技术领域
本发明属于材料领域,具体涉及光热或光声材料技术领域。
背景技术
目前,对于癌症的治疗,在传统的手术治疗、放射性治疗和化疗之外,又开 发出了免疫疗法、细胞疗法等新型疗法。但是,目前的疗法虽然能够在一定程度 上延长患者的生命,仍有其局限性,如副作用大、无法完全清除肿瘤细胞、无法 治愈等等。因此,开发新的疗法仍然是癌症领域的热点。
近年来,科学家发现半导体纳米晶体在能量转换和储存(例如光伏、热电和 电池)和纳米诊断学(例如光声(PA)成像和光热疗法(PTT))中存在着巨大 的潜力,半导体纳米晶体中空位的产生和利用引起了相当大的关注。例如,纳米 级铜缺乏铜硫属化物的Cu2-xE(E=S,硒,碲,0≤X≤1)和缺氧金属氧化物MO3-x (M=W,Mo,0≤X≤1)成功地用作PA成像和PTT的有效光热转换器,因为它 们在由空位引起的近红外(NIR)窗口中具有强烈的局部表面等离子体共振 (LSPR)。众所周知,LSPR是由于自由电荷载流子(即电子或空穴)的集体振 荡引起的。我们观察到的最常见的LSPR,其来自贵金属纳米结构,例如Au,Ag 和Cu,其具有1022-1023cm-3范围内的自由电子密度。通过增加粒径或改变其形 态,可以将金属纳米结构的LSPR调谐到NIR窗口中。与可归因于自由电子振荡 的金属中的LSPR相比,高度自掺杂的铜硫属化物(Cu2-xE,E=S,Se,Te)纳 米颗粒(NP)中的LSPR由自由空穴产生,由于缺铜,大量的空穴载体(~1021cm-3) 具有高流动性。此外,Cu2-xE中的LSPR效应与尺寸无关,并且这些纳米晶体可以 制造得尽可能小而不牺牲它们的LSPR效应。
硒化铜(Cu2-xSe)纳米颗粒由于小的粒径,在NIR窗口中具有强烈的LSPR 效应、较好的生物相容性等优点被各个领域广泛应用。
然而,关于Cu2-xSe的报道相对较少。一个原因在于难以制备高质量,水分 散性和生物相容性的Cu2-xSe纳米颗粒(NPs)。另一个可能是铜和硒的相对较 低的相互亲和力,与Cu2-xS相比,单一合成方法的尺寸、组成控制和可调性有限。
尽管硒化铜在合成上难度相对较高,但因其具有的优良性能,还是有大批科 学家投入到对硒化铜的研究中。
Cu2-xSe NPs能够在各领域的进行应用,其首先必须具有足够的稳定性,以 使其能满足对其进行各种化学操作和修饰。尽管研究者们对如何制备稳定的 Cu2-xSe NPs开展了大量的研究工作,但遗憾的是目前所能获得的Cu2-xSeNPs在 稳定性方面表现仍不尽如人意,尤其是应用于生物医学领域的水溶性Cu2-xSe NPs。
目前,高质量的Cu2-xSe NPs多通过高温油相方法合成,其特点是表面包裹 了油溶性烷基配体,因此该类型Cu2-xSe NPs不具有水溶性。而直接水相合成的 方法制备出的Cu2- xSe NPs虽然具有水溶性,目前也得到了一定的发展,但其稳 定性不高、半峰宽较宽、粒子尺寸分布不均匀等诸多始终得不到解决的缺点也限 制了其应用。因此,通过相转移法将高质量的油溶性Cu2-xSeNPs转变为水溶性, 是获得高质量水溶性Cu2-xSeNPs的一个重要途径。
传统的相转移法制备水溶性Cu2-xSe NPs通常存在着如下缺点:聚合物包裹 的水溶性Cu2-xSe NPs通常稳定性不高,且粒径较大;配体本身不够稳定,如巯 基试剂本身易被氧化,从而使包裹的Cu2-xSe NPs不稳定;试剂昂贵,配体的合 成步骤繁琐冗长,工业化生产很难实现等。
纳米材料的稳定性不仅仅取决于其无机核的自身是否稳定,很大程度上也依 赖于包裹在纳米晶表面的配体的结构与组成是否稳定,以及无机核与有机配体之 间的连接是否牢固。无机核表面有机配体的脱落、解离等情况直接影响了整个纳 米晶的稳定性。可以想象,如果纳米晶表面的配体包裹不再牢固,发生了分解, 甚至脱离了无机核,那么纳米晶将不具有分散在溶液中的能力,同时无机核将暴 露在溶液或者空气中,很容易受到环境中各种因素的侵烛损害。
