Background
Since ancient times, chickens have taken an important position in the dietary structure of humans, not only providing high quality protein, but also being rich in lysine and threonine. The chicken breeding is the most active growth point and the main supporting industry in rural economy in China and is an important component of animal husbandry. However, in the process of chicken breeding, the chicken is easily attacked by pathogenic microorganisms such as escherichia coli, staphylococcus aureus, salmonella, pasteurella, rotavirus and the like, so that diseases such as coccidiosis, bronchitis, red and white dysentery, proventriculitis and the like are caused, and the chicken grows slowly and even dies. Antibiotics are often added to chicken feed to prevent disease. The use of the antibiotic feed additive greatly improves the production efficiency of chicken and eggs and promotes the high-speed development of chicken breeding industry. However, driven by interest, more and more antibiotics are used in chicken farming and are gradually evolving to be unjustified or even abused. Antibiotics become a major problem in the global public health field, not only pollute the environment and harm human health, but also cause bacteria to generate drug resistance and the like. In order to promote the green sustainable development of the breeding industry in China, the food safety is practically guaranteed, and the addition of antibiotics is completely forbidden. Therefore, under the condition of 'banning resistance', how to keep the continuous and efficient production of the breeding industry needs to be solved urgently to find a high-efficiency and safe 'substitute resistance' product.
Disclosure of Invention
In order to solve the shortage and the deficiency of the existing 'anti' -product and develop and create a green and safe 'anti' -compound preparation of a traditional Chinese veterinary medicine, the invention aims to provide a 'anti' -compound preparation of a traditional Chinese veterinary medicine and a preparation method and application thereof.
The invention provides the following technical scheme: a veterinary drug 'Tekang' compound preparation comprises 10-20 parts of lonicera confusa extract, 20-40 parts of macleaya cordata extract, 10-20 parts of scutellaria baicalensis extract, 10-20 parts of astragalus membranaceus extract, 5-20 parts of desmodium extract and 5-20 parts of cassia twig extract.
Preferably, the composition is prepared from the following raw materials in parts by weight: 15 parts of lonicera confusa extract, 35 parts of macleaya cordata extract, 15 parts of scutellaria baicalensis extract, 15 parts of astragalus membranaceus extract, 10 parts of desmodium extract and 10 parts of cassia twig extract.
The invention also aims to provide a preparation method of the traditional Chinese medicine compound preparation, which comprises the following steps: extracting the effective components of the Chinese medicinal materials with water and/or ethanol as extraction solvent by one or more of water extraction, acid extraction and alkali precipitation, ultrasonic-assisted extraction, and water extraction and alcohol precipitation, concentrating, and drying to obtain the Chinese medicinal extract.
Preferably, the water extraction method comprises the steps of adding 5-10L of water into 1kg of medicinal materials, soaking for 1-2 h, heating and refluxing for extraction for 1-3 times, each time for 1-3 h, combining filtrates, concentrating and drying to obtain the extract.
Preferably, the acid-base precipitation method comprises the steps of adding 1kg of medicinal materials into 5-10L of dilute hydrochloric acid with the pH value of 1-3, soaking for 1-2 h, heating, refluxing and extracting for 1-3 times, wherein each time is 1-3 h, adding NaOH solution with the pH value of 12-14 to adjust the pH value to 10-12 when the extracting solution is concentrated to 10-30% of the initial volume, precipitating, carrying out suction filtration to collect solids, and drying to obtain the extract.
Preferably, the ultrasonic-assisted extraction method is that 3 to 8L of ethanol is added into 1kg of medicinal materials, and ultrasonic extraction is carried out for 1 to 3 times at the power of 25 to 55kHz and the temperature of 20 to 60 ℃, and each time lasts for 0.5 to 1.0 hour; concentrating the obtained extracting solution, standing for 10-24 h, centrifuging at 5000-10000 rpm, taking supernatant, concentrating and drying to obtain the extract.
Preferably, the water extraction and alcohol extraction method comprises the steps of adding 5-10L of water into 1kg of medicinal materials, soaking for 1-2 h, heating, refluxing and extracting for 1-3 times, wherein each time is 1-3 h, adding absolute ethyl alcohol until the alcohol content is 80-90% when the extracting solution is concentrated to 10-30% of the initial volume, standing for 10-24 h at the temperature of 2-8 ℃, precipitating, carrying out suction filtration to collect solids, and drying the extract.
