Sodium hyaluronate: sodium hyaluronate, also known as sodium hyaluronate, is an acidic mucopolysaccharide naturally present in the cornea skin and capable of absorbing 1000 times the weight of the water to achieve the effects of keeping the skin moisture, preventing the moisture from losing through the epidermis, and repairing the barrier when the skin is damaged, so that the skin does not feel dry after being used, the luster is increased, the nutrition metabolism of the skin can be improved, the skin is soft, smooth, wrinkle-removing, elasticity-increasing, aging-preventing, and the skin is a good percutaneous absorption promoter while moisture is preserved.
Sphingosine fungus glue: the sphingosine fungus glue is polysaccharide from microbial fermentation sources, has smooth skin feel, is not astringent and sticky, and has thickening and suspending properties.
White pond flower seed oil: the white pond flower seed oil is extracted from white pond flower, is a powerful natural antioxidant, and comprises the following components: 97% long carbon chain fatty acids, 20 or more carbon chains, 3 of which are not found in other vegetable oils. Has good affinity with skin, is easy to be absorbed by skin, is used for skin care products, can promote skin to be tender and elastic, and has good skin moistening and caring effects.
Urea: the urea contains nitrogen which is beneficial to human body, can promote protein synthesis, is a natural humectant in healthy skin, can reduce skin moisture loss, enhance skin moisturizing ability, enhance skin barrier and improve the penetration ability of other components.
Panthenol: the panthenol has small molecules and good permeability, has good hygroscopicity as many water-based moisturizers such as hyaluronic acid, can help to grasp moisture in the air, and can promote the permeation of water molecules to the skin, so that the skin can achieve the purposes of lasting and deep moisturizing. In addition to moisturizing effects, panthenol also stimulates the epithelial cells of the skin, promoting wound healing and relieving skin inflammation.
Lecithin: lecithin is a basic component of Natural Moisturizing Factor (NMF) of skin, and can bind moisture in skin, prevent skin from dehydration and dryness, and thus ensure skin softness, smoothness and health. Meanwhile, the phospholipid can effectively repair the damaged sebum membrane on the surface of the skin and promote the absorption of the skin.

Based on the embodiments of the present application, any changes to the present application without making any creative changes to the present application fall within the protection scope of the present application. Meanwhile, in the examples of the present application, all the preparation materials are commercially available products well known to those skilled in the art unless otherwise specified. Wherein in each embodiment, the herba Lycopi extract is prepared by drying herba Lycopi, pulverizing, sieving with 100 mesh sieve, heating and refluxing with 90% ethanol of 6 times of herba Lycopi mass, concentrating, and drying; the herb extract is prepared by drying herb, crushing, sieving with 100 mesh sieve, heating and refluxing with 95% ethanol with the mass of 8 times of herb, concentrating and drying; the pummelo peel extract is prepared by drying pummelo peel, crushing, sieving with a 100-mesh sieve, heating and refluxing an extracting solution with 70% ethanol with the mass of 5 times of the pummelo peel, concentrating and drying.
Example 1
The microcirculation-improving whitening composition comprises the following components in parts by mass: 5 parts of herba lycopi extract, 4 parts of pummelo peel extract and 2 parts of comfrey extract.
The microcirculation-improving whitening essence comprises the following components in percentage by mass: 0.25% of sphingosine bacterial gum, 0.05% of EDTA-2Na, 0.6% of p-hydroxyacetophenone, 5% of dipropylene glycol, 0.4% of 1, 2-hexanediol, 2% of urea, 0.1% of sodium hyaluronate, 0.4% of white pond flower seed oil, 0.1% of lecithin, 1.2% of panthenol, 5% of herba lycopi extract, 4% of exocarpium citri rubrum extract, 2% of comfrey extract and the balance of water.
The preparation method of the essence for improving microcirculation and whitening comprises the following steps:
(1) Stirring and heating water, dipropylene glycol, sphingosine fungus glue, EDTA-2Na, 1, 2-hexanediol, urea and sodium hyaluronate to 85 ℃, and keeping the temperature and continuing stirring until the components are completely and uniformly dissolved to obtain liquid 1;
(2) Stirring and heating the white pond flower seed oil to 75 ℃ to obtain liquid 2;
(3) Maintaining the temperature of the water phase at 885 ℃, starting stirring at 400r/min, pouring the oil phase into the water phase solution, homogenizing for 8min at 7000r/min after mixing, and stirring and cooling to obtain liquid 3.
