我国中药资源丰富,随着中药产业化的推进,中药提取过程中会产生大批量的残渣,中药渣具有多种活性成分和营养物质,有效处理可以实现“变废为宝”。例如红芪、当归可以抑制病原菌生长,提高机体抗氧化能力;益母草成分为生物碱类、黄酮和挥发油等,具有增强免疫和活血化瘀等功效;蒲公英和陈皮有消食理气,促生长等作用。中药渣经过微生物发酵可以进一步改善适口性,提高消化吸收率,增加益生代谢产物。发酵中药渣具有安全、高效、无残留等特点,在动物养殖和饲料添加剂方面极具开发利用价值。
发明内容
本发明的目的是提供一种绿色、高效的饲用抗生素替代品。
为此,本发明提供了一种中药渣发酵饲料添加剂,以重量份计包括:50-75份中药渣、5-20份复合益生菌剂和5-10份复合酶制剂;其中所述复合益生菌剂包括酵母菌、保加利亚乳杆菌、唾液乳杆菌和枯草芽孢杆菌;所述复合酶制剂包括中性蛋白酶、甘露聚糖酶、纤维素酶和木聚糖酶。
具体的,以质量百分比计,上述中药渣包括15-25%的红芪渣、10-20%的当归渣、10-25%的益母草渣、10-20%的甘草、15-30%的蒲公英渣和15-30%的陈皮渣。
具体的,上述复合益生菌剂中酵母菌的活菌数不低于7×109cfu/g、保加利亚乳杆菌的活菌数不低于2×1011cfu/g、唾液乳杆菌的活菌数不低于2×1010、枯草芽孢杆菌的活菌数不低于3×109cfu/g。
具体的,上述复合益生菌的制备方法为:将酵母菌、保加利亚乳杆菌、唾液乳杆菌和枯草芽孢杆菌接种至发酵培养基中发酵培养,得到混合菌液,离心后得到菌泥,真空冷冻干燥后得到复合益生菌剂;以质量百分比计,所述发酵培养基包括1-3%的酵母粉、2-10%的蛋白胨、10-25%的糖蜜、1%的(NH4)2SO4、2%KCl和1%的KH2PO4,余量为水。
具体的,上述复合益生菌的制备方法还包括以1:(2-10)的比例将菌泥加入到保护剂中制备菌悬浮液,平衡后低温冻结,真空冷冻干燥得到复合益生菌剂;所述保护剂包括质量比为(4-8):(3-10):(0.5-3):(4-10)的脱脂乳粉、海藻糖、甘油和山梨醇;所述保护剂pH为5-7。
具体的,上述复合酶制剂中中性蛋白酶、甘露聚糖酶、纤维素酶、木聚糖酶的质量比为(1-2):(1-2.5):(0.5-2):(0.2-1.5);其中中性蛋白酶的酶活为2000-30000U/g、甘露聚糖酶的酶活为4000-100000U/g、纤维素酶的酶活为5000-30000U/g、木聚糖酶的酶活为80000-200000U/g。
本发明还提供了上述中药渣发酵饲料添加剂的制备方法,包括以下步骤:
(1)中药渣混合均匀后粉碎过筛,备用;
(2)配制红糖水,将复合益生菌剂和复合酶制剂加入红糖水中,搅拌均匀后与中药渣混合,进行厌氧发酵;
(3)发酵结束后烘干、粉碎得中药渣发酵饲料添加剂。
具体的,上述步骤(2)具体为:配置浓度为5-25%的红糖水,复合益生菌剂和复合酶制剂加入红糖水中,搅拌均匀后与中药渣充分混合,使得益生菌和酶均匀分布在物料中,混合后物料的水分为40-45%,然后进行厌氧发酵。
具体的,上述步骤(2)中厌氧发酵条件为25-30℃,厌氧发酵4天。
本发明提供的这种中药渣发酵饲料添加剂可用于蛋鸡饲料中,添加量为基础日粮的0.5-2%。
与现有技术相比,本发明具有以下优点和有益效果:
本发明提供的这种中药渣发酵饲料添加剂以中药渣为底物,经微生物、酶混合发酵,充分利用中药渣的活性成分和营养物质,增加益生代谢产物,提高中药渣的再次利用价值,解决中药渣堆积、环境污染等问题。发酵过程中使用的复合益生菌剂包含酵母菌、保加利亚乳杆菌、唾液乳杆菌、枯草芽孢杆菌,其中酵母菌为兼性厌氧菌,发酵后产品的酒香味可以掩盖中药本身的气味,增加食欲;保加利亚乳杆菌发酵产生的半乳糖可以促进胃内容物清空,改善肠道微生态区系;唾液乳杆菌耐酸和胆盐,并具有良好的抑菌效果;芽孢杆菌消耗氧气制造厌氧环境,同时产生消化酶和益菌肽。将中药渣发酵饲料添加剂用于蛋鸡饲养时,可以提高饲料的消化利用率和蛋鸡产蛋率,改善蛋黄颜色和蛋品质,增强动物机体的免疫功能、抗氧化能力和抗病力。此外,本发明优选的中药渣底物包含6种中药成分,其中红芪、当归可以抑制金黄色葡萄球菌、大肠杆菌、沙门氏菌的生长,提高机体抗氧化能力;益母草具有增强免疫和活血化瘀等功效;甘草、蒲公英和陈皮含有理气、消食、益脾和健胃功能的药理成分等,可增加食欲,促进动物生长发育。
