Abstract
The invention relates to a composition for preventing and improving chronic diseases caused by selenium deficiency and a preparation method thereof, wherein the composition comprises the following raw material components: 6-14 parts of selenium-rich oyster mushroom powder, 15-25 parts of agrocybe aegerita powder, 18-30 parts of wheat oligopeptide, 13-17 parts of spina date seed powder and 0.01-0.03 part of vitamin E. The composition provided by the invention has the characteristics of good selenium supplementing effect and high safety, and can be used for preventing and improving various symptoms of chronic diseases caused by selenium deficiency, such as blurred vision, hypodynamia, abdominal pain, dizziness, insomnia, anorexia, chest distress, inappetence, liver discomfort, hyperglycemia, hypertension, cataract, obesity and the like.

Technical Field
The invention belongs to the technical field of foods, and particularly relates to a composition for preventing and improving chronic diseases caused by selenium deficiency and a preparation method thereof.
Background
With the aging population, the urbanization and the accelerated industrialization process and the prevalence of behavior risk factors, the cardinality of chronic disease patients in China is still continuously expanded, and thus the death rate of chronic diseases is continuously increased. The death caused by chronic diseases in 2019 accounts for 88.5% of the total death, the death rate of cardiovascular and cerebrovascular diseases, cancers and chronic respiratory diseases is 80.7%, and the prevention and control work still faces huge challenges. Among the life elements, the trace element selenium has prominent performance in preventing and treating chronic diseases. Selenium is one of essential trace elements, and has effects of promoting growth, protecting cardiovascular and myocardial health, removing heavy metal toxicity, protecting health and eyesight of visual organs, resisting oxidation, virus, cancer and aging, and enhancing immunity. However, 71% -78% of the regions in the world are deficient in selenium or are short of selenium, and 70% of the regions in China are deficient in selenium. According to the research of the world health organization, the daily intake of selenium for adults is 50-250 micrograms. The selenium intake of most people in China is about 30-40 micrograms, which is far from the requirement of 200 micrograms for preventing diseases of human bodies. The dietary structure of food alone and unscientific also affects the absorption of selenium by the human body. And as the people grow older, the absorption capacity of the human body for selenium is reduced, but the consumption of selenium is increased, so that the middle-aged and the elderly are more prone to lack of selenium nutrients. Selenium deficiency or low selenium can affect normal physiological metabolism of organism, resulting in decreased immunity, and can be used for treating more than 40 diseases, such as cancer, tumor, cataract, sciatica, asthma, diabetes, sexual disorder, etc. Properly increasing the intake of selenium has important significance for preventing certain diseases and maintaining the health.
Selenium levels in the human body are divided into three cases: the normal growth and development of the organism are maintained by the concentration of trace selenium; the medium selenium concentration maintains stable function; excessive selenium concentration can cause toxic effects, such as abnormal nail loss, skin lesions, and damaged nervous system. Selenium mainly exists in nature in the forms of selenate, selenite, elemental selenium, organic selenium after biological conversion and the like. The human body cannot synthesize selenium by itself and needs to be taken from the outside. Selenium in foods is generally absorbed into the body through the intestinal tract. The absorption efficiency of selenium is related to the form of selenium, and selenomethionine, methyl selenocysteine, selenate and selenite sequentially exist in the absorption rate from high to low. The dietary selenium supplement is mainly from plant-derived and animal-derived foods. The plant can convert the absorbed inorganic selenium into organic selenium such as selenocysteine, selenomethionine, methyl selenocysteine and the like which are easier to be utilized by human bodies, and the plant also has the capability of metabolizing toxic selenium compounds, so that the bioavailability and the safety of the plant source selenium-enriched food selenium are high. At present, most of selenium supplement products for improving chronic diseases in the market focus on the content of selenium supplement, and the absorption and utilization rate of selenium and the repair of damaged cells are neglected.
Disclosure of Invention
The invention aims to provide a composition for preventing and improving chronic diseases caused by selenium deficiency and a preparation method thereof, wherein the composition adopts selenium-enriched oyster mushroom powder, agaricus tabularis powder, wheat oligopeptide, spina date seed powder and vitamin E as main raw materials and is subjected to proper weight proportion, so that the components have synergistic effect, and the composition can achieve better effects of preventing and improving the chronic diseases caused by selenium deficiency starting from the aspects of supplementing easily absorbed organic selenium, adjusting metabolic level, repairing damaged cells, reducing free radical damage, improving immunity and the like.

