Abstract
本发明涉及一种公共场所应用喷雾复合消毒剂的制备方法,其特征在于,制备所述复合消毒剂的原料包括季铵盐、纳米银、聚合助剂、抗冻助剂、保湿助剂、惰性助剂和水,本发明通过改进,利用聚合助剂,赋予季铵盐以聚合能力,消毒剂不仅可以形成纳米笼固化到物体表面,同时将纳米银包覆在纳米网中,实现纳米银的缓慢释放。同时加入抗冻高分子聚合物,可实现超低温下显著抗菌消毒。原料来源广泛,性能稳定、超广谱杀菌、持久抗菌、无毒无腐蚀、工艺简单、成本低廉、应用范围广,特别适用于人工、消毒机、消毒机器人在常低温下的杀菌消毒,生物相容性好,对环境无二次污染。

技术领域
本发明涉及一种公共场所应用喷雾复合消毒剂的制备方法,特别涉及一种杀菌抑菌能力强、灭菌种类多、效果持久、工艺简单、成本低廉的既适用于人工喷洒又适用于消毒机地、消毒机器人的、无毒无异味地、皮肤无刺激的、环境友好型的、同时可在超低温(-20℃)作用下实现抗菌的消毒剂的制备方法。
背景技术
目前,常用的杀菌消毒方法为消毒剂喷洒、熏蒸等化学方法和紫外线照射、臭氧等物理方法,化学方法采用的药剂(次氯酸类、84类、酒精类及三氯生类)大多有刺鼻气味,对呼吸道和皮肤有剌激损伤,无论是吸附在物体的表面还是散发在空气中,都会对环境造成二次污染,轻者引起过敏性反应或诱发皮炎等现象产生,重者吸入肺部侵蚀肌肤,对人体健康造成威胁。如乳酸、过氧乙酸腐蚀性强,剌激人体呼吸道,84消毒液空气浓度较高时,对人体皮肤有较强的剌激损伤。而物理方法紫外线照射受灯管强度、洁净度、环境温湿度、遮挡物和距离影响,并且可以引起紫外线眼炎等副反应。同时,冷链消杀也面临巨大挑战。常规消毒剂无法在零下20℃的超低温环境下发挥作用。综上,目前的消毒剂存在着诸多缺点,如具有刺激性气味,杀菌能力弱、杀菌谱较窄、二次污染等。因此,研发一种抑菌、杀菌能力强,无毒无害,无刺激性气味,绿色环保的消毒剂是一项重大而艰巨的任务。
虽然已经提交的“一种纳米银/季铵盐复合消毒剂的制备方法”(CN201810170166.6)可解决上述问题,但是由于缺少缓释技术、物表聚合技术和抗冻技术,在一定程度上不能超水平发挥复合消毒剂的抗菌作用,易产生银离子初始过多释放,也无法在超低温下发挥消杀作用。本发明通过改进,利用聚合助剂,赋予季铵盐以聚合能力,消毒剂不仅可以形成纳米笼固化到物体表面,同时将纳米银包覆在纳米网中,实现纳米银的缓慢释放。同时加入抗冻高分子聚合物,可实现超低温下显著抗菌消毒。

