Description

Beverage containing eggshell membrane powder and hydrolyzed eggshell membrane component
Technical Field
The present invention relates to a beverage containing eggshell membrane component. More specifically, the present invention relates to a beverage having a skin-beautifying effect of preventing skin aging and skin roughness, an effect of promoting health, an effect of preventing diseases, and the like.
Background
The human skin is composed of the epithelial layer on the surface and the connective tissue dermis layer below the epithelial layer, and the proteins constituting the dermis layer are mainly collagen, and also consist of elastin, albumin, globulin and mucin. The collagens constituting the dermis layer are mainly α 1(I) type collagen and α 1(III) type collagen (hereinafter referred to as "type III collagen"), the type I collagen mainly functions to maintain the structure of the skin, and the type III collagen mainly functions to impart flexibility to the skin, and each of the collagen functions is different.
The ratio of type I collagen to type III collagen in skin changes with age. Just as type III collagen is also called fetal collagen, which is the most abundant in fetal stage, type III collagen in fetal skin is much but decreased from the age of ten years and so on, so if the proportion of type III collagen in skin can be increased, aging of skin due to aging can be prevented, and skin becomes smooth and moisturized.
The eggshell membrane is located at the inner side of eggshell of eggs of birds such as eggs and the like and is composed of an inner eggshell membrane and an outer eggshell membrane, wherein the outer eggshell membrane is tightly attached to the inner surface of the eggshell, and the inner eggshell membrane wraps egg white. Eggshell membrane is mainly composed of tough fibrous protein, and is known to have a function of promoting skin regeneration as it has been used for treatment of abrasion in the sumac since ancient times, and recently, it has been reported to have an action of promoting the production of type III collagen called fetal collagen.
In addition, eggshell membrane and hydrolyzed eggshell membrane obtained by hydrolyzing the eggshell membrane are known to have effects of promoting hair growth, and preventing hair loss, in addition to the above-mentioned skin beautifying effect and skin regeneration effect. Furthermore, eggshell membrane and hydrolyzed eggshell membrane have a function of reducing or eliminating active oxygen generated in the living body, and have an effect of improving or preventing various diseases or adverse physical symptoms caused by active oxygen, for example, are effective in restoring normal state of red blood cells destroyed by active oxygen, increasing hemoglobin, eliminating shoulder stiffness, stimulating appetite, eliminating fatigue, activating living body, promoting health, maintaining health, and the like.
In view of the above excellent effects of eggshell membranes and hydrolyzed eggshell membranes, orally ingested skin improving agents and skin improving beverages and foods containing eggshell membranes or hydrolyzed eggshell membranes are known (patent documents 1 to 3).
Among the eggshell membrane and the hydrolyzed eggshell membrane, the hydrolyzed eggshell membrane is water-soluble and excellent in handleability, and therefore, can be used not only by being mixed with other components to prepare solid foods or preparations (tablets, capsules, granules, etc.), but also by being dissolved in water to prepare beverages.
On the other hand, since the eggshell membrane that is not subjected to hydrolysis treatment, it can be obtained at a lower cost than the hydrolyzed eggshell membrane, but since it is insoluble in water and cannot be stably dispersed in water, it is not actually used for beverages, but is added to solid foods or used as a solid agent. However, the present inventors found that: the unhydrolyzed eggshell membrane contained in the solid food or solid agent is poor in digestion and absorption, and the above-mentioned excellent properties inherent in the eggshell membrane are not fully utilized.
In addition, collagen is one of proteins constituting dermis, ligament, tendon, bone, cartilage, and the like, is a main component of extracellular matrix of animals, and is said to account for about 30% of all proteins in human body.
Soluble collagen peptides having improved solubility, digestion and absorption properties, and dispersibility, which are obtained by reducing the molecular weight of collagen or gelatin prepared from collagen by hydrolysis treatment, are widely produced and sold. Since collagen peptides have moisture retention and water retention properties, as in collagen, and have a skin-beautifying effect such as elasticity, firmness, luster, and moisturization when ingested, various foods and beverages containing collagen peptides have been known (patent documents 4 to 6).