(4) Maintaining the stirring speed at 220r/min, and adding p-hydroxyacetophenone into liquid 3 to obtain liquid 4 when the temperature is reduced to 65deg.C.
(5) Maintaining stirring speed at 220r/min, and adding lecithin, panthenol, herba Lycopi extract, exocarpium Citri Grandis extract and herba Agrimoniae extract into the liquid 4 when the temperature is reduced to 45deg.C to obtain the microcirculation improving whitening essence.
Example 2
The microcirculation-improving whitening composition comprises the following components in parts by mass: 3 parts of herba lycopi extract, 2 parts of pummelo peel extract and 6 parts of comfrey extract.
The microcirculation-improving whitening essence comprises the following components in percentage by mass: 0.3% of sphingosine bacterial gum, 0.03% of EDTA-2Na, 0.5% of p-hydroxyacetophenone, 4% of dipropylene glycol, 0.5% of 1, 2-hexanediol, 0.5% of urea, 0.15% of sodium hyaluronate, 0.6% of white pond flower seed oil, 0.3% of lecithin, 0.5% of panthenol, 3% of herba lycopi extract, 2% of exocarpium Citri Grandis extract, 6% of comfrey extract and the balance of water.
The preparation method of the essence for improving microcirculation and whitening comprises the following steps:
(1) Stirring and heating water, dipropylene glycol, sphingosine fungus glue, EDTA-2Na, 1, 2-hexanediol, urea and sodium hyaluronate to 80 ℃, and keeping the temperature and continuing stirring until the components are completely and uniformly dissolved to obtain liquid 1;
(2) Stirring and heating the white pond flower seed oil to 78 ℃ to obtain liquid 2;
(3) Maintaining the temperature of the water phase at 80deg.C, starting stirring at 260r/min, pouring the oil phase into the water phase solution, homogenizing at 5000r/min for 10min after mixing, and stirring and cooling to obtain liquid 3.
(4) Maintaining the stirring speed at 150r/min, and adding p-hydroxyacetophenone into liquid 3 to obtain liquid 4 when the temperature is reduced to 70deg.C.
(5) Maintaining stirring speed at 150r/min, and adding lecithin, panthenol, herba Lycopi extract, exocarpium Citri Grandis extract and herba Agrimoniae extract into the liquid 4 when the temperature is reduced to 40deg.C to obtain the microcirculation improving whitening essence.
Example 3
The microcirculation-improving whitening composition comprises the following components in parts by mass: 2 parts of herba lycopi extract, 6 parts of pummelo peel extract and 8 parts of comfrey extract.
The microcirculation-improving whitening essence comprises the following components in percentage by mass: 0.2% of sphingosine bacterial gum, 0.07% of EDTA-2Na, 0.3% of p-hydroxyacetophenone, 6.5% of dipropylene glycol, 0.8% of 1, 2-hexanediol, 2.5% of urea, 0.11% of sodium hyaluronate, 0.55% of white pond flower seed oil, 0.16% of lecithin, 0.6% of panthenol, 2% of herba lycopi extract, 6% of exocarpium Citri Grandis extract, 8% of comfrey extract and the balance of water.
The preparation method of the essence for improving microcirculation of the embodiment comprises the following steps:
(1) Stirring and heating water, dipropylene glycol, sphingosine fungus glue, EDTA-2Na, 1, 2-hexanediol, urea and sodium hyaluronate to 82 ℃, and keeping the temperature and continuing stirring until the components are completely and uniformly dissolved to obtain liquid 1;
(2) Stirring and heating the white pond flower seed oil to 72 ℃ to obtain liquid 2;
(3) Maintaining the temperature of the water phase at 82 ℃, starting stirring at 260r/min, pouring the oil phase into the water phase solution, homogenizing at 8000r/min after mixing, stirring and cooling to obtain liquid 3.
(4) Maintaining the stirring speed at 160r/min, and adding p-hydroxyacetophenone into liquid 3 to obtain liquid 4 when the temperature is reduced to 68 ℃.
(5) Maintaining stirring rotation speed at 160r/min, and adding lecithin, panthenol, herba Lycopi extract, exocarpium Citri Grandis extract and herba Agrimoniae extract into the liquid 4 when the temperature is reduced to 46 deg.C to obtain the microcirculation improving whitening essence.