The oyster mushroom is edible fungus with strong suitability, high yield and dual purposes of medicine and food in artificial cultivation, contains protein, total sugar, dietary fiber, vitamin E, carotene, nicotinic acid, folic acid, pantothenic acid, vitamin A, vitamin B12, vitamin B1, vitamin B2, vitamin B6, vitamin C, vitamin D, phosphorus, potassium, sodium, copper, zinc, selenium, magnesium and other nutritional ingredients, and also contains threonine, valine, methionine, isoleucine, leucine, phenylalanine, lysine, aspartic acid, serine, glutamic acid, glycine, alanine, cystine, tyrosine, histidine, arginine and other free amino acids. The oyster mushroom contains flavonoid, organic acid, chitin and other active components beneficial to human body, and has health promotion effects of resisting tumor, resisting oxidation, lowering cholesterol, regulating immune system, resisting bacteria and virus, protecting liver, promoting brain development, etc. Selenium in the selenium-rich oyster mushroom exists in the organic forms of protein selenium and polysaccharide selenium, and the selenium-rich oyster mushroom containing selenium element with proper concentration has the functions of eliminating free radicals, resisting inflammation, resisting tumor, resisting lipid oxidation, prolonging the service life, improving the immunity of the organism and the like.
The Agrocybe aegerita contains 18 amino acids, 8 amino acids essential for human body, and multiple bioactive substances such as polysaccharide, ergosterol, and flavone, and its fruiting body polysaccharide can improve muscle fatigue state, and has obvious activity of lowering blood sugar. The Agrocybe aegerita water extract can effectively improve the activity of liver and blood superoxide dismutase and reduce the content of malondialdehyde, thereby reducing the degree of liver cell peroxidation injury.
The wheat oligopeptide is a mixture of micromolecular peptides prepared by hydrolyzing wheat protein serving as a raw material. The small peptide molecules can be absorbed and utilized by human bodies more easily and quickly through small intestinal membranes, the absorption utilization rate of protein can be effectively improved, and the small peptide molecules are more effective than the absorption of single amino acid. The wheat oligopeptide has strong antioxidant activity, can improve the absorption and utilization of protein by organisms, and can effectively protect and repair gastrointestinal mucosa. Meanwhile, the wheat oligopeptide can effectively reduce the damage of the non-steroidal anti-inflammatory drug to the gastrointestinal tract.
The spina date seed is sweet in taste and neutral in nature, and has the functions of calming heart, soothing nerves, arresting sweating and nourishing liver. Can be used for treating vexation, insomnia, palpitation, dizziness, dim eyesight, etc. The spina date seed contains saponin, triterpenes, flavonoids, alkaloids, fatty acids, steroidal compounds, daucosterol, phenolic acid micromolecules, ferulic acid, vitamin C, zizyphus jujube polysaccharide, mineral substances, amino acids essential to human bodies and plant alcohol, and has the effects of calming and hypnotizing, relieving pain, resisting convulsion, resisting arrhythmia, reducing blood fat, enhancing immunity, reducing blood pressure, resisting myocardial ischemia and the like.
Vitamin E is a fat-soluble vitamin, and has effects of resisting infertility, oxidation and cancer, improving animal organism immunity, and reducing tumor cell growth speed. The vitamin E is absorbed by animals and is mainly distributed in cell biomembranes, can prevent the oxidation of vitamin A, selenium, two kinds of sulfur amino acids and vitamin C, inhibit or slow down the oxidation and peroxidation of unsaturated fatty acid in vivo, can weaken or disappear the toxicity of free radicals when the animal brain is in ischemia, reduce the infarct area, reduce the damage of neurons and play a role in protecting the brain. Vitamin E ensures the normal function of cells by protecting and maintaining the integrity and stability of cells and organelles, and is used as an immunomodulator to regulate and control the endocrine of an animal body, such as regulating the synthesis of cell regulators, prostaglandins, prothrombin and somatotropin, and finally influences the immune performance of the body.
On the basis of the composition for preventing and improving chronic diseases caused by selenium deficiency, conventional auxiliary components including but not limited to sweeteners and essences can be added, the sweeteners are preferably sorbitol, and the adding amount can be 24-38 parts.
