Abstract
本发明公开了一种具有抗菌作用的婴儿祛湿护肤膏,包括以下重量份的原料:紫草油80.0‑85.0份、亳芍提取物5.0‑10.0份、亳丹皮提取物0.2‑0.5份、薄荷素油0.5‑1.0份、红没药醇复合抗菌剂0.05‑0.1份、蜂蜡8.0‑10.0份;其制备方法具体包括以下步骤:①将紫草油加热至85℃±5℃,加入蜂蜡融化,搅拌30‑40分钟混匀;②冷却至50℃±5℃时,依次加入亳芍提取物、亳丹皮提取物、薄荷素油、红没药醇复合抗菌剂,依次分别搅拌10‑15分钟、15‑20分钟、20‑30分钟、10‑15分钟混匀即可;制备的祛湿护肤膏具有良好的安全性,无毒副作用,具有抗敏止痒、抗炎、抑菌、皮肤屏障修复功能,适合一般人群使用,尤其适合婴幼儿皮肤外用。

技术领域
本发明涉及护肤品技术领域,具体涉及一种具有抗菌作用的婴儿祛湿护肤膏及其制备方法。
背景技术
俗称的婴儿红屁股,是指尿布疹(红臀),是婴儿中常见的皮肤病,发生在宝宝裹尿布部位,有局限性片状红斑或生水泡的病症,是一种皮肤炎性的病变。红臀为新生婴儿多见的皮肤炎症,多因小儿血热之体,皮肉娇嫩,尿布潮湿浸渍过久,湿热之气交蒸,致皮肤湮尻所致。其多发生在尿布覆盖部位,如会阴、阴囊、大腿内侧、臀部等处。其初可见片状红斑,与尿布覆盖面积相吻合,边缘清楚,因尿液浸渍,红斑上有小水泡,糜烂、渗液的现象,且日久红斑干燥,脱皮,皮肤粗糙,婴儿的啼哭吵闹其因主要是由于患部常潮湿不舒所致。
婴幼儿皮肤薄,皮脂少,屏障功能弱,对化学品、日晒、风吹、尿布摩擦和蚊虫叮咬等各种外界刺激抵御能力弱,受刺激后容易出现发热、晒伤、红苹果脸、红屁股和瘙痒等,而宝宝的抓挠会导致皮肤破损,引起皮肤感染,因此,需要开发一种具有抗菌作用的婴儿祛湿护肤膏。
红没药醇是存在于春黄菊花中的一种成份,春黄菊花的消炎作用主要来自红没药醇—一种单环不饱和半倍帖醇;红没药醇主要应用在皮肤保护和皮肤护理化妆品中,作为活性成分以保护和护理过敏性皮肤,适合于用在防晒产品,日光浴后洗澡液,婴儿产品和剃须后护理品中。
红没药醇的稳定性及很好的皮肤相容性,它很适合于用在化妆品中,长期保存不会变色,也不会渗出塑料容器,使用起来不会有什么问题,红没药醇不仅具有抗炎性能,还被证明有抑菌活性,抗刺激功效,保护皮肤功效,修复功效,但是护肤品中若直接性使用红没药醇有可能导致化妆品配方中其他有效成分的穿透性增加,穿透性增加会增加成分渗透进皮肤并从皮肤吸收进入血液的几率及含量,这样就会增加身体器官的代谢压力,因此,本发明采用红没药醇与壳聚糖进行复配得到红没药醇复合抗菌剂,在不影响抗菌消炎性能的前提下,进一步增加了皮肤锁水和修复功效,并且降低了红没药醇本身的使用量,更安全。
发明内容
本发明的目的在于提供一种具有抗菌作用的婴儿祛湿护肤膏及其制备方法。
本发明的目的可以通过以下技术方案实现:
紫草油功用凉血解毒,化腐生肌。主治水火烫伤、冻疮溃烂、久不收口等症。预防及治疗宝宝尿布疹、皮肤溃烂、湿疹、凉血、活血,解毒透疹。用于血热毒盛,斑疹紫黑,麻疹不透,疮疡,水火烫伤。在临床上,尿布性皮炎是接触性皮炎一类的皮炎,为儿科常见多发疾病,以皮肤红赤如丹、形如云片、游走不定为临床特征。如病例2,男,6个月,臀部皮疹4天就诊。两大腿内侧蔓延,成片集簇,红肿光亮,刺痒疼痛,使用紫草油,不少于4次/天,3天症状减轻,7天皮疹完全消失,嘱加强护理,保持皮肤清洁干燥,防止复发。
毫芍又称白芍,性微寒、味苦、酸,归肝、脾经。毫芍富含芍药甙、牡丹酚、芍药花甙和除虫菊素等,具有镇痛、抗炎、清热解毒、杀菌止痒以及免疫调节等作用。例如:毫芍不仅能对金黄色葡萄球菌、溶血性链球菌、草绿色链球菌、肺炎链球菌、伤寒杆菌、乙型副伤寒杆菌、痢疾杆菌、大肠杆菌、绿脓杆菌、变形杆菌、百日咳杆菌和霍乱弧菌等有不同程度的抑杀作用。而且在体外还能对堇色毛癣菌、同心性毛癣菌、许兰黄癣菌、奥杜盎小芽胞癣菌、铁锈色小芽胞癣菌、羊毛状小芽胞癣菌、腹股沟表皮癣菌、红色表皮癣菌和星形奴卡菌等皮肤真菌也有不同程度的抑杀作用。
亳丹皮,亳州地产药材,春秋季采挖3-5年的牡丹,除去根的表皮和皮肉木质,晾干,以其干燥根皮为原料萃取得到丹皮酚,性状为白色或微黄色有光泽的针状结晶,无色针状结晶,熔点49℃-51℃,气味特殊,味微辣,易溶于乙醇和甲醇中,溶于乙醚、丙酮、苯、氯仿及二硫化碳中,稍溶于水,在热水中溶解,不溶于冷水,能随水蒸汽挥发,化学式C9H10O3,分子量166.18,CAS号552-41-0,含量≥98%。质量指标符合《中国药典》(2020年版)一部牡丹皮的要求。
薄荷素油为唇形科植物薄荷Mentha haplocalyx Briq.的新鲜茎和叶经水蒸气蒸馏、冷冻、部分脱脑加工提取的挥发油,为无色或淡黄色的澄清液体;有特殊清凉香气,味初辛、后凉。质量指标符合《中国药典》(2020年版)一部薄荷素油的要求。
本发明制备的季铵盐抗菌剂不仅能够吸附于呈负电性的各类微生物上,抑制微生物分裂,使其丧失繁殖能力,而且还能形成薄膜堵塞微生物呼吸通道,加速微生物的死亡。本申请通过在抗菌制剂中加入季铵盐抗菌剂,能够使原抗菌制剂的抗菌效果更加快速高效。
蜂蜡为蜜蜂科昆虫中华蜜蜂或意大利蜂分泌的蜡。将蜂巢置水中加热、滤过、冷凝取蜡或再精制而成。为不规则团块,大小不一。呈黄色、淡黄棕色或黄白色,不透明或微透明,表面光滑。体较轻,蜡质,断面砂粒状,用手搓捏能软化。有蜂蜜样香气,味微甘。质量指标符合《中国药典》(2020年版)一部蜂蜡的要求。
一种具有抗菌作用的婴儿祛湿护肤膏,包括以下重量份的原料:紫草油80.0-85.0份、亳芍提取物5.0-10.0份、亳丹皮提取物0.2-0.5份、薄荷素油0.5-1.0份、红没药醇复合抗菌剂0.05-0.1份、蜂蜡8.0-10.0份。
进一步地,所述的紫草油提取工艺采用低温冷榨法:称取紫草药材打粉,过40目筛得到紫草细粉,加入6-8倍量茶油中,35-40℃低温浸泡提取3-5天,过滤、收集提取液,得到低温冷榨紫草油。
