技术领域
本发明涉及化妆品技术领域,特别涉及一种美白祛斑组合物及其制备方法与应用。
背景技术
皮肤是人体的第一道屏障,与外界环境接触最多。皮肤,尤其是面部皮肤容易产生一些颜色较深的斑点或斑块。引起皮肤斑点或斑块原因众多,外界环境的影响,如紫外线照射、空气污染,或者使用的化妆品重金属含量超标;内在因素,如压力增大导致人体激素水平变化或失调,肝肾功能障碍等,都会导致面部黑色素聚集,致使斑点或斑块的产生。当身体新陈代谢欠佳时,部分色素会留在皮肤表层,形成黑斑。另外由于近年来人们生活节奏加快,工作压力增大,生活不规律,睡眠质量差,从而导致越来越多的人长期处于亚健康状态,皮肤黯沉、色斑增多等现象日益加重。
由于对美的追求,人们希望通过化妆品来达到美白祛斑的效果。现有市场上祛斑美白的很多,为了达到能达到祛斑美白的效果,在产品中祛斑活性物质添加量很高,这对于皮肤敏感或轻微皮肤受损的人群,刺激性也会相应增加。或是有的美白祛斑产品,其祛斑美白效果不好。

发明内容
本发明的目的在于提供美白祛斑组合物,从而克服祛斑美白效果不好,或是对皮肤刺激性较大的缺点。
本发明的另一个目的在于提供美白祛斑组合物的制备方法。以及将该祛斑美白组合物用于制备化妆品。
为实现上述目的,本发明提供了一种美白祛斑组合物,至少包括以下原料:白术根提取物、白及根提取物、甘草根提取物、白丁香提取物、芍药根提取物和灵芝提取物。
优选地,上述技术方案中,所述的美白祛斑组合物,按重量百分数计,至少包括以下原料:白术根提取物0.1-5%、白及根提取物0.5-6%、甘草根提取物0.5-5%、白丁香提取物0.5-5%、芍药根提取物0.5-5%和灵芝提取物0.1-5%,余量为水。
优选地,上述技术方案中,所述的美白祛斑组合物,按重量百分数计,至少包括以下原料:白术根提取物1-3%、白及根提取物1.5-3%、甘草根提取物1-3%、白丁香提取物1-3%、芍药根提取物1-3%和灵芝提取物1-3%,余量为水。
优选地,上述技术方案中,所述的美白祛斑组合物,按重量百分数计,至少包括以下原料:白术根提取物1.5%、白及根提取物2%、甘草根提取物1.8%、白丁香提取物1.5%、芍药根提取物1.8%和灵芝提取物1.6%,余量为水。
优选地,上述技术方案中,所述组合物还包括以下原料:1,3丁二醇、对羟基苯乙酮和A91。
优选地,上述技术方案中,按重量百分数计,所述组合物还包括以下原料:1,3丁二醇1-5%、对羟基苯乙酮0.05-1%和A91 0.01-1%。
上述的美白祛斑组合物在制备美白祛斑化妆品中的应用。例如可用于制备爽肤水、面膜、精华液、粉底液、乳液等化妆品。
一种美白祛斑组合物的制备方法,包括以下步骤:
(1)按配比称取制备美白祛斑组合物的原料,原料包括:白术根、白及根、甘草根、白丁香、芍药根、灵芝;然后进行粉碎,得到混合原料粉末;
(2)提纯:将混合原料粉末用水浸泡一段时间,然后在真空恒温的环境下提纯一段时间,得到滤液;反复进行多次提纯,将多次提纯的滤液混合,得到混合滤液;
(3)过滤和浓缩:将混合滤液进行过滤,再进行浓缩,得到提取液,再加入1,3丁二醇、对羟基苯乙酮和A91,得到美白祛斑组合物。
优选地,上述技术方案中,步骤(2)的提纯是在真空度为-0.1MPa温度为45-55℃条件下提纯,反复提纯的次数为2-4次;提纯时,混合原料与水混合的质量比为1:20-60。
优选地,上述技术方案中,步骤(3)的过滤为,将混合滤液用200nm的陶瓷膜过滤,得到纯情液;
优选地,上述技术方案中,步骤(3)的浓缩为,在真空度-0.1MPa温度为45-55℃的条件下浓缩,得到提取液。