If the collagen peptide is ingested or vitamin C or N-acetylglucosamine is ingested together with the collagen peptide, the elasticity, firmness, luster, moisturization and the like of the skin are increased as compared with the case where the collagen peptide is not ingested, but in recent years, the degree of the skin beautifying effect required by consumers is increasing and the consumer demand is not sufficiently satisfied.

Detailed Description
The present invention relates to a beverage containing eggshell membrane powder and hydrolyzed eggshell membrane component. More particularly, the present invention relates to a beverage containing eggshell membrane powder and a hydrolyzed eggshell membrane component, and containing fish-derived collagen peptide and N-acetylglucosamine, which has effects of preventing skin aging and skin roughness, making skin smooth, soft, moist, tight, and glossy, improving intestinal flora, promoting health, and preventing diseases.
In the present invention, any powder of eggshell membrane obtained from bird eggs without hydrolysis treatment can be used as the eggshell membrane powder, and among them, eggshell membrane powder obtained from eggs is preferably used from the viewpoint of general versatility and acquisition easiness.
The eggshell membrane powder used in the present invention is preferably a fine granular eggshell membrane powder in view of dispersion stability in the beverage, digestive absorption when the beverage is ingested, and the like.
In particular, fine-particle eggshell membrane powder having a content ratio of particles of 70 mass% or more by a 150-mesh standard sieve (150Tyler sieve; pore size 104 μm) is excellent in dispersion stability in beverages, is excellent in digestion and absorption properties, and can more favorably exhibit the above-mentioned excellent functions of eggshell membrane, such as the skin beautifying effect and health promoting effect.
The eggshell membrane Powder itself has been commercialized, and for example, "ESM Powder-II" manufactured by Sanzhou food Co., Ltd, "EM Powder 300" manufactured by Kewpie Corporation [ the content ratio of particles passing through a 150-mesh standard sieve (150-mesh Tyler sieve; pore size 104 μm) is 70% by mass or more ], and the commercially available eggshell membrane Powder can be used as it is. The commercially available eggshell membrane powder may be further pulverized to prepare a fine-grained eggshell membrane powder having a content ratio of particles passing through a 150-mesh standard sieve (150-mesh Tyler sieve; pore size 104 μm) of, for example, 80% by mass or more, and further 90% by mass or more, or a further fine-grained eggshell membrane powder may be prepared by sieving (classifying) using a standard sieve having a pore size smaller than that of the 150-mesh standard sieve.
Here, in the present specification, the "content ratio of particles passing through the standard sieve of a mesh" in relation to the particle diameter of the eggshell membrane powder refers to the mass ratio (mass%) of particles passing through the standard sieve of a mesh (Tyler sieve) relative to the total mass of the eggshell membrane powder before sieving (classification) when the eggshell membrane powder is sieved (classified) using the standard sieve of a mesh (Tyler sieve).
The hydrolyzed eggshell membrane component used in the present invention is obtained by hydrolyzing eggshell membrane or the above-mentioned eggshell membrane-containing powder in a liquid having a proteolytic activity such as an aqueous acid solution, an aqueous alkaline solution, an alkaline organic solvent solution, and a protease-containing liquid. The extract may be used as an aqueous solution or an extract, but is preferably in the form of powder obtained by a drying method such as spray drying or freeze drying for the sake of handling and storage stability. The egg shell membrane-hydrolyzing component is preferably an alkali-hydrolyzing component, an enzyme-hydrolyzing component, or a mixture thereof.
The hydrolyzed eggshell membrane component is almost 100% digested and absorbed into blood, whereas approximately 54% of the eggshell membrane powder and micropowder remains in the intestine without being digested and absorbed, and the remaining components can prevent the proliferation of bad bacteria in intestinal bacteria and improve intestinal flora. Therefore, by using a hydrolyzed eggshell membrane fraction having a function of further promoting the production of type III collagen in addition to the eggshell membrane powder or micropowder which is digested and absorbed only by about 46%, the intestinal flora improving function and the type III collagen production promoting function act simultaneously, and a synergistic skin beautifying effect can be obtained.
The fish-derived collagen peptide used in the present invention is a collagen peptide obtained by subjecting collagen obtained from skin, bone and/or scale of fish or gelatin obtained from the collagen to a partial hydrolysis treatment using an enzyme or the like. Among these, fish scale-derived collagen peptides produced from collagen obtained from fish scales are preferably used, because the effects of the present invention such as a skin-beautifying effect can be more effectively exerted.
The molecular weight of the fish-derived collagen peptide used in the present invention is not particularly limited, but from the viewpoints of digestibility, solubility in water serving as a base medium of a beverage, and handleability, it is preferable to use a fish-derived collagen peptide having an average molecular weight of 500 to 15,000, more preferably a fish-derived collagen peptide having an average molecular weight of 1,000 to 10,000, and still more preferably a fish-derived collagen peptide having an average molecular weight of 1,500 to 5,000.
Among the above-mentioned fish-derived collagen peptides, various products have been manufactured and marketed as fish scale-derived collagen peptides, and any of them can be used.
N-acetylglucosamine used in the present invention is a natural amino sugar and is widely distributed in the human body as a constituent of glycoproteins, glycolipids, mucopolysaccharides, and the like. It is known that N-acetylglucosamine is produced by metabolic breakdown of glycogen in the living body and becomes a raw material of hyaluronic acid and chondroitin sulfate. In addition, in nature, N-acetylglucosamine is contained in large amounts in chitin forming the outer skin of crustaceans such as crabs and shrimps.
It has been reported that when N-acetylglucosamine is orally ingested, it is absorbed by cartilage or skin tissue, improving the skin moisturizing and smoothing effect, and preventing joint pain and friction.
Examples of the method for producing N-acetylglucosamine include a method in which chitin contained in shells of crustaceans such as crabs and shrimps is used as a raw material.
Various products of N-acetylglucosamine have been produced and marketed, and any of them can be used in the present invention as long as it is N-acetylglucosamine that is acceptable for use in food products.
The beverage of the present invention preferably contains 0.02 to 2.5% by mass of eggshell membrane powder, 0.02 to 2.5% by mass of a hydrolyzed eggshell membrane component, 0.05 to 5% by mass of a fish-derived collagen peptide, and 0.01 to 5% by mass of N-acetylglucosamine, based on the mass of the beverage (including the total mass of water as a medium). More preferably, the composition contains 0.1 to 1% by mass of eggshell membrane powder, 0.1 to 1% by mass of hydrolyzed eggshell membrane component, 0.1 to 5% by mass of fish-derived collagen peptide, and 0.05 to 5% by mass of N-acetylglucosamine, and further preferably contains 0.2 to 0.5% by mass of eggshell membrane powder, 0.2 to 0.5% by mass of hydrolyzed eggshell membrane component, 0.1 to 2% by mass of fish-derived collagen peptide, and 0.05 to 2% by mass of N-acetylglucosamine.
