具体实施方式
本公开内容涉及一种预防癌症的方法,包括向哺乳动物施用有效量的融合细胞,其中所述融合细胞通过融合树突细胞和成纤维母细胞而形成,并且所述成纤维母细胞展示至少一种特定于癌症的抗原。成纤维母细胞包含癌细胞的基因组DNA。
树突细胞
树突细胞(DC)可以从受试者的血液或骨髓,或次级淋巴器官,例如但不限于脾、淋巴结、扁桃体、肠的派尔氏斑或骨髓中分离或产生,通过本领域已知的任何方法。在一个优选的实施方案中,树突细胞是终末分化的树突细胞。在一个实施方案中,树突细胞从人血液单核细胞分化而来。在某些实施方案中,树突细胞对于要向其施用本发明的融合细胞的受试者是自体的。在替代的实施方案中,树突细胞对于要施用本发明的融合细胞的受试者是同种异体的。
从来源获得的免疫细胞通常主要包括处于分化和成熟的各个阶段的再循环淋巴细胞和巨噬细胞。树突细胞制剂可以通过标准技术进行富集(参见例如Current Protocolsin Immunology,7.32.1-7.32.16,John Wiley and Sons,Inc.,1997)。在一个实施方案中,例如,树突细胞可以通过去除T细胞和贴壁细胞,随后密度梯度离心来富集。树突细胞可以任选地通过荧光标记细胞的分选或通过使用抗-CD83 mAb磁珠进一步纯化。
或者,可以通过用细胞因子例如粒细胞-巨噬细胞集落刺激因子(GM-CSF)和白介素4(IL-4)。在这样的条件下,单核细胞分化为树突细胞而没有细胞增殖。用诸如但不限于TNFα的药剂进一步处理刺激树突细胞的终末分化。
根据标准方法从血液单核细胞获得树突细胞(参见,例如,Sallusto等,1994,J.Exp.Med.179:1109-1118)。使用Ficoll-Paque密度梯度离心和塑料粘附,通过白细胞去除术包装或血沉棕黄层制备收集来自健康献血者的白细胞。如果需要成熟的树突细胞,可以使用以下方案来培养树突细胞。允许细胞在37℃下粘附在塑料培养皿上4小时。去除未粘附的细胞并将粘附的单核细胞在含有0.1μg/ml粒细胞-巨噬细胞集落刺激因子和0.05μg/ml IL-4的培养基中培养7天。为了制备树突状细胞,在第5天加入肿瘤坏死因子-α,并在第7天收集细胞。
树突细胞表达细胞表面标志物CD83。此外,此类细胞特征性地表达高水平的MHCII类分子,以及细胞表面标志物CD1α、CD40、CD86、CD54和CD80,但不表达CD14。其他细胞表面标志物的特征包括T细胞标志物CD2和CD5、B细胞标志物CD7和髓细胞标志物CD13、CD32(FcγR II)、CD33、CD36和CD63,以及大量白细胞相关抗原。
任选地,可以使用标准技术例如形态学观察和免疫化学染色来验证树突细胞的存在。例如,树突细胞的纯度可以通过流式细胞术使用针对一种或多种上述特征性细胞表面标志物(例如CD83、HLA-ABC、HLA-DR、CD1α、CD40和/或CD54。该技术还可用于区分成熟和未成熟的树突细胞,使用针对CD14的荧光标记抗体,CD14存在于未成熟的树突细胞中,但不存在于成熟的分化树突细胞中。
成纤维母细胞
本发明的成纤维母细胞可以是带有至少一个区分癌前细胞的等位基因的任何成纤维母细胞。成纤维母细胞可以从多种来源分离,例如但不限于癌症患者的成纤维母细胞、巨噬细胞和脂肪细胞。成纤维母细胞也可以来自原代细胞培养物,其对于患者来说可以是自体的、同基因的或同种异体的,这取决于用于制备融合细胞的树突细胞的来源。
