Abstract
本发明属于配位化学技术领域,具体涉及一种多模态分子影像探针及其制备方法和应用。本发明的多模式分子影像探针具有ABA结构,中心为钆配合物磁功能单元,配合两个相同的铱配合物磷光功能单元,整体有机整合在同一个配合物分子中。该多模式分子影像探针在同一个配合物分子中同时引入了两个铱配合物光功能单元和一个钆螯合物磁功能单元,具有磁‑光双功能性质,既可以作为核磁造影剂用于磁共振成像,又可以作为光学探针用于光学成像。

技术领域
本发明属于配位化学技术领域,具体涉及一种多模态分子影像探针及其制备方法和应用。
背景技术
分子影像技术能够从生理、生化水平上为癌症的起因、发生、发展等一系列的病理生理变化提供可视化和计量化的结构和功能信息。目前,常见的分子影像技术有:X射线(X-ray)、计算机断层扫描成像(CT)、超声成像(US)、光学成像(OI)和磁共振成像(MRI)等。每一种成像技术都有其各自的优势和不足。单一的成像模式已无法为临床诊断提供足够信息。而综合应用多种成像手段必然可以获得更加全面的成像结果,进而更好地了解疾病生物学机制。因此,发展多模式分子影像探针,综合应用多种成像技术,并通过不同成像手段之间优缺点互补,将为疾病的精准治疗提供更全面深入的临床诊断信息。
近年来,磁-光双模式分子影像探针已成为分子影像学的研究热点。光学成像技术可以通过对细胞、组织病理切片等微观生物样本进行可视化观察,实现生理过程中分子水平变化的实时定性及定量检测,具有灵敏度高、选择性好等诸多优点,成为国际上公认的开展活体内分子事件研究的主流研究手段之一。不过,光学成像也存在光学穿透深度不足的问题,难以实现对较大动物模型及人的深层次成像。磁共振成像可以对大型活体样本进行全方位多角度的整体断层扫描式成像,对各种发生了解剖学改变的活体器官和组织进行更准确的成像。但磁共振成像也存在空间分辨率不够、灵敏度低、成像时间长等不足。由此可见,光学成像和磁共振成像优缺点之间存在较好的互补性。磁-光双模式分子探针可以利用两种成像手段之间的互补性,充分发挥磁共振成像无组织穿透深度限制和光学成像灵敏度高的优点,同时获得解剖学信息和分子功能性信息,完成单一成像手段难以完成的诊断任务,并提供更全面的成像信息。但是,目前,现有技术中公开的分子影像探针或具有磁共振成像的特点,或具有光学成像的特点,成像单一,限制了其应用。
发明内容
有鉴于此,本发明提供一种多模式分子影像探针及其制备方法和应用,本发明提供的多模式分子影像探针既包含了具有光学性质的铱配合物,又包含了具有磁学性质的钆配合物。
为了实现上述发明目的,本发明提供了一种多模式分子影像探针,具有式I所示结构:
所述M包括Cl-;所述R和L独立地为卤基、烷基或氢;所述Z为-O-或-S-。
优选地,所述M替换为Br-,I-,NO3 -或PF6 -。
本发明还提供了上述所述的多模式分子影像探针的制备方法,包括以下步骤:
将无机钆盐和二亚乙基三胺五乙酸衍生物溶解,进行第一配位反应,得到钆配合物;
将所述钆配合物和铱配合物前驱体溶解,进行第二配位反应,得到所述多模式分子影像探针;
所述二亚乙基三胺五乙酸衍生物具有式I-1所示结构;
所述铱配合物前驱体具有式I-2所示结构;
优选地,所述无机钆盐包括硝酸钆、氯化钆和高氯酸钆中的一种或多种。
优选地,所述无机钆盐的摩尔数与无机钆盐和二亚乙基三胺五乙酸衍生物的摩尔数之和的比例为0.2~0.8:1。
优选地,所述钆配合物的摩尔数与钆配合物和铱配合物前驱体的摩尔数之和的比例为0.3~0.7:1。
优选地,所述第一配位反应的温度为45~55℃,时间为23~25h。
优选地,所述第二配位反应的温度为40~60℃,时间为22~26h。

本发明提供了一种多模式分子影像探针,具有式I所示结构,本发明的多模式分子影像探针具有ABA结构,B为钆配合物磁功能单元,A为铱配合物磷光功能单元,整体有机整合在同一个配合物分子中。该多模式分子影像探针在同一个配合物分子中同时引入了两个铱配合物光功能单元和一个钆配合物磁功能单元,具有磁-光双功能性质,既可以用于制备核磁造影剂,又可以用于制备光学成像的光影剂。
本发明还提供了上述所述多模式分子影像探针的制备方法,包括以下步骤:将无机钆盐和二亚乙基三胺五乙酸衍生物溶解,进行第一配位反应,得到钆配合物;将所述钆配合物和铱配合物前驱体溶解,进行第二配位反应,得到所述多模式分子影像探针。本发明以二乙三胺五乙酸衍生物为原料,二乙三胺五乙酸衍生物中二乙三胺五乙酸结构可以与钆离子结合形成稳定的磁学功能单元,二乙三胺五乙酸衍生物中邻菲罗啉结构可与铱配合物前驱体结合,形成磷光功能单元,在单个配合物分子中实现了光功能和磁功能的有机统一。

