Abstract
本发明提供一种评价调节肠道屏障功能和肠道免疫能力的方法。所述方法包括以下步骤:(1)在Transwell一端小室内形成Caco‑2单层细胞;将THP‑1细胞分化为人巨噬细胞样细胞;(2)使含有Caco‑2单层细胞的一端小室完全浸没于DMEM液体培养基中,另一端小室内接种THP‑1,使其浸没于RMPI1640中,培养;(3)采用RMPI1640培养基替换后,在含有Caco‑2的一端添加待检测物孵育;在含有THP‑1的一端添加脂多糖和/或TNF‑α孵育;(4)检测Caco‑2侧培养物中脂多糖,评价免疫能力的调节作用;检测THP‑1侧培养物中TNF‑α、IL‑10或IL‑8,评价屏障功能的调节能力。

技术领域
本发明属于生物技术领域,具体涉及一种评价调节肠道屏障功能和肠道免疫能力的方法。
背景技术
慢性炎症性疾病是当今世界最重要的死亡原因,并且正在上升。我们现在知道,免疫驱动的炎症在这些疾病的病因学中是至关重要的,许多慢性炎症性疾病与肠道微生物群向炎症构型的重大转变有关,这种转变可通过诱发局部和全身炎症以及微生物群衍生的代谢物的改变而影响宿主。
免疫系统的功能是作为整个身体的“变阻器”,寻找生理干扰,并通过炎症性免疫反应,以及关键的修复/调节过程(如清除死细胞和恢复物理屏障)来纠正它们。因此,影响人类的许多慢性疾病可被视为免疫反应从修复/调节转向免疫驱动的炎症反应。其中一些疾病也与微生物群组成的重大变化有关。肠道微生物群是营养代谢物和炎症天然免疫信号的重要来源。因此,在遗传易感性和环境因素的背景下,肠道微生物群的组成变化作为疾病的调节剂和触发器具有潜在的重要性。
肠道微生物组影响包括宿主免疫在内的多器官系统。免疫系统负责维持肠道内的组织稳态,因为它必须对侵入性微生物保持警惕,同时限制对构成微生物群的大量良性有机体的明显炎症反应。为了维持这种与微生物群的危险关系,宿主发展了一系列免疫机制,限制和调节肠道上皮细胞与居住在肠腔内的微生物之间的相互作用。这一系列的“制衡”对宿主的健康至关重要,因为肠道屏障功能和免疫内环境平衡的破坏除了导致许多其他慢性疾病之外,还可能导致炎症性肠病。宿主还进化出多种产物,培育出健康多样的微生物群,包括IgA和胆汁酸的分泌以及肠上皮的岩藻糖化。这些宿主因素塑造微生物群的确切机制很复杂,因为它们既限制了某些细菌的生长,同时又有利于其他细菌,因此是维持微生物群健康一部分。
由于慢性炎症性疾病与肠道微生物群和肠道免疫相关,因此以肠道菌群为靶点的药物开发是一个热门的方向。但是目前并没有简单有效的体外模型来评价药物对肠道屏障功能和免疫能力的调节作用,这影响药物开发的效率。
CN107083364A公开了一种Caco-2和HUVEC细胞共培养体系的构建方法。将25~40代对数生长期Caco-2细胞接种到Transwell板上层小室培养至细胞完全分化;将2~6代对数生长期HUVEC细胞接种到Transwell板下层小室培养至细胞长满80~90%;将分化完全的Caco-2细胞Transwell上层小室插入到培养好的HUVEC细胞Transwell下层小室中,微调DMEM培养基,于37℃、5%CO2环境非接触共同培养。本发明构建的Caco-2/HUVEC共培养体系可同时研究药物的吸收代谢和生理活性,模拟药物经肠道吸收代谢后在循环系统的药理作用,提供一种反映药物生物利用度的细胞模型。
