Abstract
本发明公开了一种堆肥发酵方法,包括如下步骤:将花椒籽油粕、菌糠、生物炭均匀混合后,添加生防菌混合后,加水调节含水量,采用条垛式方式进行发酵,所述生防菌为枯草芽孢杆菌Tpb55(BS)和棘孢木霉菌HG1(TH),利用本发明的生防菌混合菌剂处理后,花椒籽油粕‑菌糠堆肥高温和腐熟阶段中TOC、NO3 ‑‑N含量降低,而NH4 +‑N含量增加,这促进了堆肥有机碳氮的转化,并显著增加了腐熟阶段堆肥的细菌丰度和群落多样性。BS和TH处理后主要上调了氨基酸、碳水化合物以及生物碱类代谢物水平,其中东莨菪碱、文多林、长春花碱、辣椒素和水杨酸等物质具有抑菌或增强作物抗病性的作用。

技术领域
本发明属于微生物及其应用领域,具体涉及的是一种通过添加生防菌对堆肥致病微生物和代谢物进行转化。
背景技术
花椒(Zanthoxylum bungeanum Maxim.)被人类使用已经有两三千年的历史,它被广泛用作调味品和驱虫。通常把花椒皮用来调味而把花椒籽丢弃或者榨油,花椒籽油粕常被当做废弃物未加以利用,这样会造成环境污染、温室气体排放和严重疾病发生等问题。菌糠是进行真菌菇类栽培时废弃的培养基,常被用作肥料直接施用到农田上,不仅养分利用率低还容易引入病原菌引起作物病害。这些废弃物还容易乱丢乱放被生物污染或腐化为有毒有害物质从而携带污染物和致病菌。堆肥是一种利用各种有机废弃物,经过微生物发酵将可降解有机物转化为稳定腐殖质的技术,利用堆肥技术可促进有机废弃物的养分转化、降低温室气体排放、调节微生物活性和降低抗生素抗性基因丰度等。
花椒籽和菌糠中主要含有植物生物刺激素(酚类、黄酮类、脂类、甾醇)、生物碱、氨基酸等物质,菌糠如果受到微生物污染还可能产生一些真菌毒素。在堆肥发酵或腐熟时,主要产生一些促进作物生长的代谢物,如氨基酸、碳水化合物、植物生长调节物质等。已有研究人员利用气相色谱-质谱(GC-MS)分析添加阿莫西林amoxicillin对鸡粪堆肥中代谢物的影响,结果表明amoxicillin胁迫下堆肥中促进植物生长的propanoic acid ethyl esterand oleic acid代谢物升高,而环境污染物nhexadecanoic acid and 3β,5β-Cholestan-3-ol等代谢物下降。寻求高效、相对廉价的堆肥腐熟材料是提高堆肥品质的重要热点问题。
发明内容
本发明的目的在于针对现有技术的不足,提供一种改善生物堆肥的转化代谢物和病原微生物的方法。
为了达到上述目的,本发明采用如下技术方案:
一种堆肥发酵方法,包括如下步骤:将花椒籽油粕、菌糠、生物炭均匀混合后,添加生防菌混合后,加水调节含水量,采用条垛式方式进行发酵。
进一步的,作为一个较佳的实施例,所述花椒籽油粕、菌糠:生物炭的干重比为1-5:2-10:1-5,优选为4:5.5:5。
进一步的,作为一个较佳的实施例,所述生防菌为枯草芽孢杆菌Tpb55和棘孢木霉菌HG1。
其中,枯草芽孢杆菌菌株的菌种保藏号为CGMCC NO.19276,棘孢木霉菌的菌种保藏号为CGMCC NO.2843。
进一步的,作为一个较佳的实施例,生防菌中添加枯草芽孢杆菌Tpb55和棘孢木霉菌HG1的干重比比1-5:1-5。
进一步的,作为一个较佳的实施例,所有处理原料混合后加水调节含水量至70%。
本发明的另一目的还在于提供一种延长堆肥高温持续时间的方法,包括如下步骤:将花椒籽油粕、菌糠:生物炭均匀混合后,添加生防菌混合后,加水调节含水量,采用条垛式方式进行发酵。
