Abstract
本发明公开了一种枸杞叶中亚精胺类成分的富集制备方法,包括步骤:将干燥的枸杞叶经乙醇水溶剂渗漉,渗漉液回收溶剂,加入硅胶搅拌,干燥后粉碎,得到提取物粉末;将提取物粉末倒入乙酸乙酯‑正己烷混合溶液中,常温搅拌,提取后过滤,得到不溶物粉末;将不溶物粉末重复S2,干燥,粉碎,以聚乙二醇‑水体系为溶剂,调节pH,减压蒸馏,收集馏分;将馏分经大孔树脂柱,以乙醇‑水系统洗脱,制得到富含亚精胺类的组分。本发明通过溶剂提取,树脂纯化与蒸馏手段结合,得到亚精胺类组分含量纯度相比与枸杞叶原料提升数百倍,可达8‑12%。
Description
技术领域
本发明涉及植物提取技术领域,尤其涉及一种枸杞叶中亚精胺类成分的富集制备方法。
背景技术
枸杞为茄科枸杞属的一种多年生落叶灌木,其果实枸杞子为药食同源中药材,始载于《神农本草经》。枸杞叶也在地方中药材标准中收录。
亚精胺类是植物多胺中一类参与植物DNA及激素调控的物质。近年来有研究表明,亚精胺类成分可能具有抗氧化,延缓衰老和缓解记忆障碍等活性。由于亚精胺类成分在植物中含量差异极大,稳定性较低,经过各类研究筛选后,发现枸杞子和枸杞叶均可作为亚精胺类成分的植物原料来源,其中从枸杞叶中富集亚精胺类成分的工艺鲜有报道。因此,有必要开发高效的,相对稳定的从枸杞叶中富集亚精胺类的方法。
发明内容
本发明的目的在于提供一种枸杞叶中亚精胺类成分的富集制备方法,以解决上述背景技术中所提出的问题。
为达到上述目的,本发明采用的技术方案是:一种枸杞叶中亚精胺类成分的富集制备方法,包括以下步骤:
S1)将干燥的枸杞叶在常温常压下经乙醇水溶剂渗漉,渗漉液回收溶剂,加入硅胶搅拌,干燥后粉碎,得到提取物粉末;
S2)将提取物粉末倒入乙酸乙酯-正己烷混合溶液中,常温搅拌,提取后过滤,得到不溶物粉末;
S3)将不溶物粉末重复S2一次后,干燥,粉碎,以聚乙二醇-水体系为溶剂,调节pH,减压蒸馏,收集馏分;
S4)将馏分经大孔树脂柱,以乙醇-水系统洗脱,制得到富含亚精胺类的组分。
作为进一步的优化,S1中所述乙醇水溶剂质量比为4:(1-2);所述枸杞叶与乙醇水溶剂的料液比为1:(6-10);所述硅胶与枸杞叶的质量比为1:(20-30)。通过加入硅胶可降低提取物的粘度,提高流动性,易于粉碎。
作为进一步的优化,S1中的干燥温度为60-80℃。
作为进一步的优化,S2中所述提取物粉末与乙酸乙酯-正己烷混合溶液的料液比为1:(4-8);乙酸乙酯与正己烷的质量比为1:(2-5),合适比例的乙酸乙酯-正己烷溶液可在不影响亚精胺类成分的前提下,脱去色素及脂溶性成分。
作为进一步的优化,S3中的干燥温度为40-45℃,pH值为8.0-12.0;收集馏分的条件为:蒸馏温度为120-160℃,压力为-0.08-0.1Mpa。
作为进一步的优化,S4中馏分以60-80%乙醇溶解,上样体积与树脂柱体积为1:(4-12),收集30-50%乙醇洗脱部分,减压干燥,得最终产物富含亚精胺类的组分。
经上述步骤得到的亚精胺类成分含量根据HPLC定量方法,范围在8-12%。
与已有技术相比,本发明的有益效果体现在:
1.本发明的富集工艺,通过温度的控制与溶剂的选择,在提取过程中最大保留了枸杞叶的亚精胺类成分;
2.本发明的富集工艺,通过溶剂提取,树脂纯化与蒸馏手段结合,得到亚精胺类组分含量纯度相比与枸杞叶原料提升数百倍。

具体实施方式
以下是本发明的具体实施例,结合附图对本发明的技术方案作进一步的描述,但本发明并不限于这些实施例。
实施例1:
如图1所示,将5kg干燥的枸杞叶粉碎经渗漉筒渗漉,渗漉溶剂为40L 80%乙醇,渗漉液浓缩,加入200g硅胶搅拌,70℃减压干燥4小时,粉碎,倒入8L乙酸乙酯-正己烷1:4溶液,搅拌1小时,过滤,不溶物倒入4L乙酸乙酯-正己烷1:4溶液,搅拌1小时,过滤,不溶物40℃鼓风干燥2小时,倒入蒸馏瓶,加入聚乙二醇-水 10:1溶剂5L,pH调节为9.0,蒸馏温度150℃,压力-0.1Mpa,收集蒸馏物,蒸馏物加入30mL 80%乙醇搅拌溶解,倒入200mL D101大孔树脂吸附,以20%乙醇溶液洗脱除杂,再以50%乙醇溶液洗脱,回收溶剂,减压干燥,至恒重。最终产物干重41.2g,亚精胺类成分含量根据HPLC测定结果为8.3%。
实施例2:
将2kg干燥的枸杞叶粉碎经渗漉筒渗漉,渗漉溶剂为15L 65%乙醇,渗漉液浓缩,加入80g硅胶搅拌,80℃鼓风干燥6小时,粉碎,倒入2.1L乙酸乙酯-正己烷1:3溶液,搅拌1小时,过滤,不溶物倒入1.2L乙酸乙酯-正己烷1:3溶液,搅拌1小时,过滤,不溶物40℃鼓风干燥3小时,倒入蒸馏瓶,加入聚乙二醇-水 20:1溶剂3L,pH调节为10.0,蒸馏温度155℃,压力-0.08Mpa,收集蒸馏物,蒸馏物加入10mL 80%乙醇搅拌溶解,倒入120mL D101大孔树脂吸附,以10%乙醇溶液洗脱除杂,再以30%乙醇溶液洗脱,回收溶剂,减压干燥,至恒重。最终产物干重14.6g,亚精胺类成分含量根据HPLC测定结果为9.6%。
对比例
将2kg干燥的枸杞叶粉碎经20L 80%乙醇回流提取,提取液浓缩,至粘稠状,加入20%乙醇2L搅拌,静置,沉淀,过滤,不溶物经80℃鼓风干燥4小时后,粉碎,得最终产物。最终产物干重257.9g,亚精胺类成分含量根据HPLC测定结果为0.28%。

Claims (8)
1.一种枸杞叶中亚精胺类成分的富集制备方法,其特征在于,包括以下步骤:
S1)将干燥的枸杞叶经乙醇水溶剂渗漉,渗漉液回收溶剂,加入硅胶搅拌,干燥后粉碎,得到提取物粉末;
S2)将提取物粉末倒入乙酸乙酯-正己烷混合溶液中,常温搅拌,提取后过滤,得到不溶物粉末;
S3)将不溶物粉末重复S2,干燥,粉碎,以聚乙二醇-水体系为溶剂,调节pH,减压蒸馏,收集馏分;
S4)将馏分经大孔树脂柱,以乙醇-水系统洗脱,制得到富含亚精胺类的组分。
2.根据权利要求1所述的枸杞叶中亚精胺类成分的富集制备方法,其特征在于,S1中的渗漉条件为温度为25±2℃,大气压为标准大气压。
3.根据权利要求1所述的枸杞叶中亚精胺类成分的富集制备方法,其特征在于,S1中所述乙醇水溶剂质量比为4:(1-2);所述枸杞叶与乙醇水溶剂的料液比为1:(6-10);所述硅胶与枸杞叶的质量比为1:(20-30)。
4.根据权利要求1所述的枸杞叶中亚精胺类成分的富集制备方法,其特征在于,S1中的干燥温度为60-80℃。
5.根据权利要求1所述的枸杞叶中亚精胺类成分的富集制备方法,其特征在于,S2中所述提取物粉末与乙酸乙酯-正己烷混合溶液的料液比为1:(4-8);乙酸乙酯与正己烷的质量比为1:(2-5)。
6.根据权利要求1所述的枸杞叶中亚精胺类成分的富集制备方法,其特征在于,S3中重复S2一次。
7.根据权利要求1所述的枸杞叶中亚精胺类成分的富集制备方法,其特征在于,S3中的干燥温度为40-45℃,pH值为8.0-12.0;收集馏分的条件为:蒸馏温度为120-160℃,压力为-0.08-0.1Mpa。
8.根据权利要求1所述的枸杞叶中亚精胺类成分的富集制备方法,其特征在于,S4中馏分以60-80%乙醇溶解,上样体积与树脂柱体积为1:(4-12),收集30-50%乙醇洗脱部分,减压干燥,得最终产物富含亚精胺类的组分。
