Description
技术领域
本发明涉及抗体领域,具体而言涉及人抗IL-13抗体,尤其是那些可中和IL-13活性的人抗体。本发明进一步涉及抗IL-13抗体在制备用于诊断或治疗IL-13相关疾病或失调尤其哮喘、特应性皮炎的药物中的用途。
背景技术
白细胞介素(IL)-13属于细胞因子,其主要由Th2 CD4+淋巴细胞分泌,但也可由Th1 CD4+T细胞、CD8+T淋巴细胞、NK细胞,以及诸如肥大细胞、嗜碱性粒细胞、嗜酸性粒细胞、巨噬细胞、单核细胞和呼吸道平滑肌细胞的非T-细胞群体生成。IL-13由114个氨基酸组成,分子量约为12kDa。IL-13与IL-4同属Th2细胞因子,在氨基酸水平上具有30%的序列相似性,功能上密切相关。人IL-13基因与IL-4基因相邻,共同位于染色体5q31上。
IL-13通过结合受体IL-13Rα1,进而募集IL-4Rα受体形成IL-13Rα1/IL-4Rα功能性受体复合物,从而激活JAK-STAT6信号通路。同时IL-13还可与受体IL-13Rα2结合,但IL-13Rα2所具有的胞质尾区较短,缺乏已知的信号基序,充当了IL-4Rα的引诱受体,调节自身IL-13水平。IL-13Rα1/IL-4Rα受体复合物在人B细胞、肥大细胞、单核细胞/巨噬细胞、树突细胞、嗜酸性粒细胞、嗜碱性粒细胞、成纤维细胞、内皮细胞、皮肤组织上皮细胞、呼吸道上皮细胞和呼吸道平滑肌细胞上均有表达。
哮喘又称支气管哮喘,是常见的呼吸系统炎症疾病,主要病理特征表现为多变和复发的症状、呼吸道气流阻塞和支气管痉挛。全球哮喘患者高达3亿人,中国则超过3000万人。目前,轻到中度哮喘通常可利用吸入性皮质类固醇,并结合β-增效剂或白细胞三烯抑制剂得到控制。对于重症患者,较高剂量皮质类固醇已无法有效控制症状,且长期口服皮质类固醇会导致诸如骨质疏松、体重增加、内分泌紊乱、降低儿童生长速率等副作用,迫切需要新的治疗方法。哮喘的病因被认为是由Th2淋巴细胞介导的炎症反应。哮喘患者支气管活检标本、痰和支气管肺泡灌洗(BAL)细胞中的IL-13水平均高于健康人。IL-13是哮喘靶向治疗领域的一个新兴靶点,尤其对于目前尚无有效生物疗法的非嗜酸性表型患者(占哮喘患者50%)疗效显著。
特应性皮炎(Atopic dermatitis,AD)是自身免疫疾病的一种,是最常见的炎性皮肤疾病之一,主要症状为皮肤干燥、慢性湿疹样皮损和剧烈瘙痒,严重影响患者生活质量。近二十年来,全球发病率持续上升,儿童和成人的发病率已分别高达10-20%和5-10%。2014年,我国1-7岁城市儿童特应性皮炎患病率已达12.94%。Th2型炎症是AD的基本特征,IL-13是介导AD发病的重要细胞因子之一,其高表达与特应性皮炎的严重程度呈正相关,阻断IL-13可改善特应性皮炎临床症状。常规治疗方法中针对轻度和中度患者,可根据皮损及部位选择TCS(局部皮质类固醇)/TCI(局部钙神经素抑制剂)对症治疗,必要时口服抗组胺药治疗,或湿包治疗,或使用紫外线疗法。对于重度患者可使用免疫抑制剂,及短期系统用糖皮质激素控制急性严重顽固性皮损。但是上述方法皆因不良耐受性不宜长期使用。生物制剂Dupilumab以良好的安全性和有效性已写入2020版中国特应性皮炎诊疗指南,为中和重度患者提供了治疗新选择。Dupilumab单克隆抗体靶向IL-4Rα,可同时抑制IL-4和IL-13信号通路;但已有临床试验数据表明,单纯靶向IL-13信号通路的Tralokinumab和Lebrikizumab可获得与Dupilumab相似的药效,但副作用结膜炎的发生率显著降低。相对同时靶向IL-4和IL-13信号通路,单独靶向IL-13可提供线性PK曲线的优势,从而降低给药剂量和频率,说明单独阻断IL-13信号通路可在AD中提供相当的改善和更加具体的靶向作用。
发明内容
本发明提供了一组针对IL-13的新抗体。本发明还提供了含有本发明所述的抗IL-13抗体的药物组合物,以及所述抗体在制备用于诊断或治疗特应性皮炎以及哮喘的药物中的用途。
