Abstract
The invention relates to the technical field of health care products, in particular to a preparation method of a supplement capable of relieving visual fatigue, which comprises the following components in parts by weight: natural lutein, zeaxanthin, astaxanthin, schizochytrium DHA, spirulina extract, vitamin A, vitamin E, vitamin B1, and vitamin B2; the invention adopts soft capsule preparation or tablet to effectively protect the easily oxidized components in the raw materials, has scientific composition, natural components, proper proportion, good taste and convenient taking, has the effects of supplying nutrient composition required by eyes and delaying oxidation, and is an ideal and high-end health food for relieving asthenopia.

Preferably, in step S1, the components are calculated according to the following weight portions: 30mg of natural lutein, 3mg of zeaxanthin, 3mg of astaxanthin, 400mg of schizochytrium DHA, 120mg of spirulina extract, vitamin A129ug, 12mg of vitamin E, 11.2mg of vitamin B and 21.2mg of vitamin B.
Preferably, in step S1, the natural lutein is 35 mg, the zeaxanthin is 4mg, the astaxanthin is 4mg, the schizochytrium DHA is 500mg, the spirulina extract is 130mg, the vitamin a is 130ug, the vitamin E13mg, the vitamin b 11.3mg, and the vitamin b 21.3mg.
Preferably, the method further comprises the following tablet preparation steps:
weighing 0.8g of lemon yellow, and dissolving in 120ml of distilled water;
adding 100g of gelatin, soaking overnight, fully dissolving in warm water, adding glycerol and sorbitol, and stirring to dissolve to obtain a solution A;

step S1, uniformly mixing the components according to the proportion for later use, putting 80g of linseed oil and 1g of beeswax in a proper amount into a 1L vacuum stirring barrel, heating to 70-80 ℃, dissolving, stirring and uniformly mixing for later use; linseed oil: beeswax =80: 1.
Step S2, based on step S1, as gelatin: glycerol: water = 2: 1: 2, taking gelatin, fully expanding, adding glycerol, heating, stirring to dissolve, standing and defoaming; filtering to remove impurities, mixing, maintaining the temperature of the glue solution at 55-65 deg.C, room temperature at 21-24 deg.C, and relative humidity at 40-45%;

In this embodiment, the method further comprises a tablet preparation step:

In this example, 150 g of D-sorbitol solution, 50 g of glycerol and 100g of a proper amount of purified water were placed in a 1L beaker and mixed to obtain a mixed solution;
after adding 50 g of high amylopectin to the mixture and mixing thoroughly, 100g of gelatin is added to the mixture with stirring, and the resulting mixture is heated in a water bath at about 60 ℃ to prepare a capsule shell. And (3) putting the finished product in a capsule shell according to a proper proportion to prepare the soft capsule.
Screening an optimal formula of the capsule skin:
after preliminarily determining the ranges of the ratio of glycerol to gelatin, the ratio of water to gelatin and the addition amounts of sorbitol and high amylopectin by consulting the data, the optimization is further carried out by using an L9(34) orthogonal experimental design (figure 2), and the obtained optimal formula is verified.
Product formula screening, and the used main raw materials and the use basis are as follows:
(1) astaxanthin;
(2) lutein/zeaxanthin;
(3)DHA;
(4) a vitamin;
(5) spirulina extract.
The optimal proportion of the formula of the capsule shell is as follows: as can be seen from the results of the orthogonal test and the range analysis in FIG. 3, the factor affecting the dissolution rate of the gelatin skin is glycerol/gelatin in turn>Water/gelatin>Sorbitol addition amount>The addition amount of the high amylopectin starch and the optimal formula of the soft capsule shell are A2B2C2D3。
Since the A2B3C3 combination was not found in the orthogonal test, a verification test was required. The gel was prepared according to formulation A2B3C3 and the dissolution rate of the gel was measured. The results are shown in FIG. 4.
As can be seen from FIG. 4, the verification test is closer to the prediction result, which indicates that the fitting condition of the test is better and the error of the test is small. Therefore, the analysis method meets the experimental requirements and can be used as a formula for optimizing the capsule shell. Thus, formulation A2B2C2D3, i.e. glycerol: gelatin is 0.5: 1; water: gelatin is 0.8: 1; the using amount of the sorbitol is 30 percent; the dosage of the high amylopectin is 10 percent, which is the best formula of the soft capsule shell.
In this embodiment, the method further comprises the preparation of a soft capsule:
adding gelatin, glycerol and water at a proper ratio into a gelatin barrel, heating to a proper sol temperature, stirring, vacuumizing by a vacuum pump to defoam and degas the gelatin solution, vacuumizing by the vacuum pump to defoam and degas the gelatin solution, observing the condition of the gelatin solution, filtering by a filter screen, placing into the gelatin barrel, keeping the temperature, standing, filling materials, pressing into capsules, and drying to obtain the soft capsule.
According to the result of the single-factor experiment, the optimal prescription of the content of the lutein soft capsule is optimized by adopting an orthogonal design experiment L9(34) by taking the dosage ratio (A) of the medicine and the linseed oil, the dosage (B) of the beewax and the dosage (C) of the soybean phospholipid as investigation factors. The factor level table is shown in FIG. 5:

35 mg of natural lutein, 4mg of zeaxanthin, 4mg of astaxanthin, 500mg of schizochytrium DHA, 130mg of spirulina extract, 130ug of vitamin A, 13mg of vitamin E, 11.3mg of vitamin B and 21.3mg of vitamin B.
In the above examples, the supplement product formulation is shown in figure 6. The results of the orthogonal test are shown in FIG. 7.
Referring to fig. 7, it can be known from the visual analysis and the analysis of variance that, when the sedimentation ratio is used as the inspection index, the influence on the sedimentation ratio is sequentially the dosage ratio of the drug to the linseed oil > the dosage of the beeswax > the dosage of the soybean lecithin, and the optimal process is A1B1C3, namely, the powder: medicinal flaxseed: beeswax: soybean phospholipids = 15: 80: 1: 4.