进一步地,所述的亳芍提取物的提取工艺为:亳芍中药材精选→称量→粉碎→8-10倍量水煎煮提取40-50分钟→药渣6-8倍量水煎煮提取30-40分钟→合并两次煎煮液→浓缩→干燥→粉碎、过筛。
进一步地,红没药醇复合抗菌剂的制备方法为:
第一步、改性壳聚糖的制备
称取1.0g壳聚糖于烧杯中,加入50-70ml浓度为3-5%的醋酸水溶液,在室温下搅拌1-1.5h,接着加入0.3-0.45g马来酸酐,搅拌10-15min,接着加入浓度为8-12%碳酸氢钠水溶液,调节溶液的pH为7.8-8.3,室温下继续搅拌8-10h,反应结束后,加入150-200ml无水乙醇,静置10-15min,过滤,经水洗、干燥后即得到改性壳聚糖;
第二步、红没药醇复合抗菌剂
将第一步制备的0.5g改性壳聚糖加入到烧杯中,加入30-50ml浓度为3-5%的醋酸水溶液和10-15ml乙醇,室温下搅拌溶解完全,接着加入50-60mg红没药醇,放入到水浴超声仪中,超声时间为3-5h,红没药醇上的羟基可与改性壳聚糖上的羰基形成氢键进行复配得到红没药醇复合抗菌剂。
进一步地,所述的壳聚糖的脱乙酰度为95%。
进一步地,第二步中,水浴超声仪的水浴温度为45-50℃,超声频率为20-40KHz。
一种具有抗菌作用的婴儿祛湿护肤膏的制备方法,具体包括以下步骤:
①将紫草油加热至85℃±5℃,加入蜂蜡融化,搅拌30-40分钟混匀;
②冷却至50℃±5℃时,依次加入亳芍提取物、亳丹皮提取物、薄荷素油、红没药醇复合抗菌剂,依次分别搅拌10-15分钟、15-20分钟、20-30分钟、10-15分钟混匀即可。
本发明的有益效果:
本发明公开了一种具有抗菌作用的婴儿祛湿护肤膏及其制备方法,具有良好的抗菌抗炎性能,对皮肤无刺激,且不会特别渗透肌肤到血液,具有良好的安全性,无毒副作用,具有抗敏止痒、抗炎、抑菌、皮肤屏障修复功能,适合一般人群使用,尤其适合婴幼儿皮肤外用。
当然,实施本发明的任一产品并不一定需要同时达到以上所述的所有优点。

下述实施例中:
紫草油提取工艺(低温冷榨法):称取紫草药材打粉,过40目筛得到紫草细粉,加入6倍量茶油中,40℃低温浸泡提取3天,过滤、收集提取液,得到低温冷榨紫草油。
亳芍提取工艺:亳芍中药材精选→称量→粉碎→8倍量水煎煮提取40分钟→药渣6倍量水煎煮提取30分钟→合并两次煎煮液→浓缩→干燥→粉碎、过筛。
红没药醇复合抗菌剂的制备方法为:
第一步、改性壳聚糖的制备
称取1.0g壳聚糖(脱乙酰度为95%)于烧杯中,加入50ml浓度为3%的醋酸水溶液,在室温下搅拌1h,接着加入0.35g马来酸酐,搅拌10min,接着加入浓度为10%碳酸氢钠水溶液,调节溶液的pH为7.8-8.3,室温下继续搅拌10h,反应结束后,加入200ml无水乙醇,静置10min,过滤,经水洗、干燥后即得到改性壳聚糖;如图1中A为改性壳聚糖的红外谱图,3022(-CH=CH-),1722(-CON-),1641(-COO-);
壳聚糖在碱性环境下与马来酸酐发生酰化反应,壳聚糖上的氨基与马来酸酐接枝得到的改性壳聚糖上含有羧基和酰胺基团。