发明内容
一、季铵盐在抗菌同时可在物表聚合形成纳米网,成功对纳米银实现类纳米笼包覆,极大降低了外部环境对内部纳米银的侵蚀,同时也延缓了生成的Ag+向外扩散。
二、季铵盐和银离子地双重超高正电,可对细菌和病毒表面有很强的亲和吸附力,快速破坏其包膜,进一步攻击摧毁细菌和病毒体内呼吸和代谢系统。
三、季铵盐可在物表完成快速聚合,喷洒后可牢固地固定在物体表面,保证抗菌性能更稳定、抗菌时间更持久。
四、采用抗冻高分子聚合物,可在-20℃下实现高效抗菌。
五、生物相容性好,环境无污染,无腐蚀无异味,工艺简单,成本低廉。
本发明采取以下技术方案:1、一种公共场所应用喷雾复合消毒剂的制备方法,其特征在于,制备所述复合消毒剂的原料包括季铵盐、纳米银、聚合助剂、抗冻助剂、保湿助剂、惰性助剂和水,各原料的重量份数比为季铵盐0.1~4,纳米银,0.05~5,聚合助剂0.05~20,抗冻助剂5~500,保湿助剂0.5~18,惰性助剂0.2~30,水补齐至1000,包括以下步骤:
步骤一:将季铵盐分散在溶剂中,得到溶液A;
步骤二:将纳米银加入到溶剂中快速搅拌得到溶液B;
步骤三:溶液A和溶液B混合搅拌得到溶液C;
步骤四:将溶液C与适量聚合助剂混合,得到溶液D;
步骤五:在溶液D中加入抗冻助剂,保湿助剂,惰性助剂,并加入适量水,得到性能稳定的复合消毒剂。本发明具有以下优点:原料来源广泛,性能稳定、超广谱杀菌、持久抗菌、无毒无腐蚀、工艺简单、成本低廉、应用范围广,特别适用于人工、消毒机、消毒机器人在常低温下的杀菌消毒,生物相容性好,对环境无二次污染。
所述的制备方法,其特征在于,制备溶液A和溶液B所使用的溶剂为水、乙醇、乙醚、丙酮、DMF、冰醋酸、氯仿、四氯化碳、甲苯、乙二醇、丙二醇等实验常用溶剂中的一种或几种溶剂的混合液。
所述的制备方法,其特征在于,制备溶液A和溶液B所用的溶剂水可为自来水、泉水、井水、引用纯净水、去离子水中的一种或者几种水的混合液,保证溶剂水中不含阻止实验成功的物质即可。
所述的制备方法,其特征在于,制备溶液C时,所用的季铵盐,可以是单链季铵盐如,十二烷基二甲基苄基氯化铵(苯扎氯铵)、十二烷基二甲基苯氧乙基溴化铵、十四烷基二甲基吡啶溴化铵,苯扎溴胺,十六烷基三甲基溴化铵,十六烷基三甲基氯化铵;双链季铵盐如,双癸基二甲基氯化铵、双十二烷基二甲基氯化铵,双十二烷基二甲基溴化铵,溴化(双十二烷基二甲基)乙撑二铵;聚季铵盐如,聚硅季铵盐、聚六亚甲基双胍,聚六亚甲基单胍中的一种或几种。
所述的制备方法,其特征在于,所用的纳米银粒子大小为5nm-50nm。
所述的制备方法,其特征在于,所用的聚合助剂为酯基或酰胺基,或带有酯基或酰胺基的物质,如氯化十六烷基吡啶、2-(甲基丙烯酰氧乙基)-正十二烷基-甲基溴化铵(MAE-DB)、2-(甲基丙烯酰氧乙基)-正十六烷基-甲基溴化铵(MAE-HB)等。
所述的制备方法,其特征在于,所用的抗冻助剂为为氯化钙,氯化钠,亚硝酸钠,氯化钙和氯化钠的混合物,氯化钙和亚硝酸钠的混合物,乙醇,乙二醇、异丙醇、甘油,二醇醚,1 ,2丙二醇,1 ,3丙二醇,聚乙二醇200,聚乙二醇400聚丙烯酸钠,聚乙烯吡咯烷酮, N甲基吡咯烷酮,N乙基吡咯烷酮,二甲亚砜,硝酸盐类,N-甲氧基-N-甲基丁醇等有机物中的一种或者几种。
所述的制备方法,其特征在于,所用的保湿助剂为山梨糖醇、木糖醇、尿素、聚丙二醇、1,2-丙二醇、丙三醇其中的一种或几种的混合物。
所述的制备方法,其特征在于,所用的惰性助剂为N ,N-二(3-氨基丙基)十二烷基胺、N-(3-氨基丙基)-N-十二烷基-丙二胺、十二烷基二甲基氧化胺其中的一种或几种的混合物。所述的制备方法,其特征在于,所用的纳米银可自行制备生产,也可购买成品。
所述的制备方法,其特征在于,所用的季铵盐可自行制备生产,也可购买成品。所述的制备方法,其特征在于,所用的溶剂可自行制备生产,也可购买成品。

