本发明涉及哇巴因化合物作为在制备抗病毒药物中的应用,包括:哇巴因化合物在制备抗HTLV‑2病毒、HIV‑1病毒、MMTV病毒、SARS病毒、RSV病毒或SIV病毒的药物中的应用。本发明的有益效果是:哇巴因化合物能够显著降低HTLV‑2病毒、HIV‑1病毒、MMTV病毒、SARS病毒、RSV病毒或SIV病毒的mRNA在翻译Gag‑Pol融合蛋白时程序性核糖体移码的效率,使Pol蛋白的翻译效率大幅度降低,从而导致细胞内无法产生蛋白酶、整合酶和逆转录酶,致使病毒无法有效包装,最终导致病毒的产量急剧降低,具有良好的抗病毒效果。

技术领域
本发明涉及哇巴因(Ouabain)化合物的用途,尤其涉及在制药领域中的用途。
背景技术
蛋白质由基因编码,是遗传信息表达的主要终产物。蛋白质在机体内的合成过程,实际上就是遗传信息从DNA经mRNA传递到蛋白质的过程,此时mRNA分子中的遗传信息被具体的转变成蛋白质的氨基酸排列顺序,因此这一过程也被称为翻译。由于病毒本身基因组比较小,它只能借助于宿主的翻译机器来实现自身基因的表达从而完成自身的生命过程。
在蛋白翻译过程中的主要机器就是核糖体,核糖在在翻译的起始、延伸阶段都有各种特异性的机制来保证翻译过程中氨基酸的准确排列顺序,其中之一就是密码子的连续性,即核糖体在mRNA上的转位为一个密码子,也就是核糖体都是按照连续3个核苷酸的方式向mRNA的3’端移动,直至遇见终止密码子而完成翻译的过程。在翻译过程中,如果核糖体移动序列不是3的倍数,通常会出现下游编码序列的改变,翻译出一个错误的蛋白质,从而导致蛋白的彻底失活。
但是,目前在有些病毒中,当核糖体在翻译病毒mRNA过程中遇到特殊的序列时,会特异性的让核糖体发生移码现象,这种情况被称之为程序性核糖体移码(ProgrammedRibosomal Frameshifting)。如果核糖体在翻译过程中向后移动一个核苷酸,被称之为-1PRF。目前已知的具有-1PRF调控机制的病毒有HTLV-2,MMTV,HIV-1,SARS,SARS-Covid19,RSV,SIV等7种。
程序性核糖体移码并不是发生在这些病毒mRNA的任意位置,而是需要一段特殊的RNA序列,这段序列包含了两个典型的特征,滑动序列和茎环序列,这个序列特征在目前的7种病毒中都能够被找到。首先滑动序列(Slippery Sequences)通常由一段七个核苷酸XXXY YYZ组成,其中X和Y分别指代相同的序列。茎环结构在滑动序列的下游,可以形成RNA二级结构而有效促进程序性核糖体移码的发生。
尽管目前的抗逆转录病毒治疗取得了很大进展,但HIV-1导致的艾滋病仍是一个全球性的健康问题,这种情况使得开发抗HIV-1的新药和研究这种病毒新的治疗靶点成为当务之急。HIV-1的移码效率控制着Gag:Pol的比例,这一比率的偏离,特别是移码效率的降低会导致Pol蛋白含量的减少进而使细胞内无法产生病毒所需的酶蛋白。因此,改变-1PRF的效率可以成为抗HIV-1的新靶点。
哇巴因主要来源于夹竹桃科植物苦毒毛旋花,属于强心苷类药物,可抑制细胞膜上Na+/K+-ATP酶,使细胞内Ca2+浓度增加,心肌收缩力增强,故临床上常用于心力衰竭等疾病。哇巴因的结构式为:
哇巴因在临床上常用于心力衰竭等疾病,并获得了FDA的批准在美国上市,但是目前没有相关的授权专利和文献表明哇巴因可以通过抑制程序性核糖体移码而对某些特定病毒具有治疗作用。
发明内容
本发明的目的在于提供哇巴因化合物的新用途,即在制药领域中的新应用。
实际上,本发明涉及哇巴因化合物在制备治疗因HTLV-2(人嗜T淋巴细胞II型)病毒感染所引起的淋巴瘤的药物中的应用。
在该应用中,所述药物基于抑制程序性核糖体移码阻止HTLV-2病毒的产生。
还涉及哇巴因化合物在制备治疗因HIV-1(I型艾滋病)病毒感染而所导致的艾滋病的药中的应用。
在该应用中,所述药物基于抑制程序性核糖体移码阻止HIV-1病毒的产生。
还涉及哇巴因化合物在制备抗MMTV病毒、SARS病毒、RSV病毒或SIV病毒的药物中的应用。

申请人通过查阅文献发现HTLV-2,MMTV,HIV-1,SARS,SARS-Covid19,RSV,SIV具有调控核糖体程序性移码的机制,这些调控序列如下:

其中加粗的序列为滑动序列,加粗倾斜的序列为茎环序列,下划线标记的序列为不发生-1PRF时出现的终止密码子的位置,也就是细胞内只能产生单一蛋白。只有当-1PRF发生的时候,核糖体在滑动序列的位置向后移动一个核苷酸,从而使下游的编码框发生改变,TAA/TAG终止密码子就会出现移码,进而翻译出融合的蛋白。
基于这种原理,本发明将以上病毒的调控序列构建于海肾荧光素酶(Rennilaluciferase,Rluc)和萤火虫荧光素酶(Firefly luciferase,Fluc)之间,如图1所示。由此可知,Rluc是可以正常翻译并作为内参,而FLuc只有当核糖体在移码信号区产生-1位移码才能进行翻译。因此,本发明可以利用Fluc/Rluc的比值变化来判断-1PRF效率的改变。
本发明将上述序列构建于pLVX-neomycin质粒,与pspAX2与pMD2G包装成病毒后,侵染Hela和293细胞,通过G418筛选,构建稳定细胞株。本发明采用哇巴因作用于这种双荧光素酶细胞株,实验结果表明哇巴因能够显著降低模式细胞中Fluc/Rluc的比值,进而表明药物对-1PRF效率有抑制作用,并在细胞水平进一步论证了其对病毒的产生具有显著的抑制效果。

(1)、程序性核糖体移码在特定病毒基因的表达过程中十分关键,但是目前还没有针对于此过程的有效药物筛选手段。本发明首先构建了pLVX-Rennila Luciferase-(-1PRF)-Firefly Luciferase的模式细胞,在药物处理后,通过测定Flu/Rlu的比值变化,来判断程序性核糖体移码效率的改变。这是本发明在药物筛选手段上的创新,而目前也没有相关文献或者专利发现哇巴因可以通过影响程序性核糖体移码而抑制相关病毒的复制,这也是本发明在药物作用机理上的创新。
(2)、哇巴因化学结构式如图2所示。哇巴因主要来源于夹竹桃科植物苦毒毛旋花,属于强心苷类药物,可抑制细胞膜上Na+/K+-ATP酶,使细胞内Ca2+浓度增加,心肌收缩力增强,故临床上常用于心力衰竭等疾病,并获得了FDA的批准在美国上市,但是目前没有相关授权专利和文献表明哇巴因可以通过抑制程序性核糖体移码而对某些特定病毒具有治疗作用。
(3)、哇巴因显著降低稳转细胞内HTLV-2病毒中程序性核糖体移码的效率;通过加入不同浓度的哇巴因作用于稳转了pLVX-Rennila Luciferase-(-1PRF-HTLV-2)-FireflyLuciferase的细胞,多功能酶标仪检测Rennila Luciferase和Firefly Luciferase值。结果如图3显示:相比二甲基亚砜(DMSO)处理的对照组,哇巴因处理的细胞Fluc/Rluc的值显著降低。
(4)、本发明再将HIV-1,MMTV,SARS,RSV和SIV的-1PRF调控序列构建到RennilaLuciferase和Firefly Luciferase之间,用于检测哇巴因处理后,这些病毒中程序性核糖体移码效率的改变。如图4结果可以看出,哇巴因均能降低核糖体在这些病毒中程序性移码的效率,表明哇巴因对这些病毒具有广谱的抑制程序性核糖体移码的作用,只要病毒有-1PRF的机制,哇巴因都是潜在的治疗药物。
(5)、本发明将pLVX-Rennila Luciferase-(-1PRF-HIV)-Firefly Luciferase和pLVX-Rennila Luciferase-(-1PRF-HTLV-2)-Firefly Luciferase顺转入THP1细胞和正常人外周血单核巨噬细胞48小时后,再用哇巴因处理,通过多功能酶标仪检测RennilaLuciferase和Firefly Luciferase值后发现,如图5所示,哇巴因处理的细胞Fluc/Rluc的值同样显著降低,这个结果表明:在入THP1细胞和正常人外周血单核巨噬细胞中,哇巴因同样能够抑制HIV-1和HTLV-2病毒中程序性核糖体移码。
(6)、哇巴因目前被发现可以用于强心苷类药物,是一类Na+/K+ATP酶抑制药,目前被广泛应用于治疗包括心律失常和房颤在内的心脏病,该药物没有发现可以基于阻碍程序性核糖体移码而抑制病毒的产生。为了进一步验证其特异性,我们选用了药物库中可以治疗白血病的小分子化合物氟达拉滨(Fludarabine)、米托蒽醌(Mitoxantrone)处理细胞,观察其是否能够影响HIV病毒的-1PRF机制,进而影响HIV病毒的产生。从图6的结果我们发现,相比于哇巴因(Ouabain)处理后-1PRF效率显著降低,氟达拉滨(Fludarabine)和米托蒽醌(Mitoxantrone)处理后细胞的-1PRF并未显著降低,表明其无法作用于HIV-1和HTLV-2的-1PRF调控机制,进而无法影响病毒的包装。因此,从这个结果中我们可以看出,哇巴因抑制程序性核糖体移码的作用是特异的,并不是任何药物都可以实现。
(7)、在293T细胞转染了pNL43-dE突变质粒与pHTLV质粒24小时之后,本发明用不同浓度的哇巴因处理细胞24小时再收集细胞用Western blot检测细胞内Gag-Pol融合蛋白、Gag蛋白、p24蛋白、p7蛋白的表达情况。如图7所示,当药物处理浓度逐渐增加之后,Gag-Pol融合蛋白表达量显著下降,p24和p7的蛋白水平也显著降低,但Gag蛋白没有明显变化。这个结果表明哇巴因能够显著的影响Gag-Pol融合蛋白的表达。当Pol蛋白表达下降的时候,Gag蛋白无法被有效酶切,p24和p7的含量均下降,图7中Actin作为内参。
(8)、哇巴因抑制HIV-1病毒在细胞中的复制;通过转染NL4-3突变型质粒20h后再加入哇巴因作用24h,提取细胞上清液中的总RNA,real-time PCR检测p24,pol,gp120的含量。如图8结果显示:相比对照组,加入了哇巴因的细胞其p24,pol,gp120的含量显著降低,表明上清液的病毒的含量显著降低,即哇巴因能抑制HIV-1病毒的产生。
(9)、哇巴因抑制HTLV-2病毒在症细胞中的复制;通过转染HTLV质粒20h后再加入哇巴因作用24h,提取细胞上清液中的总RNA,real-time PCR检测Gag,Pol,Env的含量。如图9结果显示:相比对照组,加入了哇巴因的细胞其Gag,Pol,Env的含量显著降低,表明上清液的病毒的含量显著降低,即哇巴因能抑制HTLV-2病毒的产生。
(10)、通过转染NL4-3突变型质粒与HTLV质粒20h后再加入哇巴因作用24h,ELISA检测上清中病毒粒子量的变化。结果如图10显示:哇巴因处理的细胞上清中,病毒颗粒的含量均显著低于对照组,这个结果表明哇巴因能够有效抑制HIV-1和HTLV-2病毒粒子的生成。
(11)、将pNL4-3-Envolope突变质粒和HTLV质粒转染293细胞后,该质粒能够包装出外膜蛋白突变的病毒颗粒,不具有感染性,但是可以表征细胞内产生病毒颗粒的情况,并用相应的ELISA试剂盒对上清液中的病毒颗粒进行检测,可以用于测定药物的EC50。通过这种方法,本发明测得哇巴因抑制HIV-1和HTLV产生的EC50浓度分别为117.82nM和132.67nM(图11)。
(12)、本发明获得了免疫缺陷的小鼠NOD.Cg-Prkdcscid Il2rgtm1Wjl/SzJ(NSG),将人的造血干细胞分离后移植到小鼠中,继续培养两周后检测造血干细胞在小鼠体内的分化情况,最终获得了造血细胞人源化的小鼠。因此本发明就可以用这个模型在体内评价药物的作用效果。本发明将HIV-dE和HTLV突变型病毒纯化后通过尾静脉注射到小鼠的血液系统,48小时后,本发明将哇巴因也通过尾静脉注射小鼠。再过48小时后,本发明检测了小鼠血液中HIV-1和HTLV-2突变型病毒颗粒的含量。图12结果表明,注射了哇巴因的小鼠其血液中HIV的病毒颗粒也显著低于对照组,这个结果表明在动物实验中,哇巴因也能显著抑制HIV-1和HTLV的复制。
本发明的有益效果是:
(1)本发明采用的哇巴因,能够有效降低HIV-1病毒、HTLV-2病毒中程序性核糖体移码的发生并能够显著抑制HIV-1病毒、HTLV-2病毒在模式细胞、THP1细胞、外周血单核细胞复制,抑制HIV-1病毒颗粒的组装。
(2)本发明发现一种哇巴因新的适应症,即有抗HIV-1病毒、HTLV-2病毒的效果。因为药物已经上市,该发明可以显著降低药物研发前期中安全性评价的资金投入,降低药物研发风险。
(3)本发明除了HIV-1和HTLV-2之外,MMTV,SARS,SARS-Covid19,RSV,SIV等病毒也具有调控程序性核糖体移码的机制,本发明发现哇巴因这个化合物具有抑制这些病毒在调控核糖体程序性移码上的广谱性。因此,针对于MMTV,SARS,SARS-Covid19,RSV,SIV等病毒,哇巴因都是潜在的治疗药物。

具体实施方式
下面结合实施例对本发明做进一步描述。下述实施例的说明只是用于帮助理解本发明。应当指出,对于本技术领域的普通人员来说,在不脱离本发明原理的前提下,还可以对本发明进行若干修饰,这些改进和修饰也落入本发明权利要求的保护范围内。
实施例1:
如图1所示,本发明建立了一种可以指示程序性核糖体移码效率的pLVX-RennilaLuciferase-(-1PRF)-Firefly Luciferase稳定细胞株。采用哇巴因作用于这种双荧光素酶细胞株,如图3所示,实验证明哇巴因能够显著降低模式细胞中Fluc/Rluc的比值,并且Fluc/Rluc的比值随哇巴因浓度的增大而减小。
进而可以证明哇巴因化合物在抑制核糖体程序性移码的作用及抗病毒应用,哇巴因化合物用于降低病毒中程序性核糖体移码的效率,致使病毒无法在细胞内包装成一个完整的颗粒,从而导致病毒产量急剧降低。
实施例2:
如图4所示,本发明将HIV-1,MMTV,SARS,RSV和SIV的-1PRF调控序列构建到Rennila Luciferase和Firefly Luciferase之间,用于检测哇巴因处理后,这些病毒中程序性核糖体移码效率的改变。从结果可以看出,哇巴因均能降低核糖体在这些病毒中程序性移码的效率,表明只要病毒有-1PRF的机制,哇巴因对这些病毒具有广谱的抑制程序性核糖体移码的作用。
实施例3:
如图5所示,哇巴因在应用时,首先将构建的pLVX-Rennila-(-1PRF-HIV)-luciferase和pLVX-Rennila-(-1PRF-HTLV)-luciferase顺转入THP1和PBMC细胞,在通过加入哇巴因作用于细胞后,多功能酶标仪检测Renilla Luciferase和Firefly Luciferase表达量的变化。结果显示:当哇巴因处理后,Firefly Luciferase和Renilla Luciferase的比值显著降低。
实施例4:
请参阅图6,利用两种可以治疗白血病的小分子化合物,氟达拉滨、米托蒽醌和哇巴因处理稳转细胞pLVX-Rennila-(-1PRF)-luciferase 293并测定荧光值的变化。结果显示:除了哇巴因,氟达拉滨和米托蒽醌处理后细胞的-1PRF并未显著降低。
实施例5:
请参阅图7,胚肾293细胞在转染pNL4-3 envelope突变质粒和HTLV质粒24h,再利用不同浓度的哇巴因化合物处理24h后收集细胞并将细胞裂解后Western blot检测细胞内Gag-Pol、Gag、p24、p7的表达情况。结果显示哇巴因化合物在能够显著降低细胞内Gag-Pol融合蛋白、p24、p7的含量,但是对于Gag蛋白的含量没有明显的抑制作用。
实施例6:
请参阅图8,通过转染NL4-3突变型质粒20h后再加入哇巴因作用24h,提取细胞上清液中的总RNA,real-time PCR检测p24,pol,gp120的含量。结果显示:相比对照组,加入了哇巴因的细胞其p24,pol,gp120的含量显著降低,表明上清液的病毒的含量显著降低,即哇巴因能抑制HIV-1病毒的产生。
实施例7:
请参阅图9,通过转染HTLV质粒20h后再加入哇巴因作用24h,提取细胞上清液中的总RNA,real-time PCR检测Gag,Pol,Env的含量。结果显示:相比对照组,加入了哇巴因的细胞其Gag,Pol,Env的含量显著降低,表明上清液的病毒的含量显著降低,即哇巴因能抑制HTLV-2病毒的产生。
实施例8:
请参阅图10,通过转染NL4-3突变型质粒与HTLV质粒20h后再加入哇巴因(Ouabain)作用24h,ELISA检测上清中病毒粒子量的变化。结果显示:哇巴因(Ouabain)处理的细胞上清中,病毒颗粒的含量均显著低于对照组,这个结果表明哇巴因(Ouabain)能够有效抑制HIV-1和HTLV-2病毒粒子的生成。
实施例9:
请参阅图11,人胚肾293细胞在转染pNL4-3 envelope突变质粒和HTLV质粒24h后,再利用不同浓度的哇巴因(Ouabain)化合物处理24h后,ELISA检测培养基中病毒颗粒的变化,用分光光度计在OD450的时候检测吸光度,并计算哇巴因化合物的EC50。
实施例10:
请参阅图12,为免疫缺陷的小鼠NOD.Cg-Prkdcscid Il2rgtm1Wjl/SzJ(NSG),将人的造血干细胞分离后移植到小鼠中两周之后,建立血细胞人源化的小鼠动物模型。将HIV和HTLV突变型病毒纯化后通过尾静脉注射到小鼠的血液系统,48小时后,本发明再将哇巴因也通过尾静脉注射小鼠。再过48小时后,本发明通过ELISA检测了小鼠血液中HIV病毒颗粒的含量。从图12中可以看出,哇巴因也能够显著抑制HIV和HTLV在人源化小鼠血液中的复制。
