背景技术
神经退行性疾病是一类因特异性神经元大量丢失而致残、致死的复杂疾病。神经退行性疾病造成中枢或外周神经系统结构和功能的进行性退化,严重降低生活质量,甚至威胁生命,给患者、家庭及社会造成了巨大的心理压力和经济负担。随着全球生育率的降低,人口老龄化逐年加剧,世界卫生组织预测,2040年神经退行性疾病将会取代癌症,成为人类第二大致死疾病。
近年来,干细胞疗法为神经退行性疾病的治疗提供了新的希望。神经退行性疾病的病理机制主要是由于中枢神经系统中的神经细胞,尤其是神经元,在退化或死亡后很难再生,从而导致永久性的功能异常。比如阿尔兹海默症病理机制之一就是脑部海马区胆碱能神经元功能异常,帕金森症则是由于中脑黑质致密带多巴胺能神经元变性坏死或缺失导致多巴胺神经递质的减少。干细胞是一类具有自我更新及多向分化潜能的细胞,例如神经干细胞(Neural Stem Cells,NSCs)能够分化为特定的神经细胞,包括神经元和胶质细胞,这些细胞可以替代退化的神经元和胶质细胞,重建神经退行性疾病中的功能性神经回路。因此,用干细胞替代病变或损伤的脑细胞对神经退行性疾病具有巨大的治疗潜力。然而,干细胞存在自发分化缓慢,且分化方向不可控等问题,这限制了其神经修复应用的安全性和有效性。探索实现干细胞快速、精准分化的有效方法是我们需要迫切解决的科学问题。
小分子化合物因其便捷性、可控性、功能多样性等特点在干预干细胞增殖分化等生物学行为中具有巨大优势。其中具有抗氧化、抗炎、神经保护作用的生物来源小分子化合物在调控干细胞神经分化方面已有广泛研究。例如维甲酸(Rtinoic Acid,RA)已证实可以促进多种干细胞包括胚胎干细胞、神经干细胞、脐带间充质干细胞向神经元甚至是多巴胺能神经元分化。然而,生物来源小分子化合物均存在稳定性差、靶向性低、半衰期短、作用机制不明确等问题,使得小分子化合物在干预干细胞分化过程中生物利用度较低。研究表明,利用纳米粒子作为药物递送载体可以改善药物生物利用度低的问题。将小分子化合物修饰到纳米粒子表面、包裹在纳米凝胶/粒子内部、封装在有机/无机纳米粒子中,可实现小分子的稳定、持续、靶向递送。如何在考虑生物来源小分子化合物固有特性前提下,实现小分子化合物的纳米化构建,需要结合其物化性质寻找可行有效的物理或化学结合途径。可知维甲酸小分子化合物具有羧酸结构,其可与多种无机金属离子(Fe3+、Ca2+、Zn2+、Cu2+等)发生络合反应得到羧酸盐固体产物,且羧酸盐在弱酸性条件下可再次缓慢降解释放出游离的药物小分子和金属离子。值得注意的是,金属离子在人体生命活动中发挥着关键作用,尤其Ca2+可维持细胞膜两侧的生物电位和正常的神经传导功能。近年来,研究人员发现Ca2+在神经发生中扮演着关键角色,尤其可参与调节干细胞的增殖分化命运。但目前有机酸钙盐对干细胞神经分化的研究未见报道,开发一种有机酸钙盐有望实现干细胞分化方向与分化速度的可控干预,可对神经干细胞的分化进行快速、精准调控。
发明内容
针对上述现有技术,本发明的目的是提供一种维甲酸-钙纳米缓释剂及其在促进干细胞向神经元分化中的应用。本发明将维甲酸小分子化合物与无机金属离子Ca2+结合构建纳米缓释剂,维甲酸-钙纳米粒子经胞吞进入细胞后,在溶酶体弱酸性微环境作用下可发生缓慢降解进而释放维甲酸小分子和Ca2+,在时间和空间维度上实现神经干细胞的快速、精准调控。本发明的维甲酸-钙纳米缓释剂有望实现神经干细胞定向分化,为干细胞定向分化提供了一种创新性的思路和方法,对开发基于干细胞疗法的神经退行性疾病的治疗具有重要临床应用价值和社会意义。
为实现上述目的,本发明采用如下技术方案:
本发明的第一方面,提供维甲酸-钙纳米缓释剂的制备方法,包括以下步骤:
(1)将钙盐溶于双蒸水中,得到钙盐溶液;将维甲酸溶于乙醇中,得到维甲酸溶液;
(2)将维甲酸溶液缓慢加入钙盐溶液中得到混合液,调整混合液pH值至8.5,加热下搅拌进行反应,反应后收集固体产物,洗涤、干燥后,得到维甲酸-钙纳米缓释剂。

本发明的第四方面,提供一种促进干细胞定向分化的培养基,所述培养基以维甲酸-钙纳米缓释剂为有效成分。
本发明的第五方面,提供一种促进干细胞定向分化的方法,所述方法为:将干细胞接种于培养基中,进行诱导分化处理。
本发明的有益效果:
(1)本发明首次利用维甲酸小分子与钙离子的络合反应,构建具有pH响应的缓释功能纳米粒子,制备的维甲酸-钙纳米材料粒径均一、形态可控,在弱酸性条件下可释放维甲酸小分子和钙离子。合成过程方法简单、成本低廉、绿色无害,可实现维甲酸-钙纳米缓释剂的重复、稳定、批量生产。
(2)本发明的维甲酸-钙纳米缓释剂具有良好的细胞相容性,经胞吞进入细胞溶酶体内释放有效组分,能够促进干细胞的定向分化。采用本发明的维甲酸-钙纳米缓释剂作用神经干细胞,维甲酸和Ca2+之间协同作用,有利于神经干细向神经元方向的分化。为干细胞定向分化提供了一种创新性的思路和方法,对开发基于干细胞疗法的神经退行性疾病的治疗具有重要临床应用价值和社会意义。

具体实施方式
应该指出,以下详细说明都是例示性的,旨在对本申请提供进一步的说明。除非另有指明,本文使用的所有技术和科学术语具有与本申请所属技术领域的普通技术人员通常理解的相同含义。
正如背景技术部分介绍的,干细胞存在自发分化缓慢,且分化方向不可控等问题,现有分化方法和技术很难实现干细胞的快速、精准分化,这限制了其神经修复应用的安全性和有效性。小分子化合物作为调控干细胞分化的一种有效方法,其应用往往是被溶解或添加到培养基中进行细胞处理,加之小分子化合物存在稳定性差、靶向性低、半衰期短、作用机制不明确等问题,这使得小分子化合物在干预干细胞分化过程中生物利用度较低。
基于此,本发明的目的是提供一种新型的能够促进干细胞定向分化的培养方法。经过发明人研究发现,只有将维甲酸溶液滴加到钙盐溶液中,调节pH值至8.5且在60℃下反应,才能制备得到维甲酸-钙纳米缓释剂。本发明在基于维甲酸固有物化特性的前提下,首次将维甲酸小分子与无机金属粒子Ca2+相结合,制备具有pH响应性的维甲酸-钙纳米缓释剂。目前,利用有机酸钙盐调控干细胞神经分化的研究未见报道。维甲酸-钙纳米粒子经胞吞进入细胞后,在溶酶体作用下可发生缓慢降解进而释放维甲酸小分子和Ca2+。最终,细胞内有效组分的释放可在时间和空间维度上对干细胞的分化进行调控。
为了使得本领域技术人员能够更加清楚地了解本申请的技术方案,以下将结合具体的实施例详细说明本申请的技术方案。
本发明实施例中所用的试验材料均为本领域常规的试验材料,均可通过商业渠道购买得到。
实施例1
(1)将0.176g醋酸钙溶于10ml ddH2O,搅拌使其充分溶解,记为溶液A,测定溶液pH值。将0.9g维甲酸溶于50ml乙醇(90%v),搅拌使其充分溶解,记为溶液B,测定溶液pH值。
(2)将溶液B以2mL/min的滴加速度加入溶液A中(边滴加边搅拌),混合液中维甲酸和醋酸钙的摩尔比为3:1,用0.1M HCl调整混合溶液的pH值为8.5。
(3)将混合液置于60℃水浴中,600rpm搅拌进行络合反应,观察产物析出情况,2h后反应结束。离心收集产物,用无水乙醇清洗三次,再用双蒸水洗涤三次,经真空冷冻干燥24h后(冷阱温度为-40℃,真空度为8Pa),获得维甲酸-钙纳米缓释剂材料。
实施例2
(1)将0.176g醋酸钙溶于10ml ddH2O,搅拌使其充分溶解,记为溶液A,测定溶液pH值。将0.6g维甲酸溶于50ml乙醇(90%v),搅拌使其充分溶解,记为溶液B,测定溶液pH值。