利用本发明的生防菌混合菌剂处理后的,向花椒籽油粕-菌糠堆肥中添加生防芽孢杆菌(BS)和棘孢木霉菌(TH)降低了堆肥高温和腐熟阶段中总有机碳(TOC)、硝态氮(NO3 --N)含量,而增加了铵态氮(NH4 +-N)含量,促进了堆肥有机碳氮的转化。BS或TH处理显著增加了腐熟阶段堆肥的细菌丰度和群落多样性,而降低了真菌群落多样性。
进一步的,本发明的BS和TH处理后主要上调了氨基酸、碳水化合物以及生物碱类代谢物,其中东莨菪碱、文多林、长春花碱、辣椒素等生物碱和水杨酸显著上调,这些物质具有抑菌或增强抗病性的作用。
一种提高堆肥中抗菌代谢物水平和消除植物病原真菌的方法,包括如下步骤:在堆肥发酵中添加生防芽孢杆菌(BS)和棘孢木霉菌(TH)。
与现有技术相比,本发明的有益效果为:

(2)BS或TH处理显著增加了腐熟阶段堆肥的细菌丰度和群落多样性,而降低了真菌群落多样性。添加生防菌处理增加了堆肥中人类致病细菌的总体丰度,但降低了除Trichothecium以外的13种植物病原真菌的丰度和种类,这意味着使用此堆肥会降低作物发生真菌病害的风险;
(3)BS和TH处理后主要上调了氨基酸、碳水化合物以及生物碱类代谢物,其中其中东莨菪碱、文多林、长春花碱、辣椒素等生物碱和水杨酸显著上调,这些物质具有抑菌或增强抗病性的作用。

参选以下本发明的优选实施方法的详述以及包括的实施例可更容易地理解本发明的内容。除非另有限定,本文使用的所有技术以及科学术语具有与本发明所属领域普通技术人员通常理解的相同的含义。当存在矛盾时,以本说明书中的定义为准。如本文所用术语“由…制备”与“包含”同义。本文中所用的术语“包含”、“包括”、“具有”、“含有”或其任何其它变形,意在覆盖非排它性的包括。例如,包含所列要素的组合物、步骤、方法、制品或装置不必仅限于那些要素,而是可以包括未明确列出的其它要素或此种组合物、步骤、方法、制品或装置所固有的要素。
一、试验材料和处理设计
1、试验材料
花椒籽油粕是花椒籽(Zanthoxylum bungeanum)榨油后的残渣,来自于山东省枣庄市山亭区,其含粗纤维62.5%,粗蛋白15.2%,脂肪5.3%,含氮2.2%,含磷0.7%,含钾2.3%;菌糠是利用秸秆、木屑等原料进行金针菇栽培,在金针菇采收后剩余的培养基,由山东平阴绿洲食用菌有限公司提供,其主要营养成分为:水分(52.35±2.31)%,总碳(43.85±1.63)%,总氮(1.06±0.04)%,C/N 41.37±1.54;生物炭是用稻壳为原料在400℃条件下制备而成,炭的风干含水率(MAD)为3.16%,风干灰分(AAD)为14.34%,风干自由基挥发分(VAD)为5.77%,购买自山东明辰生物科技有限公司;生防菌枯草芽孢杆菌菌株Tpb55(菌藏号:CGMCC NO.19276)和棘孢木霉菌HG1(菌藏号:CGMCC NO.2843)均由中国农科院烟草所提供,该菌株可用于防治烟草青枯和黑胫病等。将花椒籽油粕:菌糠:生物炭按照4:5.5:5的干重比例均匀混合,每个处理共有5吨原料混合物,采用条垛式发酵,试验处理设计如下:1)原料混合后不加任何生防菌(CK);2)原料混合物添加干重质量0.2%的枯草芽孢杆菌Tpb55(BS);3)原料混合物添加干重质量0.8%的棘孢木霉菌HG1(TH);4)原料混合物添加干重质量0.1%的枯草芽孢杆菌Tpb55和0.4%的棘孢木霉菌HG1(BS-TH)。所有处理原料混合后加水调节含水量至70%。堆肥共进行了36天,每三天取样一次,每个处理重复三次。
2、堆肥过程理化性质测定
