本发明公开了一种调节动物肠道的益生素制剂,按重量百分比计,由复合益生菌20‑30wt%、复合益生元5‑10wt%、消化酶1‑5wt%及载体酵母发酵培养物55‑70wt%所组成。通过好氧及厌氧益生菌、益生元及消化酶的科学合理复配,使三者协同作用,能有效调节动物肠道健康,减少或预防细菌性疾病的发生,同时促进动物消化吸收、促进生长,提高饲料利用率,从而达到减抗、替抗的目的。

技术领域
本发明涉及微生物制剂的技术领域,尤其涉及一种调节动物肠道的益生素制剂及其应用。
背景技术
养殖动物细菌性疾病防治,多以抗生素类抗菌物质为主,不仅造成环境污染,还能导致药物残留、提高病原菌耐药性。而益生菌能产生抗菌物质,抑制有害菌的生长,具有抗生素功能,却无抗生素所带来的弊端。所以养殖行业必将朝着减抗替抗的趋势发展,而益生菌制剂则可作为替抗的其中一种。
肠道作为机体最大的免疫器官,其附着近一半的免疫细胞,通常大部分病原菌由口进入到肠道,再由肠道进入机体各器官,如若病原菌在肠道内受到抑制,则无法在短时间内侵入到其他器官,很快就能排出体外,无法导致机体患病。且肠道除具免疫功能外,肠道还具有消化吸收、神经调节功能,故肠道健康尤为重要。
发明内容
本发明的目的在于提供一种能有效调节养殖动物肠道健康的益生素制剂,从而减少或替代抗生素的使用。该益生素制剂能有效调节动物肠道健康,减少或预防腹泻等疾病的发生,提高动物消化吸收,促进生长,且益生素制剂无毒害,为绿色、安全的新型饲料添加剂。
本发明所述一种调节动物肠道的益生素制剂,其成分主要包括复合益生菌、复合益生元、消化酶及酵母发酵培养物。其中复合益生菌采用好氧与厌氧菌株相结合,进入肠道后好氧菌快速消耗肠道内氧气,从而促进乳酸菌繁殖、抑制病原菌生长,达到调节肠道益生菌群平衡。益生元既能改善肠道健康且能刺激有益菌群的生长,达到辅助益生菌的效果。消化酶能分解大分子物质为小分子,有利于益生菌及肠道的吸收利用,达到辅助益生菌及减小消化器官负荷,且水产动物肠道短,增加消化酶能加快促进食物消化吸收。
本发明的技术方案是:一种调节动物肠道的益生素制剂,按重量百分比计,由复合益生菌20-30wt%、复合益生元5-10wt%、消化酶1-5wt%及载体酵母发酵培养物55-70wt%所组成。
优选地,所述复合益生菌,按重量比计,由枯草芽孢杆菌:凝结芽孢杆菌:植物乳杆菌:鼠李糖乳杆菌=1:2:2:3组成,原料活菌数皆为1000亿CFU/g。
优选地,所述复合益生元,按重量比计,由低聚果糖:低聚木糖=1:1组成。
优选地,所述消化酶,按重量比计,由淀粉酶:脂肪酶:蛋白酶:纤维素酶=1:1:1:1组成,原料酶活力皆为5万U/g。
优选地,所述载体酵母发酵培养物由啤酒酵母、产朊假丝酵母通过固态发酵后烘干、超微粉碎所得。
本发明提供的一种调节动物肠道的益生素制剂可运用于水产与畜禽养殖动物,作为饲料添加剂使用。
优选地,所述的一种调节动物肠道的益生素制剂,在水产动物每公斤饲料添加0.5-1g拌匀,在畜禽动物每公斤饲料添加0.5-1g拌匀。
采用上述方案后,与现有技术相比,本发明至少具有以下优点:
一、复合益生菌采用好氧与厌氧菌搭配,益生菌进入肠道后可快速使肠道处于厌氧环境,从而有利于抑制病原菌及促进乳酸厌氧菌的生长繁殖,从而调节肠道菌群平衡。
二、益生元通过上消化道时,大部分不被消化而能被肠道菌群所发酵,但到达肠道后可作为益生菌的底物,但却不能被病原微生物利用,从而促进益生菌的繁殖和抑制有害微生物。
三、消化酶可促进食物中糖、脂肪、蛋白质的水解,由大分子物质变为小分子物质,以便被体内吸收利用,从而缓解肠胃负担,促进肠胃健康。
四、酵母发酵培养物作为益生素制剂的载体,它含有大量多肽、维生素、氨基酸、有机酸、寡糖及其酶类等物质,能改善养殖动物的肠胃内环境,促进营养物质消化吸收,促进采食,增强免疫能力,从而促进动物生长、提高生产性能。且与传统矿物质载体相比,卫生指标可控,从而确保产品质量。
五、本发明的一种调节动物肠道的益生素制剂,通过益生菌、益生元、消化酶三者科学合理复配,使三者协同作用,促进肠道健康、消化吸收,从而减少或预防细菌性疾病的发生。

实施组合1-4及对照组合1-5的配方如表1所示,其中对照组合以无水葡萄糖作为替换原料,实施组合1-4的复合益生菌配比为:枯草芽孢杆菌:凝结芽孢杆菌:植物乳杆菌:鼠李糖乳杆菌=1:2:2:3。

