背景技术

医疗和美容皮肤护理领域的研究人员感兴趣的一种特定纤维化过程是在皮肤中形成胶原。胶原是胞外基质(ECM)中的主要组分并且构成人体中蛋白质的大约三分之一。在人皮肤中,胶原由真皮成纤维细胞产生,然后分泌到ECM中,其中其与现有基质聚集以形成纤维蛋白的互锁网状物。皮肤质量和外观在很大程度上取决于真皮和其胞外基质的特性。未能保持适当的胶原量被认为是皮肤老化的临床表现,诸如皱纹、下陷和松弛。

因此,期望理解与纤维化状况有关的分子和细胞过程,并且提供用于调节纤维化过程和状况的新生物分子靶标。还期望提供鉴定可以调节与纤维化状况和/或过程有关的遗传和/或生物分子靶标的化合物的新方法。进一步期望鉴定涉及抑制与皮肤中的胶原产生相关的基因的靶标。
发明内容
本公开描述了以下的新颖方法:调节纤维化状况,鉴定用于调节纤维化状况的化合物,以及制备用于调节纤维化状况的组合物。在一个实施方案中,公开了一种鉴定能够调节纤维化过程的化合物的方法。所述方法包括使多个永生化的或转化的成纤维细胞与测试化合物接触;确定与所述测试化合物接触的所述成纤维细胞的胶原抑制基因的活性水平;将所述胶原抑制基因的活性水平与对照进行比较;以及当相对于所述对照,所述胶原抑制基因的活性指示所述胶原抑制基因的上调或下调时,将所述测试化合物鉴定为能够调节纤维化过程。

对纤维化过程和状况的先前研究没有充分理解的是存在能够抑制胶原合成的出乎意料地大量的基因。这些胶原抑制基因编码蛋白质和/或RNA,所述蛋白质和/或RNA可以抑制胶原合成基因的激活和/或干扰胶原合成基因的基因产物(例如,蛋白质)的活性。现已发现,调节这些胶原调节基因中的一个或多个胶原调节基因的表达和/或由这些基因编码的蛋白质或RNA可以用于调节胶原合成。因此,可能期望靶向胶原抑制基因而不是胶原合成基因的活性。特别地,可能期望靶向先前未知为胶原抑制基因的基因的活性。

“基因表达谱分析”和“基因表达谱分析实验”意指使用任何合适的谱分析技术测量生物样本中多个基因的表达。例如,几千个基因的mRNA合成可使用微阵列技术来测定。出现的其它可使用的技术包括RNA-Seq或利用NextGen测序技术的全转录组测序。

基因调节通常涉及增加或减少由基因产生的基因表达产物的量。基因调节有时也称为基因调控,其中“上调”基因意味着促进和/或增加基因产物的产生,并且“下调”基因意味着抑制和/或减少基因产物的产生。与大多数基因一样,可以通过细胞使用的多种不同机制来在基因表达途径(例如,从蛋白质的DNA-RNA转录到翻译后修饰)中的任何步骤处调节产生胶原所涉及的基因,以增加或减少特定基因的产生。感兴趣的直接有助于胶原合成的人基因在本文中被称为“胶原合成基因”,并且示于图5中。应当理解,图5中提供的基因的子集(其未具体列出)在本文的方法中可能特别有用。
胶原由缠绕在一起以形成细长原纤维的三螺旋的蛋白质链组成。取决于矿化程度,胶原组织可以是刚性的(骨)、顺应性的(肌腱),或者具有从刚性到顺应的梯度(软骨)。在人体中发现许多不同类型的胶原,其中I型是在皮肤中发现的最常见类型。不同类型的胶原的示例性描述可见于K.Gelse等人,(2003)“Collagens–structure,function,andbiosynthesis;”Advanced Drug Delivery Reviews 55;1531–1546。胶原的功能紊乱过度产生是多种纤维化疾病(诸如硬皮病和肺纤维症)的主要原因。相反,胶原的产生不足导致老化皮肤的至少一些特征(例如,下陷、弹性损失、薄化和褶皱形成)。