The microcirculation improving whitening composition or the microcirculation improving whitening essence of this comparative example is different from that of example 1 in that: the microcirculation improving whitening composition or the microcirculation improving whitening essence of the comparative example does not contain herba lycopi extract and exocarpium citri grandis extract.

the herb extract is prepared by drying herb, crushing, sieving with a 100-mesh sieve, heating and refluxing an extracting solution with 75% ethanol of which the mass is 8 times that of the herb, concentrating and drying;
the pummelo peel extract is prepared by drying pummelo peel, crushing, sieving with a 100-mesh sieve, heating and refluxing an extract with 80% ethanol with the mass of 5 times of pummelo peel, concentrating and drying.
Experimental example 1 Effect of different test samples on AQP-1 expression
Endothelial cells were cultured in 12-well plates, each containing a polylysine-treated slide, and treated with HMGB1 or HMGB1 (100 ng/ml) and example 1, comparative example 2, comparative example 3, comparative example 4, comparative example 5, comparative example 6, comparative example 7, comparative example 8 (30 μg/ml) or controls, followed by fixation with 4% paraformaldehyde for 15 minutes, removal of the growing endothelial cells, and rinsing with PBS for 20 minutes at room temperature. After washing 3 times with PBS, cells were blocked with 5% BSA in PBS for 30min at room temperature and then incubated overnight with primary anti-AQP 1 antibody (1:200) at4 ℃. Endothelial cells were then washed twice with PBS (pH 7.4) and then exposed to FITC-labeled IgG secondary antibody (1:200) for 30min at room temperature in the dark. Nuclei were stained with DAPI for 1 min.
Western blot analysis determines AQP-1 expression: endothelial cells were washed with ice-cold PBS, then lysed with PMSF-containing RIPA lysis buffer (1:100) for 30min on ice, and then centrifuged at 12000g for 15min. Protein concentration was then determined using the diminutive acid (BCA) protein assay kit. The lysate was then heated in protein sample loading buffer at 95 ℃ for 10 minutes, and then all samples were kept at-20 ℃. Proteins (10 μl) were separated on 12.5% SDS-PAGE and transferred to PVDF membrane (on. After vigorous washing, membrane was incubated with HRP-conjugated secondary antibody (1:500) at room temperature for 1 hour at room temperature after washing three times with TBST, the bands were visualized using Omni ECL Femto-Light chemiluminescent kit and quantified using GE Amersham Imager 600 (USA), expression levels normalized to β -actin. Each of the above experiments were repeated at least 3 times.
AQP-1 (aquaporin-1) promotes the penetration of endothelial cells into water, and its overexpression induces increased vascular permeability. Thus, inhibition of AQP-1 overexpression can inhibit increased vascular permeability. As shown in the above figure 1, when the three extracts of herba Lycopi, exocarpium Citri Grandis and herba Agrimoniae are compounded, the expression of AQP-1 is significantly inhibited, so that the increase of vascular permeability is inhibited, and the inhibition effect is obviously superior to that of single use or two-by-two compounded use, and the compounded use of the three has obvious synergistic effect.
Experimental example 2 Effect of different test samples on blood flow velocity of blood vessels between zebra fish nodes
30 zebra fish were randomly selected in 6-well plates with 15 tails per well. Example 1, comparative example 2, comparative example 3, comparative example 4, comparative example 5, comparative example 6, and each well was fixed with 3mL of volume, and a normal control group was set. Then, each experimental group was incubated at 28℃for 24 hours in the absence of light. Finally, 10 zebra fish are randomly selected from each experimental group, blood flow videos of blood vessels among zebra fish nodes are recorded under a heart beat blood flow analysis system, and blood flow speeds (V) of the blood vessels among the zebra fish nodes are analyzed.
The sample improving microcirculation efficacy is calculated according to the following formula:
as can be seen from fig. 2-4, the effect of improving microcirculation when the three extracts are compounded is significantly better than that of single use or two-by-two compounding.
