Abstract
The invention provides a preparation method of a chitosan-astragalus fiber film doped with essential oil, which relates to the field of deep utilization of astragalus and the field of food fresh-keeping packaging, and comprises the steps of weighing 0.05-10 g of dried astragalus fiber, adding the dried astragalus fiber into 50-250 ml of water, and uniformly stirring; adding one or more of acetic acid, tartaric acid, malic acid, oxalic acid, citric acid, benzoic acid, salicylic acid, vitamin C, hydrochloric acid, sulfuric acid, and phosphoric acid, adjusting pH to 2-7, and preparing 10-50 ml of chitosan with mass concentration of 0.05-10%; adding one or more of sodium tripolyphosphate, polyvidone, polyethylene glycol-4000, urea, polyethylene glycol-6000, citric acid, and succinic acid, and adjusting pH to 1-10; the film packaging material with the functions of antibiosis, antioxidation, fresh keeping and health care is obtained, and is used for the fresh keeping of picked fresh vegetables, fruits, medicinal materials and the like and also can be used for the fresh keeping of fresh meat products.
Description
Technical Field
The invention relates to the field of deep utilization of astragalus membranaceus, in particular to a preparation method of an astragalus membranaceus fiber membrane.
Background
Astragalus has been used as a medicine for invigorating qi, consolidating superficial resistance, inducing diuresis, relieving swelling, expelling pus and healing sore in China for over 2000 years. With the popularization of modern health knowledge, astragalus membranaceus goes into the daily life of the public in the form of dietary therapy medicated diet or tea infusion. According to statistics, the circulation of the astragalus medicinal materials in the market is close to 3000 tons every year. During the process of processing fresh astragalus into medicinal materials and decoction pieces, only the thickest root of the fresh astragalus is generally reserved, and the utilization degree of the rest parts of the freshly harvested astragalus, such as overground parts (branches and leaves) and root sides of the astragalus, is not high.
Modern studies have shown that the leaves, branches and collateral fibrous roots of Astragalus also contain small amounts of active substances, such as Astragalus polysaccharides, astragalosides, flavones, vitamins, etc., and these fractions also contain a large amount of fibers. For a long time, the utilization of the residual part of the freshly harvested astragalus is mainly concentrated in the fields of livestock and poultry feeding, agricultural fertilizer, mushroom cultivation and the like, and in addition, the deep development is not realized. The invention prepares a high-grade biomembrane with antibacterial, fresh-keeping and health-care functions by utilizing the cellulose and other small amount of active ingredients in the overground part and the root side hair of the fresh astragalus, combining chitosan and plant essential oil, and can be used for packaging fresh foods such as fresh fruits, vegetables, meat products and the like, preventing the foods from being rotten and deteriorated and prolonging the shelf life of the foods. At present, the preparation reports of using corn straws, ginger straws, bamboo straws and the like as membrane materials are reported, but the reports of the membrane materials of astragalus membranaceus fiber fresh-keeping food are not found.
Chitosan is a polysaccharide substance extracted from natural chitin, is a known biopolymer, has antibacterial activity on gram-positive bacteria and gram-negative bacteria, and has unique polycation, chelating and film-forming properties due to the existence of active amino and hydroxyl in the structure. When the chitosan film is prepared, if only single chitosan is selected, the obtained film has low strength, poor flexibility and poor waterproofness, so that other components are often required to be added in the preparation process to prepare the composite film, for example, the method of adding fibers into the chitosan is a good method. As the molecular structure of chitosan is close to that of the fiber, the two substances are mixed together and are easy to disperse, and the mechanical property of the membrane can be greatly improved by adding cellulose. In addition, the water solubility of chitosan is poor, active amino on molecules is required to be utilized to combine protons in an acid solution, so that the chitosan becomes a cationic polyelectrolyte, the water solubility is improved, the chitosan water solution has positive charges, the surface of the astragalus fibers has negative charges, and the astragalus fibers and the chitosan have good compatibility by utilizing the charge adsorption effect. Therefore, the extracted astragalus fiber is added into the chitosan water solution to prepare the chitosan-astragalus fiber composite membrane.