发明内容
本发明第一目的在于,提供一种具有强配位能力的有机配体,从而获得小尺 寸、近单分散、形貌可控、生物相容性良好以及环境适应能力强的水溶性Cu2-xSe NPs@ODT-聚丙烯酸类聚合物复合材料。
本发明第二目的在于,提供所述的Cu2-xSeNPs@ODT-聚丙烯酸类聚合物复 合材料的制备方法。
本发明第三目的在于,提供所述的Cu2-xSeNPs@ODT-聚丙烯酸类聚合物复 合材料在作为光热材料或光声材料中的应用。

本发明提供了一种全新的Cu2-xSe NPs@ODT-聚丙烯酸类聚合物复合材料, 其创新地基于化学以及物理双作用在Cu2-xSe NPs的表面形成稳定的ODT-聚丙 烯酸类聚合物复合材料修饰包覆层。研究发现,该复合材料具有强胶束形成能力、 可有效阻止表面包覆层的脱落,提高纳米晶的稳定性。另外,包覆层与Cu2-xSe NPs 的表面原子进行强配位结合,能较好的克服颗粒粒径大、多分散性以及官能基团 少不易改性等缺点,制备出小尺寸、近单分散、形貌可控、生物相容性良好以及 环境适应能力强的纳米颗粒。
作为优选,所述的Cu2-xSeNPs具有立方晶形;优选地,其粒径为0.1~100nm; 优选地0.1nm~50微米;优选为0.1nm~10nm。
本发明研究发现,所述的ODT-聚丙烯酸类聚合物链段、侧基修饰结构及 Cu2-xSeNPs联合协同是改善复合材料稳定性、水中分散性以及应用性能的关键。
作为优选,所述的ODT侧基修饰在聚丙烯酸类聚合链的一端或者两端。
作为优选,所述的ODT-聚丙烯酸类聚合物具有式1结构式:

所述的式1-A包覆层材料(本发明也称为ODT-PMAA配体)在Cu2-xSeNPs 制备过程中主要起到两个作用:一是聚合物的羧基和Cu2-xSe NPs表面自发形成 的CuO具有很强的亲和力,在纳米颗粒生长过程中包裹在Cu2-xSeNPs表面,阻 止其进一步生长,控制形貌和大小;二是ODT-PMAA能通过十八烷基间的强疏 水作用,使ODT-PMAA自发在溶液中形成网状胶束结构,从而使ODT-PMAA 配体具有比较大的空间位阻效应,能形成网状结构将Cu2-xSe NPs分离,这样不 仅能控制Cu2-xSe NPs不发生团聚现象,而且有利于Cu2-xSe NPs形成近单分散的粒径分布。ODT-PMAA配体不仅能通过常见的静电作用和化学配位键稳定 Cu2-xSe NPs,而且还可以通过自身具有的空间位阻效应使Cu2-xSeNPs在溶剂中 稳定存在,并能适应一定范围的离子强度和p H值。
本发明还提供了所述的Cu2-xSeNPs@ODT-聚丙烯酸类聚合物复合材料的制 备方法,包括以下步骤:
步骤(1):将正十八硫醇和丙烯酸类单体进行聚合反应,制得ODT-聚丙烯酸 类聚合物;
步骤(2):ODT-聚丙烯酸类聚合物对Cu2-xSe NPs进行包覆,制得Cu2-xSe NPs@ODT-聚丙烯酸类聚合物复合材料。
本发明中,所述的丙烯酸类单体可以为常规的聚丙烯酸类单体,例如,其可 以为具有式2结构式的单体;
所述的式2中的各取代基的选取范围同式1。
本发明中,所述的聚合反应可以采用常规的丙烯酸单体交联聚合条件。
例如,所述的聚合反应在引发剂下进行,所述的引发剂为能够引发丙烯酸单 体聚合的引发剂,例如为AIBN;
优选地,步骤(1)中,聚合反应的溶剂为醇类溶剂,例如为乙醇;
优选地,步骤(1)中,聚合反应的温度为70~100℃。
本发明中,步骤(2)中,将溶解有ODT-聚丙烯酸类聚合物的溶液和形成 Cu2-xSeNPs的原料混合,反应制得所述的材料;或者,将溶解有ODT-聚丙烯酸 类聚合物的溶液直接Cu2-xSeNPs的溶液混合,制得所述的材料。
本发明中,所述的ODT-聚丙烯酸类聚合物的溶液的溶剂例如为水。
优选地,所述的形成Cu2-xSeNPs的原料包括Se源、铜源和还原剂;
优选地,所述的Se源为Se、SeO2中的至少一种;
优选地,所述的铜源为能解离出Cu+、Cu2+的水溶性盐;优选为硫酸铜、氯 化铜中的至少一种;

本发明还提供了所述的所述的Cu2-xSeNPs@ODT-聚丙烯酸类聚合物复合材 料的应用,其特征在于,将其用作光热治疗材料。
本发明还提供了所述的Cu2-xSeNPs@ODT-聚丙烯酸类聚合物复合材料在用 作光声材料中的应用。
有益效果
1、本发明提供了一种全新的Cu2-xSeNPs@ODT-聚丙烯酸类聚合物复合材 料,该材料得益于所述的包覆层材料和核材料以及二者的特殊的相互作用,能够 改善其在水中的分散性,改善其稳定性以及应用性能;
2、本发明所述的新材料有利于体内的降解和排出,具有更好的临床安全性 和更短的体内代谢时间;
3、同时具有CT和光声显像能力,可同时进行多模式成像;
4、多羧基配体,易于接枝修饰,连接各种靶向抗体;
5、水溶性和分散性好,能够在强电解质溶液中稳定,有利于储存和运输。

实施例1:配体修饰的纳米Cu2-xSe(Cu2-xSe@ODT-PMAA复合材料)的合 成
步骤(1):ODT-PMAA的合成
将AIBN(0.08g,0.5mmol)加入30mL无水乙醇中,超声溶解后加入100 mL三口烧瓶中,打开磁力搅拌器,通氮30min,然后加入MAA(50mmol)、 ODT(5mmol)。
然后开始加热,做好回流装备的摆设,打开回流水,当温度到达75℃时, 开始计时,反应5h。反应结束后,在氮气接通条件下将样品冷却到室温,加入 准备好的冰乙醚,会有白色沉淀产生,用布氏漏斗将溶液杂质的溶剂除去,重复 三次,得到白色物质,然后将物质送入真空干燥箱45℃抽真空干燥12h将多余 的溶剂以及单体去除,将得到的白色粉末。H-NMR(D6 DMSO)δ(ppm):0.9(b) CH3,1.24(CH2)91.7(b)CH2(backbone),2.4CH2(fromODT),12.3(b)CO2H
步骤(2):ODT-PMAA修饰的纳米Cu2-xSe(Cu2-xSe@ODT-PMAA复合材 料)的合成:
方法一:
1、取55ml去离子水,加入35.6mg ODT-PMAA,使之溶解;
2、加入1ml 0.2M SeO2,加入3ml 0.4MVc;
3、15min之后加入新制1ml 0.4M CuSO4·5H20与4ml 0.4MVc混合液
4、反应30h,即得,期间利用分光光度计测其吸光度。

合成的产物的结构式为:
方法二:
(1)准确称量取56.7mg硼氢化钠,加入到50ml去离子水中,充分溶解;
(2)准确称量39.5mg硒粉加入到50ml硼氢化钠溶液中,通入氮气,在室 温下磁力搅拌混合均匀,反应3小时,充分反应生成溶液A;
(3)准确称量170.5mg二水合氯化铜和89mgODT-PMAA,加入到盛有5ml 去离子水的小烧杯中,室温下超声分散使其充分溶解,形成溶液B;
(4)溶液A充分反应后,继续高速搅拌和通氮气,将溶液B迅速加入到溶 液A中,在室温下反应1小时,生成黑色溶液;
(5)待混合溶液充分反应后,将生成的黑色溶液用分子量为30KDa的超滤 离心管以5000r/min的转速用去离子水每次离心10min离心3次以去除未反应 完全的聚合物配体。
本发明中,方法一和方法二制备得到的材料性质相近。以下测试以及应用案 例中,除特别申明外,均以方法一方法制得的Cu2-xSe@ODT-PMAA复合材料为 例。