具体实施方式
下面将结合实施例对本发明中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。尽管已经详细描述了本发明的代表性实施例,但是本发明所属技术领域的普通技术人员将理解,在不脱离本发明范围的情况下可以对本发明进行各种修改和改变。因此,本发明的范围不应局限于实施方案,而应由所附权利要求及其等同物来限定。
下面通过具体实施例对本发明的中药渣发酵饲料添加剂的效果进行研究。
实施例1:
本实施例提供了一种中药渣发酵饲料添加剂,以重量份计包括:70份中药渣、10份复合益生菌剂、5份复合酶制剂。
其中,中药渣按质量百分比计包括:25%的红芪渣、15%的当归渣、10%的益母草渣、10%的甘草、15%的蒲公英渣和25%的陈皮渣。
复合益生菌剂的制备方法为:将酵母菌、保加利亚乳杆菌、唾液乳杆菌和枯草芽孢杆菌分别接种于培养基中活化12h,将活化后的各菌种以2%的接种量接种至发酵培养基中发酵培养,得到混合菌液,离心后得到菌泥,以1:10的比例将菌泥加入到保护剂中制备菌悬浮液,平衡后低温冻结,真空冷冻干燥得到复合益生菌剂。以质量百分比计,发酵培养基包括2%酵母粉,5%蛋白胨,10%糖蜜、1%的(NH4)2SO4、2%KCl和1%的KH2PO4,余量为水。保护剂包括质量比为5:3.5:2:4的脱脂乳粉、海藻糖、甘油和山梨醇;保护剂pH为7。复合益生菌剂中酵母菌的活菌数为7×109cfu/g、保加利亚乳杆菌的活菌数为2×1011cfu/g、唾液乳杆菌的活菌数为2×1010cfu/g、枯草芽孢杆菌的活菌数为3×109cfu/g。
复合酶制剂包括质量比为1:2:2:1的中性蛋白酶、甘露聚糖酶、纤维素酶、木聚糖酶,中性蛋白酶的酶活为10000U/g、甘露聚糖酶的酶活为80000U/g、纤维素酶的酶活为30000U/g、木聚糖酶的酶活为150000U/g。
本实施例提供的中药渣发酵饲料添加剂通过以下方法制备:
(1)中药渣混合均匀后粉碎至3.0mm颗粒,过筛备用;
(2)配制浓度为10%的红糖水,将复合益生菌剂和复合酶制剂加入红糖水中,搅拌均匀后与中药渣混合,使得益生菌和酶均匀分布在物料中,混合后物料的水分为40%,在25-30℃的常温下厌氧发酵4天;
(3)发酵结束后样品于流化床烘干,烘干样品粉碎至2.0mm,即得中药渣发酵饲料添加剂。
实施例2:
本实施例采用实施例1制备的中药渣发酵饲料添加剂进行蛋鸡饲喂实验,具体方法如下:
选用体重接近且健康状况良好的蛋鸡,随机分为对照组1、对照组2、实验组1、实验组2和实验组3,每组3个重复,每个重复12只,室内保持通风良好,室温维持在22-25℃,蛋鸡地面平养,自由采食饮水,免疫程序和驱虫程序按照鸡场常规同步进行,保持固定的饲喂时间,每天3次,保证水源充足、干净,其中:
对照组1:饲喂基础日粮;
对照组2:饲喂添加有0.05%市售抗生素的基础日粮;
实验组1:饲喂添加有0.5%中药渣发酵饲料添加剂的基础日粮;
实验组2:饲喂添加有1%中药渣发酵饲料添加剂的基础日粮;
实验组3:饲喂添加有2%中药渣发酵饲料添加剂的基础日粮。
基础日粮由62.5%的玉米、24%的豆粕、2%的米糠、0.5%的豆油、8%的石粉和3%的预混料组成。
观察、记载各组蛋鸡生长性能和产蛋情况,结果如表1所示。
饲喂结束后,在每个重复中随机取1只蛋鸡,测定IgA、IgM和IgG,然后屠宰,打开腹腔,迅速分离选取空肠、回肠,置于装有甲醛固定液的10ml离心管中,用于做切片检测,观察肠道形态。肠道形态及免疫性能结果如表2所示。

