Abstract

本发明提供了一种能高亲和力地识别EB病毒gp350蛋白的中和性单克隆抗体。所述抗体不仅亲和力高,而且能够阻断gp350蛋白和其配体CR2的结合。本发明所提供的中和性单克隆抗体不仅具有良好的体外中和EB病毒感染的活性,同时在体内能够清除EB病毒颗粒和预防EB病毒感染多个实体器官和组织。本发明还提供了所述中和性单克隆抗体与靶向CD89的单克隆抗体组成的双特异性抗体。该双特异性抗体能够明显的杀死EB病毒阳性的淋巴瘤细胞,抑制肿瘤细胞的生长和转移,可用于移植后淋巴增殖性疾病(post‑transplant lymphoproliferative disorders,PTLD)、淋巴增殖性疾病(lymphoproliferative disorders,LPD)、单核细胞增多症、嗜血综合征(HLH)、淋巴瘤的预防和治疗。

技术领域
本发明属于生物技术领域,具体涉及识别EB病毒gp350蛋白的高亲和力单克隆抗体及其应用。
背景技术
EB病毒(Epstein-Barr virus,EBV)对人体的感染率极高,其潜伏感染与活化与人类多种肿瘤类型的发生密切相关,可引起鼻咽癌(nasopharyngeal carcinoma,NPC)、胃癌、传染性单核细胞增多症(IM)等疾病。EBV已被证明是造成全球人类癌症负担约百分之一的病原体。尤其是,EBV感染与淋巴和上皮来源的肿瘤形成有关。淋巴来源的肿瘤包括地方性伯基特淋巴瘤(eBL)和霍奇金淋巴瘤(HL),以及上皮来源的鼻咽癌和胃癌。此外,多发性硬化症(MS)是一种涉及EBV感染的炎症性自身免疫性疾病,在经历传染性单核细胞增多症的个体患MS风险较高(Ascherio and Munger 2010)。
gp350是EBV上最丰富的病毒包膜糖蛋白,可以与B细胞表面的受体CR2结合从而引发B细胞感染,从而会形成长期的潜伏感染,从而引起相关的肿瘤疾病的发生。因此,gp350一直是用于开发EBV预防性疫苗的主要抗原。利用gp350的抗体预防和治疗EBV相关疾病的发生近年已经有相关报道,例如US2020/0055924,发明了针对于EBVgp350的单克隆抗体联合其他多肽位点来预防EBV感染和相关疾病的发生;US20200255499(昆士兰医学研究所理事会)发明了名称为“EBV抗体及其用途”,其涉及gp350重组的人源化抗体可应用于针对EBV被动免疫人类和/或治疗或预防EBV相关疾病,同时该抗体或抗体片段也可用于检测爱泼斯坦巴尔病毒;US2021/0115113,发明了名称为“EBV抗体及其用途”,同样涉及gp350重组的抗体、免疫原性肽、缀合物和药物组合物可用于治疗或预防EBV感染和EBV相关病症和疾病。EBV的感染是通过病毒表面的糖蛋白gp350与B细胞表面的受体CR2结合,阻断gp350与CR2的结合可以有效地阻断EBV感染细胞,因此,gp350可以作为靶向治疗EBV相关疾病的理想靶点。
双特异性抗体不仅可用于肿瘤治疗,还可运用于病毒的治疗,如HIV、ZiKa和登革热病毒等。双特异性抗体的靶点主要是病毒或者宿主的抗原表位,同时靶向免疫细胞的表位,有利于运用自身的免疫细胞(T细胞、NK细胞、单核巨噬细胞等)杀灭病毒和相关感染细胞。双特异性抗体在抗病毒治疗方面起步较晚,目前未发现EBV相关的双特异性抗体的研究报道,因此,研发双特异性抗体,抑制和清除EBV,治疗EBV相关的肿瘤,是治疗EBV相关疾病的重要的策略。
发明内容
本发明的目的在于提供新型的中和EB病毒侵染的单克隆抗体,所提供的抗体包括抗体片段(例如单链可变区片段(scFv))和改造的双特异性抗体。还提供了包括特异性结合gp350的抗体、编码抗体的核酸分子、包含所述核酸分子的表达载体以及表达所述核酸分子的分离的宿主细胞。
