Abstract
The invention provides a composite fermentation powder with the function of improving constipation, a composition and a preparation method, and relates to the technical field of health-care food, wherein the composite fermentation powder comprises a fructus cannabis, bunge cherry seed and prickly pear cactus composite fermentation product fermented by zymophyte; the composition with the constipation improving effect comprises 10-30 parts of polydextrose, 10-30 parts of inulin, 5-30 parts of fruit and vegetable enzyme powder, 1-30 parts of fruit juice powder, 1-10 parts of fructo-oligosaccharide, 1-10 parts of stachyose, 1-20 parts of composite fermentation powder, 0.5-8 parts of lactobacillus rhamnosus, 0.5-8 parts of lactobacillus plantarum, 0.1-0.8 part of edible essence and 0.005-0.05 part of sucralose. According to the invention, the dietary fiber, the prebiotics, the probiotics and the fruit and vegetable enzyme are combined with the composite fermentation prepared from the fructus cannabis, the bunge cherry seed and the opuntia ficus-indica, so that the constipation can be effectively relieved and treated, and the constipation recurrence can be prevented; the feed also has the effects of regulating the balance of intestinal flora, inhibiting the generation of harmful bacteria and toxins and promoting the propagation and growth of beneficial bacteria; solves the defects that the existing laxative can not radically cure constipation and has great harm to human body.
Description
Technical Field
The invention relates to the technical field of health-care food, and particularly relates to compound fermentation powder with a constipation improving effect, a composition and a preparation method.
Background
Constipation is a common intestinal disease, and the main symptoms are decreased defecation frequency, irregularity, dry and hard feces and the like. In the case of long-term constipation, toxic substances are generated in the body and cannot be discharged in time along with feces, so that the poison sac is reabsorbed into the blood, thereby causing body poisoning phenomena such as abdominal distension, anorexia, bad breath (halitosis), irritability and the like, causing anal fissure, hypertension, rectal ulcer and increasing the probability of suffering from colorectal cancer and other cancers (breast cancer and liver cancer) besides accelerating aging of the body, and also being an important induction factor of senile dementia.
Recently, the incidence of constipation is rapidly increasing, and patients with constipation are gradually becoming younger. The existing laxative drugs in the market are various in types, mainly excrete, and the drugs are discharged after being eaten, so that the effects of radical treatment and relapse prevention cannot be achieved, and the side effect is obvious after long-term administration, so that the laxative drugs can cause great damage to a human body.
Disclosure of Invention
The invention aims to provide composite fermentation powder with a constipation relieving effect, a composition and a preparation method thereof, and aims to overcome the defects that the existing laxative medicine cannot radically cure constipation and is harmful to human bodies.
In order to solve the problems, the invention firstly provides the compound fermentation powder with the effect of improving constipation, which comprises the hemp seed, bunge cherry seed and prickly pear cactus compound fermentation products fermented by zymophyte.
Furthermore, the proportion of the fructus cannabis, the bunge cherry seed and the prickly pear is (20-40): 1-5.
The invention also provides a preparation method of the composite fermentation powder with the function of improving constipation, which comprises the following steps: activating strains, inoculating bacillus subtilis natto into a nutrient broth solid culture medium for normal-temperature culture, transferring into a conical flask filled with a sterilization culture medium, and culturing to form a bacterial liquid for later use; and (3) performing compound fermentation, namely adding soaked soybeans into a beaker, adding the coarse powder of fructus cannabis, bunge cherry seeds and opuntia ficus-indica into the beaker, uniformly mixing, performing sterilization treatment, cooling, inoculating a bacterial solution, performing fermentation in a constant-temperature incubator, and drying and crushing a fermented product to obtain the compound fermentation powder.
Furthermore, the proportion of the coarse powder of fructus cannabis, bunge cherry seed and prickly pear to the soybean in the composite fermentation step is 1.
The invention also provides a composition with the function of improving constipation, which comprises the following components in parts by weight: 10-30 parts of polydextrose, 10-30 parts of inulin, 5-30 parts of fruit and vegetable enzyme powder, 1-30 parts of fruit juice powder, 1-10 parts of fructo-oligosaccharide, 1-10 parts of stachyose, 1-20 parts of composite fermentation powder, 0.5-8 parts of lactobacillus rhamnosus, 0.5-8 parts of lactobacillus plantarum, 0.1-0.8 parts of edible essence and 0.005-0.05 parts of sucralose; wherein, the composite fermentation powder is the composite fermentation powder in the technical scheme.

