背景技术
某些金属的抗微生物特性在相当长的一段时间内已经为人所知。这种独特的特性已经在各种行业(包括农业和医疗保健)中得到利用,以试图防治感染和污染。

发明内容
本发明提供了一种新方法,其中通过将普通皮肤护理制剂的有利特性与化学键合的铜和锌的抗微生物特性相结合,可在医疗保健和家庭环境中实现优越的感染防治水平。这些特性的结合完美地适合应用于一系列旨在供个人使用的产品以最小化潜在有害微生物的传播。本发明的发明人出人意料地发现,本文公开的抗微生物组合物提供了比那些已有的可用产品更持久、更广泛和更优越的保护。此外,本发明的发明人还发现,本文公开的抗微生物组合物还有一个额外的优点,其为一种有效的抗衰老剂/促血管生成剂。因此,本发明具有来自单一产品或组合物的多种益处,所述单一产品或组合物可由个人定期使用。
在第一方面中,本发明提供了一种抗微生物组合物,其包括基质和金属组分,其中金属组分包含化学键合的铜和锌,并且其中基质包含用于受试者外部使用的皮肤病学组合物。
因此,本发明提供了一种适合作为感染防治剂、抗衰老剂和促血管生成剂的抗微生物组合物。

具体实施方式
下文的描述是为了使本领域的任何技术人员能够制造和使用本发明。对本领域的技术人员而言,对本文公开的实施方式的各种修改将是显而易见的。
在第一方面中,本发明提供了一种包含基质和金属组分的抗微生物组合物,其中金属组分包含化学键合的铜和锌,并且其中基质包含用于受试者外部使用的皮肤病学组合物。
术语“抗微生物组合物”是指具有抗微生物特性的组合物,例如生物杀灭或生物停滞(biostatic)特性。在本发明的上下文中,术语“生物杀灭”理解为能够破坏、阻止病原生物,使病原生物无害或对病原生物产生防治作用的物质,而术语“生物停滞”是指能够抑制生物体(例如微生物)的生长或增殖的物质。设想本发明将对任何微生物,例如任何细菌、病毒和/或真菌有用。特别地,设想葡萄球菌属(Genus Staphylococcus)和克雷伯氏菌属(Genus Klebsiella)的细菌、念珠菌属(Genus Candida)的真菌和冠状病毒科(Coronaviridae)族的成员(例如COVID-19)将对目前描述的材料敏感。因此,本发明可被描述为防腐的、抗细菌、抗病毒、抗真菌、抗病原体或抗微生物。
本发明提供的组合物具有出人意料地高抗微生物活性。因此,在本发明的一个实施方式中,提供了一种感染防治剂。由于要求保护的组合物具有高水平的抗微生物活性,因此与已有可用的产品(例如酒精凝胶消毒剂)相比,所述产品的功效受所用产品不同用量的影响较小。掺入本发明组合物中的皮肤病学产品将通过帮助防治微生物引起的感染的传播来减少感染的发生。本发明特别适用于预防与人与人接触有关的感染(例如握手),或表面与人的传染,其中存在于物体(例如门把手和水龙头)上的微生物由于与所述物体接触而转移到人身上。设想本发明对应用于通常暴露的区域特别有用,例如,个体的四肢,特别是他们的手和/或前臂。因此,除了减少感染的传播外,设想使用这种产品可以有效地治疗已经存在的感染,例如香港脚的感染或治疗指甲真菌。
本文的“基质”是指可掺入金属组分的任何合适的结构材料,从而提供可以将金属组分分散于其上或其中的物理介质。本发明的基质包含用于受试者外部使用的皮肤病学组合物。
优选地,用于受试者外部使用的皮肤病学组合物可以是乳膏、凝胶、洗剂、喷雾或软膏。这些皮肤病学组合物也可被描述为皮肤护理制剂或外用组合物/制剂。技术人员将理解,产品采用的形式可取决于所需的稠度、所需的用途和最终用户的皮肤类型。这些皮肤护理制剂相对于基于酒精的产品而言是有利的,因为其蒸发速度更慢,从而提供在延长的时期内具有活性的产品。
本文中的“化学键合”是指铜和锌的原子、离子或分子之间由于离子键合、共价键合或金属键合而产生的任何持久吸引力。因此,这可能包括铜合金或铜化合物,包括但不限于铜盐和氧化物。
优选地,抗微生物组合物的金属组分包含铜-锌合金。合金应理解为两种元素的混合物,其中一种是金属。在这种情况下,铜-锌合金应理解为替代式合金,即两种组分的原子可以在同一晶体结构中相互替换,从而产生大量的离域电子。