对比例1:
Cu2-xSe@MSA-PMAA的制备:
和实施例1的方法一相比,进行步骤(1)和(2),区别仅在于,步骤(1)中,采 用等摩尔量的MSA替换ODT;
制得的材料标记为Cu2-xSe@MSA-PMAA。
第三部分:材料物化性能测定:
(1)Cu2-xSe@ODT-PMAA紫外吸收实验
首先将浓度为310μg/ml Cu2-xSe@ODT-PMAA母液用水分别稀释成0μg/ml、 12.5μg/ml、25μg/ml、50μg/ml、75μg/ml和100μg/ml的Cu2-xSe@ODT-PMAA 溶液,然后利用紫外分光光度计测量上述不同浓度Cu2-xSe@ODT-PMAA溶液的 吸光度情况,并绘制曲线。
(2)Cu2-xSe@ODT-PMAA光热实验
首先从上述不同浓度的Cu2-xSe@ODT-PMAA溶液中取1ml分别加到不同的 离心管中,然后在黑暗环境下,用波长为808nm的激光均匀的照射溶液,照射 时间为10min,同时通过光纤温度传感器分别记录各个浓度 Cu2-xSe@ODT-PMAA溶液的温度变化情况,并绘制温度-时间变化曲线。
(3)Cu2-xSe@ODT-PMAA的DLS和zeta电位
根据紫外吸收实验所得的曲线,我们选取浓度为75μg/ml的 Cu2-xSe@ODT-PMAA溶液作为初始溶液。然后根据这一浓度,将 Cu2-xSe@ODT-PMAA置于不同的溶液环境中,测量它们的DLS和zeta电位,并 比较不同溶液环境对DLS和zeta电位的影响。不同溶液环境如下:
(1)80mg/L的BSA溶液;
(2)含10%胎牛FBS的细胞培养基
(3)不同pH值的PBS缓冲液:分别为ph=4.0,pH=5.5,pH=6.5,pH=7.4;
(4)不同浓度的Nacl溶液:分别为1mol/L,0.01mol/L。
第四部分:物化结果和分析
(1)X射线衍射分析(XRD)测试
XRD用以表征晶体样品的晶体结构,获得材料的晶粒尺寸、面间距等参数, 是一种重要的物相分析手段。取适量Cu2-xSe@ODT-PMAA纳米颗粒干燥样品, 研磨成均匀细小的粉末,取适量研磨过的粉末载入X射线衍射仪的样品槽,尽 量使样品表面平整。测试使用Cu(Kɑ,λ=1.542A)靶,通过Jade软件分析数据, 得到的XRD测试结果如图1所示。27.1°,45.2°和53.7°处有三个的衍射峰,通 过对比Cu2-xSe@ODT-PMAA的标准XRD峰位(JCPDS cardno:06-0680),发 现它们分别对应立方晶形Cu2-xSe@ODT-PMAA的(1,1,1),(2,2,0)和(3,1, 1)晶面,该实验结果也进一步证明我们合成的Cu2-xSe@ODT-PMAA为立方晶 形。
(2)Cu2-xSe@ODT-PMAA纳米颗粒的红外分析
傅里叶红外光谱(FTIR)通常用以提供纳米材料及其修饰层物的化学结构 信息。图2是ODT-PMAA红外光谱图,可以看出3586cm-1为羧基中-OH的伸缩 振动的吸收峰,2606cm-1为羧基中-OH伸缩振动和变形振动的倍频及组合频吸收 峰,1711cm-1为羧基中C=O的伸缩振动吸收峰,1488cm-1为-CH2的变形振动吸 收峰,1450cm-1、1384cm-1为-CH3的对称变形振动的特征吸收峰,1269cm-1、 1174cm-1为C-O的伸缩振动吸收峰,966cm-1、926cm-1为(-CH2)的面内弯曲振 动吸收峰。668cm-1为C-S键的伸缩振动吸收峰。
通过对比Cu2-xSe@ODT-PMAA纳米颗粒和ODT-PMAA的红外光谱,证实 了ODT-PMAA成功包覆在Cu2-xSe@ODT-PMAA纳米颗粒上。
