Various exemplary embodiments of the invention will now be described in detail, which should not be considered as limiting the invention, but rather as more detailed descriptions of certain aspects, features and embodiments of the invention.
It is to be understood that the terminology used herein is for the purpose of describing particular embodiments only and is not intended to be limiting of the invention. In addition, for numerical ranges in this disclosure, it is understood that each intermediate value between the upper and lower limits of the ranges is also specifically disclosed. Every smaller range between any stated value or stated range, and any other stated value or intermediate value within the stated range, is also encompassed within the invention. The upper and lower limits of these smaller ranges may independently be included or excluded in the range.

The embodiment of the invention provides a composite veterinary disinfectant containing glutaraldehyde and decamethylene bromide, which comprises the following raw materials in percentage by mass: 1 to 15 percent of glutaraldehyde, 1 to 10 percent of decamethyiammonium bromide, 0.1 to 1 percent of hyperbranched gemini quaternary ammonium salt, 5 to 20 percent of ethanol, 6 to 20 percent of glycerol and the balance of water.
The invention introduces the hyperbranched gemini quaternary ammonium salt into the composite veterinary disinfectant containing glutaraldehyde and decamethy bromide, thereby greatly improving the disinfection effect (more than 4 times) and having no influence on the properties of the disinfectant.
Aiming at the problem of inconvenient low-temperature storage of glutaraldehyde decamethylene bromide disinfectant, the invention adds ethanol and glycerol which are non-toxic and have no influence on the environment into glutaraldehyde decamethylene bromide disinfectant, so that the glutaraldehyde decamethylene bromide disinfectant can maintain remarkable sterilization effect at low temperature.
In the embodiment of the invention, the hyperbranched chain gemini quaternary ammonium salt is formed by connecting two single-chain single-head common surfactants at the ion head groups through chemical bonds, so that the head group separating force of the surfactants (decamethyibromide) in the ordered aggregation process is suppressed, the surface activity is greatly improved, the action on the biological film of bacteria and viruses is enhanced, and the disinfection capacity of the disinfectant is enhanced. The hyper-branched gemini quaternary ammonium salt is preferably a hydrocarbon single chain with the carbon number of 8-16, and has the strongest disinfection synergistic effect.
In the embodiment of the invention, the hyperbranched chain gemini quaternary ammonium salt is ethylene bis (dodecyl polyoxyethylene/polyoxypropylene block group ammonium bromide), the model is 31627, and the structural formula is
In the embodiment of the invention, the decamethenamine is biquaternary ammonium salt, and the molecule of the decamethenamine is provided with two long-chain hydrophobic groups and two positively charged nitrogen ions, so that the positive charge density on the nitrogen ions is increased through induction, the adsorption of the disinfectant on the surfaces of bacteria and algae cells is facilitated, and the permeability of cell walls is changed, so that the bacteria are broken. In addition, after the disinfectant is adsorbed on the surface of the thalli, hydrophobic groups and hydrophilic groups are favorable to respectively penetrate into a lipoid layer and a protein layer of the thalli cells, so that the enzyme is deactivated and the protein is denatured, and the bisquaternary ammonium salt is hardly influenced by temperature and pH value. The addition of the synergist (hyperbranched chain gemini quaternary ammonium salt) enhances the surface activity of the decamethy bromide, enhances the affinity of the decamethy bromide to a carbon chain, is easier to adsorb bacteria and viruses, and enables the disinfectant to have strong sterilization capability due to the combined effect of the functions.
Glutaraldehyde has good bactericidal effect when the pH value is 3.2-4.6, and the disinfectant has good glutaraldehyde stability in an acidic environment and strong virus inactivating capability when the pH value is about 4.1, and is suitable for room temperature storage.
The embodiment of the invention provides a preparation method of a composite veterinary disinfectant containing glutaraldehyde and decamethenamine bromide, which comprises the following steps:
(1) Weighing all raw material components according to the formula, mixing decamethyianium bromide and hyperbranched chain gemini quaternary ammonium salt, and stirring until the mixture is completely dissolved to obtain a first solution;
(2) Mixing glutaraldehyde, ethanol, glycerol and water uniformly to obtain a second solution;
(3) And (3) adding the first solution in the step (1) into the second solution in the step (2), and stirring until the first solution is completely dissolved to obtain the water-soluble organic compound.
Example 1
A composite veterinary disinfectant containing glutaraldehyde and decamethenamine bromide comprises the following components in parts by weight per 100g of disinfectant: 5g of glutaraldehyde, 5g of decamethylene bromide, 1g of ethylene bis (dodecyl polyoxyethylene/polyoxypropylene block group ammonium bromide), 15g of glycerol, 5g of ethanol, 3g of polyethylene glycol, 1g of sodium chloride and the balance of deionized water.
The composite veterinary disinfectant containing glutaraldehyde and decamethenamine in the embodiment has stable physical properties, good fluidity and no icing phenomenon at the temperature of-15 ℃.
Example 2
A composite veterinary disinfectant containing glutaraldehyde and decamethenamine bromide comprises the following components in parts by weight per 100g of disinfectant: glutaraldehyde 5g, decamethylene bromide 5g, ethylene bis (dodecyl polyoxyethylene/polyoxypropylene block ammonium bromide) 0.5g, and the balance deionized water.
Example 3
A composite veterinary disinfectant containing glutaraldehyde and decamethenamine bromide comprises the following components in parts by weight per 100g of disinfectant: glutaraldehyde 5g, decamethylene bromide 5g, and the balance deionized water.
The preparation method of the composite veterinary disinfectant containing glutaraldehyde and decamethenamine in the embodiment is as follows: weighing the raw material components according to the component amounts, dissolving the raw material components in deionized water, uniformly stirring, and fixing the volume to a target volume.
Example 4
A composite veterinary disinfectant containing glutaraldehyde and decamethenamine bromide comprises the following components in parts by weight per 100g of disinfectant: glutaraldehyde 5g, decamethylene bromide 4.5g, ethylene bis (dodecyl polyoxyethylene/polyoxypropylene block group ammonium bromide) 0.5g, glycerol 15g, ethanol 5g, polyethylene glycol 3g, and the balance deionized water.
The composite veterinary disinfectant containing glutaraldehyde and decamethenamine in the embodiment has stable physical properties, good fluidity and no icing phenomenon at the temperature of-20 ℃.
The preparation method of the composite veterinary disinfectant containing glutaraldehyde and decamethenamine in examples 1-2 and 4 comprises the following steps:
(1) Weighing all raw material components according to the formula, mixing decamethyianium bromide and hyperbranched chain gemini quaternary ammonium salt, and stirring until the mixture is completely dissolved to obtain a first solution;
(2) Mixing other components with deionized water and stirring uniformly to obtain a second solution;
(3) Adding the first solution in the step (1) into the second solution in the step (2), adding a proper amount of deionized water for constant volume, and continuously stirring until the deionized water is completely dissolved, thus obtaining the aqueous emulsion.
Application example 1
Examples 1-4 detection of the sterilizing effect of a composite high-efficiency disinfectant containing glutaraldehyde and decamethy bromoammonium:
Referring to the national standard GB15981-1995, the test strain is prepared into a bacterial suspension of 5X 10 6 CFU/mL by PBS, and the test strain is Escherichia coli. Taking the disinfectant of each embodiment, diluting the disinfectant by different multiples (400 times, 800 times, 1600 times, 3200 times and 6400 times) to obtain disinfectant diluents with different concentrations. 2.5mL of the toxin agent diluent is canceled, 2.5mL of bacterial liquid is added and mixed uniformly, 0.05mL is taken after 10 minutes and transferred into 4.95mL of sterilized culture medium, the culture is carried out at the constant temperature of a 37 ℃ incubator, the turbidity of the liquid in the test tube is observed at regular time, and the result is recorded, and the result is shown in Table 1. The sterilizing experimental effects of the disinfectant diluents with different concentrations in examples 1-4 are shown in fig. 1-4, wherein test tube numbers A, B, C, D respectively represent examples 1, 2,3 and 4; subscript 12345 is 1: 400. 1: 800. 1:1600;1:3200;1:6400, for example, reference A 1 in FIG. 1, indicates that the disinfectant of example 1 is 400-fold diluted.