根据要预防的癌症选择成纤维母细胞的来源。优选地,成纤维母细胞对于被治疗的患者是自体的。可以使用任何成纤维母细胞,只要该细胞包含至少一种对靶细胞具有特异性的抗原。在一个实施方案中,其中树突细胞对于患者是同种异体的,成纤维母细胞可以具有至少一个MHC I等位基因,该MHC I等位基因与被治疗的哺乳动物具有相同的MHC I类单倍型。在另一个实施方案中,其中树突细胞是患者自体的,成纤维母细胞可以是被治疗的哺乳动物的同种异体或自体的。
在一个实施方案中,本发明的成纤维母细胞是从皮肤组织中分离的,该皮肤组织是从将成为融合细胞受体的哺乳动物中手术切除的。在使用之前,固体癌前组织或聚集的癌前细胞应该通过标准技术分散,最好是机械地分散到单细胞悬浮液中。酶,例如但不限于胶原酶和DNA酶也可用于分散癌细胞。在一个实施方案中,本发明的成纤维母细胞获自原代细胞培养物,即获自身体的原始细胞的培养物。在一个优选的实施方案中,成纤维母细胞与来自哺乳动物的血浆一起培养。
收集的成纤维母细胞的量应足以与树突细胞融合以制备足够用于本发明疫苗的融合细胞。在一个实施方案中,5×107成纤维母细胞用作形成融合细胞的起始材料。在一个实施方案中,大约1×106至1×109的成纤维母细胞用于形成融合细胞。在另一个实施方案中,使用5×107至2×108个成纤维母细胞。在又一个实施方案中,使用1×107至1×1010个成纤维母细胞。使用其他量的成纤维母细胞制备融合细胞也在本发明的范围内。
可以通过本领域已知的任何方法鉴定和分离含有具有癌细胞抗原性的抗原的成纤维母细胞。例如,可以通过形态学、酶分析或增殖分析来鉴定成纤维母细胞(如图3-5所示)。如果已知感兴趣抗原的特征,还可以通过本领域已知的任何生化或免疫学方法鉴定或分离成纤维母细胞。例如,可以通过手术、内窥镜检查、其他活检技术、亲和层析和荧光激活细胞分选来分离成纤维母细胞。
不要求使用克隆的或同质的或纯化的成纤维母细胞群。可以使用细胞混合物,条件是混合物中的大量细胞含有被靶向的抗原。在一个具体实施方案中,成纤维母细胞和/或树突细胞被纯化。
用来自肿瘤细胞或石蜡包埋的肿瘤组织的基因组DNA转化的成纤维母细胞
成纤维母细胞包含从具有抗原呈递细胞的肿瘤细胞或癌前细胞或石蜡包埋的肿瘤组织中提取的基因组DNA。基因组DNA可以通过技术人员已知的任何方法从不同来源获得。可以通过技术人员已知的任何方法将基因组DNA转染或显微注射到非树突细胞中。
基因组DNA可以从肿瘤组织或任何含有癌细胞基因组DNA的肿瘤样品中分离或提取。肿瘤组织或样品可以是活组织/样品或包埋在石蜡块中的组织标本。在一个实施例中,基因组DNA可以从石蜡包埋的肿瘤组织中提取。在本发明中,基因组DNA可以从石蜡块中包埋的样品中获得。因此,可以更容易、更经济地获得基因组DNA。
在本发明的方法中,用于产生融合细胞的来自皮肤组织的成纤维母细胞必须能够转化或显微注射基因组DNA,并且必须能够与树突细胞融合。可以使用本领域技术人员已知的任何方法来确定来自皮肤组织的成纤维母细胞是否适用于本发明的方法。一方面,来自皮肤组织的成纤维母细胞能够被基因组DNA转染或显微注射。另一方面,来自皮肤组织的成纤维母细胞能够与树突细胞融合。