本发明具体技术方案如下:
一种识别EBV gp350的单克隆抗体,包括重链可变区和轻链可变区,(1)所述抗体的重链可变区具有SEQ ID NO:1的27-36位、51-66位和99-111位氨基酸残基所示的互补决定区;(2)所述抗体的轻链可变区具有SEQ ID NO:2的25-35位、51-57位和90-98位氨基酸残基所示的互补决定区。
本发明所述抗体是单链抗体、双链抗体、单克隆抗体或嵌合抗体。
本发明所述的单克隆抗体可以为任何同种型。可以为例如IgM或IgG抗体,例如IgG1或IgG2。
本发明所述的抗体可以为:
(1)Fab,包含抗体分子的单价抗原结合片段的片段,其可以通过用木瓜蛋白酶消化完整抗体以产生完整轻链和一条重链的一部分而产生;
(2)Fab′,可以通过用胃蛋白酶处理完整抗体、之后进行还原以产生完整轻链和重链的一部分而获得的抗体分子片段;每个抗体分子得到两个Fab′片段;

本发明还提供了一种双特异性抗体,具有本发明所述的特异性识别EB病毒gp350蛋白的中和性单克隆抗体和细胞表面特异性抗原或受体的抗体。
所述细胞为肿瘤细胞或免疫细胞。所述免疫细胞可以为单核细胞/巨噬细胞、嗜酸性粒细胞或中性粒细胞。
所述细胞表面特异性抗原或受体选自CD89、CD3、CD16等。
本发明所述的双特异性抗体可以为IgG形态的双特异性抗体、附加在IgG形态上的双特异性及多特异性抗体、双特异性抗体片段、双特异性抗体与蛋白融合或者通过化学键连接的双特异性抗体。优选为IgG形态的双特异性抗体,抗体的抗原结合部分分别为特异性识别EB病毒gp350蛋白的单链抗体和细胞表面特异性抗原或受体的单链抗体。
所述双特异性抗体包括单链可变片段ScFv和具有铰链区、CH2结构域和CH3结构域的Fc片段,所述单链可变片段ScFv分别为特异性识别EB病毒gp350蛋白的单链抗体和细胞表面特异性抗原或受体的单链抗体。优选双特异性抗体的Fc为hFc,在其中一条重链的CH3结构域引入突变,形成一个突起的类似“杵”的结构(Knob),在另一条重链CH3结构域引入突变形成一个凹陷的类似“臼”的结构(Hole),形成Knob-hole的经典结构。利用两条重链CH3结构域域的空间位阻效应利于两种异源抗体重链的正确装配。
上述IgG形态的双特异性抗体,一个具体的示例选自Chen et al公开的hFc(Chenet al.
2020)。hFc(Knob)具有N297A/S354C/T366W的点突变,氨基酸序列如SEQ ID NO:7所示;hFc(hole)具有N297A/Y349C/T366S/L368A/Y407V的点突变,氨基酸序列如SEQ IDNO:8所示。
优选的,所述特异性识别EB病毒gp350蛋白的单链抗体的重链可变区和轻链可变区、CD89单链抗体的重链可变区和轻链可变区可以刚性连接也可以通过连接肽连接。所述连接肽为由Gly和Ser残基组成的短肽。其中使用最广泛的连接肽的序列为(Gly-Gly-Gly-Gly-Ser)n,n代表2-50的正整数,优选2-5。本发明一个具体的示例,连接肽选自(Gly-Gly-Gly-Gly-Ser)3
本发明一个具体的示例所述的双特异性抗体的特异性识别EB病毒gp350蛋白的单链抗体重链可变区氨基酸序列如SEQ ID NO:1所示,轻链可变氨基酸序列如SEQ ID NO:2所示,重链和轻链可变区之间具有连接肽(Gly4Ser)3;所述细胞表面特异性抗原或受体为CD89单克隆抗体,其重链可变区氨基酸序列如SEQ ID NO:5所示,轻链可变区氨基酸序列如SEQID NO:6所示,重链和轻链可变区之间具有连接肽(Gly4Ser)3

