背景技术
纳米治疗剂和/或诊断剂递送媒剂通常为大小介于1-1,000nm范围内的大分子或超分子多组分系统,其固有地为治疗剂(例如,不含活性医药成分)或用作治疗剂或诊断剂递送系统。迄今为止,脂质体纳米粒子和生物剂包含较大比例的FDA批准产品数量或临床试验中用于治疗和/或检测多种癌症类型的产品数量,而多种基于聚合物的粒子调配物目前处于早期试验中。
所需候选纳米治疗剂递送系统共享以受控方式纳入和释放药物化合物的共同特征,此可有利地改变药物生物利用度和药代动力学,同时最小化脱靶毒性。理想地,将成像标记纳入其中以评价其精确定位和疾病位点处的滞留。

业内仍需要可用于检测、预防和/或治疗癌症和其它疾病的平台。
发明内容
本文描述靶向诊断和/或治疗平台的靶特异性纳米粒子免疫偶联物(例如,结合到粒子表面的单链抗体片段)。在某些实施例中,纳米粒子免疫偶联物的直径小于20nm(例如,6到10nm)。发现此小大小在治疗和/或成像应用中提供优点。例如,所揭示免疫偶联物可提供经改良的患病组织靶向和减少的器官(例如,肝)非特异性摄取。较小免疫偶联物还可展示降低的免疫反应性,由此进一步改良效能。

定义

分子治疗剂(例如,抗体)可通过操纵免疫检查点(例如,单克隆抗体伊匹单抗(ipilimumab)抑制CTLA4,其是抑制免疫系统功能的负调控分子)调节针对抗肿瘤活性的免疫系统。原理是触发预先存在但静止的抗肿瘤免疫反应。其它分子和路径已用作免疫开关。还靶向在T细胞上表达的另一负调控受体PD-1。开关单一免疫检查点可不足以诱导抗肿瘤反应,此解释靶向如PD-1或CTLA4等单一免疫调控检查点的一些失败。然而,不希望受限于任何理论,可通过添加在一些情形下认为具有免疫调节性质的RT来支持治疗。在这些情形下,已发现在RT治疗领域外的肿瘤因推定的由RT引起的全身性炎症或免疫反应而皱缩,此突出显示辐射激发全身性抗肿瘤免疫反应的潜能。强化免疫活性还可加强RT的局部作用。

本文描述靶向诊断和/或治疗平台的靶特异性纳米粒子免疫偶联物(例如,结合到粒子表面的单链抗体片段)。在某些实施例中,纳米粒子免疫偶联物的直径小于20nm(例如,6到10nm)。发现此小大小在治疗和/或成像应用中提供优点。例如,所揭示免疫偶联物可提供经改良的患病组织靶向和减少的器官(例如,肝)非特异性摄取。较小免疫偶联物还可展示降低的免疫反应性,由此进一步改良效能。

C点或C’点因其物理性质且展示人类体内特征而提供独特的药物递送平台。这些粒子超小且受益于肿瘤微环境中的EPR作用,同时保留所需清除率和药代动力学性质。为此,本文描述纳米粒子药物递送系统,其中在某些实施例中,药物构筑体共价附接到C点或C’点(或其它纳米粒子)。
C点或C’点可用作高度特异性且强效的多治疗剂靶向粒子探针以在单一平台上组合抗体片段与治疗性放射性标记(例如,177Lu、225Ac、90Y、89Zr)。或者,已知具有免疫调节和抗炎症性质的靶向肽(例如αMSH)的C点或C’点偶合还可与C点或C’点放射性治疗剂(和/或基于其它粒子)平台组合以实现增强的效能。在某些实施例中,放射性同位素和/或抗体片段的浓度与诊断应用相比在治疗应用中较高。
与其它多模态平台不同,免疫偶联物可包含附接到纳米粒子自身的不同部分。例如,在某些实施例中,放射性同位素附接到纳米粒子且抗体片段附接到纳米粒子,也就是说,在这些实施例中,放射性标记并不附接到抗体片段自身。作为另一实例,免疫偶联物可包含附接到纳米粒子的靶向配体、附接到纳米粒子的放射性同位素和附接到纳米粒子的抗体片段。附接到C点的不同部分的化学计量比将影响纳米粒子免疫偶联物的生物分布。
免疫偶联物(例如C点-抗体(mAb)和C点-抗体片段(vFab)偶联物)可使用两种方法中的任一者来制备。方案1包含硫醇-马来酰亚胺化学法,如图8中所显示。方案1是围绕经修饰以含有马来酰亚胺基团的蛋白质来设计。方案2包含烯烃-四嗪化学法,如图9中所显示。