本发明中将无机钆盐和二亚乙基三胺五乙酸衍生物溶解,进行第一配位反应,得到钆配合物。
在本发明中,所述无机钆盐优选包括硝酸钆、氯化钆和高氯酸钆中的一种或多种,更优选为硝酸钆。所述无机钆盐的摩尔数与无机钆盐和二亚乙基三胺五乙酸衍生物的摩尔数之和的比例优选为0.2~0.8:1,更优选为0.5:1。
在本发明中,所述无机钆盐和二亚乙基三胺五乙酸衍生物溶解的试剂优选包括甲醇水溶液和/或乙醇水溶液;所述甲醇水溶液中甲醇和水的体积比优选为1:5~5:1,更优选为1:1~3:1;所述乙醇水溶液中乙醇和水的体积比优选为1:5~5:1,更优选为1:1~3:1。
在本发明中,所述第一配位反应的温度优选为45~55℃,更优选为50℃;时间优选为23~25h;更优选为24h。在本发明中,所述第一配位反应优选在保护气氛中进行,所述保护气氛优选包括氦气和/或氮气,更优选为氦气。
在本发明中,以硝酸钆为例,所述第一配位反应的方程式为:
所述第一配位反应后,本发明优选还包括将第一配位反应所得产物进行过滤。本发明对所述过滤不作具体限定,采用本领域技术人员熟知的操作即可。
得到钆配合物后,本发明将所述钆配合物和铱配合物前驱体溶解,进行第二配位反应,得到所述多模式分子影像探针。
在本发明中,所述钆配合物的摩尔数与钆配合物和铱配合物前驱体的摩尔数之和的比例优选为0.3~0.7:1,更优选为0.5:1。
在本发明中,所述钆配合物和铱配合物前驱体溶解的试剂优选为二氯甲烷和甲醇的混合溶剂;所述混合溶剂中二氯甲烷和甲醇的体积比优选为1:1~3,更优选为1:2。
在本发明中,所述第二配位反应的温度优选为40~60℃,更优选为50~55℃;时间优选为22~26h,更优选为24~25h。在本发明中,所述第二配位反应优选在保护气氛中进行,所述保护气氛优选包括氦气和/或氮气,更优选为氦气。
在本发明中,所述第二配位反应的方程式为:
在本发明中,当M替换为Br-,I-,NO3 -或PF6 -时,替换方法优选包括:将第二配位反应得到的产物进行离子置换反应;所述离子置换反应的试剂包括NaBr、NaI、NaNO3或六氟磷酸铵。
在本发明中,离子置换反应前,优选还包括将所述第二配位反应所得产物浓缩至干,浓缩所得产物以二氯甲烷复溶。
在本发明中,所述离子置换反应的试剂优选包括NaBr、NaI、NaNO3或六氟磷酸铵,更优选为六氟磷酸铵。在本发明中,所述第二配位反应所得产物和离子置换反应的试剂的摩尔比优选为1:1~1.5,更优选为1:1.2~1.3。
在本发明中,所述离子置换反应的温度优选为10~50℃,更优选为25~30℃,时间优选为0.5~5h,更优选为2~3h。
在本发明中,得到探针粗品后,优选还包括将探针粗品依次进行柱色谱分离和浓缩,所述柱色谱分离的洗脱剂为二氯甲烷和甲醇的混合溶剂;所述混合溶剂中二氯甲烷和甲醇的体积比优选为1:1。本发明对所述浓缩不作具体限定,采用本领域技术人员熟知的操作将洗脱液中的溶剂去除即可。
本发明还提供了所述多模式分子影像探针或上述所述的制备方法制备得到的多模式分子影像探针在制备磁共振成像造影剂或制备光学成像的光影剂中的应用。
下面将结合本发明中的实施例,对本发明中的技术方案进行清楚、完整地描述。显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
实施例1
将600mg Gd(NO3)3·6H2O、1100mg二亚乙基三胺五乙酸衍生物和80mL甲醇加入到反应器中,氮气保护下,在50℃恒温搅拌24h进行第一配位反应,过滤,收集沉淀,得到钆配合物。钆配合物的MS(ESI+)m/z为903.1219。
第一配位的反应方程为:
将144mg钆配合物和206mg铱配合物前驱体[Ir2(ppy)4Cl2]加入50mL二氯甲烷和甲醇的混合溶剂(两者体积比为1:1)中,在氮气保护下,以温度55℃,进行第二配位反应24h,得到钆配合物。
第二配位的反应方程为:
实施例2
按照实施例1的方案进行第二配位反应,将第二配位反应所得产物旋转蒸发至干,加少量二氯甲烷溶解,与58mg六氟磷酸铵在25℃进行离子置换反应1h,得到探针粗品,所得探针粗品经柱色谱分离,洗脱剂为二氯甲烷和甲醇的混合物(体积比为1:1),所得洗脱液浓缩,得到多模态分子影像探针。

