实施例1
取如下重量份的中药:麻黄10份、干姜10份、透骨草5份、红花5份、地龙4份、紫草3份、桑枝3份、杜鹃叶3份、小白蒿3份、水柏枝2份、刺柏2份置于酒坛中,在酒坛中再加入50重量份的水溶性硅元素(购自华夏瑞天(北京)生物科技有限公司)和900重量份的60°的高粱酒,混合均匀,用塞子塞紧,再用塑料薄膜裹紧密封。置于25℃下泡制30天。过滤,取滤液即为药酒。将药酒装入滚珠式药瓶中,使用时,直接涂抹病灶,涂抹后盖上保鲜膜,自动发热2min,取下保鲜膜,轻轻揉搓涂抹部位,药液即可完全被吸收。
大鼠经皮毒性试验
1.试验动物
健康大鼠200~280g。饲养温度(25±1)℃,自由饮食饮水,适应性饲养一周。
2.试验药物
实施例1制备的药酒和60%的酒精。
3.试验方法
选择皮肤健康的大鼠10只,雌雄各5只,在其背部沿中线两侧剃毛,左侧为试验区,右侧为对照区,面积均为3×3cm。将试验药物涂抹与大鼠左侧脱毛皮肤处,以60%的酒精为对照,涂抹后用保鲜膜覆盖,用无菌纱布固定,2h后取下纱布和保鲜膜,用温水清洗。重复上述操作一次。然后连续14天观察毒性反应情况。14天后,脱颈椎处死大鼠,观察大鼠主要脏器有无明显病理改变。取下左右两侧皮肤,用10%的中性甲醛固定,进行苏木精-伊红染色观察。
4.结果
各组动物无中毒反应,无死亡,未出现异常变化,试验期间活动正常,饮食饮水正常,毛发正常,与正常小鼠无显著差异。观察大鼠主要脏器(心、肝、脾、肺、肾、睾丸、卵巢、前列腺、胰腺、子宫)没有明显病理学改变,苏木精-伊红染色观察正常。因此,大鼠经皮毒性试验结果表明药液经皮给药是安全的。
Dickkopfl(DKK-1)是一种分泌性糖蛋白,是经典Wnt信号途径的可溶性拮抗剂。目前已有研究证实,DKK-1是关节重塑中的主要调控因子。因此,本研究以风湿骨病患者血清中DKK-1水平,来评价药酒对骨关节疾病的治疗作用。采用酶联免疫吸附法(enzyme-linkedimmunosorbent assay,ELISA)检测血清样本中DKK-1水平。
选择符合研究要求的48例类风湿性关节炎患者作为研究对象。48例患者中男14例,女34例;年龄35~60岁,平均年龄(47.54±5.46)岁;病程15~163个月,平均病程(72.73±27.03)月。
将实施例1制备的药酒装入规格为15ml的滚珠式药瓶中,直接涂抹于患处,1.5ml/次,一日3次,3瓶为一个疗程,治疗3个疗程。于患者治疗前后取患者空腹静脉血,采用ELISA试剂盒(BIOMEDICA,BI-20413)检测血清中DKK-1水平。结果如表1。

由表1可知,本发明提供的风湿骨痛药酒能够显著降低类风湿性关节炎患者血清中的DKK-1水平。
实施例2
取如下重量份的中药:麻黄10份、干姜10份、透骨草6份、红花6份、地龙5份、紫草5份、桑枝4份、杜鹃叶4份、小白蒿4份、水柏枝3份、刺柏3份置于酒坛中,在酒坛中再加入60重量份的水溶性硅元素(购自华夏瑞天(北京)生物科技有限公司)和880重量份的60°的高粱酒,混合均匀,用塞子塞紧,再用塑料薄膜裹紧密封。置于30℃下泡制20天。过滤,取滤液即为药酒。
实施例3
取如下重量份的中药:麻黄9份、干姜9份、透骨草4份、红花3份、地龙3份、紫草3份、桑枝3份、杜鹃叶2份、小白蒿2份、水柏枝1份、刺柏1份置于酒坛中,在酒坛中再加入40重量份的水溶性硅元素(购自华夏瑞天(北京)生物科技有限公司)和920重量份的55°的高粱酒,混合均匀,用塞子塞紧,再用塑料薄膜裹紧密封。置于20℃下泡制40天。过滤,取滤液即为药酒。
以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。
Claims (7)
1.一种风湿骨痛药酒,其特征在于,由包括如下重量份的原料制备得到:中药40~60份、水溶性硅元素40~60份、白酒800~1000份;
所述中药包括如下重量份的原料:麻黄8~10份、干姜8~10份、透骨草1~6份、红花1~6份、地龙1~5份、紫草1~5份、桑枝2~4份、杜鹃叶2~4份、小白蒿1~4份、水柏枝1~3份、刺柏1~3份。
2.如权利要求1所述的风湿骨痛药酒,其特征在于,所述白酒的度数为55~65°。
3.如权利要求2所述的风湿骨痛药酒,其特征在于,所述白酒包括以高粱、大米、糯米、小麦和玉米中的一种或几种为原料酿造的白酒。
4.一种权利要求1所述的风湿骨痛药酒的制备方法,其特征在于,将中药、水溶性硅元素和白酒混合泡制,得到风湿骨痛药酒。
5.如权利要求4所述的制备方法,其特征在于,所述泡制的时间为20~40天。
6.如权利要求5所述的制备方法,其特征在于,所述泡制的温度为20~30℃。
7.如权利要求6所述的制备方法,其特征在于,所述泡制在密封环境下进行。