上调胶原合成基因的表达通常导致胶原产生的增加。相反,抑制这些胶原合成基因的表达应导致胶原产生的减少。增强或抑制胶原合成基因的基因表达产物的活性也可以分别导致更高或更低量的胶原。另外,例如通过抑制胶原合成基因的激活和/或干扰胶原合成基因的基因表达产物,存在编码抑制胶原合成的产物的基因的相对丰度。由于尝试直接调节胶原合成的复杂性(例如,通过直接调节胶原合成基因的基因表达或抑制其基因产物的活性),可能期望调节一种或多种胶原抑制基因的表达,而不是调节一种或多种胶原合成基因的表达。例如,通过敲除胶原抑制基因(例如,经由RNA沉默)而不是上调胶原合成基因,可以更容易增加皮肤中胶原的量。如本文所用,“胶原抑制基因”是指感兴趣的抑制胶原合成并列于图6中的人基因。胶原抑制基因共享抑制胶原合成的常见功能,并且因此可以共同或单独地使用以调节胶原合成的表达和/或胶原合成基因表达产物的活性。应当理解,图6中提供的基因的子集(其未具体列出)在本文的方法中可能特别有用并被本发明所涵盖。例如,所述方法可以利用2个、3个、4个、5个、10个、15个、或甚至20个或更多个胶原抑制基因的子集。
鉴定调节纤维化过程的化合物
本文的方法涉及鉴定能够调节纤维化过程的测试化合物(“纤维化剂”)以及/或者评估化合物调节纤维化过程或治疗纤维化状况的能力。本发明方法可以用于鉴定调节胶原合成基因的表达的纤维化剂,调节胶原抑制基因的表达,以及/或者干扰胶原合成基因或胶原抑制基因的基因产物的活性。在一些实施方案中,本发明方法可以用作高通量筛选方法(即,其中可以同时测试至少25个测试化合物的筛选方法)。例如,本发明方法可以用于同时筛选多达1,000个测试化合物(例如,50个、100个、200个、500个、或750个或更多)。在一些实施方案中,所述方法涉及确定相对于对照的胶原合成的速率和/或量的变化量。胶原合成的变化可以通过蛋白质定量、报告基因活性和/或基因转录组学分析来确定。
用于本发明方法的测试化合物的一些非限制性示例是小分子、核酸(例如小干扰RNA、微RNA和小激活RNA)、抗体、植物提取物、维生素、矿物质、化妆剂和已知期望胶原调节能力的其它化合物。在一些情况下,可能期望确定测试化合物是否可以调节两种或更多种胶原抑制基因的表达(例如,3至100种基因、5至50种基因、10至30种基因、或甚至12至20种基因)。例如,可能期望确定测试化合物是否可以调节两种或更多种胶原抑制基因的表达,所述两种或更多种胶原抑制基因共享共同的作用机制,共享共同的生物化学途径,或者属于同一基因家族或超家族(例如,ZFP91、ZNF16、ZNF17和ZNF584、或者FAM107A和FAM171A2、或者CCDC57和CCDC 130)。在另一个示例中,可能期望确定测试化合物是否可以调节胶原抑制基因的表达,所述胶原抑制基因的基因产物是可成药的。“可成药的”意指已知或预测生物靶标诸如靶蛋白等以高亲和力与药物或抗体结合,这改变了靶蛋白的功能。可成药的靶标的示例可以是由胶原抑制基因编码的蛋白质。
在一些实施方案中,本发明方法涉及使多个永生化的或转化的成纤维细胞与测试化合物接触;确定所述成纤维细胞的胶原抑制基因的活性水平;将所述胶原抑制基因的活性水平与对照进行比较;以及当相对于所述对照,所测量的活性指示胶原抑制基因的上调或下调时,将所述测试化合物鉴定为能够调节纤维化过程的纤维化剂。下文更详细地描述了确定胶原抑制基因的活性的水平的方法。在一些实施方案中,所述方法还可以涉及将纤维化剂掺入组合物中以及/或者向需要治疗的个人施用纤维化剂。
本发明方法中使用的成纤维细胞不受特别限制,并且可以包括可商购获得的细胞系(例如,来自ATCC的TERT成纤维细胞;Monassas,Virginia)、从人供体获得的离体成纤维细胞、和/或这些细胞的修饰型式(例如,经永生化、经转化或经修饰以包括报告基因)。本发明方法中使用的成纤维细胞可以使用培养这种类型的细胞系的常规方法来培养,以提供测试样本。使测试样本与测试化合物接触足够量的时间以确定测试化合物是否可以调节胶原合成。测试化合物可以是对人使用(例如,经由摄入、吸入、局部施用和/或注射)安全的任何化合物,并且可以适用于药物组合物(例如,仅可通过来自专业资格人员的处方获得的组合物)、化妆品组合物(例如,非处方产品,诸如皮肤洗剂)、或药妆品组合物(例如,包括已知提供特定有益效果的活性成分的非处方产品诸如皮肤洗剂)。测试化合物可以为纯形式或者其可以与其它成分混合以促进与测试样本中的成纤维细胞接触。