具体地,本发明提供了抗IL-13抗体。本发明的优选实施方案为一种特异性结合IL-13的抗体或抗原结合片段,其包含重链CDR1、CDR2和CDR3以及轻链CDR1、CDR2和CDR3,所述重链CDR1、CDR2和CDR3以及轻链CDR1、CDR2和CDR3分别与选自下组的抗体的重链CDR1、CDR2和CDR3以及轻链CDR1、CDR2和CDR3序列具有至少80%,优选至少90%,更优选至少95%的同一性:FWB03、FWB05、FWB09、FWB10、FWB11、FWB12、FWB13、FWB14、FWB15、FWB16、FWB17和FWB19。最优选地,本发明涉及一种特异性结合IL-13的抗体或抗原结合片段,其包含选自下组的抗体的重链CDR1、CDR2和CDR3以及轻链CDR1、CDR2和CDR3:FWB03、FWB05、FWB09、FWB10、FWB11、FWB12、FWB13、FWB14、FWB15、FWB16、FWB17和FWB19。
本发明的更优选实施方案为特异性结合IL-13的抗体或抗原结合片段,其包含重链可变区和轻链可变区,其中所述重链可变区和轻链可变区与选自下组的抗体的重链可变区和轻链可变区序列具有至少85%同一性,优选至少90%同一性,更优选95%同一性,最优选98%同一性:FWB03、FWB05、FWB09、FWB10、FWB11、FWB12、FWB13、FWB14、FWB15、FWB16、FWB17和FWB19。最优选地,本发明本发明涉及一种特异性结合IL-13的抗体或其抗原结合片段,其包含选自下组的抗体的重链可变区和轻链可变区序列:FWB03、FWB05、FWB09、FWB10、FWB11、FWB12、FWB13、FWB14、FWB15、FWB16、FWB17和FWB19。

本发明还提供了一种分离的核酸,其编码本发明所述的抗体或抗原结合片段。本发明还提供了一种表达载体,其包含权利要求7所述的分离的核酸。本发明还提供了一种宿主细胞,其包含本发明所述的核酸或表达载体,优选地所述宿主细胞是真核宿主细胞,更优选所述宿主细胞是哺乳动物宿主细胞。本发明还提供了一种组合物,其包含第一核酸和第二核酸,其中所述第一核酸编码本发明所述的抗体的重链,所述第二核酸编码本发明所述的抗体的轻链。
本发明提供了一种药物组合物,其用于治疗IL-13相关疾病或失调,其包含本发明所述的抗体或其抗原结合片段、分离的核酸、表达载体、或宿主细胞,所述药物组合物还包含药用载体。优选地,所述IL-13相关疾病或失调是哮喘或特应性皮炎。

发明详述
除非另有说明,本申请中使用的术语为本领域公知常用的含义。
应当指出的是,除非另外指明,本文涉及的白细胞介素-13(IL-13)通常指人IL-13。在本文的若干处还指“抗原”。本发明提供了抗人IL-13的抗体,具体而言是与包括猕猴IL-13和恒河猴IL-13在内的非人灵长类动物IL-13以及鼠IL-13具有交叉反应性的人抗体。本发明所述的抗体可识别因130号氨基酸位的精氨酸残基被取代为谷氨酰胺后而获得的IL-13变体。本发明的另一些方面和实施方案提供了抗鼠IL-13,尤其是抗小鼠IL-13的特异性结合成员。
抗IL-13抗体分子可以是人有效的、人的、人源化的、CDR移植的、嵌合的、突变的、亲和成熟的、去免疫的、合成的或者是体外产生的抗体分子。在一个实施方案中,IL-13抗体为人源化抗体。
在一个实施方案中,IL-13抗体分子包含以下序列的重链和轻链。抗-IL-13抗体分子的重链和轻链可以是基本全长的(如抗体分子包含至少1条、优选2条重链以及至少1条、优选两条轻链)或包含抗原结合片段(如Fab、Fv或单链Fv片段)。在另一实施方案中,抗体分子具有选自以下的重链恒定区如IgG1、IgG2、IgG3、IgG4、IgM、IgA1、IgA2、IgD和IgE的重链恒定区、特别是IgG1、IgG2、IgG3和IgG4的重链恒定区,更特别是IgG4(如人IgG4)的重链恒定区。重链恒定区一般是人的或者是人恒定区的修饰形式。在另一实施方案中,抗体分子具有选自如Lambda或Kappa(优选Lambda,如人Lambda)轻链恒定区的轻链恒定区。在一个实施方案中,改变(如突变)恒定区,以修饰抗体分子的特性(如提高或降低以下一种或多种Fc受体结合、抗体糖基化、半胱氨酸残基数、效应细胞的功能或补体的功能)。例如,可以在一个或多个残基(如人IgG4的重链残基228中)突变人IgG4恒定区。

