本发明公开了一种用于治疗牙周炎的苗药固齿方及制备方法,其包括:青盐、生石膏、续断、散血飞、对叶莲、瘙羊古、艾纳香、卷柏和鸡屎藤,通过细胞学试验研究得知,苗药固齿方具有较好的促进成骨细胞增殖并促进成骨前体细胞向成熟成骨细胞分化的作用,通过将苗药固齿方制作而成的原液作为中药添加剂,制备成相关的口腔护理产品,例如苗药固齿方牙膏、苗药固齿方漱口水、苗药固齿方口腔糖及含片,解决了经常服用抗菌药物或抗炎药物导致的口腔菌群失衡和副作用大的问题。

技术领域
本发明涉及苗药的应用及其制剂领域,尤其涉及一种用于治疗牙周炎的苗药固齿方及制备方法。
背景技术
牙周炎(Periodontitis)是全球第六大流行病,是我国中老年人牙齿缺失的主要原因,严重危害着人类的健康。近一个世纪以来,世界各国学者已经做了多项研究,但牙周病的发病机制尚未完全清楚。目前已公认牙周病是细菌为主的多因素疾病,宿主免疫的保护-破坏机制是牙周病进展的重要环节。进一步揭示诱发牙周炎免疫炎症反应的关键因子,将为牙周炎病因学的研究提供新的证据。中药作为我国的传统药物对多种疾病均有较好的治疗效果,深入探索中药在牙周病方面的作用机制将为牙周炎的治疗提供新的方向。
根据中医的理论,肾虚则齿衰,肾固则齿坚。用于治疗牙周病的中药主要由补肾、滋阴、凉血等成分组成,目前研究较多的中药主要有以古方六味地黄丸为基础的固齿丸、固齿膏等。中药作为牙周病治疗中调节宿主免疫反应的一个辅助方法,有待于进一步的研究和发掘。
牙周炎的药物治疗包括全身和局部药物治疗,全身用药主要有以下三类,即抗菌类药物(如四环素类)、宿主免疫调节类药物(如非甾体类抗炎药)和中药。经常服用抗菌类药物可能导致口腔内正常菌群失调,并可能发生耐药菌株;经常服用抗炎药物,副作用较大。我国中药一贯以成本低、副作用小、疗效显著而为世界医学多称道,尤其是其中的活性物质具有通过多靶点、多环节发挥多种效应的优势。因此急需一种温和可控,副作用低的中药药物。
发明内容
本发明提供一种用于治疗牙周炎的苗药固齿方及制备方法,以克服经常服用抗菌药物及抗炎药物所引起的口腔菌群失衡及副作用大的问题。
为了实现上述目的,本发明的技术方案是:
一种用于治疗牙周炎的苗药固齿方,包括:青盐、生石膏、续断、散血飞、对叶莲、瘙羊古、艾纳香、卷柏和鸡屎藤。
进一步的,其原料组分按质量份包括为:
青盐5-15份
生石膏5-15份
续断5-12份
散血飞5-10份
对叶莲5-10份
瘙羊古5-12份
艾纳香5-10份
卷柏5-8份
鸡屎藤5-8份。
进一步的,其原料组分按质量份包括为:青盐15份、生石膏15份、续断12份、散血飞5份、对叶莲5份、瘙羊古8份、艾纳香8份、卷柏8份和鸡屎藤5份。
一种用于治疗牙周炎的苗药固齿方的制备方法,将本申请公开的一种用于治疗牙周炎的苗药固齿方加十倍质量的蒸馏水浸泡2h,大火煮至沸腾后,煎煮一段时间,取第一次药煎液置于容器内,再次向药渣中加入十倍质量的蒸馏水,大火煮至沸腾后煎煮一段时间,取第二次药煎液与第一次药煎液合并,将合并液冷却至室温后加入95%乙醇沉淀,使药液中的乙醇浓度为60%,离心后取上清液,煎煮浓缩至1g/ml作为原液,将原液过滤细菌,装入无菌瓶。