CN101914627A公开了一种评价嗜酸乳杆菌免疫调节作用的方法,涉及一种评价免疫调节作用的方法。方法:一、将待评价的嗜酸乳杆菌培养到对数生长后期,制得菌悬液;二、将极化状态的人结肠腺癌细胞Caco-2与菌悬液共同孵育;三、采用Trizol法抽提孵育后的人结肠腺癌细胞Caco-2的总RNA,再提取蛋白质;四、利用Real-Time-RT-PCR法和Western-blot法对免疫调节相关基因的表达情况分别在总RNA和蛋白质水平进行检测和比对即可。该发明通过对免疫调节相关基因的检测,可以更加客观的评价嗜酸乳杆菌对宿主可能产生的不良炎症反应等,但无法实现评价任意物质对于调节肠道屏障功能和肠道免疫能力的的目的。
CN111808910A公开了一种评价膳食多糖活性的方法。该方法包括膳食多糖加入微生物培养基作为碳源在体外厌氧条件下与粪便微生物在37℃培养24h后10000rmp离心后过0.22μm膜收集发酵液,利用代谢组学检测膳食多糖的代谢产物。利用Transwell技术上室中种植Caco-2细胞下室种植评价膳食多糖活性的细胞,并将发酵液加入上室细胞培养基中培养24h后,利用液相、气相或代谢组学等手段检测哪些代谢产物可以穿过Caco-2单层细胞模型,并测定下室细胞相关指标。利用本发明方法可以系统、科学地评价膳食多糖的生物活性。
因此,如何提供一种可以评价调节肠道屏障功能和肠道免疫能力的方法,提高药物开发效率,已成为亟待解决的问题。
发明内容
针对现有技术的不足,本发明的目的在于提供一种评价调节肠道屏障功能和肠道免疫能力的方法和应用,简单有效的体外模型来评价药物对肠道屏障功能和免疫能力的调节作用,优选提高药物开发效率,在药物及功能性食品开发方面具有巨大的应用前景。
为达到此发明目的,本发明采用以下技术方案:
第一方面,本发明提供一种评价调节肠道屏障功能和肠道免疫能力的方法,所述方法包括以下步骤:
(1)在Transwell孔板的一端小室内培养人结肠上皮细胞Caco-2,形成Caco-2单层细胞;将THP-1细胞进行培养,分化为人巨噬细胞样细胞THP-1;
(2)将步骤(1)的Transwell孔板倒置,使含有Caco-2单层细胞的一端小室完全浸没于DMEM液体培养基中,再在倒置后的Transwell孔板另一端小室内接种人巨噬细胞样细胞THP-1,并使人巨噬细胞样细胞THP-1浸没于RMPI1640培养基中,两端同时进行培养;
(3)采用RMPI1640培养基替换步骤(2)的DMEM液体培养基后,在含有Caco-2的一端添加待检测物后,进行孵育;并在含有THP-1的一端添加脂多糖和/或TNF-α后,进行孵育;
(4)检测Caco-2侧培养物中脂多糖水平,评价待检测物对肠道屏障功能的调节能力;并检测THP-1侧培养物中TNF-α、IL-10或IL-8水平,评价待检测物对肠道屏障功能的调节能力。
优选地,步骤(1)中,所述培养人结肠上皮细胞Caco-2的温度为35~40℃,例如可以是35℃、36℃、37℃、38℃、39℃、40℃等,时间为15~21天,例如可以是15天、16天、17天、18天、19天、20天、21天等。
优选地,步骤(1)中,所述形成Caco-2单层细胞后跨膜电阻为100ohm/cm2以上,例如可以是100ohm/cm2、102ohm/cm2、105ohm/cm2、110ohm/cm2、115ohm/cm2、120ohm/cm2、200ohm/cm2等。
优选地,步骤(1)中,分化为人巨噬细胞样细胞THP-1的具体步骤为:
采用160~165nM(如可以是160nM、161nM、162nM、163nM、164nM、165nM等)的佛波醇肉豆蔻酸乙酸酯溶液在35~40℃(例如可以是35℃、36℃、37℃、38℃、39℃、40℃等)下处理THP-1细胞70~74h(例如可以是70h、71h、72h、73h、74h等)后,再将其接种于RMPI1640培养基中在35~40℃(例如可以是35℃、36℃、37℃、38℃、39℃、40℃等)下培养22~26h(例如可以是22h、23h、24h、25h、26h等),使THP-1分化为人巨噬细胞样细胞THP-1。