If the content of the eggshell membrane powder and the hydrolyzed eggshell membrane component is too low, the skin-beautifying effect such as elasticity, firmness, luster, moistness, and smoothness of the skin cannot be sufficiently exerted, and the effects such as promotion of health and maintenance of health tend to be reduced. On the other hand, if the content of the eggshell membrane powder and the hydrolyzed eggshell membrane component is too high, the eggshell membrane powder cannot be stably dispersed in the beverage, precipitation, agglomeration, or the like is easily generated, and a decrease in the taste of the beverage, an increase in the cost, or the like occurs. Further, if the content of the collagen peptide derived from fish is too small, it is difficult to sufficiently exert the skin beautifying effect as described above, and it is difficult to stably disperse the eggshell membrane powder in the beverage. On the other hand, if the content of the collagen peptide derived from fish is too large, the taste of the beverage tends to be reduced, the cost tends to be increased, and a strange taste tends to occur. If the content of N-acetylglucosamine is too small, it is difficult to sufficiently exhibit the above-mentioned skin beautifying effect, and it is difficult to satisfactorily retain water and lubricate in vivo. On the other hand, if the content of N-acetylglucosamine is too large, the taste of the beverage tends to be deteriorated, the cost tends to be increased, and the flavor tends to be deteriorated.
The beverage of the present invention contains eggshell membrane powder, a hydrolyzed eggshell membrane component, a fish-derived collagen peptide, and N-acetylglucosamine, and may further contain one or more selected from hyaluronic acid, ceramide, and L-ascorbic acid as required. Hyaluronic acid, ceramide and L-ascorbic acids have been widely used as skin improving agents or food and drink products for skin improvement by oral ingestion as skin beautifying ingredients, and since there is no contraindication, the drink of the present invention preferably contains hyaluronic acid, ceramide and L-ascorbic acids.
The content of each of hyaluronic acid, ceramide and proteoglycan is preferably 0.001 to 0.5% by mass based on the total mass of the beverage.
As the L-ascorbic acid compound, one or more of L-ascorbic acid, a salt thereof, and a derivative of L-ascorbic acid (for example, a derivative in which one or more of the hydroxyl groups at the 2, 3, 5, and 6 positions of ascorbic acid are esters of phosphoric acid, polyphosphoric acid, sulfuric acid, fatty acid, and other pharmaceutically acceptable compounds) can be used.
L-ascorbic acids are one of typical antioxidants, and have an action of converting melanin into colorless reduced melanin to make spots or freckles produced on the skin inconspicuous, and also an action of assisting collagen production in the living body.
When the beverage of the present invention contains L-ascorbic acids, the content thereof is preferably 0.02 to 5% by mass, and more preferably 0.05 to 1% by mass, based on the total mass of the beverage.
The beverage of the present invention preferably contains a sweetener and/or a flavoring agent in order to improve the taste and flavor and facilitate ingestion.
As the sweetener, any conventionally used in foods and beverages can be used, and examples thereof include sucrose, glucose, fructose, maltose, lactose, trehalose, starch sugar, honey, maltooligosaccharide, fructooligosaccharide, fructoglucose liquid sugar, fruit juice, sucralose as a high-intensity sweetener, aspartame, acesulfame potassium, and the like, and one kind or two or more kinds of these can be used.
Among these, in the beverage of the present invention, one or more of maltooligosaccharide, fructoglucose liquid sugar, and a high intensity sweetener are preferably used as the sweetener, and more preferably, these three are used in combination.
High intensity sweeteners can be used in minimal amounts and without adding calories to the beverage to impart sweetness to the beverage of the present invention.
Malto-oligosaccharide has a property of acting on bifidobacterium which is a good bacterium but not on clostridium perfringens which is a bad bacterium, and therefore, it has an effect of assisting intestinal regulation, is considered to be effective for constipation or weight reduction, and is said to have an effect of improving lipid as an extension thereof, and is useful for improvement of arteriosclerosis and hyperlipidemia. When malto-oligosaccharide is contained, the content thereof is preferably 0.5 to 7% by mass based on the total mass of the beverage.
Fructose glucose liquid sugar is an isomerized sugar syrup containing 50% or more of fructose, has an instant refreshing taste, and has a property of increasing sweetness as the temperature is lower, and therefore, is suitable as a sweetener for a beverage of the present invention to be ingested at normal or low temperatures. When the fructose-glucose liquid sugar is contained, the content thereof is preferably 1 to 15% by mass, more preferably 3 to 10% by mass, based on the total mass of the beverage.