在某些实施方案中,来自皮肤组织的细胞源自与要治疗的受试者的物种不同的物种。或者,成纤维母细胞源自与待治疗受试者的物种相同的物种。在某些实施方案中,成纤维母细胞对于待治疗的受试者是异源的。在其他实施方案中,成纤维母细胞对于待治疗的受试者是自体的。在某些实施方案中,成纤维母细胞在细胞培养物中维持和/或增殖。
技术人员已知的任何方法均可用于从肿瘤、癌症或癌前病变的细胞中提取基因组DNA。用于分离基因组DNA的示例性方法是本领域众所周知的。可以使用本领域技术人员已知的任何方法将基因组DNA导入成纤维母细胞。在某些实施方案中,基因组DNA被转染到成纤维母细胞中。在更具体的实施方案中,使用脂质转染将基因组DNA转染到成纤维母细胞中。
导入成纤维母细胞的基因组DNA的最佳量可以通过技术人员熟知的标准技术确定。在某些实施方案中,从皮肤组织导入每个成纤维母细胞的基因组DNA的量对应于至少1个肿瘤细胞或癌前细胞的基因组、至少10-1个肿瘤细胞或癌前细胞的基因组的等价物。细胞,至少相当于肿瘤细胞或癌前细胞的10-2基因组,至少相当于肿瘤细胞或癌前细胞的10-3基因组,至少相当于肿瘤细胞或癌前细胞的10-4基因组,至少相当于肿瘤细胞或癌前细胞的10-5基因组,至少相当于肿瘤细胞或癌前细胞的10-6基因组,或至少相当于10-7肿瘤细胞或癌前细胞的基因组。
在某些实施方案中,使用显微注射将基因组DNA导入成纤维母细胞。在某些实施方案中,基因组DNA的片段被包装到载体中用于基因组DNA的增殖。此类载体包括但不限于噬菌体、粘粒或YAC。技术人员已知的任何方法均可用于包装和繁殖基因组DNA。
一旦将基因组DNA从皮肤组织导入成纤维母细胞,该成纤维母细胞表达一种或多种抗原,这些抗原由肿瘤细胞、赘生性细胞或从中分离基因组DNA的癌前病变细胞表达。在本发明的某些实施方案中,成纤维母细胞含有一种或多种显示肿瘤或癌前病变抗原性的分子。在某些实施方案中,抗原在患有肿瘤或癌前病变的个体其肿瘤或癌前病变中的表达水平比在任何其他组织中高至少2倍、5倍、10倍、20倍、50倍或100倍。
如图1所示,来自皮肤组织的成纤维母细胞,例如成纤维母细胞是通过众所周知的标准技术获得的。核苷酸,优选DNA被转化到成纤维母细胞中。DNA,例如癌细胞的基因组DNA,是从石蜡块中包埋的肿瘤组织中提取的。然后,可以用抗生素(例如丝裂霉素)处理用DNA转化的成纤维母细胞,以抑制癌细胞的增殖。成纤维母细胞可用于随后将详细描述的树突细胞融合。
成纤维母细胞和树突细胞的融合
本公开还包括来自皮肤组织的树突细胞和成纤维母细胞的融合细胞。融合细胞包含肿瘤细胞的基因组DNA,并且肿瘤细胞的基因组DNA编码至少一种对癌症具有特异性的抗原。
本公开还涉及融合人树突细胞和成纤维母细胞的方法,包括使树突细胞群和成纤维母细胞群经受促进细胞融合的条件。
成纤维母细胞可以如下融合到树突细胞。根据本领域中的任何细胞融合技术对细胞进行无菌洗涤和融合,只要融合技术产生适合注射入哺乳动物以预防癌症的融合细胞的混合物。优选地,使用电融合。电融合技术是本领域公知的(Stuhler and Walden,1994,Cancer Immunol.Immunother.39:342-345;参见Chang等人(编辑),Guide toElectroporation and Electrofusion.Academic Press,San Diego,1992).