本发明另一目的在于提供一种载体,含有本发明所述的多核苷酸。所述的载体包括:细菌质粒、噬菌体、酵母质粒、植物细胞病毒、哺乳动物细胞病毒如腺病毒、逆转录病毒、或其他载体。

本发明公开的特异性识别EB病毒gp350蛋白的单克隆抗体还可用于制备嵌合抗原受体(CAR;也称为嵌合T细胞受体、人造T细胞受体或嵌合免疫受体)。
本发明具体实施例中公开了高亲和力的特异性识别EB病毒gp350蛋白的单克隆抗体R1。本发明还公开gp350/CD89双特异性抗体治疗EBV相关B淋巴瘤或T淋巴瘤、NK淋巴瘤的效果。
本发明所提供的特异性识别EB病毒gp350蛋白的单克隆抗体可用于多种目的,例如用于肿瘤的分子诊断,在EBV相关肿瘤方面的胶体金试剂盒。所述样品可以为任何样品,包括但不限于来自活组织检查、尸体解剖和病理标本的组织。生物样品还包括组织的切片,例如为组织学目的获取的冷冻切片。生物样品还包括体液,例如血液、血清、血浆、痰、脊髓液或尿。生物样品一般获自哺乳动物,包括人,非人灵长类,小鼠等。同时双特异性抗体可以治疗EBV gp350相关的淋巴瘤。
本发明优点:
本发明所述的EB病毒中和性抗体R1,不仅具有高效的亲和力,可以有效降低血液中的EB病毒的滴度,还能够阻断gp350和CR2的互作,且能阻断EB病毒进入B细胞。本发明所提供的单克隆抗体具有高效的亲和力,能降低血液中的EB病毒的滴度。本发明所述EB病毒中和性单克隆抗体联合CD89抗体制备的双特异性抗体在体外能强烈杀伤EBV gp350阳性的肿瘤细胞,抑制肿瘤细胞的生长和转移,可用于EBV相关的肿瘤,如移植后淋巴增殖性疾病(post-transplant lymphoproliferative disorders,PTLD)、淋巴增殖性疾病(lymphoproliferative disorders,LPD)、单核细胞增多症、嗜血综合征(HLH)、淋巴瘤的预防和治疗。

本发明公开描述了结合gp350的单克隆抗体的制备和鉴定。具体的实施方案公开了靶向gp350的中和性单克隆抗体的表征。本发明公开的具体数据证明了这些抗体以高亲和力结合EB病毒和相关肿瘤细胞表面相关的gp350。本发明所述单克隆抗体联合CD89抗体制备的双特异性抗体在体外能强烈杀伤EBV gp350阳性的肿瘤细胞,为抗肿瘤药物开发提供了实验依据。

实施例1:本发明所述EBVgp350单克隆抗体的制备
本实施例描述了针对EB病毒的糖蛋白gp350的高亲和力中和性单克隆抗体的生成。
将2只6周左右(体重大约2kg)的新西兰大白兔用重组的gp350蛋白免疫,每14天免疫一次,免疫3次后,采集兔的脾脏,提取脾脏中的总RNA,逆转录为cDNA,构建噬菌体展示的抗体文库,经过4轮亲和筛选后,获得特异性结合gp350的单克隆R1和R9。
所述R1抗体的重链可变区氨基酸序列如SEQ ID NO:1所示,轻链可变氨基酸序列如
SEQ ID NO:2所示。

实施例2:本发明所述单克隆抗体R1的结合活性测定
将R1/R9单克隆抗体的scFv与FLAG标签序列或hFc融合,并在哺乳动物细胞293F中表达。表达产物经过纯化后,采用夹心ELISA方法测试纯化的抗体对gp350蛋白和EBV病毒颗粒的结合。将gp350-hFc或EBV颗粒蛋白包被在ELISA板上,加入梯度稀释的单抗R1/R9进行孵育,用HRP-鼠抗FLAG或HRP-羊抗人的抗体来检测R1/R9和gp350以及EBV的结合能力(图1)。结果显示R1、R9和CR2都与重组蛋白gp350和EBV颗粒同样有高度的亲和力,其中R1的结合活性优于R9和CR2