实施例1使用基因工程的方法制备抗体
对重链和轻链的CDR区氨基酸突变改造,改造后的20个候选抗体的重链可变区的框架区和可变区以及轻链可变区的框架区和可变区的氨基酸序列如下所示,所有抗体均为人IgG4 S228P抗体,即第228位氨基酸为脯氨酸。

分别根据氨基酸序列合成基因序列,并克隆至表达载体,转染至HEK293细胞,使转化的HEK293细胞表达抗体,收集纯化抗体。对20个候选抗体通过ELISA筛选得到与IL-13结合活性高的候选抗体。
实施例2通过体外实验对候选抗体进行筛选
2.1通过ELISA方法测定抗IL-13抗体的对人、猴和鼠IL-13的亲和力
首先将96孔ELISA板用50μl的1μg/ml streptavidin在4℃过夜,并用250μl的封闭液(3%BSA in TBST)在37℃下孵育2小时,之后用PBST洗板3次。在每个ELISA实验孔中加入50μl生物素标记的人源IL-13(获得自上海睿智化学研究有限公司,其序列为SEQ ID NO:181:GPVPPS TALRELIEEL VNITQNQKAP LCNGSMVWSI NLTAGMYCAA LESLINVSGC SAIEKTQRMLSGFCPHKVSA GQFSSLHVRD TKIEVAQFVK DLLLHLKKLF REGRFN)(1μg/ml),并孵育1小时,随后用PBST洗板3次。将待测20个抗体从2μg/ml进行5倍系列稀释,共8个浓度点(包括空白对照),并将系列稀释的抗体加入到ELISA孔中,在37℃下孵育1小时,随后用PBST洗板3遍。加入辣根过氧化酶结合的二抗并在37℃下孵育1小时,最后用PBST洗板3次并加入50μl TMB反应15分钟。显色反应可以用1N盐酸(50μl)进行终止,并在读板机(M5)上检测450nm的吸光度值,以此来计算各个抗体对IL-13的亲和力(EC50)。最终筛选出与人、猴和鼠IL-13结合活性强,但与人IL-4无结合的FWB03、09、11、12、13、14、15、16和FWB19继续实验。

2.2通过流式细胞仪测定抗体阻断配体IL-13与细胞表面受体hIL-13Rα结合的活性
待测抗体为FWB03、09、11、12、13、14、15、16和FWB19。微孔板中每孔收集3×105个细胞(HEK293/hIL-13Rα/hIL-4Rα),离心,用含2%FBS的PBS重悬细胞,抗体以10μg/mL为初始浓度,5倍梯度稀释,向每孔细胞加入相应浓度的抗体或空白对照100μl,设置3个复孔,放置4℃孵育0.5小时后,再加入100μl的0.32μg/mL的生物素偶联的人IL-13蛋白,放置4℃孵育1小时,用含2%FBS的PBS清洗细胞2次后,加入Alexa 488链霉亲和素,放置4℃避光孵育0.5小时,用含2%FBS的PBS清洗细胞2次,然后用100μl含2%FBS的PBS重悬细胞,最后用流式细胞仪检测细胞表面的荧光信号,检测时每孔吸取2×104个细胞。结果如表8所示。筛选出能够强力地阻断人IL-13与IL-13受体hIL-13Rα结合的FWB03、09、11、12、13和FWB16继续实验。

2.3测定抗体对TF-1细胞增殖的抑制作用