在一些情况下,可能期望通过经由报告基因测量一种或多种胶原合成基因的活性和/或胶原抑制基因的活性来确定胶原合成活性,所述报告基因被插入在感兴趣基因的内源性启动子的下游。报告基因可以编码在由特定波长范围激发时以限定的荧光波长发射的蛋白质。在一些情况下,可能期望使用连接到报告基因的外源性胶原合成基因和/或胶原抑制基因启动子。例如,可以将胶原合成基因和/或胶原抑制基因启动子克隆到质粒中,连接到报告构建体以及转染到永生化/转化的成纤维细胞系中。可以定量成纤维细胞样本中存在的荧光蛋白的量,并且该荧光蛋白的量与感兴趣基因的活性直接相关。可以使用合适的荧光光谱技术(例如,根据制造商的说明使用荧光计)来定量荧光蛋白。本文使用的荧光蛋白或编码其的基因不受特别限制,并且包括绿色荧光蛋白(“GFP”)和红色荧光蛋白(“RFP”)。RFP的一些非限制性示例是mCherry、mStrawberry、mOrange和dTomato。在更具体的示例中,可能期望使用永生化的或转化的成纤维细胞系,其经由基因编辑技术(例如,锌指核酸酶、转录激活因子样效应物核酸酶(TALEN)或规律成簇间隔短回文重复(CRISPR))修饰以包括核定位信号mCherry基因(“NLS-mCherry”),该基因当感兴趣的基因(例如,COL1A1)被激活时编码mCherry RFP。
在一些情况下,可能期望使用转录组学分析来测量相对于对照与测试化合物接触的细胞的基因活性。当测试样本和对照样本的转录谱相对于彼此对应于图5或图6中的至少一个基因在指示胶原产生的期望变化的方向上的表达变化时,将测试化合物鉴定为能够调节胶原合成的纤维化剂。例如,如果期望胶原产生增加,则测试样本应显示自图5的一个或多个基因的上调和/或自图6的一个或多个基因的下调。在一些情况下,可以测量由基因特征中的一个或多个感兴趣基因编码的信使核糖核酸(“mRNA”),并将其与对照进行比较。在一些情况下,可以使mRNA逆转录并且测量对应的互补DNA(“cDNA”)。可以使用任何合适的定量核酸测定。例如,常规定量杂交、Northern印迹和聚合酶链反应程序可以用于定量测量生物样本中mRNA转录物或cDNA的量。任选地,可以在杂交之前通过聚合酶链反应(PCR)扩增mRNA或cDNA。然后通过例如与对由一个或多个感兴趣的基因(例如,胶原抑制基因)编码的mRNA或cDNA具有特异性的寡核苷酸杂交来检查mRNA或cDNA样本,所述寡核苷酸任选地固定在基底上(例如,阵列或微阵列)。核酸与对感兴趣基因具有特异性的寡核苷酸探针的结合允许鉴定和定量所述基因的表达水平。定量基因表达的转录组学方法的合适示例公开于美国专利9,434,993;10,036,741;和10,282,514中。
在一些情况下,测试化合物可以溶解在合适的媒剂中,诸如二甲亚砜(DMSO)、水、或水性/有机组合溶剂诸如乙醇/水。在暴露于测试化合物和/或对照之后,可以从测试细胞和参考细胞中提取mRNA。从细胞中提取的mRNA可以任选地逆转录成cDNA并且用荧光染料标记(例如,如果要执行双色微阵列分析,则用红色和绿色)。在一些情况下,可以对cDNA样本进行单色微阵列分析,并且如果需要可以处理多个平行测定。cDNA样本可以与包括多个探针(例如,数十、数百或数千个探针)的微阵列共杂交。在一些实施方案中,微阵列上的每个探针具有独特的探针组标识符。由扫描仪扫描微阵列,该扫描仪激发染料并测量荧光量。计算装置分析原始图像以确定存在的cDNA的量,其代表基因的表达水平。扫描仪可以结合计算装置的功能。