Test data
Test grouping: selecting 60 healthy male SD rats with the weight of 200-220 g, adaptively feeding for 7d under the conditions of indoor temperature of 22-25 ℃ and relative humidity of 40-60%, free intake of sufficient water and food and the like, weighing (the weight is between 250-310 g), and randomly dividing into 6 groups according to the weight: blank (blank), model, example 1, example 2, example 3, aspirin positive control.
The test method comprises the following steps: the test group was continuously administered for 15 days at a dose of 30mL/60kg body weight, and the blank group and the model group were fed with equal volumes of distilled water via gavage. After the last administration, the test group and the model group duplicate migraine animal models by injecting nitroglycerin injection subcutaneously at 10mg/kg, and the blank group is injected with an equivalent amount of physiological saline. 3-5 min after molding, the phenomena of double ears reddening, back paw face beating, frequent front limb deflection, cage climbing and the like occur in the rat, the period lasts for about 3 hours, and the phenomenon of curling and lying occurs when the activity is reduced, so that the model is successfully copied. After the successful molding, 4 hours, 10% chloral hydrate was anesthetized, 3ml and 2ml of jugular blood of the rat were respectively taken and injected into a centrifuge tube containing 30. Mu.l of 10% EDTA-Na2 and 10. Mu.l of aprotinin (containing 100U/ml), and the mixture was gently shaken to mix the mixture, and the mixture was centrifuged at 3000rpm at 4℃for 10 minutes to separate plasma. Storing in a refrigerator with ultralow temperature of-80 ℃ for standby. Before measurement, putting the sample into ice water for re-melting, after mixing uniformly, centrifuging at 4 ℃ for 5min at 3000rpm again, taking supernatant, and respectively measuring the contents of CGRP and ET in the blood plasma according to the specification of the kit, wherein the results are shown in Table 1:
