The second group is formula 2, 10g of wrinkled gianthyssop herb, 10g of Chinese mosla herb, 10g of ginkgo seed, 10g of kelp, 10g of mint, 10g of lily, 10g of Chinese yam, 10g of Chinese date, 10g of white hyacinth bean, 6g of safflower, 15g of reed rhizome, 10g of raw liquorice, 6g of red yeast rice, 6g of peach kernel, 30g of fried pearl barley and 10g of houttuynia cordata, and the finally prepared health-care tea bag has the effects of regulating qi activity, tonifying qi and yin, promoting blood circulation and removing blood stasis and is suitable for patients with pulmonary fibrosis in the convalescence.
The formula of the health-care tea bag is formula 3, and the prepared health-care tea bag has the effects of tonifying qi, invigorating spleen and promoting diuresis, is suitable for prevention of common people and has wider applicability.
Fig. 3 is a table showing the taste identification of the health tea bag brewed beverages prepared according to the formulas 1, 2 and 3, and it can be seen that the drinker has a higher taste acceptance of the formula 3.
In order to further verify that the formula 3 has the effectiveness of improving immunity, improving lung function and the like, relevant immune indexes, inflammatory factors and lung tissues are detected by intervening an animal model through the formula, and experimental results are observed, wherein the specific experimental process is as follows:

Randomly dividing 60 BABL/c mice with the age of 8 weeks into a normal group, a model group and a blank control group, and dividing the formula 3 tea drink into an early intervention group, a later intervention group and a whole-course intervention group, wherein each group contains 10 mice; except the normal group, the other groups are injected with pseudomonas aeruginosa in the trachea to establish a pneumonia model.
The early stage of the drying and pre-forming is started 10 days before the molding and the stomach filling of the health care tea is started, and the drying and pre-forming is stopped when the molding is performed; after the molding is successfully performed, the later-stage intervention group starts intragastric administration until the experiment is finished; the whole-course intervention group adopts the health-care tea for intragastric administration continuously from the beginning to the end of the experiment; the normal group and the model group both adopt the same volume of physiological saline for intragastric administration and contrast; and observing and recording the weight and the breath change per minute of the mice before and after the model building and before and after the health-care tea drying.
During the experiment, diet and drinking water are monitored by videos every day for 24 hours for playback in the future; changes in body weight were recorded daily, fresh mouse faeces were collected daily (-80 ℃ storage), during which time there were deaths, and the date, body weight and pre-death clinical presentation were recorded in detail.
Second, animal specimen collection and treatment
After the experiment is finished, obtaining bronchoalveolar lavage fluid of a mouse, after the mouse is killed by ether anesthesia, taking blood from heart, dividing the blood sample into two parts, carrying out anticoagulation treatment on one part (0.4ml), centrifuging the other part to obtain a serum sample, and preserving at-80 ℃; after the experiment is finished, collecting lung, spleen, lymph tissue, intestinal tract and contents thereof of the mouse, taking a proper amount of lung, spleen and small intestine or colon tissue, fixing the lung, spleen and small intestine or colon tissue in 4% paraformaldehyde solution, then using the tissue for later stage pathological examination, quickly freezing the rest tissues by liquid nitrogen, and transferring the tissue to-80 ℃ for storage.
The immunohistochemical method is used for determining the expression level of TLR4 and CXCR4 proteins in lung tissues and the distribution condition in tissues to judge the pathological condition of the lung:
mouse lung tissue immunohistochemistry and semi-quantitative analysis: marking the lung tissue section, inserting the lung tissue section into a staining rack, and dewaxing to water; placing the slicing frame in deionized water for 5min, adding into antigen repairing solution, and placing into micro-lensCooling naturally for 7min, taking out, washing with phosphate buffer solution for 5min × 3 times, and washing with 3% H2O2Blocking endogenous peroxidase with deionized water for 20min, and washing with PBS for 5min × 3 times; sealing biological serum for 40min, and spin-drying without washing; incubating the prepared primary antibody by 30 mu l, washing by 5min multiplied by 3 times after PBS rapidly washes by 1 time, incubating by 20min for the secondary antibody by 30 mu l, washing by 5min multiplied by 3 times after PBS rapidly washes by 1 time; incubating horseradish peroxidase 30 μ l for 20min, washing with PBS for 5min × 3 times; performing color development by using DAB color development liquid, putting the DAB color development liquid into hematoxylin solution for counterstaining for 2min, and then performing blue returning by using tap water; xylene transparent, neutral resin sealing; observing each group of slices under a microscope and taking pictures; TLR4 positive staining was tan to tan, since TLR4 was mainly expressed in the cytoplasm of inflammatory cells, alveolar epithelial cells, fibroblasts, etc., and alveolar cytoplasm and/or cell membrane, semi-quantitative analysis was performed using Imageproplus software using mean optical density values; CXCR4 positive staining is tan to tan and is generally expressed on alveolar epithelial cells and on the cytoplasm and cell membranes in the alveolar mesenchyme, so the AB value quantitative scoring method is adopted according to the literature.