通常,由系统采集的基因表达数据可以包括:i)基因表达的上调(例如,与参考材料(例如,cDNA)相比,测试材料(例如,cDNA)与探针的结合更大),ii)基因表达的下调(例如,测试材料(例如,cDNA)比测试材料(例如,cDNA)与探针的结合降低),iii)非波动基因表达(例如,与参考材料(例如,cDNA)相比,测试材料(例如,cDNA)与探针的结合类似),以及iv)无可检测信号或噪声。上调和下调的基因可被称为“差异表达的”。差异表达的基因可以被进一步分析和/或分组在一起(例如,经由已知的统计方法)以鉴定代表纤维化状况或对测试化合物有生物应答的基因。

含有纤维化剂的组合物
一旦化合物被鉴定为纤维化剂,就可以将其掺入组合物中并且施用于需要治疗的个人。当然,应当理解,在一些情况下,可能期望将纤维化剂以纯形式(即,未稀释或不含载体)施用于需要治疗的个人。纤维化剂可以与合适的载体和其它任选成分混合,例如,使用常规配制和加工技术,以提供适于施用于个人的组合物。本发明组合物中所包含的任选成分不受特别限制,只要它们不会不可接受地改变组合物调节纤维化过程诸如胶原合成的能力即可。任选组分当存在时应该适用于人组织,而没有不适当的毒性、不相容性、不稳定性、变应性应答等。任选成分可以0.0001%至50%(例如,0.001%至20%、或甚至0.01%至10%)存在。本文所列的量仅用作指导,因为在组合物中使用的任选成分的最佳量将取决于所选的具体成分,这是由于它们的功效和功能可能相差很大。
组合物的形式应针对纤维化剂的期望施用途径(例如,局部施用、口服摄入或注射)进行定制。例如,组合物可以为以下的形式:溶液、悬浮液、分散液、乳液、粉末、片剂、胶囊、洗剂、乳膏、凝胶、化妆水、喷雾、气雾剂、软膏、清洁洗涤液、实心棒、洗发剂、毛发调理剂、糊剂、泡沫、粉末、摩丝、剃刮膏、擦拭物、条带、贴片、伤口敷料、胶布绷带、水凝胶、成膜产品、面部和皮肤面膜(具有和不具有不溶性片)。组合物可以被提供在包装中,所述包装的尺寸被设定为储存用于一个治疗周期的足够量的组合物。
在一些情况下,可以通过将纤维化剂与皮肤病学可接受的载体混合来配制本文的组合物以用于例如向个人的头皮、皮肤或粘膜局部施用。此类组合物还可以包含此类组合物通常所包含种类的一种或多种任选成分。“皮肤病学可接受的载体”意指载体适于局部施用于角质组织,具有良好的美学特性,与组合物中的成分相容,并且不引起任何不合理的安全或毒性问题。皮肤病学可接受的载体可以按组合物的重量计1%至95%(例如,10%至90%、30%至70%、50%至60%)存在。载体可以是含水的或无水的。例如,合适的载体可以包括水、水混溶性溶剂和油。合适的水混溶性溶剂包括一元醇、二元醇、多元醇、甘油、二醇、聚亚烷基二醇诸如聚乙二醇、以及它们的混合物。合适的油包括有机硅、烃、酯、酰胺、醚、以及它们的混合物。这些油可为挥发性的或非挥发性的。
可以添加到旨在局部施用于皮肤的组合物的任选成分可以包括但不限于抗痤疮活性物质、脱屑活性物质、抗脂肪团剂、螯合剂、类黄酮、美黑活性物质、非维生素抗氧化剂和自由基清除剂、毛发生长调节剂、抗皱纹活性物质、抗萎缩活性物质、矿物质、植物甾醇和/或植物激素、N-酰基氨基酸化合物、抗微生物剂或抗真菌活性物质、以及其它有用的皮肤护理活性物质。可以适用于本发明组合物的皮肤护理活性物质的一些非限制性示例描述于美国公布2006/0275237和US 2004/0175347以及国际化妆品成分字典和手册,第十三版。
在一些情况下,在本发明组合物中使用的任选成分可以包括已知的抗纤维化活性物质以抑制胶原产生。抗纤维化活性物质的一些非限制性示例公开于美国公布2013/0209490;美国专利7,026,283;以及Wynn等人,Journal Clin.Invest.,第117卷第3期,2007年三月,第524页。在一些情况下,组合物可以任选地包括促进胶原产生的纤维化活性物质(“促纤维化活性物质”),例如在作为抗纤维化活性物质的相同或类似的生物化学途径上起作用,但具有相反的效果。
