Description

Skin-moistening and nail-protecting composition containing modified chitosan and preparation method thereof
Technical Field
The invention relates to the technical field of health-care food, in particular to a skin-moistening and nail-protecting composition containing modified chitosan and a preparation method thereof.
Background
With the aging and the influence of environmental factors, such as ultraviolet irradiation, active oxygen radicals in human body increase, and the active oxygen can induce the peroxidation of unsaturated fatty acid (RH) in body lipids to generate lipid peroxidation free radicals. For example, malondialdehyde, a product of peroxidation of unsaturated lipids, plays an important role in the aging process of the human body. Lipid peroxidation also has a great impact on human health, and the liver is extremely vulnerable to lipid peroxide attack to cause liver disease.
The most obvious external manifestation is that the collagen of the human body tends to be lost, so that the skin loses moisture and luster, and the elasticity is deteriorated and roughened. The nails also decrease in gloss and begin to become brittle. Research has shown that the fibroblast productivity decreases from 25 years old, the loss of collagen, an organ of human body, which resists skin aging, increases, and the production and supply speed is not as high as that of human body.
Meanwhile, nowadays, in order to pursue diversification of beauty, nail beauty has become a general fashion trend. However, nails are mainly composed of keratin, and frequent use of various chemical agents for nail beautification can damage nails, cause phenomena such as nail breakage and nail brittleness, and the nails which should be pink can also be dark yellow. However, the existing research and the products on the market have little research on oral nail-care preparations, and more research focuses on nail polish. The product is generally prepared by taking essential oil as a main raw material and adding auxiliary agents such as phthalate, formaldehyde and the like and mixing, has a certain isolation effect, but the nail repair force is weakened after long-term use, and the effect is not durable.
In order to keep skin and nails young, external beauty products and medicines become choices of many people, but all the products are difficult to solve fundamentally. The theory of traditional Chinese medicine has been studied early, and the lung governs the skin and hair, and the liver governs the tendons. The stability of skin and nails is closely related to the balance of human viscera and diet, and the oral nutritional cosmetic can be used for regulating from inside to outside, and has better effect compared with the external cosmetic which is used alone, but the prior oral composition product is lacked, and has larger market blank.
In view of the above, there is a need to develop a new technical solution to provide a health product with good skin moistening and nail protection functions, so as to solve the problems in the prior art.
Disclosure of Invention
Based on the above, the invention develops the skin-moistening and nail-protecting composition containing the modified chitosan, which contains various active ingredients, and the modified chitosan is added, so that the composition has good skin-moistening, beautifying and nail-protecting effects, simultaneously has good stability and lasting effect, overcomes the defects in the existing products, and has good application prospect.
The invention aims to provide a skin-moistening and nail-protecting composition containing modified chitosan, which comprises the following components in parts by weight:
3-15 parts of peptide
0.15 to 0.75 portion of zinc element
0.2-0.6 part of propolis powder
0.05-0.3 parts of tremella extract
0.3-0.5 part of modified chitosan
0.1-0.3 part of mannitol;
wherein the content of the first and second substances,
the tremella extract comprises tremella polysaccharide;
the modified chitosan is obtained by the grafting reaction of chitosan and isonicotinic acid.
The propolis powder is a natural antioxidant, and can significantly improve the activity of superoxide dismutase (SOD) in a human body due to the fact that the propolis powder contains abundant flavonoid, terpene compounds and the like with antioxidant effect. According to the research at home and abroad, the propolis can inhibit the expression of downstream signal molecules NF-kBp65 and p53 and the level of ROS by reducing the activity of PC-PLC and the expression of TLR4 and inhibit the release of NO, thereby exerting the anti-inflammatory effect. In addition, the flavone in propolis can reduce the activity of serum alanine Aminotransferase (ALT) and aspartate Aminotransferase (AST), thereby improving the levels of SOD and GSH-Px in liver tissue, reducing the content of MDA, and protecting liver.
The tremella polysaccharide is similar to plant-derived hyaluronic acid substances, and the active center a- (1-3) -mannan can be combined with moisturizing factors such as amino acid in the skin, so that the barrier function of the skin is improved. In a mouse test, the tremella polysaccharide can obviously reduce the lipofuscin content of the myocardial tissue of a mouse and increase the activity of superoxide dismutase of the brain and the liver tissue. Meanwhile, the content of the P-450 of the liver serving as an example cell can be increased by promoting the synthesis of nucleic acid and protein.