优选地,步骤(2)中,所述培养的温度为35~40℃,例如可以是35℃、36℃、37℃、38℃、39℃、40℃等,时间为20~30h,例如可以是20h、22h、24h、26h、28h、30h等。
优选地,步骤(3)中,所述待检测物包括药物、益生菌、益生元、合生元或后生元中的任意一种或至少两种的组合。
优选地,步骤(3)中,所述待检测物在RMPI1640培养基中的含量为105~108CFU/mL,例如可以是105CFU/mL、106CFU/mL、107CFU/mL、108CFU/mL等。
优选地,步骤(3)中,在含有Caco-2的一端添加待检测物后,进行孵育的温度为35~38℃,例如可以是35℃、36℃、37℃、38℃等,时间为20~30h,例如可以是20h、22h、24h、26h、28h、30h等。
优选地,步骤(3)中,所述脂多糖在RMPI1640培养基中的含量10~1000ng/mL(例如可以是10ng/mL、20ng/mL、40ng/mL、60ng/mL、80ng/mL、200ng/mL、500ng/mL、1000ng/mL等),所述TNF-α在RMPI1640培养基中的含量10~1000ng/mL(例如可以是10ng/mL、20ng/mL、40ng/mL、60ng/mL、80ng/mL、200ng/mL、500ng/mL、1000ng/mL等)。
优选地,步骤(3)中,在含有THP-1的一端添加脂多糖和/或TNF-α后,进行孵育的温度为35~38℃,例如可以是35℃、36℃、37℃、38℃等,时间为4~8h,例如可以是4h、5h、6h、7h、8h等。
相对于现有技术,本发明具有以下有益效果:
本发明提供一种评价调节肠道屏障功能和肠道免疫能力的方法和应用,简单有效的体外模型来评价药物对肠道屏障功能和免疫能力的调节作用,提高药物开发效率,且准确度高稳定性好,步骤简单,在药物及功能性食品开发方面具有巨大的应用前景。

下面通过具体实施方式来进一步说明本发明的技术方案。本领域技术人员应该明了,所述实施例仅仅是帮助理解本发明,不应视为对本发明的具体限制。
实施例中未注明具体技术或条件者,按照本领域内的文献所描述的技术或条件,或者按照产品说明书进行。所用试剂或仪器未注明生产厂商者,均为可通过正规渠道商购获得的常规产品。
材料及方法:
Caco-2细胞和THP-1细胞购自中国科学院上海细胞库;
DEME培养基、RPMI 1640培养基购自武汉普诺赛生命科技有限公司;
胎牛血清、PBS和胰蛋白酶购自Thermo公司;
TNF-α、IL-8和LPS检测ELISA试剂盒购自南京建成生物工程研究所;
MRS固体培养基:称取蛋白胨10g、牛肉膏10g、葡萄糖20g、乙酸钠2g、酵母粉5g、柠檬酸氢二铵2g、K2HPO4·3H2O 2.6g、MgSO4·7H2O 0.1g、MnSO4 0.05g、琼脂20g和半胱氨酸氨酸盐0.5g,使用去离子水溶解,再加入1mL吐温80,定容至1L,灭菌冷却后,倒入灭菌后的培养皿中备用;
MRS液体培养基:称取蛋白胨10g、牛肉膏10g、葡萄糖20g、乙酸钠2g、酵母粉5g、柠檬酸氢二铵2g、K2HPO4·3H2O 2.6g、MgSO4·7H2O 0.1g、MnSO40.05g和半胱氨酸氨酸盐0.5g,使用去离子水溶解,再加入1mL吐温80,定容至1L,灭菌冷却后备用。
如图1所示,所述方法包括以下步骤:(1)在Transwell孔板的一端小室内培养人结肠上皮细胞Caco-2,形成Caco-2单层细胞;将THP-1细胞进行培养,分化为人巨噬细胞样细胞THP-1;(2)将步骤(1)的Transwell孔板倒置,使含有Caco-2单层细胞的一端小室完全浸没于DMEM液体培养基中,再在倒置后的Transwell孔板另一端小室内接种人巨噬细胞样细胞THP-1,并使人巨噬细胞样细胞THP-1浸没于RMPI1640培养基中,两端同时进行培养;(3)采用RMPI1640培养基替换步骤(2)的DMEM液体培养基后,在含有Caco-2的一端添加待检测物后,进行孵育;并在含有THP-1的一端添加脂多糖和/或TNF-α后,进行孵育;(4)检测Caco-2侧培养物中脂多糖水平,评价待检测物对肠道屏障功能的调节能力;并检测THP-1侧培养物中TNF-α、IL-10或IL-8水平,评价待检测物对肠道屏障功能的调节能力。
实施例1
本实施例提供了一种评价调节肠道屏障功能和肠道免疫能力模型的构建方法,具体步骤如下:
(1)人肠上皮细胞系Caco-2细胞置于补充1%非必需氨基酸、10%FBS、100U/mL青霉素和100g/mL链霉素的DEME培养基中,在37℃、5%CO2的加湿环境下培养。然后将用于共培养实验的Caco-2细胞接种于Transwell的上室培养15天,直至观察到100ohm/cm2以上的跨膜电阻(TER);将THP-1细胞在添加10%FBS、100U/mL青霉素和100μg/mL链霉素的RPMI1640中于37℃、5%CO2的加湿环境中培养。用162nM佛波醇肉豆蔻酸乙酸酯在37℃下处理THP-1细胞72h,并在完全RMPI1640培养基中在37℃下静置24h,使THP-1细胞分化为巨噬细胞样细胞;
(2)然后步骤(1)的Transwell倒置在装有DMEM培养基的容器中,在倒置后的Transwell上室中加入培养的THP-1细胞,并在RPMI1640培养基中在37℃下孵育24h;
(3)用RPMI1640替换所有培养基,并在37℃、5%CO2的加湿环境下进一步孵育24h。孵育期间,通过Millicell-ERS仪器定期测量上室的TER值,TER值无显著降低,表明整个模型建立期间Caco-2细胞层完整。
实施例2
本实施例验证了长双歧杆菌BL21【来自微康益生菌(苏州)股份有限公司】对肠道屏障功能和肠道免疫能力的调节作用,步骤如下:
S1.BL21菌株的制备:将BL21接种于MRS液体培养基中,37℃下培养18h进行活化,连续活化2次,得到活化液;将活化液按2%(v/v)的接种量接种于MRS液体培养基中,37℃下培养18h,得到菌液;将菌液在8000g下离心10min,用无菌磷酸盐缓冲盐水(PBS;pH 7.4)清洗三次,并重悬于PBS中,通过直接显微镜计数测定为108CFU/mL。
S2.按实施1所述构建评价调节肠道屏障功能和肠道免疫能力模型:,选取跨膜电阻满足要求的模型9个,分成三组(每组3个):空白对照组(CTL组),LPS刺激组(LPS组),LPS刺激+益生菌BL21干预组(BL21组)。每组的处理方式如下:
CTL组:不做任何处理,整个模型在37℃、5%CO2的加湿环境中孵育6h。
LPS组:用含100ng/mL LPS的完全RPMI1640培养基替换Transwell上室THP-1细胞的培养基,并在37℃、5%CO2的加湿环境中孵育6h。
BL21组:将1.0×108CFU长芽孢杆菌菌株加入下室的Caco-2细胞一侧,并在37℃、5%CO2的加湿环境下孵育24h。然后,用含100ng/mL LPS的完全RPMI1640培养基替换Transwell上室THP-1细胞的培养基,同时在下室并在37℃、5%CO2的加湿环境中孵育6h;