本发明的组合物可通过制备此类组合物的常规方法来制备。这些方法通常涉及在一个或多个步骤中将成分混合至相对均一的状态,可使用或不使用加热、冷却、施加真空等。通常,通过首先将水相材料与脂肪相材料分别混合,然后视情况混合两相以获得所期望的连续相,来制得乳液。组合物优选地制备成例如以使活性物质的稳定性(物理稳定性、化学稳定性、光稳定性等)和/或递送最优化。
使用方法
通常,通过本发明方法鉴定的纤维化剂可以根据使用类型的组合物的常规方法施用。包含有效量的纤维化剂的组合物可以通过向需要治疗的个人施用组合物来用于调节纤维化过程和/或治疗纤维化状况。需要治疗的个人是表现出纤维化状况的症状或被诊断患有纤维化状况的人员。当然,应当理解,在一些情况下,未表现出纤维化状况的症状或未被诊断出患有纤维化状况的个人仍然可能期望治疗。例如,使用者可能希望靶向皮肤的一部分,该部分未表现出胶原消耗的症状(例如,皱纹和松弛的皮肤),但是已知发展该状况的症状(例如,通常不被服装覆盖的皮肤表面,诸如面部皮肤)。本文的组合物可以有效治疗纤维化状况和/或调节纤维化过程的任何合适的方式施用。
含有有效量的用于调节纤维化过程的纤维化剂的组合物可以在治疗期期间一天一次、一天两次或每天更频繁地施用。理想的治疗期是纤维化剂提供期望有益效果的足够时间。例如,治疗期可以为纤维化剂提供足够的时间以在纤维化状况中提供明显和/或可测量的改善或者改变纤维化过程。治疗期可持续至少1周(例如,约2周、4周、8周或甚至12周)。在一些情况下,治疗期将延长至多个月(即,3至12个月)或多年。在一些情况下,含有有效量的纤维化剂的组合物可以在至少2周、4周、8周或12周的治疗期期间在一周的大多数天数内施用(例如,一周至少4、5或6天)、一天至少施用一次或甚至一天施用两次。
实施例
下文提供了本文所述方法的各个方面的非限制性实施例。这些实施例仅以例示性目的给出,并且不旨在解释为是对本发明的限制,因为其许多变型是可行的。
实施例1
该实施例描述了一种新颖报告系统(“Red-COL1A1”),其可以用于确定基因的活性水平。使用Red-COL1A1报告系统来鉴定感兴趣的胶原抑制基因。
在该实施例中,使用RNA干扰(“RNAi”)筛选来鉴定似乎抑制胶原合成的基因。使用锌核酸酶(TALENs)来修饰永生化的人皮肤成纤维细胞(BJhTERT,可获自ATCC,Manassas,Virginia),以将mCherry报告基因置于COL1A1的内源性启动子的下游。COL1A1在I型胶原(有时称为胶原I)的合成中是众所周知的,其是占人体中发现的胶原超过90%的纤维状胶原。报告基因编码mCherry RFP,其可以使用已知的荧光蛋白检测技术来定量。在荧光激活细胞分选(“FACS”)技术中使用转化生长因子β(“TGFβ”)、众所周知的COL1A1激活细胞因子,以选择具有低内源性表达和高水平诱导型表达的mCherry基因的细胞系。图2中示出了用于发展细胞系的过程的总结。
使用Red-COL1A1报告细胞系对大约22,000个基因进行基因组宽筛选,以鉴定具有调节胶原表达的能力的基因。基因组宽筛选涉及根据以下方法对Red-COL1A1细胞系进行RNAi筛选。利用Agilent Technologies Velocity 11将2.5μl的500nM siRNA(Dharmacon)以机器人形式预印刷到黑壁384孔微板(Greiner#781091)上。通过将0.2μl脂质体RNAiMAX与7.3μl Gibco Opti-MEM减少血清培养基预混合5分钟并将混合物分配到预印刷的siRNA测定板中以形成RNA-脂质复合物,来进行含有测试样本的每个孔的反向siRNA转染。利用Thermo Scientific Multidrop Combi Reagent Dispenser进行分配到384孔板中。在将3,000个Red-COL1A1细胞分配到每个孔中之前,使复合物形成持续20分钟。转染后3天,以10ng/mL的最终浓度向选定的对照孔(阳性对照)添加TGFβ。试验筛选是复制进行的。敲除6天后,使用含有2%蔗糖的4%多聚甲醛将细胞固定15分钟,用D-PBS洗涤一次,并且用稀释于D-PBS中的Hoechst 33342(1:10,000)染色10分钟。然后在高通量成像之前将细胞用D-PBS洗涤两次。