(3)粒径检测
Cu2-xSe@ODT-PMAA的水合粒径检测图如图4所示。对所得材料的DLS检 测表明,我们合成的聚合物配体修饰的Cu2-xSe@ODT-PMAA的水合动力学平均 粒径为86nm,这符合纳米材料尺寸的要求。在透射电镜(TEM)下可以明显地 观察到Cu2-xSe@ODT-PMAA纳米颗粒均一,形貌规则,通过Image J软件分析 得到的粒径大小范围是:1.25±0.32nm。
ODT-PMAA配体在Cu2-xSeNPs制备过程中主要起到两个作用:一是聚合物 的羧基和Cu2-xSe NPs表面CuO具有很强的亲和力,在纳米颗粒生长过程中包裹 在Cu2-xSe NPs表面,阻止其进一步生长,控制形貌和大小;二是ODT-PMAA 能通过十八烷基间的强疏水作用,使ODT-PMAA自发在溶液中形成网状胶束结 构,从而使ODT-PMAA配体具有比较大的空间位阻效应,能形成网状结构将Cu2-xSe NPs分离,这样不仅能控制Cu2-xSe NPs不发生团聚现象,而且有利于 Cu2-xSe NPs形成近单分散的粒径分布。
(4)Cu2-xSe@ODT-PMAA纳米颗粒稳定性研究
实验对Cu2-xSe@ODT-PMAA纳米颗粒在不同p H值和不同盐浓度的水溶 液中的Zeta电位和DLS粒径分布进行了测试,用以表征材料在水溶液中的稳 定性。结果如图5所示。由图5可知,在p H为4~7.4范围内,样品Zeta电 位为较大的负值;高的表面电荷归功于从溶液中吸附的离子。这些数据说明 Cu2-xSe@ODT-PMAA在水溶液中具有稳定性。图5展示了中性条件下,样品Zeta 电位和DLS随NaCl浓度的变化,随着NaCl浓度由0.01mol/L增加至1.0mol/L,样品Zeta电位逐渐减小,在浓度为1.0mol/L时,Zeta电位-3.68mV。
这些结果说明实验制备的Cu2-xSe@ODT-PMAA在一定浓度的盐溶液中同样 具有稳定性。
加入BSA蛋白后,Cu2-xSe@ODT-PMAA纳米颗粒的Zeta电位和DLS没有 发生显著的变化,说明Cu2-xSe@ODT-PMAA与血浆蛋白的主要成分BSA具有 微弱的相互作用。
ODT-PMAA配体不仅能通过常见的静电作用和化学配位键稳定Cu2-xSe NPs,而且还可以通过自身具有的空间位阻效应使Cu2-xSeNPs在溶剂中稳定存 在,并能适应一定范围的离子强度和p H值。
(5)长期储存稳定性研究
经过长达140天的持续观察,如图6所示,实施例1方法一制备的 Cu2-xSe@ODT-PMAA保持悬浮稳定和再分散性。这得益于ODT-PMAA配体中 多羧基的强配位作用,和链端十八碳烷基的强疏水缔合作用,ODT-PMAA具有 自发的胶束形成能力。临界胶束浓度是指分子在溶剂中形成胶束时的最低浓度, 可用来表示两亲性分子的自组装能力。经测,ODT-PMAA的临界胶束浓度分别 为4.2×10-3mol/L,而非烷基链端修饰的PMAA,如MSA-PMAA的临界胶束浓 度分别为8.7×10-3mol/L。ODT的链端修饰使得ODT-PMAA分子间能够通过ODT 烷基的强疏水缔合作用在水溶液中形成网状的胶束,使得原本不稳定的Cu2-xSe NPs在溶剂中长期悬浮稳定。研究发现,实施例1制备的Cu2-xSe@ODT-PMAA 可以稳定存储140天以上。而对比例MSA-PMAA配体修饰的Cu2-xSe NPs在60 天内就发生了聚沉现象。
实施例2
ODT-PMAA配体修饰的纳米Cu2-xSe(Cu2-xSe@ODT-PMAA)的光热性能及 影响因素探究
(1)纳米Cu2-xSe@ODT-PMAA紫外吸收光谱图:
为了进一步验证我们合成的材料为纳米Cu2-xSe@ODT-PMAA以及证明其作 为光热治疗剂的可行性,我们用UV-1601紫外分光光度计以水为溶剂和参比溶 液测定了材料的紫外吸收光谱,如图7所示,从图中可以看到,经水溶性聚合物 配体修饰后浓度为200μg/ml的Cu2-xSe@ODT-PMAA纳米材料有一个位于650 nm处的吸收边,所谓半导体的吸收边指的是在材料在其本征吸收过程中实现光 子能量大于等于禁带宽度的吸收边界,也即吸收光子使电子由价带激发到导带形 成电子-空穴所需的最低能量。更重要的是,材料在近红外附近(从400到1100 nm)由于LSPR效应展现出了非常强的吸收,这说明我们得到的纳米 Cu2-xSe@ODT-PMAA具有潜在的光热性能,可作为光热转换材料应用于光热治 疗领域。
(2)不同pH条件下的紫外吸收研究:
为了探究pH对Cu2-xSe@ODT-PMAA的影响,我们测试了 Cu2-xSe@ODT-PMAA在不同pH环境下的紫外吸收,如图8所示,从图中可以 看出,同一浓度的溶液在不同pH环境下的紫外吸收峰的波动很小,我们可以得 出Cu2-xSe@ODT-PMAA材料对环境的pH值不敏感。
(3)不同铜离子浓度的紫外吸收研究:
为了了解Cu2-xSe@ODT-PMAA的紫外吸收与浓度的关系,我们测试了在不 同铜离子浓度下的Cu2-xSe@ODT-PMAA的紫外吸收,分别测试15μg/ml,30 μg/ml,60μg/ml,120μg/ml,240μg/ml的紫外吸收,如图9所示,通过对比,我们 可以直观的了解到,随着浓度的升高,Cu2-xSe@ODT-PMAA的紫外吸收是越来 越强的。
(4):808nm激光照射下不同浓度的Cu2-xSe@ODT-PMAA的光热性能
为了直观的了解Cu2-xSe@ODT-PMAA的光热性能,我们对所得的材料进行 了光热转换测试。具体的测试方法为:以500μL的纯水为空白对照组,用移液 枪分别准确量取200μL不同浓度的Cu2-xSe@ODT-PMAA溶液于2ml小离心管 内,经避光处理后,采用波长808nm,功率密度为1500mW/cm2的激光光源逐 一照射各待测组分,并每间隔1min记录温度变化值。测得的结果如图10:
从图10可以看到,不同溶液浓度,纳米Cu2-xSe@ODT-PMAA表现出了相 当好的光热转换能力,它的光热转换效果随着浓度的升高而有所提升。在808nm 激光照射下,100μg/ml测试组在10min内温度上升约27℃,且在第1min时就 升温达到了3℃,在后续9min时间内升温效率慢慢降低。考虑到人体体温值为 37℃,当作为光热治疗剂使用时,在肿瘤部位能在2min内实现约10℃的升温, 在这个温度下可以实现对肿瘤细胞的杀伤。作为对照组的去离子水,温度升高仅 有4到5℃,这说明在激光光源照射下,引起温度变化的是材料本身,与溶剂无 关。
从图10我们可以得出结论,经聚合物配体修饰后的Cu2-xSe@ODT-PMAA 的光热性能与浓度有着密切的联系,即当浓度升高时,材料的光热性能会得到提 升,反之则会出现降低。另外,正常人体组织和血液的pH值在7.0到7.4之间, 而肿瘤病变部位在局部缺血时,其质液会出现异常的酸化情况,这时肿瘤组织的 pH值要低于正常组织,其值约为6.0到6.5之间,而我们经过实验测得pH对 Cu2-xSe@ODT-PMAA的光热性能影响极小,由图10,我们可以通过改变纳米 Cu2-xSe材料的溶液的浓度来实现对材料光热性能的调控,使其在应用时符合光 热治疗的要求。
(5)光热循环性能研究