Experimental example 3 tyrosinase inhibition assay
Under the action of tyrosinase, L-tyrosine generates dopa, dopa generates dopaquinone under the further action of enzyme, and finally melanin is generated through a series of reactions. Wherein the dopaquinone has a maximum absorption peak at 475nm wavelength, and the absorbance is measured in unit time, so that the enzyme activity in the reaction system is catalyzed by the indirect reaction enzyme. I.e., an increase of 0.001 in absorbance at 475nm wavelength per minute of dopa-wakeup is defined as an enzyme activity unit. Accurately sucking the reaction solutions of the L-tyrosine, PBS and the sample in the T1, T2, T3 and T4 respectively according to the surface volumes by using a micropipette, respectively placing the reaction solutions in 4 PE pipes, uniformly mixing, keeping the temperature AT 37 ℃ for L0min, then respectively adding 1mL of tyrosinase in the T2 and T4, reacting the tyrosinase with the lOmin, and rapidly measuring the absorbance ATl, AT2, AT3 and AT4 AT 475nm by using an ultraviolet spectrophotometer. The tyrosinase inhibitory activity of the samples was calculated according to the following formula:

Experimental example 4 evaluation of skin melanin and skin gloss of improving microcirculation whitening essence
The testing method comprises the following steps: the sample (example 1 improved microcirculatory whitening essence) was used twice daily, in the morning and evening, and was tried for 4 weeks. A proper amount of test sample was smeared on one side of the random pattern, and the test sample was not used as a control on the other side of the random pattern. The sunscreen product is applied to the whole face daily in the morning. The results were tested in the following 4 time periods, respectively: before the sample (D0), after 14 days of the sample (D14), and after 28 days of the sample (D28).
Detecting items:
1) The skin melanin determinator MX18 is used for detecting skin melanin;
2) The skin color analyzer CL200 is used to detect skin glossiness.
Test results:
1) Skin melanin
The subject was tested for target site melanin 28 days after the sample application (the measured value was reduced, indicating that the skin melanin content was less), the skin melanin value was significantly increased by 1.46% in the control area, while the area melanin was reduced by only 0.17% in the sample application. The prepared essence for improving microcirculation and whitening skin can effectively reduce the generation of skin melanin.

The skin glossiness of the target part of the subject is tested 28 days after the sample is used (the measured value is increased, which means that the skin glossiness is better), the skin glossiness value of the control area is reduced by 2.42%, the skin glossiness of the sample area is increased by 1.99%, and the prepared microcirculation improving whitening essence can improve the skin glossiness.

Experimental example 5Visia-CR facial ultraviolet stain analysis
A healthy female subject in China is selected with an age ranging from 30 to 40 years old. The facial photographs of the subjects were taken with a visual-CR before the subjects used the microcirculation whitening essence (D0), and the products were continuously used for 4 weeks (D28). And the larger the ultraviolet color spot area is analyzed by the image analysis of the ultraviolet color spot picture acquired and derived by the VISIA-CR, the more the skin potential pigment spots are. The subjects who had the improved microcirculatory whitening essence of example 1 applied the whitening essence after 28 days had significantly reduced and reduced uv spots, and the results are shown in figure 6.
It should be noted that the above examples are only for illustrating the technical solution of the present application and are not limiting thereof. Although the present application has been described in detail with reference to the examples given, those skilled in the art can make modifications and equivalents to the technical solutions of the present application as required, without departing from the spirit and scope of the technical solutions of the present application.

Claims (9)

1. The microcirculation-improving whitening essence is characterized by comprising the following components in percentage by mass: 0.1 to 1 percent of thickening agent, 0.01 to 0.05 percent of EDTA-2Na, 0.1 to 1 percent of preservative, 4 to 8 percent of polyalcohol, 0.5 to 5 percent of humectant, 0.1 to 1 percent of natural grease, 0.01 to 1 percent of lecithin, 0.5 to 3 percent of panthenol, 1.5 to 8 percent of herba lycopi extract, 1 to 7 percent of pummelo peel extract, 0.5 to 10 percent of comfrey extract and the balance of water;
the herba lycopi extract is obtained by concentrating and drying an extracting solution prepared by heating and refluxing pretreated herba lycopi with ethanol; the pretreatment comprises the steps of drying, crushing and sieving the herba lycopi; the mass ratio of the pretreated herba lycopi to the ethanol is 1:6; the ethanol is 90% ethanol;
the pummelo peel extract is obtained by concentrating and drying an extracting solution prepared by heating and refluxing pretreated pummelo peel with ethanol; the pretreatment comprises the steps of drying, crushing and sieving exocarpium Citri Grandis; the mass ratio of the pretreated pummelo peel to the ethanol is 1:5; the ethanol is 70% ethanol;
the extract of the polymeric grass is obtained by concentrating and drying an extracting solution prepared by heating and refluxing the pretreated polymeric grass whole grass by ethanol; the pretreatment comprises the steps of drying, crushing and sieving the comfrey; the mass ratio of the pretreated comfrey to the ethanol is 1:8; the ethanol is 95% ethanol.