The improvement of the veterinary drug 'Tekang' compound preparation is that the selected traditional Chinese medicine components are macleaya cordata extract, scutellaria baicalensis extract, astragalus mongholicus extract, desmodium extract and cassia twig extract.
The preparation method of the veterinary drug compound preparation is further improved in that one or more methods of a water extraction method, an acid-extraction-alkali precipitation method, an ultrasonic-assisted extraction method and a water extraction-alcohol precipitation method are adopted to extract the effective components of the traditional Chinese medicinal materials.
The invention further provides application of the traditional Chinese medicine extract in preparing a veterinary medicine compound preparation, wherein the compound traditional Chinese medicine 'tianti' compound preparation is used for preventing and treating common pathogenic bacteria in chicken breeding: escherichia coli, staphylococcus aureus, salmonella, and Pasteurella to improve chicken breeding efficiency.
Due to the adoption of the technical scheme, the invention has the following beneficial effects: (1) The traditional Chinese medicine components of the traditional Chinese veterinary medicine compound preparation are reasonable in compatibility, the effective components are clear, the system is stable, and the product quality is guaranteed. (2) The preparation method of the traditional Chinese medicine compound preparation is simple to operate, the technology is mature and stable, and reagents used in the whole process are safe and have no toxic or side effect. (3) The Chinese medicinal compound preparation is a pure Chinese medicinal extract, does not contain chemical medicaments and antibiotics, and is non-toxic and free from medicament residues. (4) The traditional Chinese medicinal materials applied in the invention are common in the market, are low in price and easy to obtain, have low cost and can be widely popularized and used. (5) The veterinary drug 'tianti' compound preparation has the effects of resisting bacteria and diminishing inflammation, exerts the effects similar to antibiotics, can effectively prevent common pathogenic bacteria in chicken breeding, improves the breeding efficiency of livestock and poultry, ensures food safety and the like.

Lonicera confusa (Lonicera hypoglauca) is a dried bud or a flower with initial blossom of Lonicera macranthoides, lonicera hypoglauca, lonicera confusa or Lonicera fulvescens belonging to Caprifoliaceae, and is mainly distributed in Sichuan, guizhou, guangdong, guangxi, etc. It is recorded in the miscellaneous records of famous physicians, its taste is sweet, warm and non-toxic, mainly indicated for cold and heat, swollen body, and is listed as the top grade. 2020, recorded in Chinese pharmacopoeia, mountain honeysuckle flower has effects of clearing heat, detoxicating, dispelling wind and dissipating heat, and is mainly used for treating carbuncle swelling and sore, erysipelas, wind-heat type common cold, pyretic toxicity bloody dysentery, etc. The lonicera confusa contains various bioactive substances such as organic acids, flavonoids, saponins, volatile oils and the like, wherein the organic acids have remarkable bioactivity, are considered as main active ingredients of plant-derived medicaments and are the basis for clearing heat and removing toxicity of the lonicera confusa.
Macleaya cordata (willld) r.br.) is commonly known as "earlap", haotong pole, shangchun, etc., is a perennial herb of macleaya genus of papaveraceae family, and is widely distributed in the middle and lower reaches of the Yangtze river and north of south ridge in China. The compendium of materia medica is recorded in: 'Macleaya cordata, raw Jiangnan valley' \8230thatwhen folded, people die immediately and cannot enter the mouth. Has antiinflammatory, antibacterial, and pesticidal effects. The main active component of the compound is alkaloid, and representative compounds comprise sanguinarine, chelerythrine, protopine, allocryptopine and the like.
Scutellaria baicalensis Georgi (Scutellaria baicalensis Georgi), also known as root of Camellia japonica, root of Thalictrum japonicum, is a perennial herb of the genus Scutellaria of the family Labiatae. The Chinese medicinal composition is recorded in Shennong Bencao Jing for the first time, is bitter in taste and cold in nature, and belongs to the lung channel and the gallbladder channel, and clinical practices prove that the Chinese medicinal composition has multiple effects of resisting bacteria, inflammation and viruses, resisting oxidation, reducing blood fat, protecting liver and the like, is called as a Chinese medicinal 'antibiotic', and is also widely applied to veterinary medicine products. The scutellaria baicalensis contains flavonoid, polysaccharide, volatile oil and other substances, and the most main effective component of the scutellaria baicalensis is a flavonoid compound.