The composition for preventing and improving chronic diseases caused by selenium deficiency is characterized in that selenium-rich oyster mushroom powder is obtained by drying selenium-rich oyster mushroom fruiting bodies at low temperature and carrying out superfine grinding; and/or: the agaricus tabularis powder is obtained by extracting agaricus tabularis fruiting bodies with water and precipitating with ethanol; and/or: the wheat oligopeptide is obtained by carrying out enzymolysis on wheat protein powder under the action of alkaline protease: and/or: the wild jujube kernel powder is obtained by stir-frying and crushing wild jujube seeds.
Preferably, the selenium-rich oyster mushrooms are high-quality oyster mushrooms which are planted in Chinese academy of agricultural sciences and rely on the technology of eight-number-eight organic selenium conversion in a self-built selenium-rich oyster mushroom planting base of Zhongnong and Spaceflight; drying at low temperature until the water content is less than 8%, and micronizing to powder of no less than 200 meshes.
Preferably, the dried willow mushroom powder is prepared by the following method: drying and crushing the fruiting bodies of the agaricus tabularis to obtain agaricus tabularis powder; taking the pleurotus ostreatus powder, adding water according to the feed-liquid ratio of 1g:5-20mL, and carrying out reflux extraction in a constant-temperature water bath at 100 ℃ for l-3 h; centrifuging the extracting solution for 3-5min at 5500-6500r/min to obtain supernatant and precipitate; adding water into the precipitate for uniformly mixing, repeating the extraction and the centrifugation for 1-2 times, and collecting the supernatant, wherein the water is 1g:8-12 mL; mixing the supernatants, adding 90-95% ethanol (1 mL:4-6 mL) for precipitation, centrifuging, collecting supernatant, and drying.
Preferably, the wheat oligopeptide is prepared by the following method: adding water into wheat protein powder according to a feed-liquid ratio of 1g:5-7mL, heating in a water bath at 95-100 ℃ for 5-10min, cooling to 40 ℃, adjusting the pH to 8-10, adding 5-9 wt% of trypsin, carrying out enzymolysis for 3-5h, heating to 65 ℃, adjusting the pH to 6-9, adding 5-9 wt% of trypsin, carrying out enzymolysis for 2-4h, carrying out enzyme deactivation, filtering, concentrating, spray drying and sieving to obtain the wheat protein powder.
Preferably, the wild jujube kernel powder is prepared by the following method: parching semen Ziziphi Spinosae until swelling and color is slightly darkened, pulverizing, and sieving.
The preparation of the composition for preventing and improving the chronic diseases caused by selenium deficiency is not particularly limited, and the raw material components can be uniformly mixed by referring to the conventional mixing technology.
The invention has the beneficial effects that:
1. the composition provided by the invention has the following formula mechanism: 1) the selenium in the selenium-rich oyster mushroom powder is plant-derived organic selenium, has high absorption utilization rate and high safety, and can quickly supplement selenium required by a human body under the combined action of the selenium and the polysaccharide in the pleurotus ostreatus powder and the small molecular peptide in the wheat oligopeptide, regulate carbohydrate metabolism and lipid metabolism, reduce endoplasmic reticulum stress reaction, further reduce the damage to cells and promote the absorption of nutrient substances; meanwhile, the preparation provides raw materials for the growth and repair of damaged cells, accelerates the repair of cells, and relieves the symptoms of dizziness, anorexia, chest distress, inappetence and the like. 2) The synergistic effect of organic selenium, oyster mushroom polysaccharide and flavonoid substances in the selenium-rich oyster mushroom powder, polysaccharide in the pleurotus ostreatus, wheat oligopeptide, vitamin C in the spina date seed powder, saponin and vitamin E effectively removes free radical substances in the body, reduces oxidative stress reaction of cells, and avoids oxidative damage of biological membranes and damage of free radicals to DNA; inhibiting the release of inflammatory cytokines and apoptosis of cells, thereby protecting important membrane structures in the cells from being damaged and maintaining the normal functions of the cells; promoting proliferation and differentiation of lymphocyte and production of cytokine, increasing cell cytotoxicity, enhancing phagocytosis of phagocyte, improving immunity, and effectively improving symptoms such as asthenia, fatigue, eye discomfort, abdominal pain, insomnia, etc.