进一步的,苗药固齿方经大火煮至沸腾后,煎煮30分钟后取出药煎液。

通过检索文献TCMSP(Traditional Chinese Medicine Systems PharmacologyDatabase and Analysis Platform)数据库获取青盐、生石膏、续断、散血飞、对叶莲、瘙羊古、艾纳香、卷柏、鸡屎藤的主要化学成分,设定OB≥30%,DL≥0.18为阈值,其中0B指口服生物利用度,DL指类药性,筛选中药活性成分,利用TCMSP靶点预测中药固齿方的潜在靶点;检索GeneCards,Disgenet、TTD、DrugBank数据库中牙周炎相关靶点,将得到的活性成分作用靶点与牙周炎疾病作用靶点相结合,得到疾病-药物共同作用靶点,利用STRING数据库对获得的疾病-药物靶点构建PPI网络;利用Metascape平台,对疾病-药物靶点进行GO生物功能富集分析和KEGG代谢通道富集分析,而后采用Cytoscape3.7.1软件构建“苗药固齿方-牙周炎疾病靶点-通路”网络,并根据网络拓扑学参数分别筛选固齿复方治疗牙周炎的核心成分和核心靶点,最后通过Autodock Vina和PYMOL软件进行分子对接和可视化操作。
中药固齿方治疗牙周炎的核心活性成分如表1-7所示,续断8个,卷柏1个,鸡屎藤11个,瘙羊古4个,对叶莲1个,艾纳香12个,见血飞6个。苗药固齿方作用与牙周炎相互作用的靶点有98个,表8所示为排名前11位相互作用靶点、连接度、介度及紧密度。综合文献分析,上述中药主要通过抑制牙周炎的炎症反应及促进骨形成来治疗牙周炎。

本实施例提供了一种用于治疗牙周炎的苗药固齿方及制备方法,包括青盐、生石膏、续断、散血飞、对叶莲、瘙羊古、艾纳香、卷柏和鸡屎藤,生石膏中的Ti可激活吞噬细胞,加强抗炎作用。续断能够通过PI3K/AKT信号通路调节骨重塑过程中的成骨细胞与破骨细胞活性,并能通过抑制促炎细胞因子如工L-6、工L-1β等起到抗炎作用。对叶莲通过显著减少CINC-1的表达发挥抗炎作用,并且有着抗氧化活性。卷柏能通过ERK1/2信号通路抑制炎症发生发展。鸡屎藤同样具有抗炎活性,能够使NF-kB信号通路失活,降低促炎细胞因子的水平。艾纳香挥发油具有抗肿瘤、抗菌和抗氧化活性的作用。

在具体实施例中,将各个实施例中的苗药固齿方加十倍质量的蒸馏水浸泡2h,大火煮至沸腾后,煎煮30分钟,取第一次药煎液置于容器内,再次向药渣中加入十倍质量的蒸馏水,大火煮至沸腾后,煎煮30分钟,取第二次药煎液与第一次药煎液合并,通过煎煮两次可以充分煎出苗药固齿方的有效成分,将合并液冷却至室温后加入95%乙醇沉淀,是药液中的乙醇浓度为60%,离心后取上清液,煎煮浓缩至1g/ml作为原液,将所述原液通过微孔滤膜过滤细菌后,装入无菌瓶。
采用CCK-8试验确定苗药固齿方的细胞毒性,具体操作方式为:细胞传代后,按照成骨前体细胞MC3T3-E1每孔2000个细胞,破骨前体细胞RAW264.7每孔4000个细胞,设置0g/m1-1*10-7g/ml八个浓度梯度,其中0g/ml为不添加苗药固齿方,每个浓度梯度铺设5个副孔,并设置0h、24h、48h、72h四个时间段,按照CCK-8液:培养基=1:9的比例在相应的时间段每孔加入100μL的CCK-8液,放入细胞培养箱孵育1h后在酶标仪上波长为450nm处测得0D值。
再通过茜素红染液对细胞进行染色,光镜下对细胞进行拍照,观察增长情况,具体操作方式为:
(1)配制茜素红染液:称取2g茜素红溶于100ml低氘水DDW中,滤纸过滤,用10%氢氧化钠调整其PH值至4.2,过碱则用盐酸调整;
(2)取出六孔板,用PBS缓冲液洗3次后用4%的聚甲醛固定15-20min,用低氘水DDW洗3次,随后用2%茜素红染液染色并置于37℃孵箱中30min,再用低氘水DDW洗5次,光镜下拍照。
通过CCK8对不同实施例的组分进行毒性测试,对每个浓度0h、24h、48h、72h不同时间三次采样,通过大数据统计得出结论:
如图1所示,实施例1的苗药固齿方浓度仅在1*10-1g/ml时对MC3T3-E1细胞的细胞增殖有明显的抑制作用(注:***P<0.001,****P<0.0001),而其余浓度对细胞增殖无影响。如图2所示,浓度仅为1*10-1g/ml时对RAW264.7细胞有明显抑制作用(注:**P<0.01,*P<0.05),而其余浓度对细胞增殖无影响。
如图3所示,实施例2中的苗药固齿方浓度为1×10-ag/ml及以上浓度时,在48h,72h对MC3T3-E1细胞有明显的抑制作用(注:***P<0.001,****P<0.0001),如图4至图6所示,实施例2的苗药固齿方分别在1*10-1g/ml、1*10-2g/ml、1*10-3g/ml浓度下对MC3T3-E1细胞的抑制作用(注:***P<0.001,****P<0.0001)。如图7所示,实施例2中的苗药固齿方浓度为1×10-ag/ml及以上浓度时,在48h,72h对RAW264.7细胞有明显的抑制作用(注:料P<0.01,料*P<0.001),如图8至图10所示,实施例2的苗药固齿方分别在1*10-1g/ml、1*10-2g/ml、1*10-3g/ml浓度下对RAW264.7细胞的抑制作用(注:**P<0.01,***P<0.001)。
如图11所示,实施例3的苗药固齿方浓度为1×10-2g/ml及以上浓度时,在48h,72h对MC3T3-E1细胞有明显的抑制作用(注:**P<0.01,***P<0.001),如图12所示,实施例3的苗药固齿方浓度为1×10-2g/ml及以上浓度时,在48h,72h对RAW264.7细胞有明显的抑制作用(注:****P<0.0001,***P<0.001)。
如图13所示,实施例4的苗药固齿方浓度为1×10-2g/ml及以上浓度时,在48h,72h对MC3T3-E1细胞有明显的抑制作用(注:料*P<0.001),如图14所示,实施例4的苗药固齿方浓度为1*10-1g/ml时,在48h,72h对RAW264.7细胞有明显的抑制作用(注:料P<0.01)。
如图15所示,实施例5的苗药固齿方浓度为1×10-3g/ml及以上浓度时,在48h,72h对MC3T3-E1细胞有明显的抑制作用(注:料P<0.01,料*P<0.001),如图16所示,实施例5的苗药固齿方浓度为1×10-3g/ml及以上浓度时,在48h,72h对RAW264.7细胞有明显的抑制作用(注:**P<0.01,***P<0.001)。
综合得知,实施例1至实施例5不同质量份的苗药固齿方在浓度低于1*10-3g/ml均对成骨前体细胞MC3T3-E1及破骨前体细胞RAW264.7细胞的细胞增殖有明显的增殖作用,将苗药固齿方添加到治疗牙周炎的局部药物以及保健牙膏、漱口水等中,其浓度稀释到小于1*10-3g/ml能够实现明显的细胞增殖作用,同时对比各实施例,实施例1的苗药固齿方仅在浓度为1*10-1g/ml时,对成骨前体细胞MC3T3-E1及破骨前体细胞RAW264.7细胞的细胞增殖有明显的抑制作用,而其余浓度对细胞增殖无影响,说明其细胞毒性较低,并可控,因此实施例1的组分更为安全可靠。
通过茜素红染色结果显示,如图17和图18所示,实施例1的苗药固齿方浓度为1*10-2g/m1时,对成骨前体细胞培养21天后,与不添加苗药固齿方的成骨前体细胞相比,苗药固齿方有明显的促进成骨前体细胞分化的作用,有促进成骨前体细胞形成。
最后应说明的是:以上各实施例仅用以说明本发明的技术方案,而非对其限制;尽管参照前述各实施例对本发明进行了详细的说明,本领域的普通技术人员应当理解:其依然可以对前述各实施例所记载的技术方案进行修改,或者对其中部分或者全部技术特征进行等同替换;而这些修改或者替换,并不使相应技术方案的本质脱离本发明各实施例技术方案的范围。