The plant essential oil is a pure natural, safe and reliable plant source food preservative, has the functions of resisting bacteria, fungi and oxidation and the like, and is widely applied to the preservation of fruits and vegetables, seafood, chilled meat products, processed products, grain crops and the like. However, the liquid essential oil is unstable, volatile, poor in water solubility, strong in odor, and easily affected by external light, temperature, oxygen and other environments, so that the liquid essential oil is limited in practical application. According to the invention, the plant essential oil with the antibacterial effect is added into the chitosan solution, and reacts with the astragalus fiber to prepare the membrane material with good antibacterial, fresh-keeping and health-care functions, and the membrane material can be used as a packaging material for fruits, vegetables, medicinal materials, fresh meat products and the like.
Disclosure of Invention
The invention aims to provide a preparation method of a chitosan-astragalus fiber film doped with essential oil, so as to obtain a film packaging material with antibacterial, antioxidant, fresh-keeping and health-care functions, which is used for the postharvest fresh keeping of fresh vegetables, fruits, medicinal materials and the like and also can be used for the fresh keeping of fresh meat products.
In order to solve the technical problems, the invention adopts the following technical scheme:
1. the method for preparing the astragalus fiber by using the astragalus comprises the following preparation steps:
raw material soaking, raw material high-pressure cooking and raw material post-treatment:
soaking raw materials comprises cleaning plant tissue of radix astragali (aerial parts of radix astragali include branches and leaves, underground parts include main root or side hair of main root), subdividing radix astragali into fine particles of 10-1000 meshes, and soaking in water;
the raw material high-pressure cooking comprises placing the soaked astragalus fine particles into a pressure container for high-pressure cooking;
the post-treatment of the raw materials is divided into grinding, alkali soaking, decoloring, washing and drying;
wherein, the astragalus fine particles for processing high-pressure cooking are ground to enable the astragalus fine particles to be flocculent;
alkali soaking, namely soaking the flocculent astragalus root in a 10-35% sodium hydroxide solution for 7-21 days, wherein the mass ratio of the flocculent astragalus root to the sodium hydroxide solution is 1;
washing and drying to clean flocculent radix astragali soaked in alkali, and drying in a vacuum drying oven at 80-100 deg.C to obtain radix astragali fiber.
Preferably, the soaking time of the raw materials in the soaking process is controlled within 24-48 hours.
Preferably, the cooking temperature of the raw material in the high-pressure cooking is controlled to be 100-120 ℃, the pressure is controlled to be 1-3atm, and the cooking time is 1-2 hours.
Preferably, when the raw materials are soaked in alkali, the concentration of a sodium hydroxide solution is 10-35%, and the mass ratio of the flocculent astragalus root to the sodium hydroxide is 1.
(II) the method for preparing the essential oil-doped chitosan-astragalus fiber membrane by using the astragalus fibers comprises the following preparation steps:
1) Weighing 0.05-10 g of dried astragalus fibers, adding into 50-250 ml of water, and uniformly stirring;
2) Adding 10-50 ml of 0.05-10% chitosan solution prepared from one or more of acetic acid, tartaric acid, malic acid, oxalic acid, citric acid, benzoic acid, salicylic acid, vitamin C, hydrochloric acid, sulfuric acid and phosphoric acid, and adjusting pH to 2-7;
3) Adding one or more of sodium tripolyphosphate, polyvidone, polyethylene glycol-4000, urea, polyethylene glycol-6000, citric acid, and succinic acid, and adjusting pH to 1-10;
4) Adding one or more of cinnamon essential oil, thyme essential oil, oregano essential oil, mint essential oil, garlic essential oil, clove essential oil, tea tree oil, eucalyptus oil, wormwood essential oil, ginger essential oil, pepper essential oil, lemon essential oil and citronella essential oil which are dispersed by one or more of 0.2-5% of tween-80, tween-60 and tween-20 solution, and uniformly stirring, wherein the volume ratio of the essential oil to the tween is (1);
5) Putting the mixture into an ultrasonic device for ultrasonic treatment for 5-90 minutes, uniformly mixing, measuring 5-50 ml of mixed solution, putting the mixed solution into a polytetrafluoroethylene mold, standing, and drying at 20-100 ℃ to obtain the chitosan-astragalus membranaceus fiber membrane containing the essential oil.
Preferably, the mass ratio of one or more of sodium tripolyphosphate, polyvinylpyrrolidone, polyethylene glycol-4000, urea, polyethylene glycol-6000, citric acid and succinic acid to chitosan is 1.
Preferably, the mass ratio of one or more of cinnamon essential oil, thyme essential oil, oregano essential oil, mint essential oil, garlic essential oil, clove essential oil, tea tree oil, eucalyptus oil, wormwood essential oil, ginger essential oil, pepper essential oil, lemon essential oil and citronella essential oil to chitosan is 1.
