Description

Preparation method and application of environment-friendly seed coating agent
Technical Field
The invention relates to the technical field of pesticides, and particularly relates to a preparation method and application of an environment-friendly seed coating agent.
Background
The seeds are the basis of agricultural planting production, and the sowing quality of the seeds is an important factor related to the agricultural production efficiency, so that the research on the seed protection technology is essential. The problem that the germination of seeds is easily affected by pathogenic bacteria carried by the seeds or plant diseases and insect pests in soil needs to be solved, and excessive putting of pesticides and chemical fertilizers in production can cause a large amount of accumulation, so that the problem of environmental pollution can be caused and the activity of the seeds can be affected. Therefore, the seeds are effectively treated in the early stage of sowing so as to control the occurrence of plant diseases and insect pests and improve the germination condition of the seeds and the growth condition of the seeds in the seedling period, thereby ensuring the yield of crops, solving the problem of excessive application, saving the cost and improving the economic benefit, and being a necessary way for agricultural modernization.
The seed coating is a process of treating seeds with a medicament having film-forming properties to form a film having a certain function on the surface of the seeds. The agents used for coating seeds are called seed coatings. The seed coating agent is generally prepared by mixing and processing active substances (pesticide stock solution, biological agent and the like), a dispersing agent, a film forming agent, a binding agent, a dispersing agent, a preservative, warning color and other components. Used directly or diluted for seed treatment. After the seeds are coated and sown, a protective barrier can be formed around the seeds, and the damage of the seeds by plant diseases and insect pests in soil is reduced. The coated seeds absorb water in the soil and then swell slightly, but the coating agent does not dissolve. The effective components of the seed coating agent are gradually absorbed by the plant root system in the seed germination and growth processes, are conducted to the parts of the plant such as stems, leaves and the like through the inside of plant tissues, and simultaneously have the effect of improving the microenvironment of root soil, so that the resistance of the plant to pest and disease infection in the germination stage and the seedling stage is enhanced, and simultaneously, the seed coating agent has the effects of promoting the plant growth and increasing the yield of crops. The seed coating technology has the advantage of simple and convenient operation and is widely applied to preventing and controlling crop seedling diseases and insect pests.
The biological seed coating agent is an environment-friendly preparation, and the active ingredients of the biological seed coating agent mainly take substances with antagonistic microbe effects, such as natural products, microbe living bodies, microbe metabolites and the like as main bodies, and then the biological seed coating agent is processed by adding an auxiliary agent. In the biological seed coating agent, the active substance generally has longer survival time and unchanged activity, and effectively controls plant diseases and insect pests through antagonism or competition mechanism with the diseases and insect pests. Compared with the chemical seed coating agent, the biological seed coating agent has the following advantages: firstly, after the biological seed coating agent is applied, the microenvironment of root soil can be improved, and the fusion degree of seeds and soil is improved; secondly, most of the biological seed coating agents have lower production cost and better effect; thirdly, the long-term use of chemical agents can cause the problem of '3R', and the biological seed coating agent can avoid the problems; fourthly, the biological seed coating agent is safe to people and livestock, is environment-friendly, can be applied for a long time, and is not easy to cause the problem of drug resistance of crops. The biological pesticide has the advantages of high safety to human, livestock and environment, no residue and the like, and is a hot problem in current research.

