Description

Bletilla striata medicine for treating burns and scalds and preparation method thereof
Technical Field
The invention relates to the technical field of traditional Chinese medicines, in particular to a bletilla striata burn and scald medicine and a preparation method thereof.
Background
Burn and scald are common surgical wounds in daily life, and the wounds bring certain damage to the spirit and the body of people and bring inconvenience to daily life. Burn and scald are accompanied by red, swelling and burning pain, and the wound is easy to be infected and suppurated, so that the severity of the wound is aggravated and even scars are left after poor treatment. Therefore, the key points of treating the burns and scalds are two aspects of the antibiosis and the antiphlogosis of the wound surface and the promotion of the wound surface healing.
The traditional Chinese medicine bletilla striata is a dried tuber of a perennial herb bletilla striata of the family Orchidaceae. In the field of traditional Chinese medicine, common bletilla pseudobulb compound is mostly adopted for treating burns and scalds, for example, patent CN105456722A discloses a compound burn and scald medicine prepared from lithospermum, giant knotweed rhizome, gardenia, phellodendron, garden burnet, common bletilla pseudobulb and borneol; patent CN102228623A discloses a compound burn and scald medicine prepared from sanguisorba, bletilla, callicarpa bodinieri, rheum officinale, polygonum cuspidatum, scutellaria barbata and lithospermum. But the application of the traditional Chinese medicine composition to white and single-prescription treatment of burns and scalds is less, and no medicament for treating the burns and scalds by using the common bletilla pseudobulb single-prescription is available. Aiming at the defect, the invention researches the white and single-component treatment of burns and scalds, and aims to provide a simple, noninvasive, safe and effective bletilla striata burn and scald medicine.

Experimental example 1
The influence of the bletilla striata burn and scald medicine on the scald wound surface of a rat
(1) Establishing a rat skin trauma model:
the experiment was conducted in a university of medical compliance laboratory.
64 male SD rats with the weight of 200 g-250 g are selected and are adaptively fed in a well-ventilated feeding room with the room temperature constant at 25 ℃ for one week. One week later, the rats were dehaired on their backs with an 8% sodium sulfide solution, and the area of dehairing was 4cm × 6 cm. After 24h, the anesthesia treatment was carried out with diethyl ether. After the asbestos cardboard was wetted, a square hole of 2cm × 2cm was cut out with a blade in the center. Covering the back of the rat, and exposing the area of the scald-simulating part from the hole. Setting the temperature of the constant-temperature electric iron to be 80 ℃, preheating, contacting the constant-temperature electric iron with the exposed skin for 8s, timing by a stopwatch, and removing the constant-temperature electric iron after the time to obtain a rat III-level burn and scald model.
(2) The rats with scald are randomly divided into 4 groups, each group comprises 16 rats, including a blank group, an ointment group, an oil agent group and a positive control group, and are raised in cages. Wherein the oil agent group adopts the bletilla striata oil agent burn and scald medicine prepared in the embodiment 1; the rhizoma bletillae ointment prepared in example 3 was used as the ointment group; the positive control group used commercially available Meibao moist burn cream from Meibao pharmaceutical Co., Ltd.
The medicine is not applied to the blank group, the other three groups are applied after scald for 0.5h, the application area is just covered on the affected part, the thickness is 1mm, and the medicine is applied once a day. The application was continued until the wound healed, during which the number of scab days, number of decrustation days and number of healing days were recorded for each group, and the results are shown in table 1. The healing status of the scald on days 1, 3, 7, 15 and 20 was recorded by photographing, as shown in fig. 6.
TABLE 1 healing of III-grade burn and scald in rats
Group of Number of scab formation days Number of days to decrustation Days of healing
Blank group 6.40±0.48 18.75±2.24 30.40±2.06
Paste composition 4.40±1.01 16.80±1.49 24.80±2.22
Oil agent group 4.20±0.74 15.80±1.72 27.60±2.05
Positive control group 3.40±0.80 15.00±1.41 26.40±2.57
As can be seen from table 1, compared to the blank group, the ointment group, the oil group, and the positive control group had shorter scabbing, and healing time and faster speed. Wherein, the positive control group has the fastest scabbing and scabbing speed, and the oil agent group has the second, the paste group is relatively slow; but the paste group healed the fastest, the oil group the second, the positive control group was relatively slow.
As can be seen from fig. 6, compared to the blank group, the ointment group, the oil agent group, and the positive control group had faster healing of the burn and scald and smaller wound area.
In the experiment, one rat is randomly selected from each group to be killed on the 7 th day, a back scald tissue section is taken to be subjected to He staining, and the treatment effect of each group of drugs on the third-level burn skin tissue of the rat is observed, as shown in fig. 7; one rat was randomly selected from each group on day 20 to be sacrificed and dorsal scald tissue sections were taken for He staining, and the therapeutic effect of each group of drugs on rat class III scald skin tissues was observed, as shown in fig. 8.
As can be seen from fig. 7, when the wound tissue sections were observed on day 7, a large amount of new capillaries were generated in the ointment group, the oil group, and the positive control group, and the granulation tissue was arranged more orderly with a large amount of inflammatory cell infiltration. Wherein, the cells under the tissue slices of the paste group are arranged closely and more orderly, and the observation of the number of the new capillaries and the infiltrative inflammatory cells is more obvious, which shows that the recovery effect and the anti-inflammatory effect of the paste group are better than those of the other groups.
As can be seen from fig. 8, when the wound tissue sections were observed on day 20, the cells were closely and more orderly arranged under the tissue sections of the ointment group, the oil group, and the positive control group, compared to the blank group. Wherein, the treatment effects of the ointment group and the oil group are not obviously different from the treatment effects of the positive control group.
The foregoing is only a preferred embodiment of the present invention, and it should be noted that, for those skilled in the art, various modifications and decorations can be made without departing from the principle of the present invention, and these modifications and decorations should also be regarded as the protection scope of the present invention.