通过对浓度为120μg/ml的硒化铜溶液在980nm激光照射下进行重复光照 实验,得到了如图11的升温降温曲线,在3个循环光照下,Cu2-xSe@ODT-PMAA 的光热性能依旧十分强烈,第三个循环的升温幅度与第一个循环相比较,没有任 何的减弱,这说明Cu2-xSe@ODT-PMAA是一种光热性能十分稳定,并且可以重 复进行光热实验的材料。
实施例3Cu2-xSe@ODT-PMAA光声实验
不同浓度Cu2-xSe@ODT-PMAA纳米材料脉冲光吸收情况:
为了探究Cu2-xSe@ODT-PMAA浓度与光声信号之间对应关系,采用光声成 像系统用于PA成像。该系统配备了Nd:YAG脉冲激光器(8ns脉宽,10Hz脉 冲重复频率,1064nm波长电压400V时产生258.4m J表面脉冲,波长(532nm、 1064nm);线聚焦超声探头(15MHz中心频率,8mm直径)用于接收光声 信号,扫描时样品浸入水中;数据获取/重建控制器;步进电机用于线性扫描。 多核计算机处理系统配备了可进行容积重建的高效能图形处理单元(graphics process unit);通过一种滤波投影算法(filteredbackprojection)进行图像重建。 Matlab(Matrix Laboratory,Mathworks,USA)软件用于数据处理和图像重建。我 们将装有浓度梯度依次为1.19,2.37,4.75,9.49,18.54,27.80,37.07,74.15, 152.00,304.00μg/ml的Cu2-xSe@ODT-PMAA溶液的离心管放置探测区域,同时 进行扫描,将获取到的光声信号经Matlab成像后得到图12,可知,随着从左至 右Cu2-xSe@ODT-PMAA溶液的浓度越来越大,得到的信号不断增加。
Claims (10)
2.如权利要求1所述的Cu2-xSe NPs@ODT-聚丙烯酸类聚合物复合材料,其特征在于,所述的Cu2-xSe NPs为缺铜的硒化铜颗粒;优选地,其粒径为0.1~100nm;优选地0.1nm~50微米;优选为0.1nm~10nm。
3.如权利要求1所述的Cu2-xSe NPs@ODT-聚丙烯酸类聚合物复合材料,其特征在于,所述的ODT侧基修饰在聚丙烯酸类聚合链的一端或者两端。
6.如权利要求1所述的Cu2-xSe NPs@ODT-聚丙烯酸类聚合物复合材料的制备方法,其特征在于,包括以下步骤:
步骤(1):将正十八硫醇和丙烯酸类单体进行聚合反应,制得ODT-聚丙烯酸类聚合物;
步骤(2):ODT-聚丙烯酸类聚合物对Cu2-xSe NPs进行包覆,制得Cu2-xSe NPs@ODT-聚丙烯酸类聚合物复合材料。
8.如权利要求1所述的Cu2-xSeNPs@ODT-聚丙烯酸类聚合物复合材料的制备方法,其特征在于,步骤(2)中,将溶解有ODT-聚丙烯酸类聚合物的溶液和形成Cu2-xSeNPs的原料混合,反应制得所述的材料;或者,将溶解有ODT-聚丙烯酸类聚合物的溶液直接Cu2-xSeNPs的溶液混合,制得所述的材料;
优选地,所述的形成Cu2-xSeNPs的原料包括Se源、铜源和还原剂;
优选地,所述的Se源为Se、SeO2中的至少一种;
优选地,所述的铜源为能解离出Cu+、Cu2+的水溶性盐;优选为硫酸铜、氯化铜中的至少一种;
优选地,所述的还原剂为Vc、硼氢化钠中的至少一种。
9.一种权利要求1~5任一项所述的Cu2-xSeNPs@ODT-聚丙烯酸类聚合物复合材料或权利要求6~8任一项所述的制备方法制得的Cu2-xSe NPs@ODT-聚丙烯酸类聚合物复合材料的应用,其特征在于,将其用作光热治疗材料。
10.一种权利要求1~5任一项所述的Cu2-xSe NPs@ODT-聚丙烯酸类聚合物复合材料或权利要求6~8任一项所述的制备方法制得的Cu2-xSe NPs@ODT-聚丙烯酸类聚合物复合材料的应用,其特征在于,将其用作光声材料。