Further, the preparation method of the modified chitosan comprises the following steps:
s1, dissolving chitosan in a solvent, adding isonicotinic acid and an activating agent, and stirring for reaction to obtain a crude product;
and S2, dialyzing the crude product, and freeze-drying to obtain the modified chitosan.
The isonicotinic acid grafted chitosan has better water solubility, and after the component is dissolved in water, the viscosity and the adsorption capacity of a product can be enhanced, so that active substances in the composition can be adsorbed in the solution for a long time, the action time of the product is prolonged, and the functions of long-acting skin moistening and nail protection are realized. Surprisingly, the modified chitosan has better compatibility with mannitol, tremella polysaccharide and other components, so that the active ingredients can stably exist in the composition, the phenomena of component aggregation and precipitation are effectively avoided, a good synergistic effect can be generated among various components, the absorption of the active ingredients is promoted, and the effects of inhibiting and eliminating free radicals and improving and repairing skin and nail injuries are achieved.
Further, in the step S1, the mass ratio of the chitosan to the isonicotinic acid is 1 (0.5-2).
Further, the activating agent is selected from one or more of EDC and NHS.
Further, in step S1, the chitosan has a molecular weight of 50-100 kDa and a degree of deacetylation of 80-95%.
Further, the peptide is selected from one or more of palmitoyl tripeptide, tetrapeptide, carnosine, collagen peptide, elastin peptide, antibacterial peptide, glutathione, wood frog polypeptide, soybean polypeptide, walnut polypeptide, wheat germ active peptide, fish polypeptide, scallop polypeptide, ascidian polypeptide, sea anemone polypeptide and seaweed polypeptide.
Further, the peptides are collagen peptides and scallop polypeptides.
The fish collagen peptide belongs to type I collagen, has the molecular weight of 30X 104Da, the fiber length of 280nm and the diameter of 1.5nm, is a slender needle-shaped rigid body, and forms a spiral structure by 3 alpha-peptide chains with the molecular weight of about 10X 104Da. Contains a large amount of hydrophilic groups, so that the skin moisturizing cream has a good moisturizing effect and can achieve the purpose of keeping the skin moist. It contains no cell and histocompatibility collagen, and does not induce antibody and immune response. And has very strong permeability to skin, and can rapidly supplement collagen lost by skin. The collagen is synthesized by fibroblasts under the catalysis of relevant growth factors and enzymes, is secreted to the outside of cells through exocytosis, and then forms collagen fibers under the action of extracellular enzymes, thereby achieving various skin-care and beauty-care effects.
The scallop polypeptide refers to water-soluble marine polypeptide extracted from viscera of chlamys farreri. Research reports that ultraviolet radiation damage generates superoxide anions to cause oxidative decomposition of cell membranes and enzyme protein sulfhydryl, so that cell damage is caused, and skin and nails are aged prematurely. The scallop polypeptide can effectively remove superoxide anions and hydroxyl free radicals in the skin, inhibit lipid peroxidation, increase the average thickness of fibroblasts and epidermis in skin tissues, and further delay skin aging.
Further, the collagen peptide is 3-10 parts by weight, and the scallop polypeptide is 0.05-0.5 part by weight.
Further, the content of the tremella polysaccharide is 0.1-0.2 part.
Further, the zinc element is selected from one or more of zinc sulfate, zinc glutamate, zinc acetate, zinc lactate, zinc gluconate, zinc glycinate and zinc-rich yeast.
Further, the zinc element is zinc-rich yeast.
Zinc in the zinc-rich yeast is an essential element for human body, and the lack of the element can cause the nail to have symptoms of uneven and easy to break, and the skin to be rough. In vivo, zinc has the effects of stabilizing SOD enzyme structure, scavenging free radicals, and protecting biological membrane.
It is another object of the present invention to provide a method for preparing the above skin and nail care composition containing modified chitosan, comprising the steps of: and sequentially adding the peptide, the zinc element, the propolis powder, the tremella extract and the mannitol into water, uniformly stirring, adding the modified chitosan, and stirring for 20-40 min to obtain the skin-moistening and nail-protecting composition containing the modified chitosan.
The invention has the following beneficial effects:
the skin-moistening and nail-protecting composition containing the modified chitosan adopts the isonicotinic acid grafted chitosan as an additive, the component has better water solubility, the viscosity of a solution of the composition can be improved, the release time of the active component is prolonged, and meanwhile, the active component and mannitol and other components can generate a good synergistic effect, so that the active component is uniformly distributed in the solution, the stability of the product is enhanced, and the full absorption of the active component is promoted, thereby realizing good skin-moistening, beautifying and nail-protecting effects.