第二步、红没药醇复合抗菌剂
将第一步制备的0.5g改性壳聚糖加入到烧杯中,加入40ml浓度为3%的醋酸水溶液和10ml乙醇,室温下搅拌溶解完全,接着加入52mg红没药醇,放入到水浴超声仪中,水浴温度为45℃,超声频率为30KHz,超声时间为5h,红没药醇上的羟基可与改性壳聚糖上的羰基形成氢键进行复配得到红没药醇复合抗菌剂。如图1中B为红没药醇复合抗菌剂的红外谱图,3262(-OH),3021、3007(皆为-CH=CH-,红没药醇上的双键吸收峰出峰位置略小于改性壳聚糖上的双键),1740(-CON-),1647(-COO-);
实施例1
一种具有抗菌作用的婴儿祛湿护肤膏,包括以下重量份的原料:紫草油80.0份、亳芍提取物6.0份、亳丹皮提取物0.4份、薄荷素油0.5份、红没药醇复合抗菌剂0.05份、蜂蜡10.0份。
该婴儿祛湿护肤膏的制备方法,具体包括以下步骤:
①将紫草油加热至80℃,加入蜂蜡融化,搅拌30分钟混匀;
②冷却至52℃时,依次加入亳芍提取物、亳丹皮提取物、薄荷素油、红没药醇复合抗菌剂,依次分别搅拌10分钟、15分钟、20分钟、10分钟混匀即可。
实施例2
一种具有抗菌作用的婴儿祛湿护肤膏,包括以下重量份的原料:紫草油85.0份、亳芍提取物5.0份、亳丹皮提取物0.2份、薄荷素油1.0份、红没药醇复合抗菌剂0.1份、蜂蜡10.0份。
该婴儿祛湿护肤膏的制备方法,具体包括以下步骤:
①将紫草油加热至85℃,加入蜂蜡融化,搅拌30分钟混匀;
②冷却至50℃时,依次加入亳芍提取物、亳丹皮提取物、薄荷素油、红没药醇复合抗菌剂,依次分别搅拌15分钟、20分钟、25分钟、12分钟混匀即可。
实施例3
一种具有抗菌作用的婴儿祛湿护肤膏,包括以下重量份的原料:紫草油82.0份、亳芍提取物7.0份、亳丹皮提取物0.3份、薄荷素油0.7份、红没药醇复合抗菌剂0.07份、蜂蜡9.0份。
该婴儿祛湿护肤膏的制备方法,具体包括以下步骤:
①将紫草油加热至90℃,加入蜂蜡融化,搅拌40分钟混匀;
②冷却至48℃时,依次加入亳芍提取物、亳丹皮提取物、薄荷素油、红没药醇复合抗菌剂,依次分别搅拌12分钟、18分钟、30分钟、15分钟混匀即可。
对比例1
一种具有抗菌作用的婴儿祛湿护肤膏,包括以下重量份的原料:紫草油82.0份、亳芍提取物7.0份、亳丹皮提取物0.3份、薄荷素油0.7份、红没药醇0.05份、蜂蜡10.0份。
该婴儿祛湿护肤膏的制备方法,具体包括以下步骤:
①将紫草油加热至85℃,加入蜂蜡融化,搅拌30分钟混匀;
②冷却至50℃时,依次加入亳芍提取物、亳丹皮提取物、薄荷素油、红没药醇复合抗菌剂,依次分别搅拌10分钟、20分钟、30分钟、10分钟混匀即可。
对比例2
不加入红没药醇复合抗菌剂,其余同实施例1。
对实施例1-3和对比例1-2制备的祛湿护肤膏进行性能测试
(1)抗菌性能测试
于广东省微生物分析检测中心按照《消毒技术规范(2002年版)》进行检测,检测所得的抑杀微生物试验结果如表1所示