Note that: in Table 1, "-" indicates sterility and "+" indicates sterility.
The sterilization rate results are shown in Table 2.

From the data in tables 1 and 2, it can be seen that each example exhibited a different antimicrobial effect, with the minimum kill concentration adding at least 4 times the ethylene-based bi-solvent. Among them, the sterilizing effect of examples 2 and 4 was superior to other examples.
Application example 2
Examples 1-4 detection of the content of glutaraldehyde and decamethylene bromide composite high-efficiency disinfectant components:
glutaraldehyde and decamethylene bromide contents of examples 1-4 were measured to meet the standards by reference to the detection method of veterinary drug quality standard (2017 edition).
Application example 3
Referring to GB/T38499-2020 disinfectant stability evaluation method, a constant temperature and humidity box is set to a condition temperature of 54 ℃ according to the regulations, samples of examples 1-4 are put into the box, and the content of each component is measured after 14 days. The glutaraldehyde content of each example after acceleration is shown in Table 3 and the decamethylene bromide content is shown in Table 4.

Accelerated experiments showed that example 3 had a reduced glutaraldehyde content and that the other examples were stable.
Application example 4
Low temperature stability detection
The disinfectant formulations of examples 1-4 were used for laboratory disinfection experiments, E.coli and Staphylococcus aureus were used as breeding bacteria, porcine reproductive and respiratory syndrome virus was used as viruses, and the disinfection period was 15min.
The disinfection experiment method comprises the following steps:
bacterial sheets (10 5~106 CFU/sheet) infected with specific bacteria (escherichia coli, staphylococcus aureus and porcine reproductive and respiratory syndrome virus) are placed in an autoclave sterilized sterile filter paper bag, and the bag mouth is fastened and placed in different places of a laboratory. After the sterilization is finished, viable bacteria are counted, and the sterilization rate is calculated.
The test bacteria interacted with the disinfectant for a prescribed period of time, the bacterial pieces were placed in a sterile PE tube containing 1.0mL of neutralizing agent, and mixed with an electric mixer for 20s, so that the bacteria on the bacterial pieces were eluted into the neutralizing liquid.
100. Mu.L of the above-mentioned washing solution was aspirated, and the number of viable bacteria was measured by the viable bacteria culture counting method.
Before the sprayer is replaced with disinfectant with new concentration, the disinfectant with new concentration should be cleaned and replaced with the disinfectant with new concentration.
Positive controls were established for viable counts directly with the infectious agent vector.
All test specimens were incubated at 37℃in an incubator and after 24 hours the final count was observed.
The test was repeated 3 times, the number of viable bacteria (CFU/sheet) of each group was calculated, and the sterilization rate (%).

Wherein n 0 is the number of viable bacteria in the control group before disinfection; n t is the number of living bacteria in the experimental group after disinfection. The sterilization test results are shown in Table 5.

The composite veterinary disinfectant sample states of examples 1-4 containing glutaraldehyde and decamethenamine are shown in figure 5 and are examples 1,2, 3 and 4 in sequence from left to right under the condition of freezing and storing at-20 ℃ for 50 days.

Claims (2)
1. The composite veterinary disinfectant containing glutaraldehyde and decamethylene bromide is characterized by comprising the following raw materials in percentage by mass: 1-15% of glutaraldehyde, 1-10% of decamethyiammonium bromide, 0.1-1% of a synergistic agent, 5-20% of ethanol, 6-20% of glycerol and the balance of water;
the synergistic agent is hyperbranched chain gemini quaternary ammonium salt, and the hyperbranched chain gemini quaternary ammonium salt is ethylene bis (dodecyl polyoxyethylene/polyoxypropylene block ammonium bromide).
2. A method for preparing the composite veterinary disinfectant containing glutaraldehyde and decamethyibromoammonium according to claim 1, which is characterized by comprising the following steps:
(1) Weighing the raw material components according to the formula, mixing the decamethyibromoammonium and the synergist, and stirring until the mixture is completely dissolved to obtain a first solution;
(2) Mixing glutaraldehyde, ethanol, glycerol and water uniformly to obtain a second solution;
(3) And (3) adding the first solution in the step (1) into the second solution in the step (2), and stirring until the first solution is completely dissolved to obtain the water-soluble organic compound.