在优选实施例中,使用以下程序。第一步,将大约5×107个癌前非树突细胞和5×107个树突细胞悬浮在0.3M葡萄糖中并转移到电融合比色皿中。通过在100V/cm下脉冲细胞样品5-10秒,使样品介电泳排列以形成细胞-细胞偶联物。可选地,可以通过在电穿孔试管的一个方面应用一滴介电蜡来优化对齐,以“不均匀”电场,从而将细胞引导到最高场强的区域。第二步,施加融合脉冲。各种参数可用于电融合。例如,在一个实施例中,融合脉冲可以是从单脉冲到三脉冲。在另一个实施例中,在约25μF下使用500至1500V/cm、优选地1200V/cm来完成电融合。
在优选实施例中,使用以下程序。在第一步中,非树突细胞用100μg/mL丝裂霉素C处理10小时以防止细胞生长。之后,非树突细胞用PBS洗涤,然后用0.05%胰蛋白酶-EDTA处理。接下来,将非树突细胞和树突细胞混合、洗涤并用PBS离心。然后,将沉淀的细胞加入0.5mL 50%聚乙二醇(PEG),加热至37℃,然后孵育1分钟。此外,将沉淀的细胞加入7ml无血清RPMI-1640培养基,然后加热至37℃以稀释PEG。可选地,添加8ml含10%FCS的RPMI-1640培养基进行稀释。最后,通过离心去除PEG,并将沉淀的细胞悬浮在2%灭活的自体血浆添加AIM-V培养基中,该培养基补充有rh GM-CSF(10ng/mL)、rh IL-4(10ng/mL)和rh TNF-α(10ng/mL)。
在一个优选的实施方案中,成纤维母细胞对于本发明的融合细胞将被施用于的患者是自体的。在另一个优选的实施方案中,树突细胞对于将被施用本发明的融合细胞的患者是自体的。在甚至更优选的实施方案中,癌前非树突细胞和树突细胞对于施用本发明的融合细胞的患者都是自体的。
在另一个实施方案中,树突细胞和来自皮肤组织的细胞如上所述融合。随后,融合细胞用遗传物质转化或转染,遗传物质编码刺激CTL和/或体液免疫应答的分子。在一个优选的实施方案中,遗传物质是编码IL-12的mRNA。优选的转染方法包括在阳离子聚合物存在下的电穿孔或转化或转染。
成纤维母细胞和树突细胞群内融合细胞形成的程度可以通过本领域已知的多种诊断技术来确定。例如,在一个实施方案中,杂交体的特征在于分别用红色和绿色细胞内荧光染料标记树突细胞和成纤维母细胞,并检测两种颜色的发射光。
免疫细胞活化分子
本发明提供了一种组合物,其包含通过融合来自皮肤组织的树突细胞和成纤维母细胞的融合细胞。在某些实施方案中,本发明的组合物进一步包含可刺激或诱导细胞毒性T细胞(CTL)反应和/或体液反应的细胞因子或其他分子。在一个优选的实施方案中,CTL刺激分子是IL-4、IL-12、IL-15或IL-18。

药物制备和给药方法
本公开还涉及含有公开的融合细胞的药物组合物。
本发明的组合物制剂包含有效免疫量的待施用的融合细胞。本发明药物组合物的融合细胞可以通过将抗原呈递细胞如树突细胞或通用抗原呈递细胞与皮肤组织细胞融合形成,其中皮肤组织细胞包含从癌细胞、癌前病变细胞或石蜡包埋的肿瘤组织;源自癌细胞、癌前病变细胞或石蜡包埋的肿瘤组织的cDNA或cDNA文库。在某些实施方案中,本发明的融合细胞表达待治疗或预防的癌症的一种或多种抗原。
药物组合物可以制成注射剂,作为液体溶液或悬浮液。药物组合物可以通过任何合适的应用模式施用,例如,皮内给药(i.d.)、静脉给药(i.v.)、腹膜腔内给药(i.p.)、肌内给药(i.m.)、鼻内、口服、皮下等并且在任何合适的递送装置中施用。在某些实施方案中,药物组合物被配制用于静脉内、皮下、皮内或肌肉内给药。也可以制备适用于其他给药方式的药物组合物,包括口服和鼻内应用。
药物组合物可以配制成立即释放或持续释放的制剂。此外,药物组合物可以配制用于通过免疫原截留和与微粒共同给药来诱导全身性或局部粘膜免疫。本领域的普通技术人员可以容易地确定这样的递送系统。