The composition provided by the invention has an effective rate of improving general symptoms (blurred vision, hypodynamia, bellyache, dizziness, insomnia, anorexia, chest distress, inappetence, liver discomfort) and other symptoms (hyperglycemia, hypertension, cataract and obesity) of the selenium-deficiency chronic disease by 82.9%. Has the characteristics of good selenium supplementing effect and high safety, and can be used for preventing and improving chronic diseases caused by selenium deficiency for long-term eating.

the preparation method of the composition for preventing and improving the chronic diseases caused by selenium deficiency comprises the following steps:
(1) preparing selenium-rich oyster mushroom powder: selecting high-quality oyster mushroom strains which are selected from a self-built selenium-rich oyster mushroom planting base of Zhongnong and Spirongnong and rely on the technology of eight-number cultivation and organic selenium conversion in Chinese academy of agricultural sciences, selecting and removing the root and pedicle parts of edible fungi, cleaning and drying the selenium-rich oyster mushroom, drying at low temperature until the moisture content is less than 8%, carrying out ultramicro crushing to 200 meshes, carrying out vibrating sieve, and carrying out irradiation sterilization to obtain selenium-rich oyster mushroom powder, wherein the organic selenium content of the obtained selenium-rich oyster mushroom powder is up to 220 ug/g;
(2) preparing the dried willow mushroom powder: drying and crushing the fruiting bodies of the agaricus tabularis to obtain agaricus tabularis powder; adding water into the pleurotus ostreatus powder according to the feed-liquid ratio of 1g to 5mL, and performing reflux extraction lh in a constant-temperature water bath at 100 ℃; centrifuging the extractive solution at 5500r/min for 5min to obtain supernatant and precipitate; adding water into the precipitate for uniformly mixing, repeating the extraction and the centrifugation for 1 time, and collecting supernatant, wherein the water is 1g to 8 mL; mixing the supernatants, adding 90% ethanol (1 mL:4 mL) for precipitation, centrifuging to obtain supernatant, drying, and sieving to obtain Agrocybe aegerita powder;
(3) preparing wheat oligopeptide: adding water into wheat protein powder according to a feed-liquid ratio of 1g:5mL, heating in a water bath at 95 ℃ for 10min, cooling to 40 ℃, adjusting the pH to 8, adding 5 wt% of trypsin, performing enzymolysis for 3h, heating to 65 ℃, adjusting the pH to 6, adding 9 wt% of trypsin, performing enzymolysis for 4h, inactivating enzymes, filtering, concentrating, spray-drying and sieving to obtain wheat oligopeptide, wherein the peptide content in the obtained wheat oligopeptide powder is 80%, and the average molecular weight of the wheat oligopeptide is 450 Da;
(4) preparing wild jujube kernel powder: parching semen Ziziphi Spinosae until swelling, slightly darkening, pulverizing, and sieving to obtain semen Ziziphi Spinosae powder;

(3) preparing wheat oligopeptide: adding water into wheat protein powder according to the feed liquid ratio of 1g:7mL, heating in a water bath at 100 ℃ for 5min, cooling to 40 ℃, adjusting the pH to 10, adding 9 wt% of trypsin, carrying out enzymolysis for 5h, heating to 65 ℃, adjusting the pH to 9, adding 5 wt% of trypsin, carrying out enzymolysis for 2h, carrying out enzyme deactivation, filtering, concentrating, spray drying and sieving to obtain wheat oligopeptide;
(4) preparing wild jujube kernel powder: parching semen Ziziphi Spinosae until swelling, slightly darkening, pulverizing, and sieving to obtain semen Ziziphi Spinosae powder;
(5) mixing: mixing selenium-rich Pleurotus Ostreatus powder, herba Agrocybe aegeritae powder, semen Tritici Aestivi oligopeptide, semen Ziziphi Spinosae powder, vitamin E, and sorbitol to obtain mixture.
(6) And (3) packaging: packaging the mixture, and making into 5 g/bag to obtain composition for preventing and improving chronic diseases caused by selenium deficiency.