As the seasoning, any seasoning suitable for beverages can be used as long as it does not cause deterioration, precipitation, etc. of eggshell membrane powder, fish-derived collagen peptide, N-acetylglucosamine, and other components contained in the beverage, and examples thereof include fruit juices such as apple juice, citrus juice, grape juice, pineapple juice, strawberry juice, etc.; flavors such as apple flavor, citrus flavor, grape flavor, pineapple flavor, strawberry flavor, and natural flavor; acidity regulators such as lactic acid, fermented lactic acid, and brown rice black vinegar. The content of the seasoning in the beverage may be adjusted depending on the kind and form of the seasoning, the content of the flavor and taste to be imparted to the beverage, and the like.
The beverage of the present invention may contain a dispersion stabilizer if necessary in order to stably disperse the eggshell membrane powder in the beverage. Examples of the dispersion stabilizer include thickening polysaccharides such as soybean polysaccharides, carrageenan, pectin, and gum arabic.
The beverage of the present invention may contain components other than the above components as necessary within a range not impairing the object and effect of the present invention.
The pH of the beverage of the present invention is not particularly limited as long as it does not cause precipitation, aggregation, deterioration, or a decrease in performance of components contained in the beverage, and is a pH which is easily ingested by a human, and is preferably 2 to 7, more preferably 3 to 6.5.
The method for producing the beverage of the present invention is not particularly limited, and the beverage can be produced by adding the above components to water, uniformly mixing them, dissolving the components dissolved in water, and uniformly dispersing the components insoluble in water.
The amount of water used in the production of the beverage of the present invention can be appropriately adjusted, but in order to sufficiently exhibit the above-described skin beautifying effect and other effects and to prevent precipitation, aggregation, or the like of components, the beverage is produced using water in an amount of preferably 65 to 90 mass%, more preferably 70 to 85 mass%, and particularly preferably 75 to 85 mass%, based on the mass of the finally obtained beverage.
The mixing method and mixing device used in the production of the beverage are not particularly limited, and a mixing method and a mixing device conventionally used in the production of the beverage can be used.

As seen from the results in table 2, the beverage of example 1 contained eggshell membrane powder, hydrolyzed eggshell membrane powder, fish-derived collagen peptide, and N-acetylglucosamine, and thus was excellent in storage stability, and did not cause precipitation or aggregation of components even after a long period of time.
On the other hand, the beverage of comparative example 2 contains eggshell membrane powder, hydrolyzed eggshell membrane powder, and N-acetylglucosamine, and does not contain collagen peptides derived from fish, and therefore, precipitation and/or aggregation of components occurs, and the storage stability is poor.
Further, the beverage of example 1 was superior to the beverage of comparative example 1 (beverage containing no eggshell membrane powder or hydrolyzed eggshell membrane powder), the beverage of comparative example 2 (beverage containing no fish-derived collagen peptide), and the beverage of comparative example 3 (beverage containing no N-acetylglucosamine), in the effect of beautifying the skin, and the firmness, moisturization, and luster of the skin were improved well.
Industrial applicability of the invention
The beverage of the present invention does not cause precipitation or aggregation of components, has excellent storage stability, and has a strong skin-beautifying effect with respect to imparting effects such as skin elasticity, firmness, luster, moisturization, moisture feel, and smoothness.
In addition, in the experiment using rats, about 54% of the eggshell membrane fine powder is not digested and absorbed and remains in the intestine, and the remaining components prevent the proliferation of bad bacteria in intestinal bacteria and improve the intestinal flora, so the beverage of the present invention containing the same exerts excellent effects in improving or preventing adverse physical symptoms, activating organisms, promoting health, maintaining health, and the like by improving the intestinal flora, and is therefore very effective as a beauty drink, a health promoting drink, and the like.