S3.孵育后,检测上室THP-1侧培养物中IL-8浓度,评价待测物对肠道免疫能力的调节作用;检测下室Caco-2侧培养物中脂多糖水平,从而评价待测物对肠道屏障功能的调节能力,测试结果如下表1所示:

试验结果如表1所示,结果表明与Caco-2细胞直接接触的BL21细胞共培养的THP-1细胞中,IL-8分泌显著受到抑制(p<0.01)。这一结果表明,BL21触发了Caco-2细胞产生一些对THP-1细胞免疫抑制的物质。另外,与LPS组相比,BL21组下室中LPS浓度显著降低(p<0.01),说明BL21可以显著改善肠道屏障功能。
实施例3
本实施例验证了副干酪乳杆菌LC86发酵乳【来自微康益生菌(苏州)股份有限公司】对肠道屏障功能和肠道免疫能力的调节作用,步骤如下:
S1.LC86菌株的制备:将LC86接种于MRS液体培养基中,37℃下培养18h进行活化,连续活化2次,得到活化液;将活化液按2%(v/v)的接种量接种于MRS液体培养基中,37℃下培养18h,得到菌液。
S2.将LC86接种到灭菌后的100mL新鲜牛奶中,37℃下培养48h,获得LC86发酵乳(一种后生元)。
S3.按实施1所述构建评价调节肠道屏障功能和肠道免疫能力模型,选取跨膜电阻满足要求的模型12个,分成四组(每组3个):空白对照组(CTL组),LPS刺激组(LPS组),牛奶对照组(牛奶组),LPS刺激+LC86发酵乳干预组(LC86发组)。每组的处理方式如下:
CTL组:不做任何处理,整个模型在在37℃、5%CO2的加湿环境中孵育6h。
LPS组:用含100ng/mL LPS和10ng/mL TNF-α的完全RPMI1640培养基替换Transwell上室THP-1细胞的培养基,并在37℃、5%CO2的加湿环境中孵育6h。
牛奶组:将1mL牛奶加入下室的Caco-2细胞一侧,并在37℃、5%CO2的加湿环境下孵育24h。然后,用含100ng/mL LPS和10ng/mL TNF-α的完全RPMI1640培养基替换Transwell上室THP-1细胞的培养基,同时在下室并在37℃、5%CO2的加湿环境中孵育6h。
LC86组:将1mL的LC86发酵乳加入下室的Caco-2细胞一侧,并在37℃、5%CO2的加湿环境下孵育24h。然后,用含100ng/mL LPS和10ng/mL TNF-α的完全RPMI1640培养基替换Transwell上室THP-1细胞的培养基,同时在下室并在37℃、5%CO2的加湿环境中孵育6h。
S4.孵育后,检测上室THP-1侧培养物中IL-8浓度,评价待测物对肠道免疫能力的调节作用;检测下室Caco-2侧培养物中脂多糖水平,从而评价待测物对肠道屏障功能的调节能力,测试结果如下表2所示:

试验结果如表2所示,结果表明与Caco-2细胞直接接触的LC86发酵乳共培养的THP-1细胞中,IL-8分泌显著受到抑制(p<0.01)。这一结果表明,LC86发酵乳触发了Caco-2细胞产生一些对THP-1细胞免疫抑制的物质。另外,与LPS组相比,LC86组下室中LPS浓度显著降低(p<0.01),说明BL21可以显著改善肠道屏障功能。
实施例4
本实施例验证了罗伊氏乳杆菌LR08发酵乳【来自微康益生菌(苏州)股份有限公司】对肠道屏障功能和肠道免疫能力的调节作用,步骤如下:
S1.LR08菌株的制备:将LR08接种于MRS液体培养基中,37℃下培养18h进行活化,连续活化2次,得到活化液;将活化液按2%(v/v)的接种量接种于MRS液体培养基中,37℃下培养18h,得到菌液。
S2.将LR08接种到灭菌后的100mL新鲜牛奶中,37℃下培养48h,获得LR08发酵乳(一种后生元)。
S3.按实施1所述构建评价调节肠道屏障功能和肠道免疫能力模型,选取跨膜电阻满足要求的模型12个,分成四组(每组3个):空白对照组(CTL组),TNF-α刺激组(TNF-α组),牛奶对照组(牛奶组),TNF-α刺激+LR08发酵乳干预组(LR08发组)。每组的处理方式如下:
CTL组:不做任何处理,整个模型在在37℃、5%CO2的加湿环境中孵育6h。
TNF-α组:用含100ng/mLTNF-α的完全RPMI1640培养基替换Transwell上室THP-1细胞的培养基,并在37℃、5%CO2的加湿环境中孵育6h。
LR08组:将1mL的LR08发酵乳加入下室的Caco-2细胞一侧,并在37℃、5%CO2的加湿环境下孵育24h。然后,用含100ng/mL TNF-α的完全RPMI1640培养基替换Transwell上室THP-1细胞的培养基,同时在下室并在37℃、5%CO2的加湿环境中孵育6h。
S4.孵育后,检测上室THP-1侧培养物中IL-8、TNF-α浓度,评价待测物对肠道免疫能力的调节作用;检测下室Caco-2侧培养物中脂多糖水平,从而评价待测物对肠道屏障功能的调节能力,测试结果如下表3所示:

试验结果如表3所示,结果表明与Caco-2细胞直接接触的LR08发酵乳共培养的THP-1细胞中,IL-8分泌显著受到抑制(p<0.01)。这一结果表明,LR08发酵乳触发了Caco-2细胞产生一些对THP-1细胞免疫抑制的物质。另外,与TNF-α组相比,LR08组下室中TNF-α浓度显著降低(p<0.01),说明LR08可以显著改善肠道屏障功能。
实施例5
本实施例验证了鼠李糖乳杆菌LRa05【来自微康益生菌(苏州)股份有限公司】对肠道屏障功能和肠道免疫能力的调节作用,步骤如下:
S1.LRa05菌株的制备:将LRa05接种于MRS液体培养基中,37℃下培养18h进行活化,连续活化2次,得到活化液;将活化液按2%(v/v)的接种量接种于MRS液体培养基中,37℃下培养18h,得到菌液;将菌液在8000g下离心10min,用无菌磷酸盐缓冲盐水(PBS;pH7.4)清洗三次,并重悬于PBS中,通过直接显微镜计数测定为108CFU/mL。
S2.按实施1所述构建评价调节肠道屏障功能和肠道免疫能力模型:,选取跨膜电阻满足要求的模型9个,分成三组(每组3个):空白对照组(CTL组),IL-10刺激组(IL-10组),IL-10刺激+益生菌BL21干预组(BL21组)。每组的处理方式如下:
CTL组:不做任何处理,整个模型在37℃、5%CO2的加湿环境中孵育6h。
IL-10组:用含100ng/mL IL-10的完全RPMI1640培养基替换Transwell上室THP-1细胞的培养基,并在37℃、5%CO2的加湿环境中孵育6h。
LRa05组:将1.0×108CFU长芽孢杆菌菌株加入下室的Caco-2细胞一侧,并在37℃、5%CO2的加湿环境下孵育24h。然后,用含100ng/mL IL-10的完全RPMI1640培养基替换Transwell上室THP-1细胞的培养基,同时在下室并在37℃、5%CO2的加湿环境中孵育6h;
S3.孵育后,检测上室THP-1侧培养物中IL-8浓度,评价待测物对肠道免疫能力的调节作用;检测下室Caco-2侧培养物中IL-10水平,从而评价待测物对肠道屏障功能的调节能力,测试结果如下表4所示:;