对堆肥发酵的第2h(记为Day 0),第3、6、12、18、24、27、30、36d样品进行14项理化性质测定。堆肥有机碳(OC)采用重铬酸钾氧化-分光光度法测定(TMECC,2002)。采用元素分析仪(FLASH 2000同位素比质谱仪,赛默飞世尔科技,美国)测定堆肥总碳、总氮和C/N比。采用碱熔法测定堆肥全钾和全磷;堆肥铵态氮和硝态氮采用2M KCl提取,有效磷采用0.03molL-1NH4F-0.025mol L-1HCl溶液进行提取,然后将提取液用流动分析仪(Futura连续流分析系统,Alliance仪器,法国)测定(Yan et al.,2013)。使用FP640火焰光度计(中国上海仪表集团有限公司)测定有效钾浓度。堆肥pH值按水土比2.5:1测定。用电极法按水土比5:1测定堆肥电导率。
3、堆肥病原微生物分析
根据毒力因子数据库(http://www.mgc.ac.cn/VFs/)的分类和Cai et al.,Chenet al.,and Peng et al.的报道,获得细菌病原物物种名称。然后到NCBI GenBank(http://www.ncbi.nlm.nih.gov/)获取物种的16S rRNA基因序列,构建558个致病细菌物种数据库。这些致病细菌物种信息在之前文献中有详细报道(Peng et al.2017)。使用BLASTN将16S rRNA基因序列在病原数据库中进行搜索比对。按照序列同一性≥99%、比对长度≥250bp和错配≤2bp的标准对序列进行筛选并细菌病原体注释。将高通量测序获得的真菌OTU表上传到FUNGuild数据库(http://www.stbates.org/guilds/app.php),线上分析并下载分析结果,该结果将真菌划分为12个guilds。我们将注释到植物病原菌(plantpathogens)和动物病原菌(animal pathogens)条目的物种挑选出来进行统计分析。
4、堆肥代谢物分析
将保存好的堆肥样品研磨至粉末状,准确称量100mg放入离心管中,加入80%甲醇水溶液。经过超声、提取和离心后氮气吹干,然后用溶剂复溶、低温超声提取后离心,将上清液转移至进样瓶中。每个处理重复6次。将等体积的所有样本混合成质控样本,在样本中间隔插入质控样本。样本代谢组检测在诺禾致源公司的超高效液相色谱-飞行时间质谱(UPLC-TripleTOF)平台上完成。上机完成以后将质谱原始数据导入Compound Discoverer3.1软件进行处理并搜索数据库,获得各样本代谢物定性定量结果。然后对数据进行质控保证数据的准确性和可靠性。
二、试验结果和分析
1、堆肥物质转化
堆肥温度在9-12天达到最高点,并持续了约12-14天的高温(>50℃),高温能有效地杀灭病原微生物和杂草种子。BS和BS-TH处理比TH和CK处理更快到达高温。这可能是因为BS和BS-TH中加入的枯草芽孢杆菌是嗜热细菌有关,它可以迅速分解有机物并提高温度。另外,BS和BS-TH处理延长了堆肥高温的持续时间。BS和BS-TH处理的堆肥温度在15天时迅速下降,这可能与堆肥微生物群落丰度下降有关,爆发增长的枯草芽孢杆菌种群和种间竞争导致一些微生物死亡。堆肥Moisture含量随着时间越来越小,且添加生防菌处理(BS,TH,BS-TH)的堆肥水分要比CK低,这可能是由于添加生防菌增加了堆肥的微生物活动,促进了物质转化,从而消耗了更多的水分。pH随着时间的进行越来越高,这可能是堆肥有机酸的消耗和氨化产生的氨引起的,BS和BS-TH处理的pH比CK处理更高,可能与该处理产生更多的铵态氮有关(图1)。
