20-30 parts: 10-20 parts of: 10-20 parts of: 10-20 parts of: 25-35 parts of: 5-10 parts.
Preferably: the stabilizer is one or a combination of more of calcium carbonate, calcium phosphate, beta-cyclodextrin, gelatin and the like.
Preferably: the carrier is poly [1, 3-di (p-carboxyl phenoxy) propane-sebacic acid ], polylactic acid and polyethylene glycol, and the addition weight proportion is 40-50 parts: 40-50 parts of: 10-20 parts.

a base;
the leaching barrel is of a sealed barrel structure, and the middle position of the leaching barrel is rotatably connected to the base; two filter plates are fixed in the middle of the interior of the leaching barrel, and a material cavity is formed between the two filter plates; the leaching barrel is provided with at least two feed inlets in a communicating manner, the feed inlets are communicated with the material cavity, pistons are arranged at two ends of the leaching barrel in a sliding manner, and the pistons penetrate through the end part of the leaching barrel in a sliding manner through connecting rods and are fixedly connected with pressing ends; the driving gear is rotationally connected to the base and is connected with the leaching barrel;
the reciprocating driving piece is used for lifting in a reciprocating way and driving the driving gear to rotate in a reciprocating way, so that the leaching barrel rotates 180 degrees intermittently and is in a vertical state;
the pressing piece is fixedly connected to the reciprocating driving piece and located above the leaching barrel, when the rack moves downwards, the pressing piece extrudes the pressing end to enable the piston to move downwards, so that the organic solvent above the filter plate passes through the crushed paulownia flowers under the pressurizing effect to be extracted, and the piston below is enabled to slide and reset towards the end part of the leaching barrel.

The invention has the technical effects and advantages that:
1. the paulownia flower extract has natural fragrance, so that the original pungent smell of the feed can be reduced, the bad smell of the feed can be covered, and the comfort level of the feed can be improved; the preparation method is simple, low in cost, low in energy consumption, stable in property, free of toxic and side effects, and capable of reducing irritation of the feed, improving sweet taste, improving aftertaste and increasing feed intake of animals.
2. The special carrier can regulate and control the release speed of the effective components and the additive of the paulownia flower, so that the additive can be released for a long time and has a continuous food calling effect. The top aroma gives off the natural aroma of the paulownia flower extract, then the body aroma, the apple faint scent, and finally the bottom aroma, the strong milk aroma and the apple aroma are kept for a long time.
3. The combination of the paulownia flower extract and the traditional additive overcomes the defects of single function, non-lasting fragrance, over-concentrated fragrance and the like of the traditional additive, and the effective ingredient of the paulownia flower has higher utilization degree in pigs under the synergistic effect of the additive and better antibacterial and anti-inflammatory effects.
4. The extraction is carried out by using the improved extractor, and the extraction efficiency is high and the extraction speed is high.