技术人员将认识到,为了制备所需的合金,元素铜和锌在凝固为一个新的和独特的化学实体之前以其熔融形式混合在一起。在一个实施方案中,设想将其他金属及其化合物(例如盐)掺入至材料或金属组分中。这些金属包括但不限于:锡、铁、铅、锆、铜、锌、银、金、钯、铂、铱、铝、镍、钨、钼、钽、钛、碘。可以理解的是,后面的化合物可能是所要求保护的材料的附加组分,其有助于进一步提高材料的抗微生物性能。
相对于纯金属或相关化合物的纯的形式,使用合金会产生一些与使用纯铜相比有利的特性。具体而言,铜-锌合金得益于锌的额外抗微生物特性、出色的可延展性/可铸性和高强度。
金属组分的颗粒预期测量为10-80μm,优选尺寸为15-30μm中的任何一点。与粗粉相比,细磨粉末会释放出更多的离子,其释放的离子可能是产生抗微生物效果的原因。
设想金属组分将含有至少60%的铜。这种配方将具有增强的抗微生物性能。优选地,金属组分包含75-80%的铜和相应的20-25%的锌。如上所述,金属组分还可以包含其他元素、化合物及其盐类。这些添加物可以赋予所要求保护的材料有益的特性。例如,额外的组分可以进一步增强抗微生物作用或允许所要求保护的产品的寿命延长。
在本发明的一个实施方式中,金属组分可以散布在整个基质中。本文中的“散布”是指金属组分分散在基质材料的颗粒/分子之间。这样的构造可替代地描述为“浸渍(impregnation)”。金属组分可以均匀地或不均匀地分散在整个基质材料中。技术人员会理解,散布、分散和/或浸渍的程度可能取决于要使用的基础护肤制剂和/或用于应用金属组分的方法。
设想基质可以包含软化剂、乳化剂、石油基剂、增稠剂或保湿剂。基质还可以进一步包含润湿剂、抗氧化剂、必需氨基酸、防腐剂和/或芳香剂。润湿剂的实例包括但不限于水杨酸、丙三醇、透明质酸、尿素、泛醇、乳酸钠和乙二醇。抗氧化剂的实例包括但不限于维生素C、维生素A(视黄醇)、维生素E、白藜芦醇、辅酶Q10、烟酰胺、多酚类、黄酮类和谷胱甘肽。必需氨基酸的实例包括但不限于精氨酸、组氨酸、甲硫氨酸、赖氨酸、脯氨酸、亮氨酸和甘氨酸。防腐剂的实例包括但不限于对羟基苯甲酸酯、苯氧乙醇和有机酸。基质的确切组成将取决于所需的产品的最终形式,例如,如果它是洗剂或凝胶。其他元素的包含,例如抗氧化剂和芳香剂,允许一种产品不仅具有抗微生物性,而且在美容环境中具有许多其他有益的特性。因此,本发明还提供了一种包含本文公开的抗微生物组合物的美容产品。我们所说的“美容产品”是指用于恢复或改善个体外观的任何产品。发明人未意识到有一种产品能够将令人印象深刻的高抗微生物性与此类产品相结合。
优选地,该基质在制造后可包括以下成分:
a)羧甲基纤维素钠
b)表面活性剂
c)丙三醇
d)柠檬酸
e)上文描述的任何前述组分或其混合物。
优选地,2-6重量%的基质由金属组分组成。例如,2-2.5%、2-3%、2-3.5%、2-4%、2-4.5%、2-5%、2-5.5%、2.5-3%、2.5-3.5%、2.5-4%、2.5-4.5%、2.5-5%、2.5-5.5%、2.5-6%、3-3.5%、3-4%、3-4.5%、3-4.5%、3-5%、3-5.5%、3-6%、3.5-4%、3.5-4.5%、3.5-5%、3.5-5.5%、3.5-6%、4-4.5%、4-5%、4-5.5%、4-6%、4.5-5%、4.5-5.5%、4.5-6%、5-5.5%、5-6%、5.5-6%。发明人出人意料地发现,这个范围内的金属组分可以保持产品所有合乎需要的抗微生物性能,制备效果持久可靠的产品并且在经济上是可行的。

本发明提供高水平的抗微生物活性,因此具有广泛的应用。本发明包括一种包含本发明的抗微生物组合物的感染防治产品。这样的产品可以在医疗环境中作为消毒产品使用,或在家庭环境中使用,例如,作为保持卫生的一部分,作为日常工作的一部分,或在接触特定表面后使用。本文中的“感染防治产品”是指治疗、预防或减弱感染的发展和/或传播的任何产品。