本发明用到的原料均可从市场上购得。
白术根:含挥发油1.4%,主要成分为苍术醇、苍术酮等,并含有维生素A。苦甘,温。健脾益气,燥湿利水,止汗。可以补气益血、美白润肤,适于气血虚寒导致的皮肤粗糙、萎黄、黄褐斑、色素沉着等。有延缓衰老的功效;白术对于美白皮肤有特效;白术对皮肤致病真菌如雾状表皮癣菌等有抑制作用。
白及根:苦平无毒.又苦甘微寒性涩。功效是收敛止血,消肿生肌。具有美容养颜的功效,它含有丰富的淀粉、葡萄糖、挥发油和粘液质等成分,有美白祛斑、收敛止血、消肿生肌的功效。
甘草根:其根及茎,性甘平,归心、肺、脾、胃经,主要有益气健脾、清热解毒、祛痰止咳、缓急止痛、缓和药性等作用。现代药理研究表明:甘草含甘草甜素、甘草黄酮类化合物甘草贰、甘草甙元及天冬酞胺、甘露醇等。甘草黄酮还有抑制酪氨酸酶活性、清除氧目由基,其抗氧化能力与维生素E比较接近。
白丁香:提取物主要化学成分是含挥发油、甾醇、三萜成分及酚性成分,含的多糖和多种维生素对人体皮肤有良好的营养、滋润、增白作用。能够通过活血化淤,加快基础代谢的功效,能够改进面部面色,清除脸部黄气,改进黯淡的皮肤颜色,能够超过深层次美白皮肤功效,让肌肤白里透红,光滑如玉,提升肌肤延展性。具有使皮肤收敛、柔软化、保湿、消炎、漂白的性能。还有解除硬化、角化、改善伤痕的作用,不仅能防止小皱纹、眼袋、皮肤松弛,还能保持皮肤湿润、娇嫩,同时,还可以治疗皮肤炎症,对粉刺、雀斑、痤疮以及烫伤、刀伤、虫咬等亦有很好的疗效。
芍药根:主要化学成分是芍药甙,还含有牡丹酚、安息香酸、挥发油、树脂、鞣质、糖类、淀粉、三萜类成分。有效成分芍药甙,对某些致病性细菌和真菌有抑制作用。芍药提取物对弹性蛋白酶的抑制、对自由基的消除、对氮氧化物生成的抑制以及对组织蛋白酶活性的促进等,均显示芍药提取物具有活肤抗衰的功能,并且有比较好的保湿能力。
灵芝:其化学成分含多糖、核苷类、呋喃类、甾醇类、生物碱、三萜类、油脂类、多种氨基酸及蛋白质类、酶类、有机锗及多种微量元素等。主要是通过延缓机体衰老、调节内分泌、代谢系统、消除自由基来达到美容养颜的功效。可以延缓机体衰老,可以保持和调节肌肤的水分,让肌肤充满弹性,并且保持水润,还可以抑制皮肤中黑色素的形成和沉淀,清除色斑。灵芝中的多糖,有显著的拟SOD活性,可显著清除机体产生的自由基,从而阻止自由基对机体的损伤,防止了脂体的过氧化,保护了细胞,延缓了细胞衰老。灵芝中的多肽,则具有五大护肤的功效,有效的补水、保湿、美白、修复、舒缓;而且这些灵芝营养可以直接渗透进入肌肤层,改善肌肤质低,提升肌肤修复和保湿能量,全面呵护肌肤。灵芝中的大量氨基酸,含有纯天然保湿因子,强效锁水,保持肌肤水润弹性,让肌肤变得水嫩光滑的。
与现有技术相比,本发明具有如下有益效果:
(1)本发明美白祛斑组合物,通过各种有效成分的组合协调增效作用,采用多种美白祛斑成分进行合理的复配,以白术根、白芨根、灵芝的提取物作为主要美白原料,添加白芨根提取物能促进上皮细胞修复,可以延缓衰老;添加白术根茎提取物可以控制黑色素的生成,并具有较好的保湿抗氧化效果;添加灵芝提取物可以抑制黑色素的生成和沉淀,并具有较好的延缓细胞衰老的效果。添加甘草根提取物,可以调节诸药,调节药性,使活性成分的药效得到充分发挥,同时还能抑制氨酸酶活性、清除氧目由基,使肌肤达到美白的效果。添加的白丁香和芍药根的提取物,能加快皮肤基础代谢,具有活肤抗衰的功能,同时还具有很好的保湿能力。本申请中还添加1,3丁二醇作为保湿剂及溶剂,添加对羟基苯乙酮和A91作为防腐剂,使的组合物能长时间的保存不变质,同时提高产品的保湿效果。