The feed anti-bacterial additive comprises the following components: 36 g of paulownia flower extract, 3.7g of ethyl 2-methylbutyrate, 2.3g of hexyl acetate, 2.3g of phenethylisobutyrate, 2.3g of cinnamyl isovalerate, 4.5g of 5- (6) -decenoic acid, 0.6g of calcium carbonate, 0.6g of calcium phosphate, 20g of poly [1, 3-bis (p-carboxyphenoxy) propane-sebacic acid ], 20g of polylactic acid and 7.7g of polyethylene glycol. Preparing a paulownia flower extract: putting fresh paulownia flowers into a constant-temperature oven, drying for 5 hours, naturally cooling in the oven, taking out, weighing 200 g, grinding and crushing, sieving with a 20-mesh sieve, putting into a 1000-ml round-bottom flask, adding 500 ml of each of 95% ethanol and petroleum ether, extracting for 4 hours in an extractor at 40 ℃, filtering while hot after extraction is finished, reserving filtrate, washing residues with the organic solvent for 3 times, cooling the filtrate, combining the filtrate and a washing liquor, and evaporating the solvent under reduced pressure to obtain light red viscous liquid, namely the paulownia flower extract. The yield of the paulownia flower extract is 18 percent; the paulownia flower extract is added into the rest feed anti-substitution additives to prepare the feed anti-substitution additives, the feed anti-substitution additives account for 2.5 percent of the feed and prepare the feed, and the rest are nutritional ingredients and other additives, which are not described more.
Example 2
The feed anti-bacterial additive comprises the following components: 18 g of paulownia flower extract, 10.5g of ethyl 2-methylbutyrate, 4.6g of hexyl acetate, 4.6g of phenethylisobutyrate, 4.6g of cinnamyl isovalerate, 9g of 5- (6) -decenoic acid, 1.5g of calcium carbonate, 1.2g of calcium phosphate, 20g of poly [1, 3-bis (p-carboxyphenoxy) propane-sebacic acid ], 20g of polylactic acid and 6g of polyethylene glycol. Preparing the paulownia flower extract: putting fresh paulownia flower into a constant-temperature oven, drying for 5 hours, naturally cooling in the oven, taking out, weighing 100 g, grinding and crushing, sieving by a 20-mesh sieve, putting into a 1000-ml round-bottom flask, adding 400 ml of each of 95% ethanol and petroleum ether, extracting in an extractor for 5 hours at 50 ℃, filtering while hot after extraction is finished, reserving filtrate, washing residues by the organic solvent for 4 times, merging filtrate and washing liquor after the filtrate is cooled, and evaporating the solvent under reduced pressure to obtain light red viscous liquid, namely the paulownia flower extract. The yield of the paulownia flower extract is 18 percent; the paulownia flower extract is added into the rest feed alternative anti-additive to prepare the feed alternative anti-additive, the feed alternative anti-additive accounts for 2.5 percent of the feed and prepares the feed, and the rest is nutrient components and other additives, which are not described more.
Example 3
The feed anti-bacterial additive comprises the following components: the preparation method comprises the following steps of 20g of paulownia flower extract, 9.5g of ethyl 2-methylbutyrate, 4.6g of hexyl acetate, 4.6g of phenethyl isobutyrate, 4.6g of cinnamyl isovalerate, 9g of 5- (6) -decenoic acid, 1.5g of calcium carbonate, 1.2g of calcium phosphate, 20g of poly [1, 3-bis (p-carboxyphenoxy) propane-sebacic acid ], 20g of polylactic acid and 5g of polyethylene glycol. Preparing the paulownia flower extract: putting fresh paulownia flowers into a constant-temperature oven, drying for 6 hours, naturally cooling in the oven, taking out, weighing 100 g, grinding and crushing, sieving with a 20-mesh sieve, putting into a 1000-ml round-bottom flask, adding 500 ml of each of 95% ethanol and petroleum ether, extracting in an extractor for 7 hours at 60 ℃, filtering while hot after extraction is finished, reserving filtrate, washing residues with the organic solvent for 5 times, cooling the filtrate, combining the filtrate and a washing solution, and evaporating the solvent under reduced pressure to obtain light red viscous liquid, namely the paulownia flower extract. The yield of the paulownia flower extract is 20 percent; the paulownia flower extract is added into the rest feed alternative anti-additive to prepare the feed alternative anti-additive, the feed alternative anti-additive accounts for 5 percent of the feed and prepares the feed, and the rest is nutrient components and other additives, which are not described more.
Example 4
Referring to fig. 1, 3-4, the leaching vessel includes a base 1 and a leaching barrel 2.
The base 1 can be a base and a support structure, the base is of a flat plate structure, the supports are fixed at two ends of the base, the extraction barrel 2 is rotatably connected to the supports, and the base 1 completes the rotation support of the extraction barrel 2. The leaching barrel 2 can be a sealed barrel structure, and the middle position of the leaching barrel 2 is rotatably connected on the base 1. The rotation driving member is installed on the base 1 and connected to the incomplete gear 11 for driving the incomplete gear 11 to rotate, and the rotation driving member may be a motor 15 or a combination of the motor 15 and a pulley, which is not described herein in detail. The pressing piece 7 can be fixedly connected to the rack 8 and positioned above the leaching barrel 2, when the rack 8 moves downwards, the pressing piece 7 can press the pressing end 6 to enable the piston 16 to move downwards, so that the organic solvent above the filter plate 17 passes through the crushed paulownia flowers under the pressurization effect to be extracted, and the piston 16 below is enabled to slide towards the end part of the leaching barrel 2 to be reset. 2. The method for preparing the paulownia flower extract as the feed anti-additive according to claim 1, which is characterized in that:
the proportion of the paulownia flower extract in the feed substitute antibiotic additive is 15-20%;
the feed substitute antibiotic additive accounts for 2-5% of the feed.
3. The method for preparing the paulownia flower extract as the feed anti-additive according to claim 2, which is characterized in that: the feed anti-bacterial additive comprises the following components in parts by weight:
