a Bifidobacterium longum composition comprises carrier and Bifidobacterium longum, and has bacteria content of 6 × 10 10 -10 12 CFU/g;
The carrier consists of the following components in parts by weight: 20-32 parts of lactalbumin, 10-25 parts of trehalose, 5-8 parts of chitosan, 5-8 parts of sodium alginate and 0-0.3 part of nicotinamide.
By adopting the technical scheme, the bifidobacterium longum composition consisting of the bifidobacterium longum with the thallus concentration and the carrier forms an outer wall by compounding modified and solidified whey protein with trehalose and chitosan, forms a stable microemulsion gel structure by taking the bifidobacterium longum, sodium alginate and nicotinamide as inner cores, has long-acting and durable moisturizing effect when being applied to essence, has the moisture content of 39.32-43.52 in the eighth day of use, and has the moisture content increase rate of 23.2-34.9%.
The reason for analyzing the micro-emulsion gel structure is probably that the micro-emulsion gel structure effectively reduces the loss of strains and effective substances in the inner core through the protection of the outer wall on the inner core and the oxygen isolation effect, the basic effective activity of the bacteria is higher, when the micro-emulsion gel structure is rubbed and smeared on the skin and the outer wall is broken, the effective components in the inner core are released to the surface layer of the skin, and can quickly permeate into skin tissues, and simultaneously, the free radicals and epidermic bacteria microorganisms can be effectively reduced, so that the activity and the hydration degree of the epidermis are obviously improved, and in addition, a lubricating protective film can be formed on the surface of the skin, and the water loss of the epidermis can be effectively prevented.
Preferably, the carrier consists of the following components in parts by weight: 20-26 parts of lactalbumin, 15-20 parts of trehalose, 5-6 parts of chitosan, 5-6 parts of sodium alginate and 0.1-0.3 part of nicotinamide.
By adopting the technical scheme, the carrier with the optimized components has a better compounding effect with the bifidobacterium longum, and the sodium alginate and the nicotinamide in the carrier can be combined with the bifidobacterium longum to obviously inhibit the generation of free radicals, so that the vitality and the hydration degree of the skin are ensured.
Preferably, the bifidobacterium longum is one or more of BBMN68, BB536 and BL 21.
By adopting the technical scheme, the bifidobacterium longum strain is 6 x 10 10 -10 12 CFU/g has high effective activity, and can rapidly permeate into skin tissue, and simultaneously can be compounded with other components to reduce the generation of free radicals and epidermic bacteria microorganisms, thereby improving the activity and the hydration degree of the epidermis.

and A2, cooling the gel mixture to 30-50 ℃, adding bifidobacterium longum and nicotinamide, mixing, filtering and washing in a sterile manner, and freeze-drying to obtain the bifidobacterium longum composition.
By adopting the technical scheme, the preparation steps are simple, various conditions are easy to control, and meanwhile, the obtained bifidobacterium longum composition has high effective activity of strains, stable and uniform performance, can effectively form a microemulsion gel structure, and improves the activity and the hydration degree of epidermis.
Preferably, the Bifidobacterium longum is further subjected to ultrasonic wall breaking treatment, i.e. treatment at a power of 80-120W for 10-30S, before being added into the gel-like mixture.
By adopting the technical scheme, the broken product of the bifidobacterium longum subjected to the ultrasonic wall-breaking treatment contains a large amount of effective components capable of promoting the activity and the hydration degree of the epidermis, is easier to be absorbed by the epidermis, and can fully permeate into skin tissues to play a role in maintaining and moisturizing the skin.
In a third aspect, the present application provides an application of a bifidobacterium longum composition, which adopts the following technical scheme:

Detection test
The essences prepared in each example and comparative example are used as detection objects, the change value of moisture and the increase rate of moisture content after the application are tested, the test group is composed of 100 volunteers with healthy skin and 20-45 years old, the test group is randomly divided into 25 groups, each group comprises 4 persons, and the specific test method and the detection steps are as follows:
before the test, the test subject needs to use Corneometer CM825 PC to obtain blank value (MMV) of the test part (i.e. the area to be coated) 0 ) Measuring, continuously using corresponding essence for 8 days, respectively in the morning and evening, 3-5 drops (2 ml) each time, and testing water content (MMV) at the same time period after 8 days t ) And the average value of each detection result is recorded in the following table.
Wherein the moisture content increase rate of skin = (MMV) t -MMV 0 ) 0 x 100% of MMV, namely using the average value of final detection after 8 days to subtract a blank value to obtain the change value of the MMV in the period, and then removing the blank value to calculate the growth rate of the MMV.
Examples
Examples 1 to 6
An essence comprises the components and corresponding parts by weight shown in Table 2, and is prepared by stirring at 800r/min for 30 min;
wherein the bifidobacterium longum composition is prepared by the preparation example 1; the pH regulator is citric acid; the chelating agent is EDTA; the antioxidant is BHT; the emulsifier is sorbitan monopalmitate.
Table 2 the components and weights (kg) of the essences of examples 1-6
The essences prepared in the above examples 1 to 6 were extracted, the moisture content and the skin moisture content increase rate thereof after eight days of use were measured according to the above measurement method, and the measurement results were averaged and recorded in the following table.
As can be seen from the above table, the blanks (MMV) of the essences obtained in examples 1-6 were only 30.18-33.08; after trial for 8 days, the water content (MMVt) is increased to 39.32-42.78, and the increase rate of the water content is as high as 23.2-31.2%;
the essence with the components can quickly permeate into skin tissues, remarkably reduce the generation of free radicals and epidermic bacteria microorganisms through the compounding effect of multiple components, and form a lubricating protective film on the surface of the skin, so that the long-acting and durable skin care effect is achieved.
It can also be seen from the table that examples 3-4 are preferred examples, and it can be seen that the skin moisturizing effect of the essence with the above components is optimal, and the skin moisturizing effect does not continue to increase as expected with the increase of the amount of the bifidobacterium longum composition, and the reason for analyzing this may be that the excessive amount of the bifidobacterium longum composition increases the respiratory load of the skin surface layer instead because the production of free radicals and epidermal bacterial microorganisms on the skin surface has been suppressed to the minimum threshold, thereby slowing down the absorption of moisture by the skin, see examples 5-6.
Examples 7 to 11
The essence is different from the essence of example 1 in the using condition of the used bifidobacterium longum composition, and the specific corresponding relation is shown in the table below.
Table: EXAMPLES 7-11 use of Bifidobacterium longum compositions
Group of | Bifidobacterium longum composition |
Example 7 | Prepared from preparation example 2 |
Example 8 | Prepared from preparation example 3 |
Example 9 | Prepared from preparation example 4 |
Example 10 | Prepared from preparation example 5 |
Example 11 | Prepared from preparation example 6 |
The essences prepared in the above examples 7 to 11 were extracted, and the moisture content and the skin moisture content increase rate thereof after eight days of use were measured according to the above measurement method, and the measurement results were averaged and recorded in the following table.
As can be seen from the above table, the serum obtained in examples 7 to 11 had a blank value (MMV) of only 31.16 to 32.38; after trial for 8 days, the moisture content (MMVt) is increased to 41.58-43.52, and the moisture content increase rate is as high as 33.4-34.9%;
therefore, the bifidobacterium longum composition with the components can endow the essence with long-acting and long-lasting moisturizing and skin moistening effects when being applied to the essence, and the bifidobacterium longum composition exists in the essence in a microemulsion gel structure, so that the activity of effective components of thalli can be guaranteed.
In addition, from examples 7 to 11, it is also known that nicotinamide can significantly inhibit the generation of free radicals by being combined with bifidobacterium longum and sodium alginate, thereby further improving the moisturizing effect of the essence.
As can be seen from the above table, example 9 is a preferred example, and its blank value (MMV) is only 32.22; after trial for 8 days, the moisture content (MMVt) of the product is increased to 43.48, and the moisture content increase rate is as high as 34.9%; the component can be seen as the bifidobacterium longum composition with the optimal proportion.
When the essence needs to be used, the bifidobacterium longum composition with the microemulsion gel structure is broken on the outer wall in the process of rubbing and smearing the composition on skin, the active ingredients in the inner core of the composition are released to the surface layer of the skin and quickly permeate into skin tissues, and free radicals and epidermal bacterial microorganisms are effectively reduced, so that the activity and the hydration degree of the epidermis are obviously improved, and in addition, a lubricating protective film can be formed on the surface of the skin, so that the moisture loss of the epidermis can be effectively prevented, and a long-acting moisturizing effect is achieved.

Comparative example 1 | From comparative preparation example 1 |
Comparative example 2 | From comparative preparation example 2 |
The essences prepared in the above examples 12 to 13 and comparative examples 1 to 2 were extracted, the moisture content and the skin moisture content increase rate after eight days of use thereof were measured according to the above measurement methods, and the measurement results were averaged and recorded in the following table.
As can be seen from the above table, the serum obtained in examples 1, 12-13 had a blank value (MMV) of only 30.18-32.28; after trial for 8 days, the water content (MMVt) is increased to 39.32-40.03, and the increase rate of the water content is as high as 23.2-24.1%;
it can be seen that the above-mentioned Bifidobacterium longum bacteria have high effective activity, and can quickly permeate into skin tissue, and at the same time can be compounded with other components so as to reduce production of free radical and epidermic bacteria microorganism, so that it can raise epidermic vitality and hydration degree, and when the bacteria content is too low, its moisture-retaining effect is reduced, and when the bacteria content is continuously increased, its moisture-retaining and skin-moistening effect is not increased as expected, see comparative examples 1-2, so that 8 x 10 is comprehensively considered 9 -10 12 The bacterial content of CFU/g was set as the optimum value.