(2)本发明美白祛斑组合物的原料配方合理,祛斑美白效果显著,且制备方法简单,适用于工业化生产。制成的组合物可广泛应用于不同类型的化妆品中。
具体实施方式
下面结合具体实施例,对本发明的具体实施方式进行详细描述,但应当理解本发明的保护范围并不受具体实施方式的限制。
实施例1
一种美白祛斑组合物,按重量百分数计,包括以下原料:白术根提取物1.5%、白及根提取物2%、甘草根提取物1.8%、白丁香提取物1.5%、芍药根提取物1.8%、灵芝提取物1.6%、1,3丁二醇2%、对羟基苯乙酮0.3%和A910.4%,余量为水。
一种美白祛斑组合物的制备方法,包括以下步骤:
(1)按配比称取制备美白祛斑组合物的原料,原料包括:白术根、白及根、甘草根、白丁香、芍药根、灵芝;然后用破壁机进行粉碎,得到混合原料粉末;
(2)提纯:将混合原料粉末用纯净水浸泡60min,混合粉末和纯净水的质量比为1:50,然后在真空度为-0.1MPa温度为50℃条件下提纯60min,取出第一次滤液;再加入纯净水,混合粉末与纯净水的质量比为1:41,在真空度为-0.1MPa温度为50℃条件下提纯60min,取出第二次滤液;再加入纯净水,混合粉末与纯净水的质量比为1:33,在真空度为-0.1MPa温度为50℃条件下提纯60min,取出第三次滤液;将多次提纯的滤液混合,得到混合滤液;
(3)过滤:用200nm的陶瓷膜对混合滤液进行过滤,得到纯清液;
(4)浓缩:将纯清液在真空度为-0.1MPa温度为50℃条件下浓缩,得到提取液,再加入1,3丁二醇、对羟基苯乙酮和A91混合均匀,得到美白祛斑组合物。
将上述的美白祛斑组合物用于制备美白祛斑化妆品,例如可用于制备爽肤水、面膜、精华液、粉底液、乳液等化妆品。
实施例2
一种美白祛斑组合物,按重量百分数计,至少包括以下原料:白术根提取物0.1%、白及根提取物6%、甘草根提取物0.5%、白丁香提取物5%、芍药根提取物5%、灵芝提取物0.1%、1,3丁二醇1%、对羟基苯乙酮1%和A910.01%,余量为水。
一种美白祛斑组合物的制备方法,包括以下步骤:
(1)按配比称取制备美白祛斑组合物的原料,原料包括:白术根、白及根、甘草根、白丁香、芍药根、灵芝;然后用破壁机进行粉碎,得到混合原料粉末;
(2)提纯:将混合原料粉末用纯净水浸泡30min,混合粉末和纯净水的质量比为1:60,然后在真空度为-0.1MPa温度为55℃条件下提纯90min,取出第一滤液;再加入纯净水,混合粉末与纯净水的质量比为1:35,在真空度为-0.1MPa温度为50℃条件下提纯90min,取出第二次滤液;将多次提纯的滤液混合,得到混合滤液;
(3)过滤:用200nm的陶瓷膜对混合滤液进行过滤,得到纯清液;
(4)浓缩:将纯清液在真空度为-0.1MPa温度为55℃条件下浓缩,得到提取液,再加入1,3丁二醇、对羟基苯乙酮和A91混合均匀,得到美白祛斑组合物。
将上述的美白祛斑组合物用于制备美白祛斑化妆品,例如可用于制备爽肤水、面膜、精华液、粉底液、乳液等化妆品。
实施例3
一种美白祛斑组合物,按重量百分数计,至少包括以下原料:白术根提取物5%、白及根提取物0.5%、甘草根提取物5%、白丁香提取物0.5%、芍药根提取物0.5%、灵芝提取物5%、1,3丁二醇5%、对羟基苯乙酮0.05%和A911%,余量为水。
一种美白祛斑组合物的制备方法,包括以下步骤:
(1)按配比称取制备美白祛斑组合物的原料,原料包括:白术根、白及根、甘草根、白丁香、芍药根、灵芝;然后用破壁机进行粉碎,得到混合原料粉末;
