优选的,一种制备方法,用于制备自体胶原再生组合物,其包括以下步骤:
S1、将芦荟原液溶于氯化钠水溶液中,在机械搅拌条件下加入交联葡聚糖,获得基础液;
S2、将阿胶分解所得的,不含过敏原的多肽、氨基酸和微量元素的液体加入到基础液中,充分搅拌获得填充液;
S3、将自体成纤维悬浮液及自体胶原蛋白纤维加入到填充液中并进行充分搅拌即得自体胶原再生组合物。
优选的,自体成纤维悬浮液的制备方法包括以下步骤:
S21、在含有成熟的成纤维细胞的培养瓶内加入胰蛋白酶溶液于35~38℃条件下消化3~6min;
S22、用弯头吸管吸取培养瓶内的培养液,反复吹打瓶底壁,使已经消化的细胞脱离培养瓶底壁;
S23、离心800~1200r/min,3~5min,除去上清液;
S24、将细胞沉淀用生理盐水悬浮,细胞浓度为1×107个/ml,即制成自体成纤维细胞悬浮液。
优选的,成纤维细胞的培养方法包括:
S31、采用机械解离细胞法从皮肤组织中分离出成纤维前体细胞;
S32、对成纤维前体细胞进行破碎至糊状;
S33、将糊状的成纤维前体细胞加入到含培养液到烧杯中,通过振捣15秒,静至5至10分钟后,去除上清液,保留剩余组织;
S34、将所述剩余组织移入培养瓶内进行培养;
S35、在培养瓶内加入胎盘血清。
优选的,所述自体胶原蛋白纤维制备方法包括如下步骤:
S31、提取真皮成纤维细胞,通过体外扩增培养,获得源组织;
S32、对所述源组织进行去脂;

S24、将细胞沉淀用生理盐水悬浮,细胞浓度为1×107个/ml,即制成自体成纤维细胞悬浮液。
而成纤维细胞的培养方法包括:
S31、采用机械解离细胞法从皮肤组织中分离出成纤维前体细胞;
S32、对成纤维前体细胞进行破碎至糊状;
S33、将糊状的成纤维前体细胞加入到含培养液到烧杯中,通过振捣15秒,静至5至10分钟后,去除上清液,保留剩余组织;
S34、将所述剩余组织移入培养瓶内进行培养;
S35、在培养瓶内加入胎盘血清。
另外,所述自体胶原蛋白纤维制备方法包括如下步骤:
S31、提取真皮成纤维细胞,通过体外扩增培养,获得源组织;
S32、对所述源组织进行去脂;
S33、将蛋白酶加入至去脂后的所述源组织中去除胶原端肽;
S34、将碱灭活酶活性剂酸复溶加入至去除胶原端肽的所述源组织中,经纯化沉淀,获得所述自体胶原蛋白纤维。
以上所述,仅为本发明的具体实施方式,但本发明的保护范围并不局限于此,任何熟悉本技术领域的技术人员在本发明揭露的技术范围内,可轻易想到变化或替换,都应涵盖在本发明的保护范围之内。因此,本发明的保护范围应所述以权利要求的保护范围为准。
Claims (5)
1.一种能改善面部凹陷皱纹的自体胶原再生组合物,其特征在于,包括:自体成纤维悬浮液、自体胶原蛋白纤维、阿胶、芦荟原液、交联葡聚糖、氯化钠水溶液、胎盘血清。
2.一种制备方法,用于制备权利要求1中所述的自体胶原再生组合物,其特征在于,包括以下步骤:
S1、将芦荟原液溶于氯化钠水溶液中,在机械搅拌条件下加入交联葡聚糖,获得基础液;
S2、将阿胶分解所得的,不含过敏原的多肽、氨基酸和微量元素的液体加入到基础液中,充分搅拌获得填充液;
S3、将自体成纤维悬浮液及自体胶原蛋白纤维加入到填充液中并进行充分搅拌即得自体胶原再生组合物。
3.根据权利要求2所述的制备方法,其特征在于,所述自体成纤维悬浮液的制备方法包括以下步骤:
S21、在含有成熟的成纤维细胞的培养瓶内加入胰蛋白酶溶液于35~38℃条件下消化3~6min;
S22、用弯头吸管吸取培养瓶内的培养液,反复吹打瓶底壁,使已经消化的细胞脱离培养瓶底壁;
S23、离心800~1200r/min,3~5min,除去上清液;
S24、将细胞沉淀用生理盐水悬浮,细胞浓度为1×107个/ml,即制成自体成纤维细胞悬浮液。
4.根据权利要求3所述的制备方法,其特征在于:所述成纤维细胞的培养方法包括:
S31、采用机械解离细胞法从皮肤组织中分离出成纤维前体细胞;
S32、对成纤维前体细胞进行破碎至糊状;
S33、将糊状的成纤维前体细胞加入到含培养液到烧杯中,通过振捣15秒,静至5至10分钟后,去除上清液,保留剩余组织;
S34、将所述剩余组织移入培养瓶内进行培养;
S35、在培养瓶内加入胎盘血清。
5.根据权利要求2所述的制备方法,其特征在于,所述自体胶原蛋白纤维制备方法包括如下步骤:
