本发明提供了一种氮硫掺杂碳点的制备方法,具体为:将对氨基苯磺酸和有机溶液混合,水热反应后,得到氮硫掺杂碳点。本申请还提供了基于氮硫掺杂碳点检测Fe3+的方法。本申请中采用同时作为碳源、氮源、硫源的对氨基苯磺酸通过水热法制备碳点,碳点的表面含有丰富的磺酸基团,该基团中的硫原子容易失去最外层的电子,由此使得磺酸基团和Fe3+之间非常容易发生配位作用,然后就一起结合到了碳点表面,使得荧光猝灭,实现Fe3+快速、高效的荧光检测。

技术领域
本发明涉及检测分析技术领域,尤其涉及氮硫掺杂碳点的制备方法和检测Fe3+的方法。
背景技术
随着现代社会和日常生活生产的不断进步,人们对于大自然中存在的各种各样的物质也有了越发广泛的应用,与此同时不可避免地也造成了很多问题并且已经严重危害人类健康和人们赖以生存的自然环境,例如重金属离子的污染问题,它们无法降解,只能通过大气、食物、水体一直不断地传播,在环境中始终存在着,最终对人体和其他生物的生命健康带来了不可逆的伤害。因此,急需发展一种快捷和绿色的方法实现对重金属离子的有效检测。
现在有很多可以检测金属离子的方式,例如火焰原子吸收光谱法、原子发射光谱法以及电感耦合等离子体光谱法等,这些方法主要存在着时间长、方法复杂、仪器贵重等一系列问题;当然也发展了许多荧光探针检测金属离子的方法,如金属有机骨架探针以及金/银纳米簇等。
碳点在荧光纳米探针的大家族中,是一个具有众多优点的新成员,现在碳点在化学检测方面的应用已经多种多样,碳点在水溶液中和不同的重金属离子相遇时会发生不同类型的电子转移,这种反应机理会导致碳点的荧光猝灭、荧光增强或者碳点的荧光颜色发生肉眼可见的变化,很多科研人员们便是通过这些光学信号发生的变化而对重金属离子的种类进行辨别和判断,这种方法一旦发展成熟必定能在实际应用上发挥重大作用,将给环境安全和生物安全带来新的保障。
发明内容
本发明解决的技术问题在于提供一种氮硫掺杂碳点的制备方法及其应用,本申请制备的碳点可以实现高效快捷检测Fe3+。
有鉴于此,本申请提供了一种氮硫掺杂碳点的制备方法,包括以下步骤:
将对氨基苯磺酸和有机溶液混合,水热反应后,得到氮硫掺杂碳点。
优选的,所述有机溶液为乙醇和水的混合溶液。
优选的,所述反应的温度为200~400℃,时间为3~5h。
优选的,所述氮硫掺杂碳点的尺寸为0.8~2.0nm。
本申请还提供了一种检测Fe3+的方法,包括以下步骤:
将碳点溶液调整pH值后再采用水稀释,得到碳点水溶液;
将所述碳点水溶液与含有Fe3+的溶液混合再进行荧光检测;
所述碳点溶液中的碳点为权利要求1~4任一项所述的制备方法所制备的氮硫掺杂碳点。
优选的,所述碳点溶液的浓度为0.30~0.50g/L。
优选的,所述pH值为3~5,所述调整pH值的试剂为PBS缓冲液。
优选的,所述碳点溶液与水的体积比为1:(2~4)。
优选的,所述荧光检测的激发波长为350~440nm。

具体实施方式
为了进一步理解本发明,下面结合实施例对本发明优选实施方案进行描述,但是应当理解,这些描述只是为进一步说明本发明的特征和优点,而不是对本发明权利要求的限制。
本申请提供了一种氮硫掺杂碳点的制备方法,同时提供了利用氮硫掺杂碳点检测Fe3+的方法,基于Fe3+氮硫掺杂碳点能够猝灭碳点荧光,本申请提供了基于氮硫掺杂碳点检测Fe3+的方法,其具有共存干扰物的低影响、检测目标特异性强的特点。具体的,本发明实施例公开了一种氮硫掺杂碳点的制备方法,包括以下步骤:
将对氨基苯磺酸和有机溶液混合,反应后,得到氮硫掺杂碳点。
在上述制备过程中,同时作为碳源、氮源和硫源的对氨基苯磺酸通过水热法制备的了氮硫掺杂碳点。具体的,所述有机溶液选自乙醇和水的混合溶液,所述乙醇和水的体积比为1:(1~3);在具体实施例中,所述乙醇和水的体积比为1:1。所述反应的温度为200~400℃,时间为3~5h;更具体的,所述反应的温度为400℃,所述反应的时间为4h。
本申请制备的氮硫掺杂碳点的尺寸为0.8~2.0nm。
本申请还提供了一种基于氮硫掺杂碳点检测Fe3+的方法,包括以下步骤:
将碳点溶液调整pH值后再采用水稀释,得到碳点水溶液;
将所述碳点水溶液与含有Fe3+的溶液混合再进行荧光检测。
在检测Fe3+的过程中,本申请所述碳点初始溶液的浓度为0.30~0.50g/L,在具体实施例中,所述碳点初始溶液的浓度为0.45g/L。本申请优选采用PBS缓冲液调整所述碳点溶液的pH至3~5,为了使得检测效果更显著,具有较好的荧光强度,所述pH值选用3。所述碳点溶液与水的体积比为1:(2~4);为了使得碳点溶液具有较强的荧光强度,所述碳点溶液与水的体积比为1:2。
在检测含有Fe3+的溶液,本申请优选采用激发波长为350~440nm的光进行激发,在具体实施例中,所述激发波长选自410nm。
本申请提供的氮硫掺杂碳点荧光体系对Fe3+的响应效果很好,具有很好的选择性,且干扰性小。
为了进一步理解本发明,下面结合实施例对本发明提供的氮硫掺杂碳点的制备方法和检测Fe3+的方法进行详细说明,本发明的保护范围不受以下实施例的限制。
实施例1
试剂
实验试剂:对氨基苯磺酸(C6H7NO3S),乙醇(C2H6O),磷酸二氢钾(KH2PO4),磷酸氢二钠(Na2HPO4),六水合三氯化铁(FeCl3·6H2O),均为分析纯,去离子水。
碳点的合成
水热法制备氮硫掺杂碳点,流程如图1所示,具体步骤为:
称量0.2g对氨基苯磺酸,溶于30ml去离子水和30ml乙醇中,并机械搅拌至完全溶解呈透明溶液,将该溶液转移至反应釜中,将反应釜放置干燥箱中并设置温度为200℃,时间为4h,程序升温至200℃时开始计时,时间到后待温度降至室温后取出反应釜,将反应产物收集到烧杯中。
不同反应条件对碳点荧光的影响
研究反应时间对氮掺杂型碳点荧光的影响时,反应时间分别为3h、4h、5h,其余制备方法不变;研究pH对氮掺杂型碳点荧光的影响时,pH=3/4/5/6/7/8五种不同实验条件;研究浓度对氮掺杂型碳点荧光的影响时,直接将碳点原液和水以不同比例(1:1、1:2、1:3、1:4、2:1、3:1)稀释;最后通过观察碳点荧光强度的变化,并以410nm的激发波长对碳点进行激发,记录荧光光谱仪测定的荧光强度,并绘制成曲线图。
实施例2
碳点荧光检测Fe3+
取一定量固体碳点0.2g的实施例1制备得到的碳点,利用PBS缓冲溶液调整为pH=3、浓度为0.45g/L的碳点溶液中,然后进行稀释,碳点溶液与去离子水以1:2比例混合成所需的碳点水溶液,混合成的碳点溶液与不同体积的Fe3+标准溶液以1:1的比例各取1.5mL混合于4mL的离心管中,震荡均匀混合后进行荧光检测,激发波长为410nm。
透射电子显微镜(TEM)表征
首先如图2氮、硫共掺杂碳点的透射电镜照片(a)和尺寸分布图(b)所示,可以从中得知,采用对氨基苯磺酸通过水热法合成制备出的碳点具有良好的分散性,粒径尺寸也较为均匀。并对碳点粒径统计分析,如下图(b),碳点尺寸主要分布在0.8nm~2.0nm左右,说明碳点非常小。
红外吸收光谱(FTIR)表征
如图3所示,1040cm-1和1210cm-1处的峰都是磺酸基团(SO3-)的伸缩振动峰;在1500cm-1处的吸收带是表示这里为C-NH-C键的弯曲振动峰,说明该碳点中含有C-N基团也就是表明N原子已成功引入碳点;在1630cm-1处有一个吸收带是属于-C=O-的特征伸缩振动峰,说明碳点表面具有羧基;在3400cm-1~3500cm-1处有一个小的吸收带,这一小段是与O-H、N-H的伸缩振动峰相关;在2950cm-1处有一个属于C-H的伸缩振动的吸收峰;通过红外光谱吸收图谱可知,该碳点的表面具有较多的磺酸基团,同时也具有羧基、氨基和羟基,这些基团赋予了该碳点良好的水溶性。
紫外可见光吸收光谱和荧光光谱分析
从图4(A)中可以清楚看见,碳点溶液的紫外吸收光谱在235nm处有一个非常明显的强吸收带,这是芳香族苯环的典型吸收峰,是由于sp2杂化芳香族π→π*体系的电子跃迁所导致的,主要归因于碳核本身;除此之外,还可以发现碳点在242nm处有个较强吸收峰,这可能是C=O中π→π*跃迁吸收峰。碳点的最大激发波长是410mm。另外,在图4(A)中的插图中,左边是表示碳点溶液在日光灯下淡黄色的,右边表示碳点溶液在410nm紫外光的照射下时能够发出明亮的绿色荧光。
如图4(B)所示,从图中得知,激发波长为410nm时,最大荧光峰值位于483nm处,最佳激发波长是410nm。当激发波长从350nm到440nm时,荧光峰位一直在483nm左右,这表明碳点荧光光谱与激发波长之间没有依赖性。但在350nm~370nm、370nm~410nm、410nm~440nm这三个激发波长区域时,其荧光强度的变化趋势是先增加再降低然后再增加。因此从总体上来看,当以不同的激发波长对碳点的荧光进行激发时,由于CQDs的粒径尺寸都分布较为均匀,由同一粒径的CQDs受到激发而发出荧光,荧光发射峰几乎不随激发波长而变化,发射峰始终位于455nm左右,没有表现出明显的激发波长依赖性。氮硫掺杂碳点的荧光量子产率为64%。
碳点荧光稳定性分析
为了研究存储时间对于氮掺杂型碳点荧光强度的影响,通过荧光光谱图对其荧光稳定性进行了分析,如图5所示,曲线1表示最初的氮硫掺杂碳点的荧光强度,曲线2表示同一碳点溶液的样品在存放三个月之后的荧光强度,两者均是在410nm的激发波长条件下进行激发的,可以看出曲线2的荧光强度并没有明显的下降;同时插图中是在410nmUV灯的照射下,左边是最初的碳点溶液的荧光亮度,右边是存放三个月后的碳点溶液的荧光亮度,可以发现亮度上只有很小一部分的降低。由此可以说明,采用对氨基苯磺酸通过水热法制备的氮硫掺杂碳点在存放三个月后仍然能够保持较高的荧光强度,具有优良的荧光稳定性。
实施例3
碳点“turn-off”荧光检测Fe3+
构建碳点荧光探针“turn-off”检测Fe3+之前,为了使碳点和Fe3+之间有较好的作用效果,分别研究了反应时间不同、碳点溶液pH不同和浓度不同时对碳点荧光强度的影响,对碳点和Fe3+之间的响应效果进行了优化。如图6所示,下图(A)是表示以不同反应时间来制备碳点时对碳点荧光强度的影响,可以明显看出当反应时间为4h时,其荧光强度最高;图(B)是表示当溶液中的pH不同时对碳点荧光强度的影响,可以得知当pH=3时,也就是溶液处于酸性条件下,其荧光强度最高。图(C)是表示碳点原液以水以不同比例稀释时碳点荧光强度的变化,可以看出当碳点原液与水以1:2的比例稀释时,荧光强度最高,而以1:3、1:4的比例混合时,碳点的荧光强度越来越低,这可能是由于浓度越来越低导致溶液中碳点的含量也越来越少,但是随着碳点溶液的浓度越高,其荧光强度反而越来越低,可能是由于浓度越高,碳点粒子在溶液中的分散性越差。
因此综上所述,后续实验会在反应时间为4h、pH=3、碳点原液与水1:2的浓度这样一个最佳条件下进行。
基于碳点“turn-off”荧光检测Fe3+
在溶液浓度是碳点原液与水的比例为1:2和pH=3的实验条件下,研究Fe3+浓度不同对碳点荧光强度的影响。将不同浓度的Fe3+溶液加入到碳点水溶液中,结果如图7所示,从图(A)中可以看出,Fe3+能够明显猝灭碳点的荧光,而且随着溶液中Fe3+浓度的增加,Fe3+对碳点荧光的猝灭程度也随之增加。