实施例3:本发明的单克隆抗体R1的病毒中和活性测试
1、本发明的单克隆抗体R1/R9阻断gp350和CR2的结合活性
ELISA验证单抗体外的中和活性。将gp350-hFc包被在板底,添加梯度稀释的R1-hFc进行结合,加入2μg/ml的重组CR2蛋白,通过检测CR2的结合来判断抗体能否有效的阻断两者结合,结果表明R1/R9能够有效地阻断CR2和gp350的结合,其中R1的中和活性优于R9(图2)。
流式细胞术测试R1/R9阻断gp350和CR2的结合。在CR2阴性的上皮细胞A431基础上构建CR2超表达的细胞系G4。将gp350-His标记上生物素,然后与G4共孵育,加入不同浓度的R1抗体进行阻断,最后加入PE标记的抗生物素二抗,检测结合在细胞表面的gp350-His-Biotin。结果表明,R1抗体能剂量依赖性地阻断gp350和CR2的结合,R1抗体浓度越高,阻断效果越明显(图3)
2、本发明的单克隆抗体R1/R9阻断EB病毒颗粒和CR2的结合活性
通过Cy5.5 NHS ester标记EBV,将Cy5.5-EBV和单克隆抗体R1/R937℃共孵育1小时,然后将其与G4和Raji细胞共孵育45min,将细胞收集并用PBS洗两次,通过流式检测Cy5.5-EBV结合细胞的能力。结果显示R1、R9和CR2均有中和EBV与A431(CR2)和Raji结合的能力,R1相较于R9或CR2更能有效地阻断EBV与CR2阳性细胞的结合(图4)3、本发明的单克隆抗体R1/R9阻断EB病毒颗粒感染CR2阳性的细胞
将Cy5.5-EBV与不同浓度的R1-hFc 37℃共孵育30min,将1*10^6Raji加入到相应孵育好的EBV/R1-hFc混合物中,37℃共孵育60min。将细胞收集并用PBS洗两次,再用新的RPMI1640培养48h。经流式细胞分析(A)和qPCR分析(B)感染效率。(A)通过流式检测Raji细胞中的细胞中的荧光强度。(B)qPCR技术检测单抗中和EBV感染Raji细胞。通过检测细胞基因组中EBV的拷贝数来确定中和活性。结果显示R1相对于未加抗体阻断的荧光密度降低了58.09%,而CR2和R9中和能力则依次降低,同时通过qPCR分析显示了R1在100μg/ml的浓度下能够有效地阻断EBV感染Raji细胞。结果表明,R1能够明显降低EB病毒感染Raji细胞,且活性优于R9(图5)。
实施例4利用本发明的单克隆抗体R1构建gp350/CD89双特异性抗体R1-KH
1、双特异性抗体的制备与表达
双特异性抗体为IgG形态,可变区分别为本发明的单克隆抗体R1的scFv和CD89特异性抗体的scFv。所述CD89单克隆抗体的VH氨基酸序列如SEQ ID NO:5所示,VL氨基酸序列如SEQ ID NO:6所示,重链和轻链可变区之间具有连接肽(Gly4Ser)3;hFc(knob)的氨基酸序列如SEQ ID NO:7所示,hFc(hole)的氨基酸序列如SEQ ID NO:8所示。CD89单链抗体的VHCD89和VLCD89片段之间引入(Gly4Ser)3(简写作(G4S)3)氨基酸序列。编码R1的scFv与hFc(knob)融合,编码CD89抗体的scFv与hFc(hole)融合,如图6A所示。
将编码R1-hFc(knob)和CD89抗体-hFc(hole)的表达载体质粒按1:1混合,瞬时转染到HEK-293F细胞中。每6×108个细胞转染600μg混合质粒。转染后培养细胞约6-8天,期间需要每日检测细胞存活率,当细胞存活率低于85%时收取培养基上清。离心,收集上清液,然后用Protein A柱子亲和层析,纯化双特异性抗体,SDS-PAGE检测双特异抗体的纯度,结果显示,双特异性抗体两者单体大小约为50Kd,形成二聚体蛋白约为100Kd,结果表明所表达的双特异性抗体大小符合预期蛋白大小(图6B)。
