背景
多瘤病毒是普遍存在的病毒,其感染许多各种不同的哺乳动物物种。目前,已经鉴别了超过12种不同的人类多瘤病毒物种(包含BK多瘤病毒(BKV/人类多瘤病毒1)、约翰坎宁安(John Cunningham)多瘤病毒(JCV/人类多瘤病毒2)、以及默克尔细胞多瘤病毒(MCV/人类多瘤病毒5))。

在一些实施方案中,APC是B细胞、抗原呈递T细胞、树突状细胞、或人工抗原呈递细胞(例如,aK562细胞)。可以通过从患者样品中取出PBMC并且将其粘附至塑料来制备用于过程的树突状细胞。通常,单核细胞群体粘着,并且全部其他细胞可以被清洗掉。然后用IL-4和GM-CSF分化粘附的群体,以产生单核细胞来源的树突状细胞。这些细胞可以通过添加IL-1β、IL-6、PGE-1和TNF-α(其上调树突状细胞的表面上的重要的共刺激分子)来成熟化,并且然后与本文所描述的重组腺病毒接触。

T细胞

药物组合物

制备这些制剂或组合物的方法包含使本文所描述的剂与载体和可选地一种或更多种助剂缔合。通常,通过将本文所描述的剂与液体载体或精细粉碎的固体载体或两者均匀地且密切地缔合,并且然后使产品成型(如果必要的话)而制备制剂。

治疗方法

实施例3:JCV特异性T细胞扩增和表征
在用合并的JCV表位刺激后,JCV特异性T细胞在体外扩增。具体地,用合成的JCV肽(表1)刺激来自健康志愿者的PBMC达1小时,并且然后在存在不同细胞因子组合(包含IL-2(10ng/ml)、IL7(10ng/ml)、IL12(10ng/ml)和/或IL15(10ng/ml))的情况下培养12-14天。使用标准细胞内细胞因子测定评估经扩增的T细胞的JCV特异性(表6)。

实施例4:JCV表位与BKV表位之间的T细胞交叉反应性

实施例5:分析健康个体和移植受者中JCV特异性T细胞的功能特征和表型特征
近年来,T框转录因子(T-bet)和脱中胚蛋白(Eomesodermin)(Eomes)已经被证明在确定感染期间CD8+T细胞的命运中起重要作用。高水平的T-bet与抗原特异性细胞中的细胞毒性T细胞分化以及穿孔素和颗粒酶B的上调相关。高水平的脱中胚蛋白与长期记忆形成相关。已经在各种研究中看出,其合作表达对于感染控制是关键性的。在小鼠研究中,还已经表明任一转录因子的缺失造成无法控制感染。因此,研究这些转录因子的表达是关键性的,其能够帮助理解在急性病毒感染和慢性病毒感染两者的过程中T细胞和T细胞分化的表型特征。尚未理解JCV特异性T细胞中T-bet和脱中胚蛋白的表达模式,并且这样的T细胞上的这些转录因子的分析可以使得对JCV特异性T细胞的分化的更深入的理解成为可能。对于T细胞的功能特征的详细研究还能够导致对于JCV相关性疾病有效的免疫疗法的开发。一组初步的实验研究调节其分化的T细胞上的转录因子。使用ICS在JCV特异性T细胞和CMV特异性T细胞上测定T-bet、脱中胚蛋白、穿孔素和颗粒酶B的表达。初步分析示出JCV特异性T细胞中中等至低水平的T-bet表达,而在CMV特异性T细胞内看到高水平的T-bet。相比于CMV特异性T细胞,在JCV特异性T细胞内发现非常低的脱中胚蛋白表达。同样地,还在JCV特异性T细胞中看到低水平的穿孔素和颗粒酶B。这表明,相对于CMV特异性T细胞,JCV特异性T细胞在效应子功能方面功能低。因此,驱动JCV特异性CTL的效应子功能是有助于开发有效过继性T细胞免疫疗法的研究的关注点。
实施例6:利用建议的肽库的T细胞扩增的可行性:
随机选择来自15个健康供者的PBMC,而不考虑其HLA类型,并且JCV特异性T细胞在用包括本文公开的肽(即,包括SEQ ID NO.1-21中所示的氨基酸序列的肽)的肽库刺激后扩增。使T细胞扩增17天,并且使用细胞内细胞因子染色测定评估JCV应答。来自15个供者中的13个供者的T细胞具有JCV特异性T细胞应答,其通过在用肽库重新刺激后IFN-γ的产生被证明(参见图5)。
实施例7:JCV特异性T细胞产物的功能表征:
为了确定使用肽库扩增的JCV特异性T细胞的多功能性,通过细胞内染色针对T细胞的IL-2、TNF、IFN-γ和CD107的表达对T细胞进行分析(参见图6a)。JCV特异性T细胞(连同其他细胞因子)显示出较高的TNF表达。