Claims (6)

1.一种用于治疗牙周炎的苗药固齿方,其特征在于,原料组分包括:青盐、生石膏、续断、散血飞、对叶莲、瘙羊古、艾纳香、卷柏和鸡屎藤。
2.根据权利要求1所述的一种用于治疗牙周炎的苗药固齿方,其特征在于,其原料组分按质量份包括为:
青盐5-15份
生石膏5-15份
续断5-12份
散血飞5-10份
对叶莲5-10份
瘙羊古5-12份
艾纳香5-10份
卷柏5-8份
鸡屎藤5-8份。
3.根据权利要求2所述的一种用于治疗牙周炎的苗药固齿方,其特征在于,其原料组分按质量份包括为:青盐15份、生石膏15份、续断12份、散血飞5份、对叶莲5份、瘙羊古8份、艾纳香8份、卷柏8份和鸡屎藤5份。
4.一种用于治疗牙周炎的苗药固齿方的制备方法,其特征在于,将权利要求1至3任意一项所述的一种用于治疗牙周炎的苗药固齿方加十倍质量的蒸馏水浸泡2h,大火煮至沸腾后,煎煮一段时间,取第一次药煎液置于容器内,再次向药渣中加入十倍质量的蒸馏水,大火煮至沸腾后煎煮一段时间,取第二次药煎液与第一次药煎液合并,将合并液冷却至室温后加入95%乙醇沉淀,使药液中的乙醇浓度为60%,离心后取上清液,煎煮浓缩至1g/ml作为原液,将原液过滤细菌,装入无菌瓶。
5.根据权利要求4所述的一种用于治疗牙周炎的苗药固齿方的制备方法,其特征在于,所述原液通过微孔滤膜过滤细菌。
6.根据权利要求5所述的一种用于治疗牙周炎的苗药固齿方的制备方法,其特征在于,苗药固齿方经大火煮至沸腾后,煎煮30分钟后取出药煎液。