本发明还提供了一种作为抗衰老剂或作为促血管生成剂使用的抗微生物组合物。因此,本发明可用于治疗衰老的症状和/或改善皮肤的外观或与衰老有关的皮肤状况。因此,本发明提供了一种用于恢复或改善个人外观的美容产品。例如,该抗微生物组合物可用于预防、减少或延缓皱纹的形成、肤色和弹性的丧失、粉刺和黑头的形成。抗微生物组合物还可用于预防、减少或延缓皮肤变色如黄褐斑、老年斑或雀斑的形成,使干燥或受刺激的皮肤恢复活力,闭合或收紧毛孔,改善皮肤质地、光滑度或紧实度,并创造具有改善弹性的光滑和柔软的皮肤。后面的状况可通过本发明还是一种适合促进血管生成的剂而进一步改善。因此,本发明提供了一种通过促进血管生成来治疗衰老的抗微生物组合物。本发明还提供了一种通过促进血管生成来治疗、预防、减少或推迟与衰老有关的症状的抗微生物组合物。此类症状可包括皱纹、肤色丧失、弹性丧失、粉刺和黑头的形成、皮肤变色(如黄褐斑、老年斑或雀斑)、皮肤干燥、皮肤受刺激、毛孔张开、肤色粗糙或皮肤弹性降低。术语“血管生成”理解为指新的血管从先前存在的血管形成的生理过程。这一过程已建议作为一种可以治疗/改善/减少/延缓衰老症状的方法。因此,本发明提供了一种抗微生物组合物,其除了抗微生物特性外,还具有许多有利的特性。
在第二方面中,本发明提供了一种制造包含基质和金属组分的抗微生物组合物的方法,其中金属组分包含化学键合的铜和锌,基质包含用于受试者外部使用的皮肤病学组合物。该方法包含以下步骤:a)将铜和锌结合以制备所述金属组分;b)将该金属组分加热至熔融状态;c)用高速气体碎裂所述熔融状态,且;d)将被碎裂的金属组分与基质结合,其中基质包含本文所述的皮肤病学组合物。优选地,该皮肤病学组合物是一种乳膏、凝胶、洗剂、喷雾或软膏。
因此,制备本发明的金属组分的一种方法可能涉及等离子体或气体雾化方法。设想在本发明的方法中使用粉末状的金属,但其他形式的金属也可以是适当的,其可被本领域的技术人员所理解。
设想等离子体或气体雾化方法将产生粉末状的金属组分,其可酌情与基质结合,如会被本领域的技术人员所理解的那样。
在一个优选的实施方案中,在开始等离子体或气体雾化方法之前,可任选地通过使用机械磨碎工艺来减小金属组分的尺寸。本文中的“机械磨碎”是指结果是将金属组分逐渐分解成更小的元素的任何方法。该方法可以通过使用一些磨碎设备来实现,包括但不限于:磨碎机、卧式磨、1D振动磨、3D振动磨和行星磨。所有上述设备都由于研磨介质之间的撞击过程中传给样品的能量而导致尺寸减小。因此,在本发明的方法中金属形式的铜和锌可以被磨碎成对我们而言适当的形式。
一旦铜和锌已经结合,就可以进行雾化工艺。如本领域的技术人员所理解的,将铜和锌结合起来的方法可能会随所使用的雾化工艺而不同。
等离子体雾化要求金属组分呈丝的形式,以作为原料使用。如本领域技术人员所理解的,这通常是金属组分的合金丝。与传统的气体雾化相反,等离子体雾化使用等离子体火炬在单一步骤中瞬间熔化和雾化金属丝。然后使用冷却塔将形成的液滴转化为球形粉末。
替代地,可以使用传统的气体雾化。这可能涉及将铜-锌金属组分加热到约2000℃,以产生所述组分的熔融状态。本文中的“熔融状态”是指所述金属组分在暴露于高温下时的液体形式。如本领域技术人员所理解的,高速气流可以流经膨胀喷嘴,将熔化的金属组分从输入室虹吸。在该方法中可以使用的气体的实例包括氮气、氩气、氦气或空气。技术人员将认识到,在该方法中可以使用一种以上的气体,而优选的气体或气体混合物将是惰性/非活性的。所用气体的选择将取决于所需的最终被碎裂的金属(粉末)特性。为了提供适合用于本发明材料的金属组分,可能需要高速度的惰性气体。技术人员将认识到,所需的速度将随所使用的气体而有所不同,但可能在100-2000m/s的范围内。该方法破碎了熔融金属的液流,并导致产生精细颗粒,最终达到所需的粉末形式的金属组分。通过上述方法获得粉末形式有许多优点;制备高度球形的颗粒、低氧含量和对铜和锌的制备的适应性。技术人员还将认识到,可能存在制备金属粉末的替代性方法,其可用于实现同样的效果。
