Abstract
The invention belongs to the technical field of selenium-rich tea planting and processing, and provides selenium-rich green tea with a blood pressure lowering effect and a selenium-rich planting and re-drying aroma-improving processing method thereof. The selenium element in the tea is greatly improved by applying the glucosamine selenium fertilizer aqueous solution to the leaf surfaces or the roots of the tea trees; the picked fresh tea leaves are made into the selenium-rich green tea through the working procedures of withering, steam de-enzyming, rolling and deblocking, re-baking and aroma raising and the like. The selenium fertilizer glucosamine selenium adopted by the invention is organic selenium, has no accumulated toxicity to human body, and is safer compared with inorganic selenium fertilizer; compared with the prior art of enriching the selenium in the tea, the selenium enrichment capacity of the tea is stronger. In-vitro experiments show that the selenium-rich green tea has a remarkable blood pressure reducing effect, and the blood pressure reducing activity of the selenium-rich green tea is obviously higher than that of common green tea. The selenium-rich green tea prepared by the invention not only supplements selenium element necessary for human body, but also can assist in maintaining blood pressure balance after being drunk for a long time, and has good application prospect in hypertension patient groups.

The invention belongs to the technical field of selenium-rich tea planting and processing, and relates to selenium-rich green tea with a blood pressure reducing effect and a selenium-rich planting and re-drying aroma-improving processing method thereof.
Background
Tea is the second largest beverage in the world next to water, mainly comprises tea polysaccharide, tea polyphenol, caffeine, mineral substances, amino acids and carbohydrates, has multiple functions of exciting central nerves, improving immunity, resisting oxidation, regulating glycolipid metabolic disturbance and the like, and meets the requirements of different people. Tea consumption helps to prevent or reduce the risk of cardiovascular disease, especially hypertension. Polyphenols extracted from green tea can reduce blood pressure of spontaneous hypertensive rats. The tea rich in gamma-aminobutyric acid can lower blood pressure of rats fed with high-salt diet, and prevent hypertension.
Selenium (Se) is a trace element necessary for human body, is closely related to health, and can also enhance the capabilities of tea in resisting diseases and insect pests, reducing fluorine, resisting heavy metal pollution and the like. Selenium cannot be synthesized in the human body and cannot be stored in the human body for a long time, and needs to be continuously taken from the outside. Relevant researches show that the recommended intake of the adults in China to the selenium is 60-400 mug per day, the average intake of the residents in China is only 44.6 mug per day, the intake of the selenium in 1 hundred million multi-person diets in China is insufficient, and the health level of the people is seriously threatened. The selenium-rich tea can effectively supplement the trace element selenium required by human body by drinking.
The natural selenium-rich tea is produced only in a few selenium-rich areas on the earth, and has the defects that the content of selenium in tea is difficult to control, and standardized production cannot be carried out. in this embodiment, the common green tea obtained by the same processing method without applying selenium fertilizer in the same experimental tea garden is used as a control, and the selenium-rich green tea prepared in embodiment 1 is used as an experimental example to verify the blood pressure lowering activity of the product of the present invention:
1. the content of selenium in the selenium-enriched green tea is measured by adopting a hydride-atomic fluorescence method (GB 5009.93-2017):
the analysis result shows that the selenium content of the selenium-rich green tea is 2.09 +/-0.12 mg/kg, which is obviously higher than the selenium content of the common green tea by 0.23 +/-0.05 mg/kg.
2. The in vitro blood pressure lowering activity experiment of the selenium-rich green tea adopts an Angiotensin Converting Enzyme (ACE) activity inhibition experiment: respectively pulverizing common green tea and selenium-rich green tea, ultrasonic extracting with 30 times of hot water (100 deg.C) for 15min, and repeating the extraction for 2 times. Centrifuging, mixing the supernatants, rotary evaporating for concentration, and lyophilizing to obtain common green tea extract and selenium-rich green tea extract. Common green tea extract and selenium-rich green tea extract samples to be tested are dissolved in HEPES buffer solution (pH 8.3), buffer solution is used as blank, and ACE enzyme activity A and blank ACE enzyme activity B of the samples are respectively determined.
ACE inhibition was calculated according to the following formula:
the results are shown in tables 1 and 2.

Angiotensin converting enzyme plays an important role in the regulation of hypertension, and angiotensin converting enzyme can convert angiotensin i, which has a very low activity, into angiotensin ii, which has a strong vasomotor activity. Inhibition of angiotensinConvertase helps to inhibit the activity of angiotensin converting enzyme and reduce the production of angiotensin, thus causing vasodilatation and lowering blood pressure. From table, the ACE inhibition ratio IC of the selenium-rich green tea water extract50Is far lower than the common green tea water extract, has obvious ACE inhibiting effect and has potential blood pressure lowering effect.