药物组合物也可以配制成合适的剂量单位形式。在一些实施方案中,药物组合物含有每公斤体重约0.5μg至约1mg的tau肽免疫原构建体。药物组合物的有效剂量取决于许多不同的因素,包括给药方式、目标部位、患者的生理状态、患者是人还是动物、给药的其他药物以及治疗是预防性的还是治疗性的。通常,患者是人类,但也可以治疗非人类哺乳动物,包括转基因哺乳动物。当以多剂量递送时,可方便地将药物组合物分成合适的每剂量单位形式。正如治疗领域所熟知的,给药剂量将取决于受试者的年龄、体重和总体健康状况。
此外,如果需要,组合物制剂还可包括少量辅助物质,例如润湿剂或乳化剂、pH缓冲剂和/或增强组合物有效性的化合物。辅助物质的有效性可以通过测量针对融合细胞的抗体的诱导来确定。在一些实施方案中,药物组合物含有佐剂或载体,例如矿物盐,包括明矾凝胶、磷酸铝或油包水乳剂。
给予组合物的哺乳动物优选是人,但也可以是非人动物,包括但不限于牛、马、羊、猪、家禽(例如鸡)、山羊、猫、狗、仓鼠、小鼠和大鼠。

实施例1
含有基因组DNA的成纤维母细胞的制备
皮肤活检运输和洗涤培养基和皮肤活检培养基的制备
制备“皮肤活检运输和洗涤培养基”和“皮肤活检培养基”。皮肤活检培养基是个体化的。
一种皮肤活检转运和洗涤介质,包括以下成分:RPMI1640、胎牛血清、青霉素链霉素和庆大霉素溶液。
一种皮肤活检培养基,包括以下成分:HFDM-1(+)、哺乳动物血浆、青霉素链霉素。哺乳动物的血浆对患者来说是自体的。
成纤维母细胞的制备
从皮肤组织获得成纤维母细胞。取决于用于制备融合细胞的树突细胞的来源,皮肤组织对于患者可以是自体的、同基因的或同种异体的。皮肤组织通过皮肤活检运输和洗涤介质保持和洗涤。皮肤组织用匀浆器匀浆。成纤维母细胞在皮肤活检培养基中维持和培养。使用来自美天旎生物技术公司(Miltenyi biotec)的全皮肤分离试剂盒(人)(WholeSkin Dissociation Kit)(订单号:130-101-540)分散成纤维母细胞。
制备成纤维母细胞的方法包括以下步骤:
(a)获得皮肤组织;
(b)在皮肤活检运输和清洗介质中保持和清洗皮肤组织;
(c)用匀浆器匀浆皮肤组织,得到成纤维母细胞;
(d)在皮肤活检培养基中培养成纤维母细胞;
(e)在皮肤活检培养基中传代培养成纤维母细胞;
(f)收获或冷冻成纤维母细胞。

表4.原代培养基(皮肤活检培养基)的制备:
原代培养操作:
(1)将14.5mL培养基倒入15mL管中。
(2)从培养箱中取出酶分散容器(C管),从15mL管中取出。加入500μL培养基并盖紧盖子。
(3)将C管设置为gentleMACS(自动组织破碎破碎机)并启动程序。
(4)程序完成后,加入2mL培养基。通过70μm网孔过滤器收集在15mL管中。
(5)再次加入2mL培养基,离心(冲洗)。通过70μm网孔过滤器收集在15mL管中。
(6)充分搅拌后,取30μL细胞溶液,混合1.5mL计算管。装入血细胞计数器。
(7)相差显微镜计数细胞数。
(8)离心(1200rpm,5分钟)。
(9)吸上清液后,悬浮于10mL培养基中,转移至T25培养瓶中。
(10)在37℃、5%CO2培养箱中培养。
3.成纤维母细胞自T25培养瓶到T225培养瓶的继代培养

成纤维母细胞继代培养的制备
将培养基、PBS、胰蛋白酶EDTA恢复至室温
继代操作:
(1)将10mL培养上清液转移到T225平方厘米(cm2)培养瓶中。
(2)加入5mL PBS后除去,加入2mL胰蛋白酶EDTA,室温消化1分钟,然后除去。
(3)吸取30mL培养基到T225 cm2培养瓶中。
(4)轻弹培养瓶使细胞剥离,加入5mL培养基。加入15mL试管。