为了制备最终的抗微生物组合物,将金属组分加入至基质中。具体而言,加入少量金属粉末,直到全部产品转移到基质中。所得组合物在室温(20-22℃)下以350rpm混合2小时。
本发明还提供了一种预防或治疗感染的方法,其包括在医疗或兽医环境中使用本发明的抗微生物材料。
为了更清楚地理解本发明,现在将通过实施例的方式描述其实施方式。
实施例1
对两种不同的细菌菌株进行抗微生物组合物测试,所述菌株为:金黄色葡萄球菌(Staphylococcus aureus)和肺炎克雷伯菌(Klebsiella pneumoniae)。
在0.85%NaCl中,将每种试验生物制备为约1×105菌落形成单位(CFU)/mL。对于每个样品,使用每种试验生物进行5次平行接种。在接种点使用胰蛋白胨大豆琼脂(TSA)的倾注平板(pour plate)对接种物进行计数。接种的样品在24℃±1℃和>95%的湿度下保持24小时。暴露时间过后,将接种的试样无菌地移到9ml稀释液中。将其剧烈摇晃,以确保彻底重新悬浮任何余留的试验生物。将所得的悬浮液置于TSALT(补充有0.3%的大豆卵磷脂和3%Tween 80的TSA)中。平板在31℃±1℃下孵育至少5天。

对于样品“3%CuZn”和“15%CuZn”,在24小时的接触时间内,两种细菌菌株的数量都减少了>4log。与样品“0%CuZn”相比,它对两种试验生物都没有显示出明显的抗细菌活性。
实施例2
对真菌白色念珠菌(Candida albicans)的抗微生物组合物的测试。
在0.85%NaCl中,将每种试验生物制备为约1×106CFU/mL。对于每个样品,用适当体积的试验生物对试样进行5次平行接种(表2)。在接种点使用沙氏葡萄糖琼脂(SDA)的倾注平板对接种物进行计数。然后,将接种的样品置于24℃±1℃的孵育箱中,在>95%的湿度下孵育1、8或24小时。在规定的暴露时间过后,将接种的试样无菌地移到9ml稀释液中。将其剧烈摇晃,以确保彻底重新悬浮任何余留的试验生物。将所得的悬浮液置于SDALT(补充有0.3%大豆卵磷脂和3%Tween 80的SDA)中。平板在24℃±1℃下孵育至少5天。对于阴性对照,用适当体积(表2)的无菌0.85%NaCl接种样品,以与试样相同的方式进行孵育和分析。

对于样品“0%CuZn”,在24℃下接触1、8或24小时的时间后,未观察到白色念珠菌数量的明显减少。对于样品“2%CuZn”,在1小时的接触时间后,观察到白色念珠菌的数量减少超过3log;在24℃下接触8或24小时的时间后,观察到白色念珠菌的数量减少超过4log。对于样品“3%CuZn”,在1小时的接触时间后,未观察到白色念珠菌数量的明显减少;在24℃下8小时后,观察到白色念珠菌的数量减少超过3log;在24℃下24小时后,观察到白色念珠菌的数量减少超过5log。
实施例3
对牛冠状病毒(BCV)菌株L9的抗微生物组合物的测试。
为了制备材料,在无菌条件下切成1×1cm的碎片,经过折叠步骤后转移到Eppendorf杯中。为了制备测试病毒溶液,培养U373细胞。为了制备病毒,将BCV菌株L9加入至制备的单层中。在24-48小时的孵育期后,通过快速冷冻/解冻循环裂解细胞。去除细胞碎片并将上清液直接用作测试病毒悬浮液。通过使用微量滴定法通过终点稀释滴定测定传染性。通过计算与未处理材料的杀病毒活性相比滴定度的下降来评估处理材料的杀病毒活性。
表8-60分钟的暴露时间后,经处理(新的绿色/白色尼龙铜浸渍织物)和未经处理
的材料(参考:Tork Premium Special Tücher)的10倍试验的病毒滴度(牛冠状病毒)。
在60分钟的接触时间之后,使用新的绿色/白色尼龙铜浸渍织物,只能测出一种材料的残留病毒。相比之下,检查未处理的材料,在所有情况下都可以检测到残留病毒。得出以下平均值:≤1.55±0.04(新的绿色/白色尼龙铜浸渍织物)和2.98±0.12(参考)。根据60分钟暴露时间后的10倍测定,两种材料之间的1.43log10差异等级是明显的。