本发明对实施例2制备的多模态分子影像探针进行了光学性质研究。多模态分子影像探针的紫外可见吸收光谱和磷光发射光谱均在水溶液中进行,其中磷光发射光谱采用380nm紫外光激发,测试结果见图1和图2。从图1可知:该多模态分子影像探针在紫外区(230~320nm)范围内有较强的吸收,在320~450nm范围内具有相对较弱的吸收。从图2可知:该多模态分子影像探针在380nm紫外光激发下,在520~700nm范围内具有磷光发射,发射峰位于580nm左右。这种紫外吸收和磷光发射性质,为该探针用于光学成像奠定了基础。
本发明对实施例2制备的多模态分子影像探针的活细胞光学成像研究。实验过程为:将处于对数生长期的EMT6细胞分至共聚焦35mm共聚焦培养皿中,在37℃下含有10%胎牛血清的1640培养液中贴壁生长24h,然后加入30μM多模态分子影像探针,孵育2h,再加入细胞核染料Hochest 33258、线粒体红色荧光探针及溶酶体绿色荧光探针,共同孵育15min,用PBS洗涤3次,然后立即进行共聚焦显微成像。细胞光学成像是在蔡司激光共聚焦显微镜LSM900上完成,实验结果如图3所示,从图3可知:该多模态分子影像探针能够进入细胞,并用于活细胞成像。将该探针与线粒体、溶酶体、细胞核商业化染料进行共染,发现该探针与线粒体商业化染料具有很高的重叠性,表明该探针进入细胞中的线粒体,具有线粒体靶向性。
本发明对多模态分子影像探针弛豫率进行了测定。测试方法为:将多模态分子影像探针用体积浓度为50%的乙腈水溶液配制总体积为1.5mL的溶液,浓度分别为0.4;0.2;0.1;0.05;0.025以及0mM,用MesoMR核磁共振分析与成像系统(上海纽迈电子科技有限公司生产)对上述样品进行T1加权成像,共振频率23.314MHz,磁体强度0.5T,线圈直径为60mm,磁体温度为32℃。实验结果如图4所示,由图4可知:该多模态分子影像探针浓度和1/T1成正比,即探针浓度越高,T1加权图像越亮,弛豫率高达10mM-1s-1,远高于商业化钆类造影剂。
本发明对多模态分子影像探针活体磁共振成像进行了探究,实验步骤为:取一只20g左右的荷瘤小鼠,用8wt%的水合氯醛溶液150μL腹腔注射,待荷瘤鼠麻醉后,成像作为空白图像。随后将多模态分子影像探针注射到实验鼠肿瘤内。在注射30min、1h两个时间点,用MesoMR23-060H-I中尺寸核磁共振分析与成像系统(苏州纽迈电子科技有限公司生产)对上荷瘤小鼠进行磁共振成像,共振频率23.313MHz,磁体强度0.55T,线圈直径为40mm,磁体温度为32℃。使用核磁共振成像软件及MSE序列采集裸鼠的冠状面和横断面图像。实验结果如图5所示,从图5可知:裸鼠的横断面面图中,可以观察到肿瘤。对比不同的时间点,可以看出该多模态分子影像探针注射到肿瘤内,肿瘤有明显的造影效果,随着造影时间的增加,肿瘤亮度也增加,30min时造影效果最好,然后肿瘤亮度开始减小,60min时造影剂已经开始代谢,造影效果减弱。
以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和修饰也应视为本发明的保护范围。

3.根据权利要求2所述的制备方法,其特征在于,所述无机钆盐包括硝酸钆、氯化钆和高氯酸钆中的一种或多种。
4.根据权利要求2所述的制备方法,其特征在于,所述无机钆盐的摩尔数与无机钆盐和二亚乙基三胺五乙酸衍生物的摩尔数之和的比例为0.2~0.8:1。
5.根据权利要求2所述的制备方法,其特征在于,所述钆配合物的摩尔数与钆配合物和铱配合物前驱体的摩尔数之和的比例为0.3~0.7:1。
6.根据权利要求2或4所述的制备方法,其特征在于,所述第一配位反应的温度为45~55℃,时间为23~25h。
7.根据权利要求2或5所述的制备方法,其特征在于,所述第二配位反应的温度为40~60℃,时间为22~26h。
8.权利要求1所述的多模式分子影像探针或权利要求2~7任一项所述的制备方法制备得到的多模式分子影像探针在制备磁共振成像造影剂或制备光学成像的光影剂中的应用。