具体实施方式
为了使本领域的技术人员可以更好地理解本发明,下面结合实施例对本发明技术方案进一步说明。
实施例一:
步骤一:将2 g苯扎氯氨加入到100g1 ,2丙二醇中,得到溶液A;
步骤二:将0.5g纳米银加入到100g1 ,2丙二醇中,得到溶液B;
步骤三:溶液A和溶液B混合搅拌得到溶液C;
步骤四:将溶液C与2g氯化十六烷基吡啶混合,得到溶液D;
步骤五:在溶液D中加入350g聚乙二醇200,10g十二烷基二甲基氧化胺,并加入437.9g水得到性能稳定的复合消毒剂。
实施例二:
步骤一:将2 g苯扎溴铵加入到150g乙二醇中,得到溶液A;
步骤二:将0.6g纳米银加入到80g1 ,2丙二醇中,得到溶液B;
步骤三:溶液A和溶液B混合搅拌得到溶液C;
步骤四:将溶液C与2g 2-(甲基丙烯酰氧乙基)-正十二烷基-甲基溴化铵(MAE-DB)混合,得到溶液D;
步骤五:在溶液D中加入300g聚乙二醇400和200g聚乙二醇200,10g N-(3-氨基丙基)-N-十二烷基-丙二胺,并加入257.9g水得到性能稳定的复合消毒剂。
实施例三:
步骤一:将2 g苯扎溴铵加入到200g1 ,2丙二醇中,得到溶液A;
步骤二:将0.4g纳米银加入到100g乙二醇中,得到溶液B;
步骤三:溶液A和溶液B混合搅拌得到溶液C;
步骤四:将溶液C与2g 2-(甲基丙烯酰氧乙基)-正十六烷基-甲基溴化铵(MAE-HB)混合,得到溶液D;
步骤五:在溶液D中加入500g聚乙二醇400,10g N ,N-二(3-氨基丙基)十二烷基胺,并加入257.9g水得到性能稳定的复合消毒剂。
应当说明的是,各个组分可以选择发明内容列举的多种化合物或者其组合,至于使用集中化合物时,各个化合物的具体含量,是本领域技术人员根据使用情况和制备环境进行选择和调整的。往往为了制备方便,选取其中一种或者两种进行组合,上述实施例仅仅是结合申请人实验进行列举说明。
上述实施例方式不应理解成对本发明的保护范围的限制,凡是不背离本发明构思的改变或等同替代均包括在本发明的保护范围内。
本发明所得的成品低温消毒材料进行如下检测:
1、悬液法中和剂鉴定试验
检测依据《消毒技术规范》第2002版第2 .1 .1 .5 .5,判断所选中和剂能否完全中和消毒成分,试验分组如下:
第1组消毒剂 + 菌悬液 →培养
观察消毒剂对试验菌有无杀灭或抑制能力。
第2组(消毒剂 + 菌悬液)+ 中和剂 → 培养
观察残留消毒剂被中和后受到清毒剂作用后的试验菌是否能恢复生长。
第3组中和剂 + 菌悬液 → 培养
观察中和剂是否抑菌。
第4组(消毒剂 + 中和剂)+ 菌液 → 培养
观察中和产物,或未被完全中和的残留消毒剂对试验菌的生长繁殖是否有影响。
第5组稀释液 + 菌悬液 → 培养
作为菌数对照。
第6组稀释液 + 中和剂 + 培养基 → 培养
作为阴性对照。
2、悬液定量杀菌试验检测
依据《消毒技术规范》第2002版第2 .1 .1 .7 .4,以实施例1和实施例2原液进行测试,消毒作用时间分别为1min、5min、10min、30min,中和作用10min,在-20℃条件下,以悬液定量杀灭试验中,对金黄色葡萄球菌杀灭对数值均≥5 .00,完全可以在低温下实现高效杀菌的预期效果。
研究结果
2 .1 中和剂鉴定试验
试验浓度为有实施例1和实施例2原液,杀菌时间1min,中和时间10min,中和剂为D/E中和肉汤,试验温度为-18℃~-20℃冰箱冷冻,中和剂为含0 .5%硫代硫酸钠的PBS时,第2组平均生长菌落数为0cfu/ml,第3、4、5组三组间误差率为5 .37%,第6组无菌生长。
D/E中和肉汤在实施例1原液实施例1原对金黄色葡萄球菌悬液定量杀菌试验中和剂鉴定试验中,可以有效中和该消毒液对试验菌的残留毒性,该中和剂及中产物对试验菌无毒性,对培养基无不良影响,实施例1结果证明如表1所示。