2. The improved microcirculatory whitening essence according to claim 1, wherein the mesh size of each of herba lycopi, exocarpium Citri Grandis and comfrey is 100 mesh.
3. The improved microcirculatory whitening essence according to claim 1, wherein the thickener comprises sphingosine gel;
the preservative comprises p-hydroxyacetophenone;
the polyalcohol is any one or more of dipropylene glycol, glycerol, butanediol and 1, 2-hexanediol;
the humectant comprises one or more of sodium hyaluronate, urea and betaine;
the natural oil comprises one or more of linseed oil and white pool flower seed oil.
4. The improved microcirculatory whitening essence according to claim 3, wherein the polyalcohol is dipropylene glycol and 1, 2-hexanediol;
the humectant is sodium hyaluronate and urea;
the natural oil is white pool flower seed oil.
5. The improved microcirculation whitening essence as claimed in claim 1, which is characterized by comprising the following components in percentage by mass: 0.2 to 0.8 percent of sphingosine bacterial gel, 0.03 to 0.07 percent of EDTA-2Na, 0.3 to 0.8 percent of p-hydroxyacetophenone, 3.5 to 6.5 percent of dipropylene glycol, 0.2 to 0.8 percent of 1, 2-hexanediol, 0.5 to 2.5 percent of urea, 0.08 to 0.3 percent of sodium hyaluronate, 0.25 to 0.8 percent of white pond flower seed oil, 0.02 to 0.8 percent of lecithin, 0.5 to 1.5 percent of panthenol, 3 to 6 percent of herba lycopi extract, 1.5 to 5.5 percent of exocarpium citri grandis extract, 1 to 7 percent of polymeric grass extract and the balance of water.
6. The method for preparing the microcirculation-improving whitening essence as set forth in claim 5, which is characterized by comprising the following steps:
s1, stirring and heating water, dipropylene glycol, sphingosine fungus glue, EDTA-2Na, 1, 2-hexanediol, urea and sodium hyaluronate, and then keeping warm and continuously stirring until the components are completely and uniformly dissolved to obtain liquid I;
s2, stirring and heating the white pond flower seed oil to obtain liquid II;
s3, maintaining the temperature of the water phase at a high temperature state, starting stirring, pouring the oil phase into the water phase solution, homogenizing after mixing, and stirring and cooling to obtain liquid III;
s4, continuing stirring, and adding the p-hydroxyacetophenone into the liquid III when the temperature is reduced to obtain a liquid IV;
and S5, maintaining the stirring rotation speed, and adding lecithin, panthenol, herba lycopi extract, exocarpium citri rubrum extract and comfrey extract into the liquid IV when the temperature is further reduced, so as to obtain the microcirculation improving and whitening essence.
7. The method of claim 6, wherein steps S1 and S2 are not sequentially separated;
in the step S1, the stirring and heating temperature is controlled to be 80-85 ℃;
in the step S2, the stirring and heating temperature is controlled to be 75-80 ℃;
in the step S3, the temperature of the water phase is kept at 80-85 ℃, the stirring rotation speed is started to be kept at 200-400 r/min, the oil phase is poured into the water phase solution, after the mixing is finished, the homogenization is carried out for 5-10 min at 5000-8000 r/min, and then the stirring and the cooling are carried out;
in the step S4, stirring is continued, the stirring rotation speed is kept at 100-250 r/min, and p-hydroxyacetophenone is added into the liquid III when the temperature is reduced to 65-70 ℃;
in the step S5, the stirring rotation speed is kept at 100-250 r/min, and when the temperature is reduced to 40-48 ℃, lecithin, panthenol, herba lycopi extract, exocarpium citri rubrum extract and comfrey extract are added into the liquid IV.
8. Use of the concentrate according to any one of claims 1 to 5 for the preparation of a whitening product for improving microcirculation; the product is a cosmetic.
9. The use according to claim 8, wherein the cosmetic product comprises a facial cleanser, a facial mask, a face cream and a sun cream.