TN含量在堆肥开始至24天时下降,但后期有所上升,前期下降是因为以NH3排放造成的氮损失,后期增加是因为堆肥含碳化合物的干质量净损失和降解速率快于氮损失,则TN含量将作为浓度效应增加。随着堆肥的进行,NH4 +-N含量迅速上升,在18天后则持续下降。NH4 +-N含量上升是因为前期高温高湿增强了微生物活动,促进了堆肥含氮有机物分解。NH4 +-N含量下降主要是因为NH3的排放和NH4 +-N向NO3 --N的转化作用。在堆肥成熟期,添加生防菌处理的NH4 +-N含量要高于CK,主要是因为添加生防菌提高了堆肥微生物的活性,促进了有机氮的分解。NO3 --N含量在前期变化不大,在24天后则持续上升。主要是因为前期堆肥高温抑制了堆肥硝化细菌的生长和活性,后期堆肥温度下降,硝化细菌活性上升。添加生防菌的NO3 --N含量要比CK升高的慢,原因可能是添加生防菌抑制了将NH4 +-N转化为NO3 --N的微生物活性(图1)。
堆肥总碳随着时间持续下降,主要是微生物分解堆肥有机物导致CO2和CH4排放,引起碳损失,BS和BS-TH处理的总碳要比CK下降的快,堆肥有机碳也呈现与总碳类似的变化,但有机碳下降更明显,有机碳除了被微生物分解排放CO2和CH4外,还有一部分转化为溶解性的无机碳。生防菌处理增加了堆肥总碳和有机碳的损失。原因可能是因为添加生防菌增加了堆肥微生物活性,一方面是嗜热微生物促进了有机碳的分解,另一方面是一些碳水化合物、蛋白质和脂肪等被微生物作为能源利用,C/N是衡量堆肥成熟的一个基本指标。在堆肥第24到第36天,所有处理C/N快速下降,达到13.4-15.2,满足堆肥成熟度的要求。
堆肥中总磷含量中期含量上升,成熟期下降至初始水平。TH和BS-TH提高了堆肥TP含量。堆肥AP含量整体呈下降趋势,但添加生防菌处理的AP含量要高于CK。堆肥TK含量一直呈上升趋势。堆肥AK一直呈下降趋势,添加生防菌处理的AK在前期比CK下降慢,在堆肥成熟期比CK下降快。TK随着堆肥发酵而不断增加,但其绝对含量不会发生变化,只会在不同形式之间发生变化。EC在堆肥过程中先减少后增加,且添加菌处理的EC要高于CK,说明添加菌处理的堆肥有机物矿化程度更高(表1)。

2、堆肥微生物多样性变化
Shannon指数可以表示堆肥微生物群落多样性程度,Shannon指数越大表示微生物群落多样性越高。BS处理的Shannon指数在第27和36天时相比于CK处理显著上升,表明在堆肥降温和腐熟阶段,BS处理的细菌群落多样性增加。TH处理的Shannon指数在第6和36天时相比于CK处理显著上升,表明在堆肥升温和腐熟阶段,TH处理的细菌群落多样性增加。在第12和18天(高温阶段),TH处理的Shannon指数要显著高于BS处理。TH处理的真菌群落Shannon指数在第12和18天(高温阶段)比CK、BS、和BS-TH处理显著增加,表明TH处理的真菌多样性在高温阶段显著增加。添加生防菌处理(BS,TH,BS-TH)的真菌群落Shannon指数在第36天时显著下降。各处理细菌群落Chao1指数在升温、高温阶段变化不大,而在第36天(腐熟阶段)添加生防菌处理的细菌群落Chao1指数显著高于CK,表明添加生防菌处理增加了堆肥腐熟阶段的细菌丰度。真菌群落Chao1指数在堆肥过程中逐渐下降,表明堆肥过程中真菌丰度下降。PCoA分析表明取样时间和添加生防菌处理是堆肥微生物群落差异的决定因素。不同取样时间对堆肥微生物群落的影响大于不同生防菌处理。堆肥第6、27和36天的BS和TH样本的细菌群落与CK样本存在差异。堆肥第18和36天的BS或TH样本的细菌群落与CK样本存在差异(图2)。
3、堆肥致病微生物变化