使用自动图像采集和分析来定量荧光蛋白。在来自Molecular Devices的IMAGEXPRESS牌微高含量成像软件上以10倍放大率按顺序获取每个孔的四个位点(每个位点在405nm和561nm的两个激发波长下成像)。使用来自Molecular Devices的METAXPRESS牌高含量图像分析软件来分析图像。使用“Translocation-Enhanced”模块来确定具有高于背景的阳性核定位mCherry信号的细胞的百分比。参考siNT(n=8)的平均值对每个样本孔的阳性mCherry细胞的平均倍数变化百分比进行归一化。使用来自BMC Bioinformatics的SCREENSIFTER牌RNAi数据筛选软件来处理和分析原始数据,并且确定筛选平行测定的平均值±SD。使用数据筛选软件中可用的衍生方法得出阳性命中(>2)的阈值。类似地,将每个样本孔的平均细胞计数归一化为同一板中siNT孔中的平均细胞计数,其中阈值任意设定为80%。
基因组宽筛选致使鉴定322个基因(Z分数>10),这些基因抑制胶原产生,如通过其在敲除时增加Red-COL1A1细胞中m-cherry表达的能力所展示的。为了排除脱靶RNAi效果,进行去卷积RNAi筛选,其中针对可供购自Dharmacon的297个基因,测试了针对每个基因的单独siRNA。以2个平行测定来进行筛选,每个平行测定具有技术复制。与汇集的siRNA相比,单独siRNA往往会引发不太有效的效果。因此,使用2.5倍阈值信号,通过至少两个独立的siRNA来确认被鉴定为抑制胶原产生的322个基因中的146个基因。评估敲除此146个基因对胶原产生的效果展示了m-cherry蛋白和胶原1A1蛋白两者在Red-COL1A1 BJ成纤维细胞细胞系中的诱导。确认在另外的成纤维细胞细胞系中进行,并且方法经由Western印迹分析和定量逆转录聚合酶链反应(qRT-PCR)来验证。在该测试中鉴定的胶原抑制基因示于图6中。
实施例2
该实施例展示了可以通过降低由胶原抑制基因编码的蛋白质的活性来增加胶原合成基因表达。为了展示列于图6中的基因的蛋白质的小分子抑制与这些基因的表达的siRNA敲除具有类似的效果,选择了ADRA1B受体蛋白的拮抗剂以用于评估其对COL1A1表达的效果。结果的总结示于图3A和图3B中,其示出了在无毒剂量下增加COL1A1表达的ADRA1B拮抗剂α-麦角隐亭剂量增加。图3A示出了增加浓度的ADRA1B拮抗剂alpha-麦角隐亭增加了由在永生化的tertBJ成纤维细胞中位于内源性COL1A1启动子下游的mCherry报告分子构建体的表达增加产生的荧光量(参见图2)。这种增加的荧光与增加的胶原合成直接相关。图3A中还示出了以下的结果:阳性对照(TGFβ)、TGFβ的媒剂对照(培养基对照)、以及α-麦角隐亭的媒剂对照(DMSO对照)。如图3B所示,增加浓度的α-麦角隐亭对永生化的tertBJ成纤维细胞没有毒性,如通过细胞计数所确定的通过用4′,6-二脒基-2-苯基吲哚(DAPI)(其是与DNA中富含腺嘌呤-胸腺嘧啶的区域强烈结合的荧光染色剂)染色细胞核来确定细胞计数。DAPI染色是众所周知的技术,其用于确定荧光显微镜中的细胞计数。
实施例3