The real-time fluorescent quantitative PCR technology is used for measuring the expression quantity of IL-6 and TNF-alpha mRNA in the cells:
selecting and dividing THP-1 cells after passage into blank group, control group and drug-adding group, dividing all groups into three multiple wells, adding 20 μ l culture medium into the blank group, adding 20 μ l filtered drug (125 μmolL) into the drug-adding group-1) Terminating the culture after 24 hours; then adding 800 mu L Trizol into the cell culture hole, and then sequentially adding cell lysate chloroform, isopropanol and 75% alcohol to extract total RNA; dissolving RNA in deionized water added with DEPC (diethyl pyrocarbonate), then measuring the purity and concentration of A260 and A280 quantitative analysis RNA by using an ultraviolet spectrophotometer, and further performing reverse transcription to form cDNA according to the total RNA concentration and a reverse transcription kit, wherein the reaction conditions are as follows: annealing at 25 deg.C for 5 min; extending at 42 ℃ for 60 min; inactivating at 70 deg.C for 5 min.

the qPCR reaction system comprises 10 mu L of SYBR Green Premix Ex Taq II, 1.6 mu L of forward and reverse primers (5 pmol/. mu.L), and ddH2O4.8. mu.L and cDNA 2. mu.L; the reaction conditions are pre-denaturation at 95 ℃ for 30s, denaturation at 95 ℃ for 5s, 60 ℃ for 30s and cycle number of 40; dissolution curve: 95 ℃, 15s, 60 ℃, 30s, 95 ℃, 15 s. And (5) correcting the Ct value of each sample by taking GAPDH as an internal reference in detection to obtain a delta Ct value.
SPSS25.0 statistical software is adopted for processing, data measurement values are expressed by mean +/-standard deviation (x +/-s), homogeneity of variance is firstly tested before each group of data are compared, single-factor variance analysis is adopted when normal distribution and homogeneity of variance are met, independent sample t test is adopted for mean comparison between two groups of samples, and the difference is considered to have statistical significance when P is less than 0.05.
Fig. 4 is a graph showing the effect of tea drink of the present invention on the expression of the TLR4 protein positive cells in lung tissue of mice infected with pseudomonas aeruginosa (immunohistochemistry, x 200), wherein the graph shows from left to right: A. blank group; B. a model group; C. a model control group; D. early intervention group; E. a later intervention group; F. and (4) performing whole-process intervention group.
Fig. 5 is a graph showing the effect of tea drink of the present invention on the expression of CXCR4 protein positive cells in lung tissue of mice infected with pseudomonas aeruginosa (immunohistochemistry, x 200), from left to right: A. blank group; B. a model group; C. a model control group; D. early intervention group; E. a later intervention group; F. and (4) performing whole-process intervention group.
The results in FIG. 6 and FIG. 7 show that the blank group and the control group have no significant difference in the expression level of TNF-alpha mRNA in the THP-1 cells; however, the expression level of IL-6mRNA was decreased in the tea drink-treated group (MCD group), i.e., the tea-treated group treated with Yi Yang tea, compared with the blank and the control group, and the difference was statistically significant (P < 0.05).
The research results show that the tea drink group has low observation score of lung pathological tissues, the expression of TLR4 and CXCR4 proteins is low, the influence on congestion and edema of mouse lung tissues, lung structural damage and fiber mass formation is obviously reduced compared with a model group and a control group, and the research results show that the tea drink can effectively prevent and treat the mouse lung damage caused by LPS (lipopolysaccharide), and the treatment effect on the lung damage is achieved by mainly inhibiting TLR4 and CXCR4 signal channels to play a role in anti-inflammatory response.