所有样本中共检测到了26种致病细菌属,17种真菌致病真菌属。其中,检测出10种优势细菌病原菌,包括Staphylococcus、Pseudomonas、Acinetobacter、Bordetella、Corynebacterium、Escherichia.Shigella、Salmonella、Bacillus、Hungateiclostridium、Enterococcus。优势细菌病原菌在堆肥第18天左右丰度有所下降,在腐熟期丰度恢复。Clostridium_sensu_stricto1、Clostridium_sensu_stricto_15、Clostridium_sensu_stricto_18在堆肥后期丰度上升,而Legionella、Aeromonas、Burkholderia、Caballeronia、Paraburkholderia丰度下降。腐熟堆肥比初始时增加了优势细菌病原菌的总体丰度,这可能是因为花椒籽油粕、菌糠等经过微生物分解释放了大量养分物质,有利于细菌包括病原体的生长。生防菌处理后腐熟堆肥的病原细菌总体丰度相比于CK处理增加了。其中Staphylococcus、Pseudomonas、Bordetella、Corynebacterium、Salmonella、Enterococcus、Yersinia等细菌病原菌丰度在添加生防菌后丰度上升,而Acinetobacter、Escherichia.Shigella丰度在添加生防菌后丰度下降。
除Trichosporon是动物病原真菌外,其他的都为植物病原真菌。堆肥中优势病原真菌主要有Trichosporon、Trichothecium、Cutaneotrichosporon、Clonostachys、Gibberella、Strelitziana、Pseudocercospora、Acrophialophora等。随着堆肥的进行,真菌病原菌的种类大幅降低。Gibberella、Strelitziana、Pseudocercospora、Curvularia、Bipolaris、Microdiplodia、unclassified_f__Elsinoaceae、Plectosphaerella、Acrophialophora、Marssonina、Lasiodiplodia病原菌在堆肥腐熟时基本消失。这可能是因为堆肥高温期温度达到60度,能将大部分病原真菌杀灭,所以堆肥过后真菌病原菌种类大幅下降。添加生防菌处理比初始时降低了腐熟堆肥真菌病原菌的种类但增加了丰度。在堆肥腐熟期,添加生防菌处理比CK处理的Trichothecium丰度增加,这导致病原真菌总丰度增加,而Trichosporon、Clonostachys丰度降低。Trichothecium丰度增加可能是因为添加生防菌促进了有机碳、有机氮等养分的分解和转化,使得在堆肥高温期存活下来的病原真菌利用这些养分大量繁殖。Trichothecium丰度增加则与其他微生物竞争或抑制其他微生物生长,这可能是Trichosporon、Clonostachys丰度降低的原因(图3)。
总体来说,添加生防菌处理增加了堆肥中人类致病细菌的总体丰度,但降低了除Trichothecium以外的所有植物病原真菌的丰度和种类。这表明使用添加生防菌的堆肥可能会增加农民感染疾病的风险,但会降低作物发生真菌病害的风险。堆肥中的病原细菌主要通过伤口和口鼻吸入感染使用者,因此,使用本堆肥产品时最好佩戴口罩和手套。
3、堆肥中代谢物的变化