The invention also provides a preparation method of the composition with the function of improving constipation, which comprises the following steps: preparing fruit and vegetable enzyme powder: cleaning and crushing fruits and vegetables, pulping to form fruit and vegetable pulp, adding glucose powder to form a mixed solution, performing three-time fermentation, separating and filtering to obtain a fermentation solution, and performing low-temperature freeze drying on the fermentation solution to obtain fruit and vegetable enzyme powder; sieving and mixing: sieving fruit and vegetable ferment powder, inulin, fructo-oligosaccharide, stachyose, juice powder, composite fermentation powder, edible essence and sucralose with 40-60 mesh sieve, and mixing to obtain a first mixture; mixing probiotics: uniformly mixing lactobacillus rhamnosus and lactobacillus plantarum to obtain a second mixture; equal incremental mixing: mixing the second mixture with polydextrose and inulin in an equal incremental manner to obtain a third mixture; total mixing: mixing the first mixture and the third mixture in a mixing device according to the formula amount for 20-40min to obtain a composition finished product; wherein, the composite fermentation powder is the composite fermentation powder in the technical scheme.
Further, the preparation of the fruit and vegetable ferment powder comprises the following steps: first-stage fermentation, adding yeast into the mixed solution for anaerobic fermentation, controlling the fermentation temperature to be 25-35 ℃ and the fermentation time to be 150-180 days; fermenting for the second time, adding yeast, lactobacillus plantarum and Bifidobacterium longum, and fermenting at 25-35 deg.C for 150-180 days; fermenting for three periods, continuously adding yeast, and continuously fermenting at 25-35 deg.C for 150-180 days.
According to the composite fermentation powder and the composition with the function of improving constipation and the preparation method thereof, the dietary fiber, the prebiotics, the probiotics and the fruit and vegetable enzyme are combined with the composite fermentation prepared from the fructus cannabis, the bunge cherry seed and the prickly pear, so that the constipation can be effectively relieved and treated, and the constipation relapse can be prevented; the feed also has the effects of regulating the balance of intestinal flora, inhibiting the generation of harmful bacteria and toxins and promoting the propagation and growth of beneficial bacteria; regulating and recovering gastrointestinal function, and enhancing gastrointestinal peristalsis; promoting the discharge of feces and toxins in the intestinal tract; meanwhile, the health-care food has the effects of promoting digestion, promoting metabolism, improving body functions and the like. The raw materials are compatible with each other, and experiments prove that the Chinese medicinal composition has good effect of promoting defecation without toxic or side effect.

Detailed Description
In order to make the aforementioned objects, features and advantages of the present invention comprehensible, embodiments accompanied with figures are described in detail below. It should be understood that the specific embodiments described herein are merely illustrative of the invention and are not intended to limit the invention.
Example 1
1. The raw material components are as follows: 30 parts of polydextrose, 20 parts of inulin, 20 parts of fruit and vegetable enzyme powder, 20 parts of fruit juice powder, 10 parts of fructo-oligosaccharide, 10 parts of stachyose, 15 parts of composite fermentation powder, 5 parts of lactobacillus rhamnosus, 5 parts of lactobacillus plantarum, 0.2 part of edible essence and 0.01 part of sucralose.
2. Preparation method of compound fermented powder (as shown in figure 1)
(1) Activating strains: inoculating Bacillus subtilis natto in nutrient broth solid culture medium, culturing at 37 deg.C for 1d, transferring into 250mL conical flask containing 30mL sterilized nutrient broth liquid culture medium, and shake-culturing at 37 deg.C for 20h at 180r/min to obtain bacterial solution;
(2) Establishment of fermentation conditions: adopting a single-factor rotation method and an orthogonal test design method, taking fructus cannabis coarse powder, bunge cherry coarse powder, prickly pear coarse powder and soybean as culture medium components, inoculating the prepared bacterial liquid into the culture medium components, uniformly mixing, and culturing in a constant-temperature incubator; the solid fermentation conditions comprise that the mixture ratio of fructus cannabis coarse powder, bunge cherry seed coarse powder, prickly pear coarse powder and soybean is 1, the material-liquid ratio is 5;
(3) And (3) composite fermentation: weighing 25 parts of soaked soybeans, putting the soybeans into a 1000mL beaker, adding 25 parts of fructus cannabis coarse powder, bunge cherry seed coarse powder and pear fruit cactus coarse powder, uniformly mixing, sterilizing, naturally cooling, taking 5mL of bacterial liquid from a super clean bench, inoculating the bacterial liquid into the bacterial liquid, putting the bacterial liquid into a 37 ℃ incubator, culturing for 48 hours, and finishing fermentation.
The complex baking powder mentioned in the following examples was prepared by this preparation method.
3. Preparation method of composition with constipation relieving effect (as shown in figure 2)
(1) Preparing fruit and vegetable enzyme powder: first-stage fermentation, cleaning fruits and vegetables, pulverizing, pulping to obtain fruit and vegetable pulp, mixing with glucose powder, adding yeast, performing anaerobic fermentation at 25-35 deg.C for 150-180 days; and (3) secondary fermentation: adding yeast, lactobacillus plantarum and Bifidobacterium longum, and fermenting at 25-35 deg.C for 150-180 days; and (3) three-stage fermentation: adding yeast, and fermenting at 25-35 deg.C for 150-180 days; separating and filtering to obtain fermentation liquor, and freeze-drying the fermentation liquor at low temperature to obtain fruit and vegetable enzyme powder;
(2) Sieving and mixing: respectively weighing fruit and vegetable enzyme powder, inulin, fructo-oligosaccharide, stachyose, juice powder, composite fermentation powder, edible essence and sucralose, sieving with a 40-60 mesh sieve, and mixing to obtain a first mixture;
(3) Mixing probiotics: mixing lactobacillus rhamnosus, lactobacillus plantarum and the like to obtain a second mixture;
(4) Equal amount of incremental mixing: mixing the second mixture with polydextrose and inulin in an equivalent incremental manner to obtain a third mixture;
(5) Total mixing: mixing the first mixture and the third mixture in a mixing device according to the formula amount for 20-40min to obtain the composition finished product.
The compositions having an effect of improving constipation mentioned in the following examples are prepared by this preparation method.
Example 2
The composition with the function of improving constipation comprises the following raw material components: 15 parts of polydextrose, 15 parts of inulin, 20 parts of fruit and vegetable enzyme powder, 15 parts of fruit juice powder, 8 parts of fructo-oligosaccharide, 8 parts of stachyose, 10 parts of fructus cannabis, bunge cherry seed and pear fruit cactus composite fermentation powder, 5 parts of lactobacillus rhamnosus, 5 parts of lactobacillus plantarum, 0.2 part of edible essence and 0.01 part of sucralose.
Example 3
The composition with the function of improving constipation comprises the following raw material components: 20 parts of polydextrose, 30 parts of inulin, 25 parts of fruit and vegetable enzyme powder, 20 parts of fruit juice powder, 10 parts of fructo-oligosaccharide, 10 parts of stachyose, 15 parts of fructus cannabis, bunge cherry seed and pear cactus composite fermentation powder, 8 parts of lactobacillus rhamnosus, 8 parts of lactobacillus plantarum, 0.2 part of edible essence and 0.02 part of sucralose.
Example 4 bowel relaxing function test
1. Materials and methods
1.1 test animals: SPF grade healthy kunming mice, male. Provided by Beijing Wittiulihua laboratory animal technology Co., ltd, the production license of laboratory animals: SCXK (Jing) 2007-0001. The breeding environment is of a barrier level, the test environment temperature is 23-24 ℃, and the humidity is 54-58%.
1.2 test methods
1.2.1 animal groups
Adult male Kunming mice with the weight of 18-22g are selected and divided into 6 groups, 10 mice are selected in each group, and the groups are respectively a low dose group, a medium dose group, a high dose group, a blank control group, a model group and a positive control group. The test substances of the low-dose group, the middle-dose group and the high-dose group are 0.4, 0.8 and 1.2g/kg of the composition prepared in the example 1, the test substance of the positive control group is 0.8g/kg of the Bishengyuan intestinal clear tea, the blank group and the model group are physiological saline with the same volume, 0.2mL of the test substance is given to each 10g of body weight, and the test sample is given for 14 days.
1.2.2 Small bowel movement test
After the test sample was given for 14d, each group of mice was fasted for 16h without water deprivation. The model group, the positive control group and the 3 dose groups were administered with compound diphenoxylate (5 mg/kg. BW) by gavage, and the blank group was administered with physiological saline. After the compound diphenoxylate is given for 0.5h, the ink containing the tested sample is respectively given to the low, medium and high dosage groups, and the ink is given to the positive control group for intragastric administration. After 25min, the cervical vertebrae were immediately removed to sacrifice the animal, the abdominal cavity was opened to separate mesentery, the intestine tube from the pylorus, the lower end to the ileocaecal region at the upper end was cut and placed on a tray, the small intestine was gently pulled into a straight line, the length of the intestine tube was measured as "length of small intestine", and the length of the ink pushed from the pylorus to the front edge of the ink was measured as "length of ink pushed".
1.2.3 measurement of defecation time, stool particle count and stool quality
After 14d of the test sample, each group of mice was fasted for 16h without water deprivation. The blank group was given physiological saline, and the model group, the positive control group and the 3 dose group were given compound diphenoxylate (10 mg/kg. BW) by gavage. After 0.5h of compound diphenoxylate, the mice of the blank group, the model group and the positive control group are perfused with ink, the low, medium and high dose groups are fed with the ink containing the tested sample, and the animals are all raised in a single cage and normally drink water to eat. Starting from the ink filling, the time of discharging black feces of the first grain of each animal and the number and the quality of the discharged black feces within 5 or 6 hours are recorded.
1.3 data statistics: and performing data statistical analysis on the detection result by adopting SPSS software.
The test results are shown below, where table 1 shows the effect of the composition on the small intestine propulsion test, and table 2 shows the effect of the composition on the defecation time and the number of defecation granules in the mouse.