The invention has the beneficial effects that:
1. the invention utilizes the advantage that the plant tissue of the astragalus is rich in cellulose, and the astragalus fiber, the plant essential oil and the chitosan are mixed to prepare the astragalus fiber-chitosan membrane containing the plant essential oil with excellent performance, wherein the membrane is made of biological materials, is nontoxic and harmless and is easy to degrade;
2. the main components of the film, namely chitosan and essential oil, have antibacterial effects, can effectively prevent packaged objects from being rotten and deteriorated, and the astragalus fibers also retain part of pharmacological components of astragalus, so that the film has antibacterial and health-care functions;
3. chitosan and cellulose have a moderate isolation effect on oxygen, can maintain the oxygen consumption required by the respiration of the packaged object, and simultaneously have high moisture and oxygen permeability due to the biological characteristics, so the chitosan and cellulose have a good fresh-keeping effect on the packaged object;
4. the invention utilizes the characteristics of rich fiber, softness, fineness and good strength of astragalus to obtain the astragalus fiber through the procedures of soaking, high-pressure cooking and grinding, alkali soaking, washing, drying and the like, and the astragalus fiber, chitosan, essential oil and other components are added into the astragalus fiber, chitosan and other components according to a certain mass ratio and are mixed, stood and dried to prepare the membrane material.

Example 1
In this example, a preparation method of a chitosan-astragalus membranaceus fiber membrane containing essential oil is provided, and the preparation method comprises the following preparation scheme:
soaking raw materials, steaming and boiling the raw materials under high pressure, grinding, soaking in alkali, washing, drying and preparing a film;
the preparation method of the essential oil-chitosan-astragalus membranaceus fiber membrane is detailed in the following way by combining the preparation scheme:
cleaning radix astragali, slicing into 0.5-20 mm pieces with a slicing machine, or pulverizing into 10-1000 mesh granules with a pulverizer, and soaking in water for 24-48 hr;
adding the soaked radix astragali into a pressure container, adding water to soak radix astragali, adjusting temperature to 100-120 deg.C and pressure to 1-3atm, and steaming for 1-2 hr;
grinding the raw materials after high-pressure cooking in a grinder to break the wall and form flocculent;
filtering the ground raw materials, adding 10-35% sodium hydroxide solution, and soaking at normal temperature for 7-21 days;
filtering the raw material soaked in the alkali, and washing with water until the solution is nearly neutral;
drying the washed raw materials in a vacuum drying oven at 80-100 deg.C for 1-2 hr to obtain radix astragali fiber, and packaging in a sealed bag for storage;
weighing 0.05-10 g of dried astragalus fibers, adding into 50-250 ml of water, and uniformly stirring;
adding 10-50 ml of chitosan solution with mass concentration of 0.05-10% prepared from one or more of acetic acid, tartaric acid, malic acid, oxalic acid, citric acid, benzoic acid, salicylic acid, vitamin C, hydrochloric acid, sulfuric acid and phosphoric acid, and adjusting pH to 2-7;
adding one or more of sodium tripolyphosphate, polyvinylpyrrolidone, polyethylene glycol-4000, urea, polyethylene glycol-6000, citric acid and succinic acid, wherein the mass ratio of the sodium tripolyphosphate, the polyvinylpyrrolidone, the polyethylene glycol-4000, the urea, the polyethylene glycol-6000, the citric acid and the chitosan is (1-10000), and adjusting the pH value to be 1-10;
adding one or more of cinnamon essential oil, thyme essential oil, oregano essential oil, mint essential oil, garlic essential oil, clove essential oil, tea tree oil, eucalyptus oil, wormwood essential oil, ginger essential oil, pepper essential oil, lemon essential oil and citronella essential oil which are dispersed by 0.2-5% of one or more of tween-80, tween-60 and tween-20 solution, and uniformly stirring, wherein the volume ratio of the essential oil to the tween is 1;
putting the mixture into an ultrasonic device for ultrasonic treatment for 5 to 90 minutes, and uniformly mixing;
measuring 5-50 ml of mixed solution, putting the mixed solution into a polytetrafluoroethylene mold, standing the mixed solution, and drying the mixed solution at 20-100 ℃ to obtain the essential oil-containing chitosan-astragalus membranaceus fiber membrane which has stronger tensile strength, higher elongation at break, higher water vapor transmission coefficient and better antibacterial effect.