2、gp350/CD89双特异性抗体R1-KH与靶细胞的结合特异性测试
将纯化的双特异性抗体R1-KH分别与EBV阳性的细胞(Raji和B95-8)和CD89阳性的巨噬细胞THP1以及未激活的人外周血单形核细胞(PBMC)进行结合,加入PE标记的羊抗人的二抗,检测结合至细胞表面上的抗体。结果如图7A-B所示,R1-KH能很强地结合Raji、B95-8、THP1和PBMC,说明gp350/CD89双特异性抗体R1-KH既能特异性地识别并结合细胞表面的gp350蛋白,又能结合到含有CD89蛋白的巨噬细胞。
3、gp350/CD89双特异性抗体R1-KH介导的对肿瘤细胞的杀伤作用
未激活的PBMC作为免疫效应细胞(Effector cell),不同的肿瘤细胞作为靶细胞(target cell),将效应细胞:靶细胞按10:1的比例混匀,再加入不同浓度的R1-KH双特异抗体,共孵育36h,利用CCK-8测定活细胞中的脱氢酶含量来定量分析肿瘤细胞的存活率。双特异抗体R1-KH在一定浓度范围内对肿瘤细胞Raji和B95-8均具有明显的杀伤活性(图8A-B)。
实施例5本发明的单克隆抗体R1和双特异性抗体R1-KH均能有效降低血液和实体器官中的EB病毒载量
采用小鼠感染模型,将EBV(B95-8株,10μg/mice)和10×106PBMC通过尾静脉注射到小鼠体内,然后通过单抗R1-hFc(50mg/Kg)和双特异抗体R1-KH(5mg/Kg)进行治疗,并在接毒后第6天和第18天,通过尾静脉取血检测EB病毒在外周血中的载量,并于接毒后第42天处死小鼠,分析各组织器官中的病毒载量。结果如图9所示,单抗R1-hFc和双特异抗体R1-KH均能有效地降低血液和各组织器官中的EB病毒载量,双特异性抗体R1-KH在注射浓度低的情况下效果优于单克隆抗体R1。
实施例6本发明的双特异抗体R1-KH对EBV阳性淋巴瘤的治疗活性
实验采用NSG荷瘤小鼠治疗模型。通过尾静脉注射EBV阳性的Raji和B95-8淋巴瘤细胞。2小时后,尾静脉注射5×106人PBMC细胞,3小时后,再注射R1-KH(5mg/kg)进行治疗,并在接瘤后第1、7、14和21天以尾静脉注射的方式补充PBMC,同时每隔3天注射双特异抗体R1-KH进行治疗。在结瘤后第7和14天,尾静脉取血,分析血液及各组织器官中的EB病毒载量(gp350的基因拷贝数),并统计治疗后小鼠的生存期。结果如图10所示,双特异抗体R1-KH能够有效地清除血液和各组织器官中的EBV病毒,并能抑制淋巴瘤细胞的生长和转移。

Claims (20)

1.特异性识别EB病毒gp350蛋白的单克隆抗体,包括重链可变区和轻链可变区,其特征在于:
(1)所述抗体的重链可变区具有SEQ ID NO:1的27-36位、51-66位和99-111位氨基酸残基所示的互补决定区;
(2)所述抗体的轻链可变区具有SEQ ID NO:2的25-35位、51-57位和90-98位氨基酸残基所示的互补决定区。
2.根据权利要求1所述的单克隆抗体,其特征在于:
(1)所述抗体的重链可变区氨基酸序列如SEQ ID NO:1所示;
(2)所述抗体的轻链可变氨基酸序列如SEQ ID NO:2所示。
3.根据权利要求1所述的单克隆抗体,其特征在于所述抗体是单链抗体、双链抗体或嵌合抗体。
4.根据权利要求1所述的单克隆抗体,其特征在于所述抗体是VH单结构域抗体、Fab片段、Fab'片段、F(ab)'2片段、单链可变片段(scFv)或二硫键稳定的可变片段(dsFv)。