试验结果如表1所示,结果表明与Caco-2细胞直接接触的LRa05细胞共培养的THP-1细胞中,IL-8分泌显著受到抑制(p<0.01)。这一结果表明,LRa05触发了Caco-2细胞产生一些对THP-1细胞免疫抑制的物质。另外,与IL-10组相比,LRa05组下室中IL-10浓度显著升高(p<0.01),说明LRa05可以显著改善肠道屏障功能。
综上所述,本发明提供了一种评价调节肠道屏障功能和肠道免疫能力的方法和应用。该方法简单有效,在药物和功能性食品开发评价方面具有很高的应用价值。
申请人声明,本发明通过上述实施例来说明本发明的评价调节肠道屏障功能和肠道免疫能力的方法,但本发明并不局限于上述工艺步骤,即不意味着本发明必须依赖上述工艺步骤才能实施。所属技术领域的技术人员应该明了,对本发明的任何改进,对本发明所选用原料的等效替换及辅助成分的添加、具体方式的选择等,均落在本发明的保护范围和公开范围之内。
Claims (7)
1.一种评价调节肠道屏障功能和肠道免疫能力的方法,其特征在于,所述方法包括以下步骤:
(1)在Transwell孔板的一端小室内培养人结肠上皮细胞Caco-2,形成Caco-2单层细胞;将THP-1细胞进行培养,分化为人巨噬细胞样细胞THP-1;
(2)将步骤(1)的Transwell孔板倒置,使含有Caco-2单层细胞的一端小室完全浸没于DMEM液体培养基中,再在倒置后的Transwell孔板另一端小室内接种人巨噬细胞样细胞THP-1,并使人巨噬细胞样细胞THP-1浸没于RMPI1640培养基中,两端同时进行培养;
(3)采用RMPI1640培养基替换步骤(2)的DMEM液体培养基后,在含有Caco-2的一端添加待检测物后,进行孵育;并在含有THP-1的一端添加脂多糖和TNF-α后,进行孵育;
所述待检测物为益生菌或后生元中的任意一种或两种的组合;
所述待检测物在RMPI1640培养基中的含量为105~108CFU/mL;
所述脂多糖在RMPI1640培养基中的含量10~1000ng/mL,所述TNF-α在RMPI1640培养基中的含量40~1000ng/Ml;
(4)检测Caco-2侧培养物中脂多糖水平,评价待检测物对肠道屏障功能的调节能力;并检测THP-1侧培养物中TNF-α、IL-10或IL-8水平,评价待检测物对肠道屏障功能的调节能力。
2.根据权利要求1所述的评价调节肠道屏障功能和肠道免疫能力的方法,其特征在于,步骤(1)中,所述培养人结肠上皮细胞Caco-2的温度为35~40℃,时间为15~21天。
3.根据权利要求1所述的评价调节肠道屏障功能和肠道免疫能力的方法,其特征在于,步骤(1)中,所述形成Caco-2单层细胞后跨膜电阻为100ohm/cm2以上。
4.根据权利要求1所述的评价调节肠道屏障功能和肠道免疫能力的方法,其特征在于,步骤(1)中,分化为人巨噬细胞样细胞THP-1的具体步骤为:
采用160~165nM的佛波醇肉豆蔻酸乙酸酯溶液在35~40℃下处理THP-1细胞70~74h后,再将其接种于RMPI1640培养基中在35~40℃下培养22~26h,使THP-1分化为人巨噬细胞样细胞THP-1。
5.根据权利要求1所述的评价调节肠道屏障功能和肠道免疫能力的方法,其特征在于,步骤(2)中,所述培养的温度为35~40℃,时间为20~30h。
6.根据权利要求1所述的评价调节肠道屏障功能和肠道免疫能力的方法,其特征在于,步骤(3)中,在含有Caco-2的一端添加待检测物后,进行孵育的温度为35~38℃,时间为20~30h。
7.根据权利要求1所述的评价调节肠道屏障功能和肠道免疫能力的方法,其特征在于,步骤(3)中,在含有THP-1的一端添加脂多糖和TNF-α后,进行孵育的温度为35~38℃,时间为4~8h。