(5)使用自动细胞计数器计算细胞数,同时保持未染色。
(6)将细胞溶液全部转移到T225 cm2细胞培养中。
(7)37℃、5%CO2培养箱培养。
4.冷冻T225培养瓶上的成纤维母细胞

成纤维母细胞冷冻的制备:
将培养基、PBS、胰蛋白酶EDTA恢复至室温。并为血清管签发冷冻管理条码标签。
冷冻操作:
(1)吸出培养上清液。
(2)加入15mL PBS后移去,再加入5mL胰蛋白酶EDTA。在室温下保持1分钟。
(3)将3个培养瓶翻转并剥离,加入11mL培养基。转移到50mL管中。
(4)将1mL转移到样品储存管中并储存在-20℃。
(5)使用自动细胞计数器计算细胞数,同时保持未染色。
(6)冷冻储存细胞浓度1x 106个细胞/mL。
(7)离心(1200rpm,5分钟)。
(8)准备所需数量的血清管,粘贴条形码标签,输入细胞数。
(9)吸出上清液后,用1mL冷冻细胞保存液悬浮并计算溶液。暂停到所需的数量。
(10)倒入血清管中,并放入细胞冻结处理容器中。
(11)在-80℃冰箱中冷冻。
图3-5为原代培养第0-13天成纤维母细胞形态,原代培养第0-9天成纤维母细胞增殖率。
基因组DNA的分离
石蜡块包埋的肿瘤组织作为肿瘤样品从医疗机构获得。肿瘤样本用苏木精和伊红(H&E)染色,然后由医生确定以确认肿瘤组织的抗原性。石蜡块用二甲苯脱蜡。用苯酚和乙醇处理肿瘤组织,提取和沉淀肿瘤组织的基因组DNA。使用聚合酶链反应(PCR)扩增基因组DNA并储存在-20℃。
制备基因组DNA的方法包括以下步骤:
(a)获得包埋在石蜡块中的肿瘤组织;
(b)用苏木精和伊红(H&E)染色石蜡块并确认肿瘤组织的抗原性;
(c)用二甲苯对石蜡块进行脱蜡;
(d)苯酚和乙醇提取和沉淀肿瘤组织的基因组DNA;
(e)使用聚合酶链反应(PCR)扩增基因组DNA;
(f)将基因组DNA储存在-20℃。
图2显示了肿瘤组织的切片和苏木精伊红(H&E)染色的结果。
下表7显示了来自癌症患者的基因组DNA样品的O.D.260/280检测结果。

含有肿瘤细胞基因组DNA的成纤维母细胞的制备
从癌症患者获得皮肤组织。皮肤组织用皮肤活检运输和洗涤介质洗涤,并使用超声破碎以获得真皮成纤维母细胞。真皮成纤维母细胞在皮肤活检培养基中传代培养数小时。成纤维母细胞用从通过脂质转染包埋在石蜡块中的肿瘤组织中获得的基因组DNA转化。转化的成纤维母细胞在转染培养基中培养并用丝裂霉素处理以抑制癌细胞的增殖。
用于产生癌抗原(成纤维母细胞+癌基因组核酸)的方案

(1)将9mL培养基移入15mL试管中。
(2)37℃下迅速融化冷冻细胞,将细胞溶液转移至15mL试管中。
(3)未染色时,使用自动细胞计数器计算细胞数。
(4)离心(1200rpm,5min,室温(RT))。
(5)弃上清液,悬浮于培养基体积,计算为0.1x 106cell/mL。
(6)6孔板(0.2×106个细胞/2mL/孔)各接种2mL。
(7)37℃、5%CO2培养箱培养。
2.转染操作(Transfection operation)

(1)将125μL优化培养基移入1.5mL试管(L1,T1)。
(2)将7.5μL lipofectamin 3000移入1.5mL试管(L1)中。
(3)将4.0μL P3000试剂倒入1.5mL试管(T1)中。
(4)向1.5mL试管(T1)中加入2μg DNA。
(5)在总体积中加入1.5mL试管(L1)至1.5mL试管(T1)。静置10分钟。
(6)观察细胞板有无异常。
(7)弃去培养上清液,加入2mL 10%FBS-optiMEM。
(8)静置后,将1.5ml试管(T1)加入成纤维母细胞,每种抗原。
3.丝裂霉素处理(Mitomycin treatment)(癌细胞生长抑制)
表10.仪器和试剂制备:
(1)观察细胞板有无异常。