由表1可知,实施例1-3和对比例1制备的祛湿护肤膏均具有优良的抗菌性能。
(2)抗过敏测试
参考《化妆品卫生规范》(2007年版)皮肤刺激反应评分标准评分;取皮肤状态良好的家兔5只,在各自背部脊柱两侧用电剃须刀进行剪毛,去毛范围左、右各2cm×2cm,取实施例1-3和对比例1-2制备的祛湿护肤膏各1g分别涂抹于对应的家兔脊背左侧上,右侧不涂抹,连续涂抹10天,每次涂抹前均用电剃须刀进行剪毛;
测试结果如表2所示

由上表2可知,实施例1-3和对比例1-2制备的祛湿护肤膏在家兔皮肤上均无过敏性反应。
(3)祛湿抗炎性测试
对年龄0-6周岁湿疹患者60例随机进行分组,分为6组,每组10人。1-5组分别对应实施例1-3和对比例1-2制备的祛湿护肤膏,第6组采用用氢化可的松软膏,早晚各一次,每次0.5g,涂抹7天进行回访查看,并每天对尿液进行进行拍照进行颜色对比;结果如表3所示:

1.一种具有抗菌作用的婴儿祛湿护肤膏,其特征在于,包括以下重量份的原料:紫草油80.0-85.0份、亳芍提取物5.0-10.0份、亳丹皮提取物0.2-0.5份、薄荷素油0.5-1.0份、红没药醇复合抗菌剂0.05-0.1份、蜂蜡8.0-10.0份;
所述的红没药醇复合抗菌剂的制备方法为:
第一步、改性壳聚糖的制备
称取1.0g壳聚糖于烧杯中,加入50-70ml浓度为3-5%的醋酸水溶液,在室温下搅拌1-1.5h,接着加入0.3-0.45g马来酸酐,搅拌10-15min,接着加入浓度为8-12%碳酸氢钠水溶液,调节溶液的pH为7.8-8.3,室温下继续搅拌8-10h,反应结束后,加入150-200ml无水乙醇,静置10-15min,过滤,经水洗、干燥后即得到改性壳聚糖;
第二步、红没药醇复合抗菌剂

2.根据权利要求1所述的一种具有抗菌作用的婴儿祛湿护肤膏,其特征在于,所述的紫草油提取工艺采用低温冷榨法:称取紫草药材打粉,过40目筛得到紫草细粉,加入6-8倍量茶油中,35-40℃低温浸泡提取3-5天,过滤、收集提取液,得到低温冷榨紫草油。
3.根据权利要求1所述的一种具有抗菌作用的婴儿祛湿护肤膏,其特征在于,所述的亳芍提取物的提取工艺为:亳芍中药材精选→称量→粉碎→8-10倍量水煎煮提取40-50分钟→药渣6-8倍量水煎煮提取30-40分钟→合并两次煎煮液→浓缩→干燥→粉碎、过筛。
4.根据权利要求1所述的一种具有抗菌作用的婴儿祛湿护肤膏,其特征在于,所述的壳聚糖的脱乙酰度为95%。
5.根据权利要求1所述的一种具有抗菌作用的婴儿祛湿护肤膏,其特征在于,第二步中,水浴超声仪的水浴温度为45-50℃,超声频率为20-40KHz。
6.一种如权利要求1-5所述的具有抗菌作用的婴儿祛湿护肤膏的制备方法,其特征在于,具体包括以下步骤:
①将紫草油加热至85℃±5℃,加入蜂蜡融化,搅拌30-40分钟混匀;
②冷却至50℃±5℃时,依次加入亳芍提取物、亳丹皮提取物、薄荷素油、红没药醇复合抗菌剂,依次分别搅拌10-15分钟、15-20分钟、20-30分钟、10-15分钟混匀即可。