The astragalus root in the embodiment comprises astragalus root, astragalus stem and leaf and dregs after extracting the active ingredients of the astragalus root.
The process for extracting astragalus membranaceus fibers in the embodiment comprises the working procedures of soaking, high-pressure cooking, grinding, alkali soaking, washing, drying and the like, and the sequence of the working procedures can be partially adjusted.
The invention relates to a preparation method of a chitosan-astragalus fiber membrane doped with essential oil, which comprises the steps of mixing a chitosan aqueous solution, an essential oil solution and an astragalus fiber solution, and drying the mixture to prepare the chitosan-astragalus fiber membrane doped with the essential oil.

Claims (6)
1. The method for preparing the astragalus fiber from the astragalus is characterized by comprising the following preparation steps:
soaking raw materials, steaming the raw materials at high pressure and carrying out post-treatment on the raw materials;
soaking the raw materials comprises cleaning radix astragali, finely dividing radix astragali into 10-1000 mesh fine particles, and soaking in water;
the raw material high-pressure cooking comprises placing the soaked astragalus fine particles into a pressure container for high-pressure cooking;
the post-treatment of the raw materials is divided into grinding, alkali soaking, washing and drying;
wherein, the astragalus fine particles for processing high-pressure cooking are ground to enable the astragalus fine particles to be flocculent;
soaking in alkali, soaking flocculent radix astragali in 10-35% sodium hydroxide solution;
washing and drying to clean the flocculent astragalus root soaked by the alkali, and placing the flocculent astragalus root in a vacuum drying oven at 80-100 ℃ for drying to obtain the astragalus fiber.
2. The method for preparing astragalus membranaceus fiber from astragalus membranaceus as claimed in claim 1, wherein the method comprises the following steps: the soaking time in the raw material soaking is controlled within 24-48 hours.
3. The method for preparing astragalus fiber from astragalus root as claimed in claim 1, wherein: the cooking temperature of the raw material in the high-pressure cooking is controlled to be 100-120 ℃, the pressure is 1-3atm, and the cooking time is 1-2 hours.
4. The method for preparing the essential oil-containing chitosan-astragalus fiber membrane by using the astragalus fibers is characterized by comprising the following preparation steps:
1) Weighing 0.05-10 g of dried astragalus fiber, adding into 50-250 ml of water, and uniformly stirring;
2) Adding one or more of acetic acid, tartaric acid, malic acid, oxalic acid, citric acid, benzoic acid, salicylic acid, vitamin C, hydrochloric acid, sulfuric acid, and phosphoric acid, adjusting pH to 2-7, and preparing 10-50 ml of chitosan with mass concentration of 0.05-10%;
3) Adding one or more of sodium tripolyphosphate, polyvidone, polyethylene glycol-4000, urea, polyethylene glycol-6000, citric acid, and succinic acid, and adjusting pH to 1-10;
4) Adding one or more of cinnamon essential oil, thyme essential oil, oregano essential oil, mint essential oil, garlic essential oil, clove essential oil, tea tree oil, eucalyptus oil, wormwood essential oil, ginger essential oil, pepper essential oil, lemon essential oil and citronella essential oil which are dispersed by 0.2-5% of one or more of tween-80, tween-60 and tween-20 solution, and uniformly stirring, wherein the volume ratio of the essential oil to the tween is 1;
5) Putting the mixture into an ultrasonic device for ultrasonic treatment for 5-90 minutes, uniformly mixing, measuring 5-50 ml of mixed solution, putting the mixed solution into a polytetrafluoroethylene mold, standing, and drying at 20-10 ℃ to obtain the chitosan-astragalus membranaceus fiber membrane containing the essential oil.
5. The method of claim 4, wherein the essential oil-doped chitosan-astragalus membranaceus fiber membrane comprises: one or more of sodium tripolyphosphate, polyvinylpyrrolidone, polyethylene glycol-4000, urea, polyethylene glycol-6000, citric acid and succinic acid, wherein the mass ratio of the sodium tripolyphosphate, the polyvinylpyrrolidone, the citric acid and the chitosan is 1.
6. The method of claim 4, wherein the essential oil-doped chitosan-astragalus membranaceus fiber membrane comprises: the mass ratio of one or more of cinnamon essential oil, thyme essential oil, oregano essential oil, mint essential oil, garlic essential oil, clove essential oil, tea tree oil, eucalyptus oil, wormwood essential oil, ginger essential oil, pepper essential oil, lemon essential oil and citronella essential oil to chitosan is 1.