由表2结果可知,与未使用添加剂的对照组1相比,添加本发明提供的中药渣发酵饲料添加剂能显著提高平均蛋重和产蛋率,改善蛋黄颜色,并降低料蛋比,且对生长性能及蛋品质的改善效果接近添加抗生素的对照组2,采用本发明的饲料添加剂可以提高饲料转化率和蛋品质。添加量梯度试验表面1%的添加量效果相对较好。

小肠是主要的吸收场所,肠绒毛高度与肠消化吸收呈正相关,小肠绒毛高度增加,有利于肠道对营养物质的吸收利用,进而促进蛋鸡的生长发育。由表3可知,相较未使用添加及的对照组1,添加本发明提供的中药渣发酵饲料添加剂后,蛋鸡肠道绒毛高度增加,接近添加抗生素的对照组2。同时实验组1-3增加了IgA、IgM、IgG的含量,表明中药渣发酵饲料添加剂对蛋鸡免疫性能有积极地影响。

Claims (10)
1.一种中药渣发酵饲料添加剂,其特征在于,以重量份计包括:50-75份中药渣、5-20份复合益生菌剂和5-10份复合酶制剂;其中所述复合益生菌剂包括酵母菌、保加利亚乳杆菌、唾液乳杆菌和枯草芽孢杆菌;所述复合酶制剂包括中性蛋白酶、甘露聚糖酶、纤维素酶和木聚糖酶。
2.如权利要求1所述的中药渣发酵饲料添加剂,其特征在于:以质量百分比计,所述中药渣包括15-25%的红芪渣、10-20%的当归渣、10-25%的益母草渣、10-20%的甘草、15-30%的蒲公英渣和15-30%的陈皮渣。
3.如权利要求1所述的中药渣发酵饲料添加剂,其特征在于:所述复合益生菌剂中酵母菌的活菌数不低于7×109cfu/g、保加利亚乳杆菌的活菌数不低于2×1011cfu/g、唾液乳杆菌的活菌数不低于2×1010、枯草芽孢杆菌的活菌数不低于3×109cfu/g。
4.如权利要求1所述的中药渣发酵饲料添加剂,其特征在于,所述复合益生菌剂的制备方法为:将酵母菌、保加利亚乳杆菌、唾液乳杆菌和枯草芽孢杆菌接种至发酵培养基中发酵培养,得到混合菌液,离心后得到菌泥,真空冷冻干燥后得到复合益生菌剂;以质量百分比计,所述发酵培养基包括1-3%的酵母粉、2-10%的蛋白胨、10-25%的糖蜜、1%的(NH4)2SO4、2%KCl和1%的KH2PO4,余量为水。
5.如权利要求4所述的中药渣发酵饲料添加剂,其特征在于,所述复合益生菌剂的制备方法还包括:以1:(2-10)的比例将菌泥加入到保护剂中制备菌悬浮液,平衡后低温冻结,真空冷冻干燥得到复合益生菌剂;所述保护剂包括质量比为(4-8):(3-10):(0.5-3):(4-10)的脱脂乳粉、海藻糖、甘油和山梨醇;所述保护剂pH为5-7。
6.如权利要求1所述的中药渣发酵饲料添加剂,其特征在于:所述复合酶制剂中中性蛋白酶、甘露聚糖酶、纤维素酶、木聚糖酶的质量比为(1-2):(1-2.5):(0.5-2):(0.2-1.5);其中中性蛋白酶的酶活为2000-30000U/g、甘露聚糖酶的酶活为4000-100000U/g、纤维素酶的酶活为5000-30000U/g、木聚糖酶的酶活为80000-200000U/g。
7.如权利要求1-6任意一项所述的中药渣发酵饲料添加剂的制备方法,其特征在于,包括以下步骤:
(1)中药渣混合均匀后粉碎过筛,备用;
(2)配制红糖水,将复合益生菌剂和复合酶制剂加入红糖水中,搅拌均匀后与中药渣混合,进行厌氧发酵;
(3)发酵结束后烘干、粉碎得中药渣发酵饲料添加剂。
8.如权利要求7所述的中药渣发酵饲料添加剂的制备方法,其特征在于,所述步骤(2)具体为:配置浓度为5-25%的红糖水,复合益生菌剂和复合酶制剂加入红糖水中,搅拌均匀后与中药渣充分混合,使得益生菌和酶均匀分布在物料中,混合后物料的水分为40-45%,然后进行厌氧发酵。
9.如权利要求7所述的中药渣发酵饲料添加剂的制备方法,其特征在于:所述步骤(2)中厌氧发酵条件为25-30℃,厌氧发酵4天。
10.如权利要求1-6任意一项所述的中药渣发酵饲料添加剂的在蛋鸡饲料中的应用,其特征在于:所述中药渣发酵饲料添加剂的添加量为基础日粮的0.5-2%。