(2)提纯:将混合原料粉末用纯净水浸泡90min,混合粉末和纯净水的质量比为1:50,然后在真空度为-0.1MPa温度为45℃条件下提纯30min,取得第一滤液;再加入纯净水,混合粉末与纯净水的质量比为1:40,在真空度为-0.1MPa温度为50℃条件下提纯30min,取出第二次滤液;再加入纯净水,混合粉末与纯净水的质量比为1:30,在真空度为-0.1MPa温度为50℃条件下提纯60min,取出第三次滤液;再加入纯净水,混合粉末与纯净水的质量比为1:20,在真空度为-0.1MPa温度为50℃条件下提纯30min,取出第四次滤液;将多次提纯的滤液混合,得到混合滤液;
(3)过滤:用200nm的陶瓷膜对混合滤液进行过滤,得到纯清液;
(4)浓缩:将纯清液在真空度为-0.1MPa温度为45-55℃条件下浓缩,得到提取液,再加入1,3丁二醇、对羟基苯乙酮和A91混合均匀,得到美白祛斑组合物。
将上述的美白祛斑组合物用于制备美白祛斑化妆品,例如可用于制备爽肤水、面膜、精华液、粉底液、乳液等化妆品。
实施例4-7美白祛斑组合物配方及制备方法如表1所示。各组分的占比以重量百分数计%。实施例4-5采用实施例1的制备方法进行制备,实施例6采用实施例2的制备方法进行制备,实施例7-8采用实施例3的制备方法进行制备。对比例1-7美白祛斑组合物配方如表1所示,制备方法采用实施例1的制备方法进行制备。

一、测试不同实施例和对比例对抑制黑色素生成的影响
1、试剂和仪器
RPMI1640,南宁生航生物技术有限公司;小牛血清(FCS),江苏依莱萨生物技术有限公司;乳酸脱氢酶试剂盒,上海酶联生物科技有限公司;B16细胞,武汉普诺赛生命科技有限公司;
BBHF-16CO2培养箱,上海力申公司;TDL80-2B离心机,上海安亭科学仪器厂;DG5031酶联免疫检测仪,济南来宝科学仪器有限公司。
2,测试方法:
选择对数生长期的同一代细胞,常规胰酶消化处理后吹打成单细胞悬液细胞计数定量接种于24孔培养板。24小时后,弃去上清液,分别加入含有下表2中所示试验材料的培养液。在37℃,体积分数5%的CO2饱和湿性条件下培养,对照孔加RPMI1640培养液,培育4天,中间更换一次培育液。弃去上清液,常规胰酶消化处理后,计数细胞密度,离心得到细胞沉淀,用PBS冲洗两次,离心弃去上清液,定量加含10%的DMSO的1N的NaOH液,75℃水浴45分钟,然后在酶标仪450nm处测定吸光值。
黑色素生成的抑制率(%)=100(各试验材料的吸光度/对照组的吸光度),具体结果见表2。

二、测试不同实施例和对比例的麦拉宁值
试验方法::取人体皮肤三维模型试验盒(MEL-300B)。按照该试验操作流程将三维模型移至24孔细胞培养板中,用长期维持培养基(EPI-100LLMM)置于37℃,5%CO2下恒温箱预培养1小时后,加入不同实施例和对比例的试验样品,分别添加配制好试验样品500μL至角质层侧,空白组加入等量的1,3-丁二醇水溶液(50%)。基底层侧使用长期维持培养基继续培养。2天或3天的间隔交换培养基和受试物共进行20天的培养。培养后,用PBS(-)缓冲洗皮肤组织后,利用MX18(Courage+Khazaka公司制造)测试不同组别的麦拉宁值。测试结果如表3所示。

如表3所示,已知麦拉宁值越低,说明抑制黑色素的能力越强。全部含有本发明原料的药物组合物,其降低麦拉宁值的效果较好;部分含有本发明原料的药物组合物,其降低麦拉宁值的效果不好。
三、美白祛斑效果临床试验
受试群体:28-55岁志愿者170名,其中男性40人,女性130人。随机分成17组,每组10人。接受试验志愿者均做致敏试验,无过敏反应。