2 .2 悬液法定量杀菌试验
试验浓度为有实施例1和实施例2原液(检测报告应为载体法),分别在20℃、-20℃条件下进行杀菌试验,中和时间10min,试验结果表面,实施例1和实施例2原液作用时间1min,对金黄色葡萄球菌的平均杀灭对数值均>5 .00。20℃、-20℃条件下杀菌试验结果无差异,-1稀释梯度的菌落计数结果均为0,说明悬液状态下的消毒剂在低温条件下具有显著的消毒能力。

注:阳性对照组平均菌落数为1 .76×107cfu/ml。
阴性对照组无菌生长。

1.一种公共场所应用喷雾复合消毒剂的制备方法,其特征在于,制备所述复合消毒剂的原料包括季铵盐、纳米银、聚合助剂、抗冻助剂、保湿助剂、惰性助剂和水,各原料的重量份数比为季铵盐0.1~4,纳米银,0.05~5,聚合助剂0.05~20,抗冻助剂5~500,保湿助剂0.5~18,惰性助剂0.2~30,水补齐至1000,包括以下步骤:
步骤一:将季铵盐分散在溶剂中,得到溶液A;
步骤二:将纳米银加入到溶剂中快速搅拌得到溶液B;
步骤三:溶液A和溶液B混合搅拌得到溶液C;
步骤四:将溶液C与适量聚合助剂混合,得到溶液D;
步骤五:在溶液D中加入抗冻助剂,保湿助剂,惰性助剂,并加入适量水,得到性能稳定的复合消毒剂,本发明具有以下优点:原料来源广泛,性能稳定、超广谱杀菌、持久抗菌、无毒无腐蚀、工艺简单、成本低廉、应用范围广,特别适用于人工、消毒机、消毒机器人在常低温下的杀菌消毒,生物相容性好,对环境无二次污染。
2.根据权利要求1所述公共场所应用喷雾复合消毒剂的制备方法,其特征在于,制备溶液A和溶液B所使用的溶剂为水、乙醇、乙醚、丙酮、DMF、冰醋酸、氯仿、四氯化碳、甲苯、乙二醇、丙二醇等实验常用溶剂中的一种或几种溶剂的混合液。
3.根据权利要求2所述公共场所应用喷雾复合消毒剂的制备方法,其特征在于,制备溶液A和溶液B所用的溶剂水可为自来水、泉水、井水、引用纯净水、去离子水中的一种或者几种水的混合液,保证溶剂水中不含阻止实验成功的物质即可。
4.根据权利要求1所述公共场所应用喷雾复合消毒剂的制备方法,其特征在于,制备溶液C时,所用的季铵盐,可以是单链季铵盐如,十二烷基二甲基苄基氯化铵(苯扎氯铵)、十二烷基二甲基苯氧乙基溴化铵、十四烷基二甲基吡啶溴化铵,苯扎溴胺,十六烷基三甲基溴化铵,十六烷基三甲基氯化铵;双链季铵盐如,双癸基二甲基氯化铵、双十二烷基二甲基氯化铵,双十二烷基二甲基溴化铵,溴化(双十二烷基二甲基)乙撑二铵;聚季铵盐如,聚硅季铵盐、聚六亚甲基双胍,聚六亚甲基单胍中的一种或几种。
5.根据权利要求1所述公共场所应用喷雾复合消毒剂的制备方法,其特征在于,所用的纳米银粒子大小为5nm-50nm。

9.根据权利要求1所述公共场所应用喷雾复合消毒剂的制备方法,其特征在于,所用的惰性助剂为N ,N-二(3-氨基丙基)十二烷基胺、N-(3-氨基丙基)-N-十二烷基-丙二胺 、十二烷基二甲基氧化胺其中的一种或几种的混合物。