Astragalus (Astragalus membranaceus (fisch.) bungee) is a dried root of the leguminous plant Astragalus membranaceus capsular or Astragalus mongholicus, and was originally described in "shennong herbal" in: "mainly abscess deep-rooted carbuncle, long-term decay and wound, pus discharge and pain relief, wind and scab disease". Radix astragali has sweet taste and mild nature, enters spleen and lung meridians, has the effects of invigorating qi and invigorating yang, inducing diuresis to alleviate edema, benefiting defense and consolidating exterior, is mainly used for treating diseases such as spleen and stomach qi deficiency, exterior deficiency and spontaneous perspiration, chronic ulcer and unhealing, and has the effects of tonifying deficiency and assisting qi, expelling toxin, consolidating exterior and the like. The main effective components of radix astragali include saponins, polysaccharides and flavonoids.
Lysimachia christinae Hance is dry whole plant of primula family plant Lysimachia christinae Hance, cold in nature, sweet and salty in flavor, and enters liver, gallbladder, kidney and bladder meridians. Has the effects of clearing heat, promoting diuresis, treating stranguria, clearing liver and gallbladder damp-heat, and clearing heat and detoxicating, and is mainly used for treating heat stranguria, stranguria with stone, liver and gallbladder calculus, jaundice, toxic heat, carbuncle and swelling, etc. The main components are kaempferol and quercetin, and the product has pharmacological effects of promoting bile secretion and excretion, promoting urination and removing urinary calculus, promoting bile secretion and excretion, resisting inflammation, and resisting bacteria.
Ramulus Cinnamomi (Cinnamomum cassia Presl) is dry twig of Cinnamomum cassia Presl of Lauraceae, and has warm nature, pungent and sweet taste, and meridian tropism for heart, lung and bladder. The preparation method of the traditional Chinese veterinary medicine composition comprises the following steps:
removing impurities from the above 6 medicinal materials, respectively, and pulverizing into coarse powder with particle size of about 0.4 cm.
Extracting the effective component alkaloid in macleaya cordata by using an acid-extraction-alkali precipitation method, taking 1kg of medicinal materials, adding 5-10L of dilute hydrochloric acid with the pH value of 1-3, soaking for 1-2 h, then heating and refluxing for extraction for 1-3 times, wherein each time lasts for 1-3 h, adding NaOH solution with the pH value of 12-14 to adjust the pH value to 10-12 when the extracting solution is concentrated to 10-30% of the initial volume, precipitating, carrying out suction filtration to collect solid, and drying to obtain the macleaya cordata extract.
Extracting honeysuckle and scutellaria baicalensis by using an ultrasonic-assisted extraction method, adding 3-8L of ethanol into 1kg of medicinal materials, and ultrasonically extracting for 1-3 times at the power of 25-55 kHz and the temperature of 20-60 ℃ for 0.5-1.0 h each time; concentrating the obtained extracting solution, standing for 10-24 h, centrifuging at 5000-10000 rpm, taking supernatant, concentrating and drying to obtain the extract.
Extracting astragalus membranaceus by a water extraction and alcohol precipitation method, adding 5-10L of water into 1kg of medicinal materials, soaking for 1-2 hours, heating, refluxing and extracting for 1-3 times, wherein each time is 1-3 hours, concentrating an extracting solution to 10-30% of the initial volume, adding absolute ethyl alcohol until the alcohol content is 80-90%, standing for 10-24 hours at 2-8 ℃, precipitating precipitates, performing suction filtration, collecting solids, and drying the extract.
Extracting the desmodium and the cassia twig by a water extraction method, adding 5-10L of water into 1kg of medicinal materials, soaking for 1-2 h, heating and refluxing for extraction for 1-3 times, each time for 1-3 h, combining filtrates, concentrating and drying to obtain the extract.

Comparative example 1
The macleaya cordata powder serving as a traditional Chinese veterinary medicine 'tiaozhi' product sold in the market is selected as a contrast agent.
The effect of the compound preparation is compared and tested.
Selecting common pathogenic bacteria in chicken breeding: escherichia coli (CMCC 44102), staphylococcus aureus (CMCC 26003), salmonella (CMCC 50041) and Pasteurella (CVCC 393) are used as test objects, and the in vitro bacteriostatic activity of each Chinese medicinal extract is screened by using a plate method. The main operation steps are as follows:
preparation of a liquid medicine: dissolving the effective components of the traditional Chinese medicine by using water and dimethyl sulfoxide to prepare different concentration gradients.
Preparation of a culture medium: 9g of agar-free culture medium powder is accurately weighed, dissolved in 500mL of distilled water, adjusted to pH 7.2-7.4 by 10% NaOH, sterilized at high temperature and high pressure, and cooled for later use (2% of agar needs to be added into a solid culture medium).
Activating strains: taking 1mL of culture medium in a sterile centrifuge tube, unfreezing the strains stored in glycerol, sucking 10 mu L of bacterial liquid into the centrifuge tube, shaking up the bacterial liquid sufficiently, placing the centrifugal tube at 37 ℃ and culturing the bacterial liquid for 10 to 12 hours at 180r/min in a shaking way for later use.
And (3) amplification culture: after the strain is activated, pouring the strain into a conical flask containing 9mL of liquid culture medium, sealing the conical flask, and then placing the conical flask at 37 ℃ for shake culture at 180r/min for 3-4 h.
Determining the number of bacteria: sucking 1mL of the bacterial liquid after the amplification culture and 4mL of the liquid culture medium, uniformly mixing, taking the liquid culture medium as a blank, measuring an OD value at 600nm by adopting a spectrophotometer, and calculating the colony content (CFU/mL) in the bacterial liquid.
Dilution culture: taking the sterilized solid culture medium, airing until the solid culture medium is not hot and is in an unsolidified state, sucking a proper amount of bacterial suspension into the culture medium to enable the bacterial colony content to be 106CFU/mL, fully mixing uniformly, and pouring into a culture dish plate after being tilted.
Punching and dosing: after the culture medium is solidified, a hole (the diameter is about 3 mm) is punched on the culture medium by a puncher, the traditional Chinese medicine solution passes through a 0.22 mu L filter membrane to remove mixed bacteria, a 120 mu L injection hole is sucked, the injection hole is sealed and placed in a refrigerator at 4 ℃ to enable the liquid medicine to diffuse for 4h, the mixture is transferred into an incubator at 37 ℃ to be cultured for 18h, a flat plate is taken out, and the size of a bacteriostatic circle is observed and measured by a vernier caliper.
Data processing: each set of experiments was performed in 3 replicates and averaged, with the results shown in table 1.

The preparation method of the traditional Chinese medicine composition according to claim 3, wherein the water extraction method comprises the steps of adding 5-10L of water into 1kg of medicinal materials, soaking for 1-2 h, heating and refluxing for extraction for 1-3 times, each time for 1-3 h, combining filtrates, concentrating and drying to obtain the extract.

The method for preparing a Chinese medicinal composition according to claim 3, wherein the acid-base precipitation is carried out by adding 1kg of the medicinal material into 5-10L of dilute hydrochloric acid with pH value of 1-3, soaking for 1-2 h, heating, refluxing, extracting for 1-3 times, each time for 1-3 h, concentrating the extract to 10-30% of the initial volume, adding NaOH solution with pH value of 12-14, adjusting pH value to 10-12, precipitating, vacuum filtering, collecting solid, and drying.

The preparation method of the traditional Chinese medicine composition according to claim 3, wherein the ultrasonic-assisted extraction method comprises adding 3-8L ethanol into 1kg of medicinal materials, and ultrasonically extracting for 1-3 times at a power of 25-55 kHz and a temperature of 20-60 ℃ for 0.5-1.0 h each time; concentrating the obtained extracting solution, standing for 10-24 h, centrifuging under the condition of 5000-10000 rpm, taking supernatant, adjusting, concentrating and drying to obtain the extract.

The preparation method of the traditional Chinese medicine composition according to claim 3, wherein the water extraction and alcohol extraction method comprises the steps of adding 5-10L of water into 1kg of medicinal materials, soaking for 1-2 h, then heating and refluxing for extraction for 1-3 times, wherein each time lasts for 1-3 h, concentrating an extracting solution to 10% -30% of the initial volume, adding absolute ethyl alcohol until the alcohol content is 80% -90%, standing for 10-24 h at 2-8 ℃, precipitating, carrying out suction filtration to collect solids, and drying the extract.
8. Use of the Chinese medicinal composition of any one of claims 1-2 in the preparation of a veterinary drug "tikan" compound preparation.
9. The method as claimed in claim 8, wherein the said application is the control of pathogenic bacteria commonly found in chicken breeding processes: escherichia coli, staphylococcus aureus, salmonella, and pasteurella.
10. According to claim 8, the application object includes, but is not limited to, chicken breeding industry, and can be any breeding industry or a class of breeding industries.