此预测性实施例使用转录分析技术展示了本发明方法鉴定调节胶原合成的纤维化剂的能力。在该实施例中,使用含有用于图6中基因的探针的合适的微阵列(例如,Affymetrix等)来鉴定胶原抑制基因的基因表达的变化。单独的实验(称为批次)通常包括使用选定微阵列分析的30至96个样本。每个批次含有6个平行测定的媒剂对照(例如,DSMO),2个平行测定的阳性对照样本,所述阳性对照在使用的细胞类型(例如,TGF-β)和测试化合物的样本中产生强烈可重现的效果。测试化合物的平行测定在单独的批次中进行以补偿批次效果。在6孔板中进行体外测试以提供足够的RNA以用于分析(2-4μg总RNA产率/孔)。
在正常细胞培养瓶和细胞培养板(Corning,Lowell,Mass.)中于补充有10%胎牛血清(HyClone,Logan,Utah)的伊格尔最低限度必需培养基(ATCC)中培养人永生化成纤维细胞(例如,BJ细胞系,来自ATCC)。将细胞在具有5%CO2的加湿培养箱中于37℃下温育。在暴露前二十四小时,将细胞自T-75烧瓶进行胰蛋白酶化,并接种到6孔板中的基础生长培养基中。在t=0时,除去培养基并且按照实验设计用适当的加药溶液替换。加药溶液是前一天在无菌4ml Falcon按扣盖管中制备的。纯测试材料可以1-200μM的浓度制备,并且植物提取物可以按加药溶液的重量计0.001%至1%的浓度制备。在化学暴露6至24小时之后,观察细胞并使其成像。在细胞裂解和RNA分离之前,用显微镜检查孔,以评估毒性的形态证据。如果形态变化足以表明细胞毒性,则测试较低浓度的测试化合物。然后将细胞用含有β-巯基乙醇(Qiagen,Valencia,Calif.)的350μl/孔的RLT缓冲液裂解,转移到96孔板中,并储存在-20℃下。
根据制造商的说明,使用RNAdvance组织结合磁珠(BeckmanCoulter)从RLT缓冲液中分离来自细胞培养物批次的RNA。根据制造商的说明,使用AmbionMessage AmpTM II生物素增强试剂盒(Applied Biosystems Incorporated)标记每个样本的1μg总RNA。使所得带生物素标记和片段化的cRNA与选定微阵列杂交(例如,AffymetrixHG-U133A 2.0),然后使用制造商提供的方案(例如,Affymetrix)将其洗涤、染色和扫描。以统计上显著的方式(例如,p值≤0.1、0.05或0.01),在微阵列数据上进行统计分析(例如,t检验)以鉴定由测试化合物调节(上调或下调)的胶原抑制基因,并且当胶原抑制基因中的至少一个胶原抑制基因上调或下调时,将测试化合物鉴定为能够调节纤维化过程。
实施例4
该实施例展示了可以通过降低胶原抑制基因的mRNA和蛋白质的水平(什么-mRNA/表达?的水平)来增加胶原合成基因表达以及可以通过提高胶原抑制基因的mRNA和蛋白质的水平来减少胶原合成基因表达。为了展示降低列于图6中的基因的mRNA水平并且因此减少蛋白质增加了COL1A1 mRNA的表达,使用对5个基因(包括QPCT、FNDC11、TGFBI、ADAMTS5、ADRA1B)中的每一个基因具有特异性的siRNA来减少这些基因的表达,如实施例1中所述。结果的总结示于图4A-E中,其示出了使用对QPCT、FNDC11、TGFBI、ADAMTS5、ADRA1B mRNA具有特异性的siRNA敲除这些基因的表达而单独增加的COL1A1 mRNA。相反,通过在降低COL1A1蛋白质水平的成纤维细胞中过表达QPCT、FNDC11、TGFBI、ADAMTS5、ADRA1B蛋白质来提高这些蛋白质的水平。将siRNA转染到成纤维细胞中-根据制造商说明,用RNAiMAx(Invitrogen-Thermo Fisher Scientific,Waltham MA)在无血清OPTIMEM中进行siRNA转染。简而言之,将2μL RNAiMax试剂与指示的siRNA(Dharmacon Inc.,Lafayette CO;Thermo FisherScientific)在OPTIMEM中复合20分钟。