The comparative example provides a composition for preventing and improving chronic diseases caused by selenium deficiency, which is prepared from the following raw materials: 20 parts of agaricus tabularis powder, 25 parts of wheat oligopeptide, 15 parts of wild jujube kernel powder, 0.02 part of vitamin E and 40 parts of sorbitol. The preparation method is the same as example 3.
Comparative example 3
The comparative example provides a composition for preventing and improving chronic diseases caused by selenium deficiency, which is prepared from the following raw materials: 10 parts of selenium-rich oyster mushroom powder, 20 parts of agaricus tabularis powder, 25 parts of wheat oligopeptide, 0.02 part of vitamin E and 45 parts of sorbitol. The preparation method is the same as example 3.
At least one of the general symptoms (blurred vision, hypodynamia, bellyache, dizziness, insomnia, anorexia, chest distress, inappetence and liver discomfort) and other symptoms (hyperglycemia, hypertension, cataract and obesity) which are collected with the selenium-deficiency chronic disease totals 140 people, the age is between 30 and 70 years old, 75 men and 65 women are randomly divided into 4 groups, namely a test group, a control group 1, a control group 2 and a control group 3, and 35 people are respectively selected. And (4) invalidation: 13 main clinical symptoms of blurred vision, hypodynamia, bellyache, dizziness, insomnia, anorexia, chest distress, inappetence, discomfort in liver area, hyperglycemia, hypertension, cataract and obesity are not obviously relieved or disappeared or have adverse reactions.
The effective rate is the percentage of the sum of the improved and improved people and the total people of each group.

The effective rates of the test group (composition of example 3), the control group 1 (composition of comparative example 1), the control group 2 (composition of comparative example 2), and the control group 3 (composition of comparative example 3) were 82.9%, 68.6%, 65.7%, and 77.1%, respectively. Compared with the control group 1, the effective rate of the test group is improved by 14.3 percent, compared with the control group 2, the effective rate of the test group is improved by 17.2 percent, and compared with the control group 3, the effective rate of the test group is improved by 5.8 percent. In the test group, the improvement of dizziness, hypodynamia, insomnia and blurred vision is particularly prominent, and the number of the effective people is 54.3 percent. The results show that the compositions provided by the invention can synergistically improve the effect of preventing and improving chronic diseases caused by selenium deficiency through interaction.
Typical case of using effect

8-12 parts of selenium-rich oyster mushroom powder, 18-23 parts of willow pine mushroom powder, 20-28 parts of wheat oligopeptide, 14-16 parts of spina date seed powder and 0.01-0.02 part of vitamin E.

and/or: the agaricus tabularis powder is obtained by extracting agaricus tabularis fruiting bodies with water and precipitating with ethanol;
and/or: the wheat oligopeptide is obtained by carrying out enzymolysis on wheat protein powder under the action of alkaline protease:
and/or: the wild jujube kernel powder is obtained by stir-frying and crushing wild jujube seeds.

The composition for preventing and improving chronic diseases caused by selenium deficiency as claimed in claim 5, wherein the selenium-rich Pleurotus Ostreatus is selected from high quality Pleurotus Ostreatus based on the technology of planting No. eight transformed organic selenium in Chinese farm courtyard at self-built selenium-rich Pleurotus Ostreatus planting base of Zhongnong; drying at low temperature until the water content is less than 8%, and micronizing to powder of no less than 200 meshes.

adding water into wheat protein powder according to a feed liquid ratio of 1g:5-7mL, heating in a water bath at 95-100 ℃ for 5-10min, cooling to 40 ℃, adjusting the pH to 8-10, adding 5-9 wt% of trypsin, performing enzymolysis for 3-5h, heating to 65 ℃, adjusting the pH to 6-9, adding 5-9 wt% of trypsin, performing enzymolysis for 2-4h, inactivating enzyme, filtering, concentrating, spray drying, and sieving to obtain the wheat protein powder.

The composition for preventing and improving chronic diseases caused by selenium deficiency according to claim 5, wherein the Zizyphi Spinosae semen powder is prepared by the following method: parching semen Ziziphi Spinosae until swelling and color is slightly darkened, pulverizing, and sieving.