(2)去除培养上清液,加入1mL 10%FBS-RPMI。
(3)加入200μL丝裂霉素(500μg/mL)(处理浓度100μg/mL)。
(4)在37℃、5%CO2的培养箱中放置至少10小时。
4.癌抗原细胞的采集与冷冻
表11.仪器和试剂制备:
(1)将7mL 10%FBS-RPMI分配到15mL管中。
(2)观察细胞板有无异常。
(3)收集培养上清液,洗涤1mL PBS后收集细胞。加入1mL 0.05%胰蛋白酶EDTA,在37℃下放置5分钟。
(4)剥离细胞并收集在15mL管中。
(5)离心(1200rpm,5min,RT)。
(6)弃上清液,用2mL PBS悬浮。
(7)用相差显微镜目测计数(台盼蓝2倍稀释)后计算细胞数。
(8)离心(1200rpm,5min,RT)。
(9)弃上清液,用1000μL冷冻细胞保存液悬浮,加入冷冻管。等分并放置在细胞冻结处理容器中,并在-80℃下冷冻。
成纤维母细胞包含癌细胞的基因组DNA,并且基因组DNA编码至少一种对癌症具有特异性的抗原。为了确认结果,使用以下分析方法。
通过脂质转染导入成纤维母细胞的基因的微阵列分析mRNA表达。基因组DNA和通过随机引物扩增的基因组通过脂质转染转染成纤维母细胞。导入核酸,通过微阵列(单色法)从RNA进行表达分析。

1.一种重组细胞的制备方法,其包含下述步骤:
(a)提供成纤维母细胞其为取自哺乳动物的结缔组织;
(b)从所述哺乳动物的肿瘤组织中萃取癌细胞的基因体(genome)DNA,其中所述基因体DNA编码至少为一种对癌症具有特异性的抗原,其中取自所述哺乳动物的所述肿瘤组织不是新鲜分离的;以及
(c)转化所述癌细胞基因体(genome)DNA到所述成纤维母细胞。
2.如权利要求1所述的方法,其特征是,所述哺乳动物是人。
3.如权利要求1所述的方法,其特征是,所述结缔组织为皮肤组织。
4.如权利要求1所述的方法,其特征是,取自所述哺乳动物的所述肿瘤组织不是新鲜分离的。
5.如权利要求1所述的方法,其特征是,所述哺乳动物的所述肿瘤组织包埋在石蜡块中。
6.如权利要求3所述的方法,其特征是,所述皮肤组织其保持和洗涤是通过皮肤活检运输和洗涤介质。
7.如权利要求1所述的方法,其特征是,所述成纤维母在皮肤活检培养基中维持和培养,所述皮肤活检培养基包含来自哺乳动物的血浆。
8.一种融合树突细胞和重组细胞的方法,包括利用来自哺乳动物的树突细胞群和如权利要求1所述的重组细胞群通过促进细胞融合的条件。
9.如权利要求8所述的方法,其特征是,所述树突细胞和所述重组细胞是来自哺乳动物的自体细胞。

11.一种重组细胞,其中所述重组细胞包含癌细胞基因体,且所述癌细胞基因编码至少对任一癌症具有一特异性抗原;该癌细胞基因体从包埋在石蜡块中的肿瘤组织中分离;所述重组细胞是来自哺乳动物皮肤组织的成纤维母细胞;所述重组细胞和所述成纤维母细胞为哺乳动物自体的。
12.如权利要求11所述的重组细胞,其特征是,所述的重组细胞是通过权利要求1的方法制备获得。
13.一种融合细胞,其包含
(a)树突细胞;以及
(b)如权利要求11所述重组细胞
其中,所述树突细胞和所述重组细胞来自哺乳动物的自体细胞;所述融合细胞包含只少一种癌症特异性抗原,所述融合细胞包含CD83、CD1α、CD40、CD86、CD54、CD80、MHC classII。
14.如权利要求13所述的融合细胞,其特征是,所述的融合细胞是通过权利要求10的方法制备获得。
15.一种药物组合物,其包含权利要求13的融合细胞和有效量的佐剂或载体。
16.如权利要求15所述的药物组合物,进一步包含刺激免疫应答的分子,所述免疫应答选自体液免疫应答、细胞毒性T细胞免疫应答及其组合。
17.如权利要求16所述的药物组合物,所述分子为细胞激素。