将反应混合物添加到具有30,000个胰蛋白酶化细胞的24孔板中。所有转染用最终浓度为25nM的siRNA进行。Graphical Nomenclature.siCON–对照siRNA;siQPCT、siFNDC11、siTGFBI、siADAMTS5、siADRA1B–对这些基因中的每个基因具有特异性的siRNA。通过电穿孔过表达蛋白质-将30ug质粒电穿孔到具有转染系统的800,000个BJ-Tert成纤维细胞中。使用P100尖端在1600V 10ms 2脉冲设定下进行电穿孔。将细胞接种到6孔板中并在蛋白质和定量之前温育48小时。CON–无蛋白质插入的对照表达质粒;QPCT、FNDC11、TGFBI、ADAMTS5、ADRA1B–含有所指示蛋白质的表达质粒。

Claims (15)
1.一种鉴定能够调节纤维化过程的纤维化剂的方法,包括:
a)使多个永生化的或转化的成纤维细胞与测试化合物接触;
b)确定与所述测试化合物接触的所述成纤维细胞的胶原抑制基因的活性水平;
c)将所述胶原抑制基因的活性水平与对照进行比较;以及
d)当相对于所述对照,所述胶原抑制基因的活性指示所述胶原抑制基因的上调或下调时,将所述测试化合物鉴定为能够调节纤维化过程的纤维化剂。
2.根据权利要求1所述的方法,其中所述胶原抑制基因的基因产物是可成药的。
3.根据权利要求1或2所述的方法,其中确定两种或更多种胶原抑制基因的活性水平。
4.根据权利要求3所述的方法,其中所述两种或更多种胶原抑制基因来自同一基因家族。
5.根据前述权利要求中任一项所述的方法,其中所述测试化合物是选自以下的核糖核酸(RNA):小干扰RNA、微RNA和小激活RNA。
6.根据前述权利要求中任一项所述的方法,其中所述胶原抑制基因的活性是通过蛋白质定量和转录组学分析中的至少一者来确定的。
7.根据权利要求6所述的方法,其中所述胶原抑制基因的活性是通过测量报告基因活性来确定的,并且其中所述蛋白质是由报告基因编码的荧光蛋白,所述报告基因被插入在处于所述成纤维细胞中的所述胶原抑制基因的内源性启动子的下游。
8.根据权利要求7所述的方法,其中所述荧光蛋白是选自以下的红色荧光蛋白:mCherry[SEQ ID NO:215]、mStrawberry[SEQ ID NO:216]、mOrange[SEQ ID NO:217]和dTomato[SEQ ID NO:218]。
9.根据权利要求6所述的方法,其中所述胶原抑制基因的活性是通过转录组学分析来测量的,所述转录组学分析包括:
a)生成与所述测试化合物接触的所述多个成纤维细胞的转录谱,其中所述转录谱包括与胶原抑制基因的转录有关的数据,
b)将所述成纤维细胞的转录谱与对照进行比较,以及
c)当相对于所述对照,所述成纤维细胞的转录谱指示所述胶原抑制基因的上调或下调时,将所述测试化合物鉴定为能够调节纤维化过程的纤维化剂。
10.根据权利要求9所述的方法,其中所述转录组学分析还包括从所述成纤维细胞中分离RNA,由所分离的RNA产生带标记的cRNA或cDNA,以及使所述带标记的cRNA或cDNA与包含针对所述胶原抑制基因的探针的微阵列杂交。
11.根据前述权利要求中任一项所述的方法,还包括向需要治疗的个人施用所述纤维化剂。
12.根据权利要求11所述的方法,其中与安慰剂相比,施用所述纤维化剂导致胶原产生的增加。
13.根据前述权利要求中任一项所述的方法,还包括将所述纤维化剂与载体混合以提供治疗组合物。
14.根据权利要求13所述的方法,其中所述治疗组合物包含选自以下的附加成分:皮肤护理活性物质、抗纤维化活性物质、促纤维化活性物质、以及它们的组合。
15.根据权利要求13所述的方法,其中所述治疗组合物处于适于由个人局部施用、摄入或注射中的至少一种的形式。