5.一种双特异性抗体,其特征在于具有权利要求1-4任一项所述的特异性识别EB病毒gp350蛋白的单克隆抗体和细胞表面特异性抗原或受体的抗体。
6.根据权利要求5所述的双特异性抗体,其特征在于所述细胞为肿瘤细胞或免疫细胞。
7.根据权利要求6所述的双特异性抗体,其特征在于所述细胞表面特异性抗原或受体选自CD89、CD3或CD16。
8.根据权利要求7所述的双特异性抗体,其特征在于为IgG形态的双特异性抗体、附加在IgG形态上的双特异性及多特异性抗体、双特异性抗体片段、双特异性抗体与蛋白融合或者通过化学键连接的双特异性抗体。
9.根据权利要求8所述的双特异性抗体,其特征在于为所述双特异性抗体包括单链可变片段ScFv和具有铰链区SG、CH2结构域和CH3结构域的Fc片段,所述单链可变片段ScFv分别为特异性识别EB病毒gp350蛋白的单链抗体和细胞表面特异性抗原或受体的单链抗体。
10.根据权利要求9所述的双特异性抗体,其特征在于双特异性抗体的Fc为hFc,hFc的CH3结构域形成Knob-hole的结构。
11.根据权利要求10所述的双特异性抗体,其特征在于所述特异性识别EB病毒gp350蛋白的单链抗体的重链可变区和轻链可变区、CD89单链抗体的重链可变区和轻链可变区经连接肽连接。
12.根据权利要求11所述的双特异性抗体,其特征在于连接肽为(Gly-Gly-Gly-Gly-Ser)n,n代表2-50的正整数。
13.根据权利要求12所述的双特异性抗体,其特征在于所述特异性识别EB病毒gp350蛋白的单链抗体重链可变区氨基酸序列如SEQ ID NO:1所示,轻链可变氨基酸序列如SEQ IDNO:2所示,重链和轻链可变区之间具有连接肽(Gly4Ser)3;所述细胞表面特异性抗原或受体为CD89单克隆抗体,其重链可变区氨基酸序列如SEQ ID NO:5所示,轻链可变区氨基酸序列如SEQ ID NO:6所示,重链和轻链可变区之间具有连接肽(Gly4Ser)3;具有Knob突变的hFc的氨基酸序列如SEQ ID NO:7所示,具有hole突变的hFc的氨基酸序列如SEQ ID NO:8所示。
14.一种重组蛋白,其特征在于,所述的重组蛋白包括权利要求1-4任一项所述的单克隆抗体或权利要求5-13任一项所述的双特异性抗体和协助表达和/或纯化的标签序列。
15.一种免疫缀合物,其特征在于包含权利要求1-4任一项所述的单克隆抗体或权利要求5-13任一项所述的双特异性抗体和效应分子。
16.一种多核苷酸,其特征在于其编码根据权利要求1-4任一项所述的单克隆抗体或权利要求5-13任一项所述的双特异性抗体,或权利要求14所述的重组蛋白,或者权利要求15所述的免疫缀合物。
17.一种载体,其特征在于含有权利要求16所述的多核苷酸。
18.一种遗传工程化的宿主细胞,其特征在于含有权利要求17所述的载体,或基因组中整合有权利要求16所述的多核苷酸。
19.一种药物组合物,其特征在于包括根据权利要求1-4任一项所述的单克隆抗体或权利要求5-13任一项所述的双特异性抗体,或权利要求14所述的重组蛋白,或者权利要求15所述的免疫缀合物,权利要求16所述的多核苷酸,权利要求17所述的载体或者权利要求18所述的遗传工程化的宿主细胞中的一种或几种。
20.根据权利要求1-4任一项所述的单克隆抗体或权利要求5-13任一项所述的双特异性抗体,或权利要求14所述的重组蛋白,或者权利要求15所述的免疫缀合物,权利要求16所述的多核苷酸,权利要求17所述的载体或者权利要求18所述的遗传工程化的宿主细胞在制备自身免疫疾病、病毒感染或癌症的治疗药物或诊断试剂中的应用。