试验方法:测量涂抹待测样品前受试群体面部的黑色素含量、白度及水分含量,志愿者每天早晚各使用一次,早晚洁面后涂抹对应各组的待测样品,全脸涂抹,连续使用60天。试验期间不使用其他任何保湿祛斑美白相关的护肤产品,使用60天后检测受试者的面部变化情况,试验结果如表4所示。

如表4所示,使用本发明原料制备的药物组合物,黑色素减少率、白度变化率、水分含量变化率明显高于对比例。由此可知,使用本发明各组分及含量具有相互协同作用,当缺少单一或多种组分时,会对配方协同发生一定情况的失衡,弱化组合物的美白祛斑效果。
前述对本发明的具体示例性实施方案的描述是为了说明和例证的目的。这些描述并非想将本发明限定为所公开的精确形式,并且很显然,根据上述教导,可以进行很多改变和变化。对示例性实施例进行选择和描述的目的在于解释本发明的特定原理及其实际应用,从而使得本领域的技术人员能够实现并利用本发明的各种不同的示例性实施方案以及各种不同的选择和改变。本发明的范围意在由权利要求书及其等同形式所限定。
Claims (10)
1.一种美白祛斑组合物,其特征在于,至少包括以下原料:白术根提取物、白及根提取物、甘草根提取物、白丁香提取物、芍药根提取物和灵芝提取物。
2.根据权利要求1所述的美白祛斑组合物,其特征在于,按重量百分数计,至少包括以下原料:白术根提取物0.1-5%、白及根提取物0.5-6%、甘草根提取物0.5-5%、白丁香提取物0.5-5%、芍药根提取物0.5-5%和灵芝提取物0.1-5%,余量为水。
3.根据权利要求1所述的美白祛斑组合物,其特征在于,按重量百分数计,至少包括以下原料:白术根提取物1-3%、白及根提取物1.5-3%、甘草根提取物1-3%、白丁香提取物1-3%、芍药根提取物1-3%和灵芝提取物1-3%,余量为水。
4.根据权利要求1所述的美白祛斑组合物,其特征在于,按重量百分数计,至少包括以下原料:白术根提取物1.5%、白及根提取物2%、甘草根提取物1.8%、白丁香提取物1.5%、芍药根提取物1.8%和灵芝提取物1.6%,余量为水。
5.根据权利要求1所述的美白祛斑组合物,其特征在于,所述组合物还包括以下原料:1,3丁二醇、对羟基苯乙酮和A91。
6.根据权利要求5所述的美白祛斑组合物,其特征在于,按重量百分数计,所述组合物还包括以下原料:1,3丁二醇1-5%、对羟基苯乙酮0.05-1%和A91 0.01-1%。
7.一种如权利要求1-6任一所述的美白祛斑组合物在制备美白祛斑化妆品中的应用。
8.一种美白祛斑组合物的制备方法,其特征在于,包括以下步骤:
(1)按配比称取制备美白祛斑组合物的原料,原料包括:白术根、白及根、甘草根、白丁香、芍药根、灵芝;然后进行粉碎,得到混合原料粉末;
(2)提纯:将混合原料粉末用水浸泡一段时间,然后在真空恒温的环境下提纯一段时间,得到滤液;反复进行多次提纯,将多次提纯的滤液混合,得到混合滤液;
(3)过滤和浓缩:将混合滤液进行过滤,再进行浓缩,得到提取液,再加入1,3丁二醇、对羟基苯乙酮和A91,得到美白祛斑组合物。
9.根据权利要求8所述的美白祛斑组合物的制备方法,其特征在于,步骤(2)的提纯是在真空度为-0.1MPa温度为45-55℃条件下提纯,反复提纯的次数为2-4次;提纯时,混合原料与水混合的质量比为1:20-60。
10.根据权利要求8所述的美白祛斑组合物的制备方法,其特征在于,步骤(3)的过滤为,将混合滤液用200nm的陶瓷膜过滤,得到纯情液;
步骤(3)的浓缩为,在真空度-0.1MPa温度为45-55℃的条件下浓缩,得到提取液。