待测抗体为FWB03、09、11、12、13和FWB16。TF-1细胞(人髓系白血病细胞,购买自ATCC)的生长增殖依赖于IL-13。使用含有10%胎牛血清FBS的RPMI1640培养基培养TF-1细胞,并添加2ng/mL粒细胞-巨噬细胞集落刺激因子GM-CSF。96孔板中每孔收集2×105个TF-1细胞,使用50μL含有2%FBS的RPMI1640培养基培养。将人IL-13抗原分别与FWB03、09、11、12、13或FWB16抗体预混合。每孔中加入相应浓度的抗原抗体混合液50μl,抗原终浓度为10ng/mL,抗体终浓度分别为10μg/mL、2μg/mL、0.4μg/mL、0.08μg/mL、16ng/mL、3.2ng/mL、0.64ng/mL、0.128ng/mL、0.0256ng/mL。37℃5%CO2培养箱中培养3天,然后每孔加入50μLCell-Titer Glo,并在读板机(M5)上检测荧光强度。获得50%抑制效果所需的抗体浓度为IC50值。

2.4通过BIAcore分析抗IL-13抗体与人IL-13的亲和力
通过利用BIAcore 2000Biosensor(BIAcore AB)进行的表面等离子共振技术测定FWB03、FWB09、FWB11、FWB12、FWB13、FWB16作为人抗IL-13的IgG4抗体与人IL-13的亲和力。简言之,利用胺偶联试剂盒(BIAcore)使抗体与CM5传感器芯片偶联,表面密度约为500Ru,用HBS-EP缓冲液系列稀释IL-13,所获的系列稀释液(50nM-0.78nM)通过该传感器芯片表面。利用BIA评估3.1软件评估由上所获传感器记录,获得动力学数据。

实施例3抗IL-13抗体在小鼠哮喘模型中的药效
本实施例评价了FWB13在通过气管注射人类重组IL-13诱发Balb/c小鼠呼吸道反应模型上的体内药效学研究。
将29只小鼠分为5组,分别为
1.G1)Naive,n=5;
2.G2)IL-13+人IgG4组(不能特异性识别和结合IL-13的抗体),3mpk,n=8;
3.G3)IL-13+川隆单抗组(Tralokinumab),3mpk,n=8;
4.G4)IL-13+FWB13组,3mpk,n=8;
G2、G3、G4组小鼠在第一天和第三天腹腔注射0.2mL待测抗体,浓度为3mg/kg。第二天和第四天气管给药人重组IL-13,每只小鼠给药25μg,溶于PBS,体积为50μL。G1 Naive对照组小鼠气管喷雾施用PBS,不施用IL-13和待测抗体。
体内实验:
在实验的第五天,用浓度递增的乙酰胆碱(分别为0mg/mL、1.25mg/mL、2.5mg/mL、5mg/mL、10mg/mL、20mg/mL和40mg/mL)对小鼠进行雾化吸入刺激,并对其肺功能进行检测。Penh(增强停顿)是一个无量纲指标,通常用于评估气流进出呼吸道模式的改变。Penh已被广泛接受用于评估肺功能的变化,以及评价气道反应性的方法。测得的Penh值结果如表11所示。结果显示,在20mg/mL的乙酰胆碱刺激后,模型组G2与G1组相比统计学上即有显著性差异(P<0.001),显示人重组IL-13诱发的小鼠呼吸道反应模型构建成功。而在乙酰胆碱上升到40mg/mL浓度后,阳性对照抗体给药G3组与模型组G2组相比,Penh值显著降低(P<0.001),同时待测抗体FWB13 G4组与模型组G2组相比Penh值也显著降低(P<0.001),表明FWB13在体内实验中对小鼠被高浓度乙酰胆碱诱发的呼吸道反应penh值有显著的抑制作用。

病理实验:
第六天安乐死处死小鼠,取小鼠肺组织放到10%中性福尔马林中固定,保存。石蜡包埋后切成5μm厚度的切片进行PAS染色,并做病理分析。
PAS(Periodic Acid-Schiff stain)染色即过碘酸希夫氏反应,其化学反应的基本过程是通过过碘酸的氧化作用,使多糖暴露出醛基,醛基与无色碱性品红结合反应,于多糖存在的部位形成新的紫红色复合物,通过显微镜观察而对组织细胞内的糖原、糖蛋白或粘多糖等化学成分进行定位、定性和定量的研究。对于哮喘(气道炎症)模型来说,炎症导致粘多糖分泌增加,PAS阳性反应表明该部位的气道诱发了炎症反应。
染色结果用ImageScope软件进行分析,用来计算单个末端细支气管中的PAS阳性细胞即染成红色的杯状细胞和柱状上皮细胞的比例。
组织切片显示,对照组G1的PAS染色显示阴性,而模型组的炎症细胞染色明显,且细支气管中的粘液分泌量也明显增多(图1)。
如表13所示,组别G1(空白对照组)的PAS染色完全是阴性的;最强的信号(粘液分泌)出现在组别G2(有诱导物IL-13而无中和抗体组,PAS阳性细胞占比28.39%);川隆单抗组(Tralokinumab,组别G3)和FWB13组(组别G4)与疾病模型组(组别G2)相比,粘液分泌下降明显,PAS阳性细胞占比分别为15.09%和13.36%。病理实验结果表明,FWB13(G4组)能显著抑制PAS染色阳性细胞数的增高。

实施例4抗IL-13抗体在小鼠特应性皮炎模型中的药效
本实施例评价了FWB13在可诱导IL-13过表达的小鼠皮炎模型中的体内药效。R26-(2xIns-TRE-CMV-Il13-SV40-pA)F1代(内部构建)与转基因小鼠品系Tg(KRT5-rtTA)(购自JAX)交配,子代小鼠于6周前完成IL-13+和KRT5+双阳性Tg(+)鉴定,可通过饲喂含有Doxycycline(DOX,1mg/mL)的水诱导IL-13在表皮组织过表达从而引起特应性皮炎症状。
转基因小鼠品系R26-(2xIns-TRE-CMV-Il13-SV40-pA)由内部构建,首先构建ROSA26-(2xIns-TRE-CMV-IL13-SV40-pA)供体同源重组载体。该载体通过一对同源重组臂定点整合入小鼠C57 BL/6J基因组的ROSA26基因的内含子中,并在四环素应答元件(TRE)和CMV融合启动子的驱动下过量表达IL-13基因。该表达载体通过受精卵注射到F0代小鼠,鉴定后进一步与野生小鼠交配从而得到F1代小鼠。F1代小鼠与皮肤组织特异性(keratin5,角蛋白启动子)转基因小鼠品系Tg(KRT5-rtTA)(FVB背景)交配,子代小鼠可通过饲喂Doxycycline,特异性驱动IL-13基因在皮肤表皮组织中表达。
将Tg(+)雄性小鼠和雌性小鼠分别作为两种药效评估模型。
4.1 FWB13在Tg(+)雄性小鼠中的药效评估
将48只Tg(+)雄性小鼠分为6组,分别为
1.normal组,n=8;
2.vehicle组,PBS,i.p.,BIW(每周两次),n=8;
3.Tralokinumab,3mg/kg组(阳性对照低剂量组),i.p.,BIW,n=8;
4.Tralokinumab,10mg/kg组(阳性对照高剂量组),i.p.,BIW,n=8;
5.FWB13,3mg/kg组(待测抗体低剂量组),i.p.,BIW,n=8;
6.FWB13,10mg/kg组(待测抗体高剂量组),i.p.,BIW,n=8;
第1天、第4天、第8天和第11天,第3-6组实验组腹腔注射抗体药,vehicle组腹腔注射PBS。从第1天起给除normal组外的所有组小鼠持续饲喂Doxycycline(1mg/mL)水溶液,并于第13天停止饲喂Doxycycline水溶液。Normal组饲喂灭菌饮用水。于第7天、第11天、第13天和第15天进行表型评分,包括红斑(0-3分)、结痂(0-3分)和厚度(0-3分)三项加和,总分为0-9分。并用Two way ANOVA进行统计分析。评分(图2)显示阳性对照抗体药低剂量组Tralokimumab 3mg/kg与vehicle组在评分上没有显著性差异,高剂量组10mg/kg相比vehicle组在第7天和第10天有显著性差异,相对vehicle组分别可以降低评分56%和27%,因此确定以下实施例4.2实验中使用的阳性对照组抗体的剂量为10mg/kg。待测抗体FWB13低剂量组3mg/kg在第7天、第11天和第15天与vehicle组相比均有显著性差异,可分别降低评分59%、25%和22%。由此可知,3mg/kg剂量下,FWB13药效优于阳性对照抗体药Tralokinumab。高剂量组10mg/kg在第7天、第11天和第15天与vehicle组相比均有显著性差异,可降低评分72%、33%和23%;但与3mg/kg剂量组药效相似并无显著性差异,因此确定以下实施例4.2实验中使用的最高剂量组的抗体剂量为3mg/kg。第11天和第15天的各组小鼠的表型如图3所示。