3. The in vivo blood pressure lowering activity experiment of the selenium-rich green tea comprises the following steps:
(1) all animal experiments are strictly carried out according to the ethical standards of experimental animals of the Shanghai university Committee for the ethical and use of experimental animals (approval No. A2020080) and the national guidelines for welfare of experimental animals, so as to improve the welfare of animals and reduce the pain of experimental animals to the greatest extent. 24 (6/group) 8-week-old male Wistar rats were purchased from Shanghai Spiker laboratory animals GmbH and housed in Shanghai university of transportation animal laboratories. All rats had free access to food and water and the rat environment was kept at 25 + -1 deg.C, 70-75% humidity and 12h alternating light and dark.
Rats were acclimatized for 1 week prior to the experiment and then randomly divided into 4 groups, a blank group (normal diet + distilled water), a model group (8% high salt diet + distilled water), a normal green tea group (8% high salt diet +500mg normal green tea extract/kg rat/day) and a selenium-rich green tea group (8% high salt diet +500mg selenium-rich green tea extract/kg rat/day).
(2) The weight of the rats was measured every 3 weeks (initial, middle and late) during the feeding period using an electronic scale, and the results are shown in Table 3. The blood pressure of the rat is measured by a noninvasive caudal artery pressure method, the rat is placed in a 35 ℃ cage for 30min, after the rat is stabilized, a pulse and pressure sensor is placed at the tail of the rat to pressurize, the blood pressure value corresponding to the first pulse appearing after the pulse disappears is the systolic pressure, 3 continuous readings with the blood pressure value less than or equal to 10mmHg are recorded, the average value is taken, and the result is shown in table 4.
TABLE 3 mean body weight of rats in each group
Note: the values marked with different letters (a-b) in the same column differ significantly (p < 0.05).
TABLE 4 mean blood pressure values of rats in each group
Note: the values marked with different letters (a-c) in the same column differ significantly (p < 0.05).
As shown in table 3, tea leaves continuously intervened in rats for 9 weeks without significant effect on their body weight. The late blood pressure of the model group increased by 20.90% compared with the early blood pressure, while the late blood pressure of the common green tea group increased by 10.13% and 5.80% compared with the selenium-rich green tea group (Table 4). Compared with the model group, the blood pressure of the common green tea and the selenium-rich green tea group is obviously reduced, and the blood pressure regulating effect of the selenium-rich green tea group is superior to that of the common green tea group.
(3) Elevated blood pressure can affect cellular oxidative stress and inflammation and can lead to vascular dysfunction. After 9 weeks of continuous feeding of aqueous tea extract to rats, 8% chloral hydrate was used for anesthesia, blood was taken from the abdominal aorta, and serum oxidative stress and inflammatory factor levels including oxidative stress (GPX), inflammation (CRP), endothelial function (eNOS) were analyzed. As shown in table 5, high salt intake resulted in a decrease in GPX levels in serum and an increase in CRP levels, indicating an increase in oxidative stress and inflammation; at the same time, high salt diet resulted in a decrease in eNOs content, indicating that endothelial dysfunction was more severe.
TABLE 5 serum indices of rats in each group
Note: the values marked with different letters (a-c) in the same column differ significantly (p < 0.05).
Compared with a model group, the level of GPX is respectively increased by 20.64 percent and 37.86 percent through the intervention of water extracts of common green tea and selenium-rich green tea; the level of eNOS increased by 28.34%, 80.06%, respectively; the level of CRP was reduced by 10.94%, 46.56%, respectively. These results indicate that the selenium-rich green tea can effectively relieve oxidative stress, inflammation and endothelial dysfunction caused by blood pressure rise, and the relieving effect of the selenium-rich green tea is stronger than that of common green tea.
Example 6:
in the embodiment, the fresh tea leaves with selenium fertilizer applied in the embodiment 1 are compared with the traditional selenium-rich green tea prepared by a common green tea processing technology (green removing, kneading and drying), and the redried selenium-rich green tea prepared by a redrying technology is used as a test example to perform sensory evaluation (GB/T23776-.
As shown in fig. 2, sensory evaluation results show that both the traditional selenium-rich green tea and the redried selenium-rich green tea have strong fragrance, the traditional selenium-rich green tea mainly has faint scent with a little grass smell, and the redried selenium-rich green tea mainly has sweet fragrance with a little caramel fragrance. On the whole, compared with the traditional selenium-rich green tea, the re-baked selenium-rich green tea has stronger fragrance and more lasting tea fragrance.