实施例1:抑菌试验
抑菌试验采用管碟法,上述组合物采用无菌生理盐水稀释1000倍后静置5min,吸取上清备用。取1mL病原指示菌悬液加入50℃ 100mL营养琼脂培养基中摇匀(菌终浓为105CFU/mL),倒入培养皿中(厚度约为3mm),待冷却凝固后将牛津杯置于平板中,往牛津杯中各加入0.20mL上清液,放置4℃冰箱2h后于36℃下培养24h,观察有无抑菌圈产生,并用游标卡尺测量抑菌圈直径大小。

由表3实施组合与对照组合的数据对比可知,复合益生菌对上述6株病原菌的抑制起主要作用,而复合益生元、消化酶、酵母发酵培养基无抑菌效果。且随着复合益生菌含量的增加,抑菌效果更显著,说明本发明的益生素制剂能有效抑制上述6种病原菌。
实施例2对虾饲喂试验
福建某对虾养殖场,选取同期放苗养殖的13个虾池,饲喂日粮按1g/kg比例添加实施组合1-4及对照组合1-9,连续试验30天后,检测对虾肠道及养殖池水的总弧菌数、增重率。弧菌检测采用TCBS培养基涂布,30℃培养48h,增重率(%)=(末重M2-初重M1)/初重M1*100%。

由表4可知,有复合益生菌成分的实施组合1-4及对照组合2-4,6-9能有效减少对虾肠道、养殖水体的弧菌总数,尤其实施组合3效果最为明显,说明复合益生菌对抑制弧菌起重要作用,且在复合益生菌的复配比例以实施组合3的为最优。在对虾增重方面,实施组合1-4的效果较优于对照组合1-5,且以实施组合3最佳,与对照组合5相比增重17.84%。由对照组合1-4的数据可知,影响对虾增重的因素依次为复合益生菌(对照组合1)>消化酶(对照组合3)>载体酵母发酵培养物(对照组合4)>复合益生元(对照组合2)。故由表3可知,本发明的的益生素制剂在1g/kg用量情况下能促进对虾增重,减少肠道弧菌总数,从而保证肠道健康。
实施例3仔猪饲喂试验
选取约30日龄仔猪180只,体重相近,健康。随机分为9组,每组20只,公母对半,分开饲养避免交叉污染。分别在平常饲粮中按0.5g/kg比例添加实施组合1-4及对照组合1-5,9组管理一致,饮水自由,试验天数为30天,采样新鲜粪便检测大肠菌群,并检测、计算增重率以及腹泻率等相关数据。其中大肠菌群采用饲料中大肠菌群的测定GBT18869-2019方法进行检测。

为期30天试验,无仔猪死亡,由表5可知,有复合益生菌成分的实施组合1-4及对照组合2-4能有效减少仔猪肠道大肠菌群数量,降低腹泻。实施组合1-4在0.5g/kg用量情况下对仔猪的增重效果优于对照组合1-5,以实施组合3效果最佳,与对照组合5相比增重55.87%。
通过上述对虾及仔猪的试验结果表明,本发明提供的一种调节动物肠道的益生素制剂能有效抑制肠道病原菌,预防或减少细菌性疾病发生,改善肠道健康,促进肠道消化吸收,从而增加机体重量,且能够适用于水产及畜禽动物。
以上详细描述了本发明的优选实施方式,但是,本发明并不限于上述实施方式中的具体细节,在本发明的技术构思范围内,可以对本发明的技术方案进行多种简单变型,这些简单变型均属于本发明的保护范围。
另外需要说明的是,在上述具体实施方式中所描述的各个具体技术特征,在不矛盾的情况下,可以通过任何合适的方式进行组合。为了避免不必要的重复,本发明对各种可能的组合方式不再另行说明。
Claims (7)
1.一种调节动物肠道的益生素制剂,其特征在于,按重量百分比计,由复合益生菌20-30wt%、复合益生元5-10wt%、消化酶1-5wt%及载体酵母发酵培养物55-70wt%所组成。
2.根据权利要求1所述的一种调节动物肠道的益生素制剂,其特征在于,所述复合益生菌,按重量比计,由枯草芽孢杆菌:凝结芽孢杆菌:植物乳杆菌:鼠李糖乳杆菌=1:2:2:3组成,原料活菌数皆为1000亿CFU/g。
3.根据权利要求1所述的一种调节动物肠道的益生素制剂,其特征在于,所述复合益生元,按重量比计,由低聚果糖:低聚木糖=1:1组成。
4.根据权利要求1所述的一种调节动物肠道的益生素制剂,其特征在于,所述消化酶,按重量比计,由淀粉酶:脂肪酶:蛋白酶:纤维素酶=1:1:1:1组成,原料酶活力皆为5万U/g。
5.根据权利要求1所述的一种调节动物肠道的益生素制剂,其特征在于,所述载体酵母发酵培养物由啤酒酵母、产朊假丝酵母通过固态发酵后烘干、超微粉碎所得。
6.根据权利要求1-5所述的一种调节动物肠道的益生素制剂,运用于水产与畜禽养殖动物,作为饲料添加剂使用。
7.根据权利要求6所述的一种调节动物肠道的益生素制剂,在水产动物每公斤饲料添加0.5-1g拌匀,在畜禽动物每公斤饲料添加0.5-1g拌匀。