the thickening agent is xanthan gum;
the antifreezing agent is propylene glycol;
the warning agent is acid bright red.
The preparation method comprises the following steps: the same as preparation 1.
Preparing a bacillus licheniformis fermentation liquid: the bacillus licheniformis is inoculated in a liquid culture medium (2 percent of corn flour, 1.0 percent of peptone, 0.5 percent of yeast powder, 2.0 percent of sucrose, 2.0 percent of sodium chloride, 0.02 percent of magnesium sulfate and 0.05 percent of manganese sulfate) for fermentation preparation.
Example 3: control effect of different seed coating agents on wheat sharp eyespot
And (3) testing crop diseases: sheath blight of wheat
The test method comprises the following steps: reference GB/T17980.108-2004 pesticide field test guidelines (second) part 108: the bactericide can be used for preventing and treating wheat sharp eyespot. The field plot experiment is adopted, the plot area is 50m2, the arrangement is carried out by blocks at any time, and the operation is repeated for four times. Soaking seeds before sowing the wheat (seed coating agent is diluted by 100 times: 1L seed coating agent, water is added for 99L, the seeds are placed in the diluent to be stirred and soaked for 4h, and the seeds can be sowed after being fished out and dried); disease investigation is carried out in the jointing stage, five points of diagonal lines of each cell are sampled, 100 plants are investigated at each point, and disease grading is carried out:
level 0: the disease is not developed;
level 1: the leaf sheath is diseased but the stem is not diseased;
and 3, level: the leaf sheath is diseased and invades the stem, but the stalk lesion has insufficient ring stem 1/2;
and 5, stage: the stigma canephora exceeds 1/2, but does not fall or break;
and 7, stage: withering, lodging and white ear.
The disease index [ ∑ (number of diseased plants at each level × relative level value)/(number of total investigated plants × 7] × 100;
the prevention and treatment effect is [ (control disease index-treatment disease index)/control disease index ] × 100%.
And (3) testing results: as can be seen from Table 1, when the Trichoderma harzianum fermentation broth is used in combination with three or more components of plant growth regulators such as humic acid, alginic acid, S-abscisic acid or chitosan, the control effect of the seed coating agent on wheat sharp eyespot can be remarkably improved. In formulas 1-3, when the trichoderma harzianum fermentation broth and the plant growth regulator (humic acid, alginic acid, S-abscisic acid and chitosan) are used in combination, the control effects on wheat sharp eyespot are 90.2%, 85.5% and 81.6% respectively. Compared with the formula 4-10, the formula 1 has the advantage that the composition of the plant growth regulator has obvious influence on the control effect of the wheat sharp eyespot. In addition, the combination of the trichoderma harzianum fermentation liquor and the bacillus subtilis fermentation liquor can improve the control effect of the seed coating agent on wheat sharp eyespot.

Example 3 prevention and treatment Effect of different seed coating agents on maize head smut
And (3) testing crop diseases: head smut of maize
The test method comprises the following steps: reference GB/T17980.106-2004 PestIcIde field test guidelines (second) part 106: the bactericide can be used for preventing and treating maize head smut. The field plot experiment is adopted, the plot area is 60m2, the arrangement is carried out by blocks at any time, and the process is repeated for four times. Soaking seeds before sowing the corns (the seed coating agent is diluted by 150 times and added with 149L of water in 1L of the seed coating agent; the seeds are put into the diluent to be stirred and soaked for 6 hours, and the seeds can be sowed after being fished out and dried); disease investigation is carried out when symptoms are obvious after the fruit clusters are full, investigation is carried out on all the plants except the side rows in each cell, the total number of plants and the number of diseased plants are recorded, and the disease rate and the control effect are calculated
The disease rate is the number of diseased plants/the total number of investigated plants x 100%;
the control effect is [ (control plant disease rate-treatment plant disease rate)/control plant disease rate ] × 100%.
And (3) testing results: as can be seen from Table 2, when the Trichoderma harzianum fermentation broth is used in combination with three or more of humic acid, alginic acid, S-abscisic acid or chitosan, the prevention and treatment effect of the seed coating agent on the maize head smut can be significantly improved. In formulas 1-3, when the trichoderma harzianum fermentation broth and the plant growth regulator combination (humic acid, alginic acid, S-abscisic acid and chitosan) are used in combination, the control effects on the maize head smut are 89.2%, 86.0% and 84.6% respectively. Compared with the formula 4-10, the formula 1 has the advantage that the composition of the plant growth regulator has obvious influence on the prevention and treatment effect of the maize head smut. In addition, the combination of the trichoderma harzianum fermentation liquor and the bacillus subtilis fermentation liquor can improve the control effect of the seed coating agent on the maize head smut.