As can be seen from Table 1, there was no significant difference (p > 0.05) between the sample dose-administered groups and the positive control group. Compared with the model group, the animal ink propelling length and the ink propelling rate of each dosage group of the sample have the rising trend along with the increase of the dosage and have significant difference (p is less than 0.01), which indicates that the sample has a certain effect on the small intestine propelling function of the test animal.

As can be seen from Table 2, there was no significant difference (p > 0.05) between the sample dose-administered groups and the positive control group. Compared with the model group, the number of the grains of the excrement of each dosage group of the sample tends to increase, the quality of the excrement tends to rise, and the obvious difference (p is less than 0.01) shows that the sample has a certain effect on the function of relaxing the bowels of the animals.
It can be seen that the small intestine propulsion function is significantly enhanced and the stool grain count and stool quality are significantly improved in the low, medium and high dose groups compared to the blank group, indicating that the composition prepared in example 1 has the function of facilitating defecation.
EXAMPLE 5 comparative test of the composition in cathartic efficacy

As can be seen from table 3, the ink propulsion rate, the first defecation time, the number of defecation granules and the defecation quality of the administration group, the control group 1 and the control group 2 were significantly improved (p < 0.01) compared with the model group; indicating that the administration group, the control group 1 and the control group 2 all had a laxative effect. Compared with the control group 1 and the control group 2, the administration group has significant differences in ink propulsion rate, first defecation time, defecation number and defecation quality (p is less than 0.01); indicating that the cathartic effect of the administration group is better than that of the control group 1 and the control group 2. Example 6 human feeding trial test
1.1 test methods: 100 constipation patients are selected, and constipation occurs in all cases; treatment groups and control groups were randomized. Treatment groups were 50, 25 men and 25 women; the age is 20-40 years. Control group 50, 25 men and 25 women; the age is 20-40 years old. The difference between the age, sex and disease course of the two groups has no statistical significance (p > 0.05). The composite fermentation powder is compared with 10% of fructus cannabis powder, 10% of bunge cherry seed powder and 0.5% of opuntia ficus-indica powder, the control group and the administration group are given with the same dosage, the blank group is replaced by physiological saline with the same volume, the efficacy verification method is according to the embodiment 4, and the test results are as follows:

As can be seen from Table 5, compared with 10% of fructus cannabis powder, 10% of semen pruni, and 0.5% of opuntia ficus-indica powder, the composite fermentation powder has significant differences (p < 0.01) in the propulsion rate of the fermentation liquid ink, the first defecation time, the number of defecation granules, and the defecation quality, and shows that the defecation effect of the composite fermentation powder is superior to that of 10% of fructus cannabis powder, 10% of semen pruni, and 0.5% of opuntia ficus-indica. In conclusion, the composite fermentation powder has the function of relaxing bowels and has obvious effect.
Example 8 Effect of Complex fermentations on laxation with functional food addition
Adding composite fermentation powder on the basis of inulin, fruit and vegetable enzyme powder, stachyose and lactobacillus rhamnosus to serve as a test group 1; inulin, fruit and vegetable enzyme powder, stachyose, lactobacillus rhamnosus, fructus cannabis powder, semen pruni powder and prickly pear powder are taken as a test group 2; the control group is a composition prepared from inulin, fruit and vegetable enzyme powder, stachyose and lactobacillus rhamnosus, the preparation formulation is powder, and the preparation formula is shown in table 2. The control group and the administration group were administered at the same dose, the efficacy verification method was as in example 4, and the test results are shown in table 6:

As can be seen from table 7, the liquid ink propelling rate, the first defecation time, the number of defecation granules and the defecation quality of the fermentation liquor in the test group 1 are significantly different from those in the control group and the test group 2 (p is less than 0.01); the composite fermentation powder test group 1 is proved to have better defecation effect than the test group 2 and the reference group. In conclusion, the composite fermentation powder is added into the functional food, so that the constipation relieving effect of the functional food can be enhanced, and the effect is obviously improved.

Claims (10)
1. A composite fermentation powder with the function of improving constipation is characterized by comprising a fructus cannabis, bunge cherry seed and prickly pear cactus composite fermentation product fermented by zymophyte.
2. The composite baking powder of claim 1 wherein the ratio of fructus cannabis, bunge cherry seed and opuntia ficus-indica is (20-40): (1-5).
3. A preparation method of composite fermentation powder with the function of improving constipation is characterized by comprising the following steps:
activating strains, inoculating bacillus subtilis natto into a nutrient broth solid culture medium for normal-temperature culture, transferring into a conical flask filled with a sterilization culture medium, and culturing to form a bacterial liquid for later use;
and (3) performing compound fermentation, namely adding soaked soybeans into a beaker, adding the coarse powder of fructus cannabis, bunge cherry seeds and opuntia ficus-indica into the beaker, uniformly mixing, performing sterilization treatment, cooling, inoculating a bacterial solution, performing fermentation in a constant-temperature incubator, and drying and crushing a fermented product to obtain the compound fermentation powder.
4. The method for preparing composite fermentation powder according to claim 3, wherein the ratio of the crude powder of fructus cannabis, bunge cherry seed and prickly pear to the soybean in the composite fermentation step is 1.
5. The composition with the effect of improving constipation is characterized by comprising the following components in parts by weight: 10-30 parts of polydextrose, 10-30 parts of inulin, 5-30 parts of fruit and vegetable enzyme powder, 1-30 parts of fruit juice powder, 1-10 parts of fructo-oligosaccharide, 1-10 parts of stachyose, 1-20 parts of composite fermentation powder, 0.5-8 parts of lactobacillus rhamnosus, 0.5-8 parts of lactobacillus plantarum, 0.1-0.8 part of edible essence and 0.005-0.05 part of sucralose;
wherein the composite fermentation powder is the composite fermentation powder of any one of claims 1 to 4.
6. The composition with the effect of improving constipation according to claim 5, which comprises the following components in parts by weight: 20-30 parts of polydextrose, 20-30 parts of inulin, 10-30 parts of fruit and vegetable enzyme powder, 10-30 parts of fruit juice powder, 5-10 parts of fructo-oligosaccharide, 5-10 parts of stachyose, 10-20 parts of composite fermentation powder, 2-8 parts of lactobacillus rhamnosus, 2-8 parts of lactobacillus plantarum, 0.1-0.5 part of edible essence and 0.005-0.03 part of sucralose.
7. The composition with constipation relieving effect of claim 5, wherein the fruit juice powder is one or more of strawberry powder, apple powder, lemon powder and grapefruit powder.

The composition with constipation relieving effect according to claim 5, wherein the fruit and vegetable ferment powder is composed of one or more of banana, sweet potato, cabbage, strawberry, tomato, lemon, kumquat, eggplant, broccoli, mustard, water spinach, mung bean sprout, lettuce, carambola, guava, asparagus, wax gourd, corn, towel gourd, coconut, mustard, cucumber, mung bean, white radish, cherry, mushroom, sweet orange, strawberry, balsam pear, kiwi, cabbage, needle mushroom, soybean, black bean, sweet potato, pineapple, apple, pumpkin, mulberry, grape, watermelon, chinese cabbage, taro, melon, brown rice.
9. The preparation method of the composition with the effect of improving constipation is characterized by comprising the following steps:
preparing fruit and vegetable enzyme powder: cleaning and crushing fruits and vegetables, pulping to form fruit and vegetable pulp, adding glucose powder to form a mixed solution, performing three-time fermentation, separating and filtering to obtain a fermentation solution, and performing low-temperature freeze drying on the fermentation solution to obtain fruit and vegetable enzyme powder;