(2)将溶液B以2mL/min的滴加速度加入溶液A中(边滴加边搅拌),混合液中维甲酸和醋酸钙的摩尔比为2:1,用0.1M HCl调整混合溶液的pH值为8.5。
(3)将混合液置于60℃水浴中,600rpm搅拌进行络合反应,观察产物析出情况,2h后反应结束。离心收集产物,用无水乙醇清洗三次,再用双蒸水洗涤三次,经真空冷冻干燥24h后(冷阱温度为-40℃,真空度为8Pa),获得维甲酸-钙纳米缓释剂材料。
实施例3
(1)将0.176g醋酸钙溶于10ml ddH2O,搅拌使其充分溶解,记为溶液A,测定溶液pH值。将0.3g维甲酸溶于50ml乙醇(90%v),搅拌使其充分溶解,记为溶液B,测定溶液pH值。
(2)将溶液B以2mL/min的滴加速度加入溶液A中(边滴加边搅拌),混合液中维甲酸和醋酸钙的摩尔比为1:1,用0.1M HCl调整混合溶液的pH值为8.5。
(3)将混合液置于60℃水浴中,600rpm搅拌进行络合反应,观察产物析出情况,2h后反应结束。离心收集产物,用无水乙醇清洗三次,再用双蒸水洗涤三次,经真空冷冻干燥24h后(冷阱温度为-40℃,真空度为8Pa),获得维甲酸-钙纳米缓释剂材料。
对比例1
与实施例1的区别在于:步骤(2)中,将A溶液滴加到B溶液中。
对比例2
与实施例1的区别在于:步骤(2)中,调整混合溶液的pH值为7.0。
对比例3
与实施例1的区别在于:步骤(3)中,将混合液置于室温下。
以上实施例为三种不同摩尔比(维甲酸和醋酸钙的摩尔比为3:1,2:1,1:1)条件下,制备维甲酸-钙纳米缓释剂材料。对比例1与实施例1的区别在于将溶液A缓慢滴加入溶液B中,对比例2与实施例1的区别在于调整混合溶液的pH值为7.0,对比例3与实施例1的区别在于反应混合液置于室温。对比例1,对比例2,对比例3均未有固体产物析出;实施例1,实施例2,实施例3均有固体产物析出。
对实施例1的样品进行分析表征(形貌、化学结构和pH响应性)。如图1,SEM成像显示维甲酸-钙纳米粒子的尺寸为300-400nm,呈现规则的球状形貌。如图2,FTIR光谱显示维甲酸-钙纳米粒子出现了维甲酸化学官能团的特征吸收峰,并且羧基官能团中-OH的伸缩振动峰消失,这暗示Ca2+与羟基发生了结合。图3结果表明,当体系pH值由7.0降为5.5时,维甲酸-钙纳米粒子可以在弱酸性条件下缓慢释放维甲酸分子和钙离子。
试验例
1、提取大鼠来源神经干细胞,具体步骤如下:
a、取孕13-15天的SD大鼠,乙醚麻醉后引颈处死。用医用碘伏和75%医用酒精对其腹部进行消毒,打开孕鼠腹部取出子宫,移入含有无菌PBS的培养皿中。用手术剪打开子宫取出胚胎,剪下其头部,于体视镜下分离取出海马区组织。
b、将海马组织剪碎成1mm3的碎块,然后将组织碎块移入含有培养基的离心管中,反复轻柔吹打数次。向组织块中加入适量Accutase酶,37℃水浴震荡15min,酶解后反复轻柔吹打,静置后收集上清液。离心获取单细胞,加入新鲜培养基进行重悬,接种于T型瓶中培养获得神经干细胞。
2、将神经干细胞分别接种于含有10μg/mL、25μg/mL、50μg/mL、100μg/mL、200μg/mL实施例1所制备材料的增殖培养基中(增殖培养基主要成分包括:Neurobasal培养基、2%B27因子、20ng/mL bFGF、20ng/mL EGF、1%双抗),并以增殖培养基作为空白对照组。分别培养1、3、5天后,检测细胞活率,并对细胞进行死活染色成像。如图4A所示,CCK8检测结果发现,不同浓度材料处理组的细胞活性与空白对照组无显著性差异。细胞死活染色结果显示(图4B),细胞贴壁状态良好,且无明显死细胞,说明材料具有很好的生物相容性。