4.2 FWB13在Tg(+)雌性小鼠中的药效评估
将47只Tg(+)雌性小鼠分为5组,分别为
1.normal组,n=7;
2.vehicle组,PBS,i.p.,BIW,n=10;
3.Tralokinumab组(阳性对照剂量组),10mg/kg,i.p.,BIW,n=10;
4.FWB13组,1mg/kg(待测抗体低剂量组),i.p.,BIW,n=10;
5.FWB13组,3mg/kg(待测抗体高剂量组),i.p.,BIW,n=10;
在第0天、第3天、第6天、第10天、第13天、第17天,第3-5组实验组腹腔注射抗体药,vehicle组腹腔注射PBS。从第1天起至第12天给除normal组外的所有组小鼠每天持续饲喂Doxycycline(1mg/mL)水溶液,并于第13天停止饲喂Doxycycline水溶液。Normal组饲喂灭菌饮用水。于第3天、第5天、第7天、第10天、第12天、第14天、第17天和第20天进行表型评分,包括红斑(0-3分)、结痂(0-3分)和厚度(0-3分)三项加和,总分为0-9分。并用Two wayANOVA进行统计分析。评分(图4)显示阳性对照高剂量组Tralokinumab 10mg/kg与vehicle组相比在第10天具有显著性差异,可降低评分29%,但随着病症的加重在其他时间点并无显著性差异。FWB13低剂量组1mg/kg组在第10天、第12天、第14天、第17天和第20天与vehicle组相比均有显著性差异,可分别降低评分28%、17%、22%、16%和22%。FWB13高剂量组3mg/kg组在第10天、第12天、第14天、第17天和第20天与vehicle组相比也具有显著性差异,可降低评分36%、32%、33%、33%和32%。FWB13 1mg/kg剂量组与阳性对照抗体药高剂量组Tralokinumab10mg/kg药效相似,FWB13 3mg/kg剂量组在第12天、第14天、第17天和第20天药效显著优于阳性对照Tralokinumab 10mg/kg剂量组。
第14天时小鼠表型如图5所示。除评分外,分别于第3、7、10、12、14天测定经皮水分流失(Trans-epithelial water loss,TEWL)值,如图6所示,经Two way ANOVA统计分析,Tralokinumab 10mg/kg与vehicle组相比无显著性差异。FWB13 1mg/kg与vehicle组相比,在第10天具有显著性差异,可减少TEWL值36%;FWB13 3mg/kg与vehicle组相比,在第7天、第10天和第20天显著减少TEWL 59%、63%和47%。与Tralokinumab 10mg/kg组相比,FWB133mg/kg剂量组可更加显著地改善表皮水分流失的状况。
实施例5通过ELISA方法测定抗IL-13抗体与人IL-13的亲和力
首先用包被液(PBS)稀释人IL13(Sino10369-HNAC,购买自北京义翘神州,其氨基酸序列为SEQ ID NO:182:MALLLTTVIA LTCLGGFASP GPVPPSTALR ELIEELVNIT QNQKAPLCNGSMVWSINLTA GMYCAALESL INVSGCSAIE KTQRMLSGFC PHKVSAGQFS SLHVRDTKIE VAQFVKDLLLHLKKLFREGR FN)至1μg/mL,每孔加入50μL,4℃孵育过夜。然后吸去抗原溶液,每孔加入200μL的3%BSA溶液,室温度孵育1小时;之后用PBST洗板3次。将表13所示的12个待测抗体(其重链恒定区和轻链恒定区以及重链和轻链可变区的框架区均相同)从50μg/mL进行10倍梯度稀释,共8个浓度点(包括空白对照),并将系列稀释的抗体加入到ELISA孔中,在37℃下孵育1小时,随后用PBST洗板3遍。加入辣根过氧化酶结合的二抗并在37℃下孵育1小时,最后用PBST洗板3次并加入50μL TMB反应15分钟。显色反应可以用1N盐酸(50μl)进行终止,并在读板机(M5)上检测450nm的吸光度值,以此来计算各个抗体对IL-13的亲和力(EC50)。实验结果如下表14以及图7-12所示。实验结果表明,FWB1313与人IL-13抗原的亲和力高于FWB1313B。