然后为了得到氮硫掺杂碳点对Fe3+的检测范围和检出限,经过Oringe将相关数据进行了线性拟合之后,我们得到了碳点的荧光强度和Fe3+浓度之间的线性方程,从图7(B)中可见在Fe3+浓度为400μM时获得了最小的F/F0值,F和F0分别表示碳点溶液中存在Fe3+和不存在Fe3+时碳点的荧光强度,而且能够从图中看出Fe3+对碳点荧光的猝灭效果非常好。碳点的荧光强度与Fe3+的浓度之间存在较好的线性关系,其数学表达式为F/F0=0.71-0.0008[Fe3 +](μM),线性范围为25~400μM,相关系数R2为0.993。检出限(LOD)为2.549μM,欧盟(EC)要求Fe3+在饮用水中浓度的最大允许值为3.57μM,所以该检测方法是有效可行的。
关于Fe3+猝灭氮硫掺杂碳点的荧光的原理解释,猜测可能是由于碳点的表面含有丰富的磺酸基团,该基团中的硫原子容易失去最外层的电子,主要是因为其电负性较低且原子半径较大;由此就使得磺酸基团和Fe3+之间非常容易发生配位作用,然后就一起结合到了碳点表面,使得荧光猝灭。如图8中所示,在没有向碳点溶液中引入Fe3+时,当受到紫外光照射时,基态电子吸收能量跃迁至激发态后又回到基态时,形成电子-空穴对,碳点产生荧光;再引入Fe3+后,与磺酸基团结合附着在碳点表面,导致碳点荧光被猝灭。
选择性研究
为了评估该检测方法是否对Fe3+有着很好的选择性检测效果,研究了碳点对于Fe3 +的选择性。在和上述实验条件相同的情况下,选择了十三种重金属离子作为干扰物质,研究测试了将上述13种干扰金属离子和Fe3+(浓度均为2mM)在相同条件下加入到碳点溶液中后的响应效果。
结果可以由图9得知,只有加入Fe3+时,对碳点溶液的荧光造成了明显的猝灭现象,其次是Fe2+也对碳点溶液的荧光有一定程度的猝灭,可能是由于Fe2+不稳定,在检测过程中有一部分的Fe2+氧化成了Fe3+;而其余的十二种常见重金属离子对碳点的荧光影响很小,几乎在视觉上无法看出有猝灭现象。由此可以说明,该碳点荧光体系对于Fe3+的响应效果很好,具有较好的选择性。
以上实施例的说明只是用于帮助理解本发明的方法及其核心思想。应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以对本发明进行若干改进和修饰,这些改进和修饰也落入本发明权利要求的保护范围内。
对所公开的实施例的上述说明,使本领域专业技术人员能够实现或使用本发明。对这些实施例的多种修改对本领域的专业技术人员来说将是显而易见的,本文中所定义的一般原理可以在不脱离本发明的精神或范围的情况下,在其它实施例中实现。因此,本发明将不会被限制于本文所示的这些实施例,而是要符合与本文所公开的原理和新颖特点相一致的最宽的范围。
Claims (9)
1.一种氮硫掺杂碳点的制备方法,包括以下步骤:
将对氨基苯磺酸和有机溶液混合,水热反应后,得到氮硫掺杂碳点。
2.根据权利要求1所述的制备方法,其特征在于,所述有机溶液为乙醇和水的混合溶液。
3.根据权利要求1所述的制备方法,其特征在于,所述反应的温度为200~400℃,时间为3~5h。
4.根据权利要求1所述的制备方法,其特征在于,所述氮硫掺杂碳点的尺寸为0.8~2.0nm。
5.一种检测Fe3+的方法,包括以下步骤:
将碳点溶液调整pH值后再采用水稀释,得到碳点水溶液;
将所述碳点水溶液与含有Fe3+的溶液混合再进行荧光检测;
所述碳点溶液中的碳点为权利要求1~4任一项所述的制备方法所制备的氮硫掺杂碳点。
6.根据权利要求5所述的方法,其特征在于,所述碳点溶液的浓度为0.30~0.50g/L。
7.根据权利要求5所述的方法,其特征在于,所述pH值为3~5,所述调整pH值的试剂为PBS缓冲液。
8.根据权利要求6所述的方法,其特征在于,所述碳点溶液与水的体积比为1:(2~4)。
9.根据权利要求5所述的方法,其特征在于,所述荧光检测的激发波长为350~440nm。