添加生防菌处理(BS、TH、BS-TH)显著影响了碳水化合物、氨基酸和生物碱类代谢物。对于半胱氨酸和蛋氨酸代谢,BS和TH处理显著上调了蛋氨酸(Methionine)、L-胱氨酸(L-Cystine)和O-磷酸-L-丝氨酸(O-phospho-L-serine)的代谢,而下调了谷胱甘肽(Glutathione)代谢。在甘氨酸、丝氨酸和苏氨酸代谢中,单独添加生防菌处理显著上调了L-天冬氨酸(L-Aspartate),L-苏氨酸(L-Threonine)、甜菜碱醛(Betaine aldehyde)等代谢物,而下调了甜菜碱(Betaine)。单独添加生防菌处理显著上调了组氨酸代谢中1-甲基-L-组氨酸(1-Methyl-L-histidine)、L-组氨酸(L-Histidine)、尿酸(Urocanate)、L-谷氨酸(L-Glutamate)等代谢物(图4)。这表明添加生防菌显著增加了堆肥中氨基酸类代谢物。
添加菌处理对碳水化合物代谢影响较大的途径是嘌呤代谢和嘧啶代谢。在嘧啶代谢中,添加生防菌除了下调胞嘧啶(Cytosine)和5-甲基胞嘧啶(5-Methylcytosine)外,其他代谢物如尿苷(Uridine)、胞苷(Cytidine)、胸苷(Thymidine)和Thymine(胸腺嘧啶)等都显著增加。此外,在嘌呤代谢中,添加生防菌处理的所有差异代谢物都是上调的(图4)。
生物碱类物质主要存在于植物体内,还有部分存在于动物和微生物中。生物碱是生物在长期的生态环境适应过程中为抵御微生物、病毒和其他动植物攻击或侵染而形成的一类次生代谢产物。本研究发现在托烷、哌啶和吡啶生物碱生物合成中,单独添加生防菌处理的堆肥L-赖氨酸(L-Lysine)、L-哌醋甲酯(L-Pipecolate)、L-糖苷(L-Saccharopine)、烟酸盐(Nicotinate)、东莨菪碱(Scopolamine)、L-苯丙氨酸(L-Phenylalanine)代谢物上调,而托品(Tropine)下调。此外,添加生防菌处理还上调了烟酸、哌可酸、甲羟戊酸、水杨酸、泛酸等生物碱或酸性物质,而BS处理还上调了白皮杉醇、月桂酸、文朵灵、长春花碱、3-甲氧基酪胺、辣椒素生物碱或酸性代谢物,TH处理还上调了月桂酸,香豆素生物碱或酸性代谢物(图4)。东莨菪碱、白杉醇、文多林、长春花碱和辣椒素等生物碱一般来源于植物或者微生物,用作医药,具有抗肿瘤、抑菌等作用。哌可酸是人体和动物所必须的一种物质,常用作饲料添加剂使用。外施水杨酸通过调控作物光系统和抗氧化酶活性增强作物系统抗病性,防御病原菌的侵染。
以上依据本发明的理想实施例为启示,通过上述的说明内容,相关人员完全可以在不偏离本项发明技术思想的范围内,进行多样的变更以及修改。本项发明的技术性范围并不局限于说明书上的内容,必须要根据权利要求范围来确定技术性范围。
Claims (7)
1.一种堆肥发酵方法,其特征在于,包括如下步骤:将花椒籽油粕、菌糠、生物炭均匀混合后,添加生防菌混合后,加水调节含水量,采用条垛式方式进行发酵。
2.根据权利要求1所述的一种堆肥发酵方法,其特征在于,所述花椒籽油粕、菌糠:生物炭的干重比为1-5:2-10:1-5。
3.根据权利要求2所述的一种堆肥发酵方法,其特征在于,所述花椒籽油粕、菌糠:生物炭的干重比4:5.5:5。
4.根据权利要求1所述的一种堆肥发酵方法,其特征在于,所述生防菌为枯草芽孢杆菌Tpb55和棘孢木霉菌HG1。
5.根据权利要求4所述的一种堆肥发酵方法,其特征在于,所述生防菌中枯草芽孢杆菌Tpb55和棘孢木霉菌HG1的干重比为1-5:1-5。
6.一种降低堆肥发酵中TOC含量而增加NH4 +-N含量进而促进堆肥有机碳氮转化的方法,其特征在于,向花椒籽油粕-菌糠堆肥中添加生防芽孢杆菌和棘孢木霉菌。
7.一种提高堆肥中抗菌代谢物水平和消除植物病原真菌的方法,包括如下步骤:在堆肥发酵中添加生防芽孢杆菌和棘孢木霉菌。