Claims (15)
1.一种特异性结合IL-13的抗体或其抗原结合片段,其包含重链CDR1、CDR2和CDR3以及轻链CDR1、CDR2和CDR3,所述重链CDR1、CDR2和CDR3以及轻链CDR1、CDR2和CDR3分别与选自下组的抗体的重链CDR1、CDR2和CDR3以及轻链CDR1、CDR2和CDR3序列具有至少80%,优选至少90%同一性:FWB03、FWB05、FWB09、FWB10、FWB11、FWB12、FWB13、FWB14、FWB15、FWB16、FWB17和FWB19。
2.根据权利要求1所述的特异性结合IL-13的抗体或其抗原结合片段,其包含选自下组的抗体的重链CDR1、CDR2和CDR3以及轻链CDR1、CDR2和CDR3:FWB03、FWB05、FWB09、FWB10、FWB11、FWB12、FWB13、FWB14、FWB15、FWB16、FWB17和FWB19。
3.根据权利要求1或2所述的特异性结合IL-13的抗体或其抗原结合片段,其包含重链可变区和轻链可变区,其中所述重链可变区和轻链可变区与选自下组的抗体的重链可变区和轻链可变区序列具有至少85%同一性,优选至少90%同一性,更优选95%同一性,最优选98%同一性:FWB03、FWB05、FWB09、FWB10、FWB11、FWB12、FWB13、FWB14、FWB15、FWB16、FWB17和FWB19。
4.根据权利要求3所述的特异性结合IL-13的抗体或其抗原结合片段,其包含选自下组的抗体的重链可变区和轻链可变区序列:FWB03、FWB05、FWB09、FWB10、FWB11、FWB12、FWB13、FWB14、FWB15、FWB16、FWB17和FWB19。
5.根据权利要求1或2所述的特异性结合IL-13的抗体或其抗原结合片段,其包含重链和轻链,其中所述重链和轻链分别与选自下组的抗体的重链和轻链序列具有至少80%,优选至少85%、更优选90%,最优选至少95%同一性:FWB03、FWB05、FWB09、FWB10、FWB11、FWB12、FWB13、FWB14、FWB15、FWB16、FWB17和FWB19。
6.根据权利要求5所述的特异性结合IL-13的抗体或其抗原结合片段,其包含选自下组的抗体的重链和轻链序列:FWB03、FWB05、FWB09、FWB10、FWB11、FWB12、FWB13、FWB14、FWB15、FWB16、FWB17和FWB19。
7.权利要求1-6中任一项的抗体,其中所述抗体是人抗体、人源化抗体、或嵌合抗体。
8.权利要求1-6中任一项的抗体,其中所述抗体是IgA、IgG和IgD。
9.权利要求1-6中任一项的抗原结合片段,其中所述抗原结合片段选自Fab片段、F(ab')片段、Fv片段、F(ab')2片段、单链抗体(scFV)和双抗体。
10.一种分离的核酸,其编码权利要求1-9中任一项中所述的抗体或其抗原结合片段。
11.一种表达载体,其包含权利要求10所述的分离的核酸。
12.一种宿主细胞,其包含权利要求10所述的分离的核酸或权利要求11所述的表达载体,优选地所述宿主细胞是真核宿主细胞,更优选所述宿主细胞是哺乳动物宿主细胞。
13.一种药物组合物,其用于治疗IL-13相关疾病或失调,其包含权利要求1-9所述的抗体或其抗原结合片段、权利要求10所述的分离的核酸、权利要求11所述的表达载体、或权利要求12所述的宿主细胞,所述药物组合物还包含药用载体。
14.一种组合物,其包含第一核酸和第二核酸,其中所述第一核酸编码权利要求1-9中任一项所述的抗体的重链,所述第二核酸编码权利要求1-9中任一项所述的抗体的轻链。
15.权利要求1-9中任一项所述的抗体或其抗原结合片段在制备用于治疗IL-13相关疾病或失调的药物中的用途,优选地,其中所述IL-13相关疾病或失调是哮喘或特应性皮炎。