3、将神经干细胞分别接种于含有200μg/mL实施例1所制备材料(实施例1组)、对比1组(等量维甲酸,RA)、对比2组(等量Ca2+)、对比3组(等量RA+Ca2+,其中维甲酸和Ca2+的摩尔比为3:1)的增殖培养基中,并以增殖培养基作为空白对照组。在培养第3天,检测细胞活性。如图5所示,CCK8结果表明,维甲酸-钙纳米材料组与空白对照组无显著性差异,而维甲酸组、Ca2+组、维甲酸+Ca2+组的细胞活性显著低于空白对照组。该结果说明在等质量浓度条件下,维甲酸-钙纳米材料因缓释能力对细胞活性并未造成影响,而维甲酸组、Ca2+组、维甲酸+Ca2+组不具备胞内缓释能力,瞬时高质量浓度组份的暴露造成细胞死亡,显著降低了细胞的活性。
4、用含有50μg/mL实施例1所制备材料(实施例1组,NPs)、对比1组(等量维甲酸,RA)、对比2组(等量Ca2+)、对比3组(等量RA+Ca2+,其中维甲酸和Ca2+的摩尔比为3:1)的分化培养基处理神经干细胞(分化培养基主要成分包括:Neurobasal培养基、2%B27因子、2%FBS、1%双抗),并以分化培养基作为空白对照组。在诱导分化的第7天,分析分化细胞的表型,分化细胞中神经元(Tuj-1阳性细胞)所占百分比如表1和图6所示。

由表1可以看出,实施例1组中分化细胞中神经元所占百分比远高于对比1~3组,对比实施例1组、对比1组和对比2组可以看出,维甲酸与钙离子之间可以协同促进神经干细胞向神经元分化。
如图6所示,细胞免疫荧光染色结果表明,处理组的神经元细胞数量明显多于维甲酸+Ca2+组,且更趋于形成细胞间网络结构。
综上所述,本发明制备的维甲酸-钙纳米缓释剂材料对神经干细胞具有良好的生物相容性,维甲酸与钙离子可协同促进神经干细胞向神经元方向的分化。

Claims (10)
1.一种维甲酸-钙纳米缓释剂的制备方法,其特征在于,包括以下步骤:
(1)将钙盐溶于双蒸水中,得到钙盐溶液;将维甲酸溶于乙醇中,得到维甲酸溶液;
(2)将维甲酸溶液缓慢加入钙盐溶液中得到混合液,调整混合液pH值至8.5,加热下搅拌进行反应,反应后收集固体产物,洗涤、干燥后,得到维甲酸-钙纳米缓释剂。
2.根据权利要求1所述的制备方法,其特征在于,步骤(1)中,所述钙盐为醋酸钙;所述钙盐溶液的浓度为17.6g/L。
3.根据权利要求1所述的制备方法,其特征在于,步骤(1)中,所述乙醇的体积浓度为90%;所述维甲酸溶液的浓度为6~18g/L。
4.根据权利要求1所述的制备方法,其特征在于,步骤(2)中,所述混合液中维甲酸和钙盐的摩尔比为(3~1):1;采用浓度为0.1M HCl溶液调整混合液pH值至8.5。
5.根据权利要求1所述的制备方法,其特征在于,步骤(2)中,所述加热为水浴加热,所述加热的温度为60℃,所述搅拌速度为600rpm;通过离心收集固体产物,离心的速度为10000rpm,离心的时间为10min;所述洗涤为:依次用乙醇和双蒸水分别洗涤2~5次。
6.权利要求1~5任一项所述制备方法制备得到的维甲酸-钙纳米缓释剂,其特征在于,所述维甲酸-钙纳米缓释剂的粒径为300-400nm;在弱酸性环境中,所述维甲酸-钙纳米缓释剂释放维甲酸分子和钙离子。
7.权利要求6所述的维甲酸-钙纳米缓释剂在促进干细胞定向分化或制备促进干细胞定向分化的培养基中的应用。
8.根据权利要求7所述的应用,其特征在于,所述促进干细胞定向分化为促进神经干细胞定向神经元分化。
9.一种促进干细胞定向分化的培养基,其特征在于,所述培养基以权利要求6所述的维甲酸-钙纳米缓释剂为有效成分。
10.一种促进干细胞定向分化的方法,其特征在于,所述方法为:将干细胞接种于权利要求9所述的培养基中,进行诱导分化处理。