实验实例
C点-(Cy5)-PEG-马来酰亚胺的制备:
将马来酰亚胺和NHS酯官能化聚乙二醇(mal-dPEG12-NHS)与氨基甲硅烷(APTES)于DMSO中偶联(摩尔比mal-PEG-NHS:APTES:DMSO 1:0.9:60)。将反应混合物在室温下在氮下静置48小时以产生甲硅烷官能化mal-dPEG(mal-dPEG-APTES)。使马来酰亚胺官能化Cy5(mal-Cy5)与硫醇-甲硅烷(MPTMS)于DMSO中反应(摩尔比Cy5:MPTMS:DMOS 1:25:1150)。将反应物在室温下在氮下静置24小时以产生甲硅烷官能化Cy5(Cy5-MPTMS)。然后将TMOS和Cy5-MPTMS滴定到氢氧化铵溶液(约pH 8)中(摩尔比TMOS:Cy5:NH3:H2O 1:0.001:0.44:1215)。将溶液在室温下在600rpm下搅拌24小时以形成均质的Cy5囊封的二氧化硅纳米粒子。然后将mal-dPEG-APTES和甲硅烷官能化聚乙二醇(PEG-甲硅烷,MW约500,盖勒斯特(Gelest))添加至合成溶液中以聚乙二醇化且表面官能化粒子(PEG-甲硅烷:TMOS:mal-PEG-APTES 1:2.3:0.006)。将溶液在室温下在600rpm下搅拌24小时,然后在80℃下在不搅拌下再培育24小时。将溶液在2000mL去离子水中透析两天(10k MWCO),用200nm针筒过滤器过滤,且最后色谱纯化(Superdex 200),产生所需mal-C点。
C点免疫偶联物的制备
进行研究以将单链抗体片段(scFv)偶联到C点核心二氧化硅纳米粒子。在大肠杆菌中表达结合基质金属蛋白酶12(MMP-12)的scFv。构筑体含有C末端His和FLAG标签用于镍亲和色谱和免疫检测。构筑其中多肽链的最后一个胺基酸转化成半胱氨酸(Cys)的突变体scFv。通过对突变体基因测序来确认所述改变。通过十二烷基硫酸钠(SDS)聚丙烯酰胺凝胶电泳(PAGE)确认野生型scFv和含C末端Cys突变体的表达和镍亲和纯化,通过在与scFv一致的分子量下用考马斯蓝(Coomassie blue)染色可视化。用抗FLAG标签HRP偶联物进行scFvSDS PAGE凝胶的西方墨点(western blot)分析。西方墨点分析确认凝胶条带的属性为scFv。
scFv是经含有双功能性连接体的叠氮化物修饰的克隆。野生型scFv经N-羟基-琥珀酰亚胺(NHS)酯-聚乙二醇(PEG)4-叠氮化物修饰。不希望受限于任何理论,用NHS酯-PEG4-叠氮化物修饰野生型scFv可将PEG4-叠氮化物随机纳入表面赖氨酸刺激上的游离胺上。将C末端scFv Cys构筑体与马来酰亚胺-PEG3-叠氮化物偶联以将位点特异性PEG3-叠氮化物引入Cys巯基上。通过与二苯并环辛炔(DBCO)-PEG-Cy5荧光探针反应分析scFv构筑体的叠氮化物纳入。通过不含金属的点击化学反应使叠氮化物与DBCO反应以形成共价键联。通过40kDa截止大小排阻旋转柱自反应混合物移除未反应的DBCO-Cy5染料。通过使用伯乐(BioRad)Versa-Doc成像仪使野生型和C末端Cys scFv-PEG-Cy5荧光染料构筑体可视化来确认将叠氮基成功引入scFv的表面上。
然后使叠氮化物偶联的scFv与含有1-3个DBCO的C点在其表面上反应。使反应在室温下持续12h。使用多种技术自偶联的scFv-C点纯化未偶联的scFv,所述技术包括50,000分子量截止旋转柱中的磷酸盐缓冲盐水洗涤、G-200大小排阻柱色谱或大小排阻旋转柱和通过蔗糖垫的速率沉降。速率沉降和大小排阻色谱似乎是最可缩放的纯化方法。通过点墨点scFv免疫检测/粒子荧光分析、凝胶电泳和使用固定化MMP-12的荧光ELISA分析经纯化的scFv C点偶联物。
这些方法可适用于其它类型的抗体片段,例如sdAb。
图1显示示意性图解说明,其显示使用基于螯合剂的放射性标记技术合成89Zr标记的C'点放射性免疫偶联物。首先使聚乙二醇化和马来酰亚胺官能化C’点(C’点-PEG-Mal,1)与还原谷胱甘肽(GSH)反应以为后续生物偶联物引入-NH2基团,形成C’点-PEG-GSH(2)。然后使纳米粒子与DBCO-PEG4-NHS酯和DFO-NCS偶联,分别形成C’点-PEG-DBCO(3)和DFO-C’点-PEG-DBCO(4)。基于应变促进的叠氮化物-炔环加成使叠氮化物官能化小靶向配体(例如单链可变片段(scFv-叠氮化物))(或单结构域抗体sdAb-叠氮化物)偶联到纳米粒子,形成DFO-C’点-PEG-scFv(5)。通过用草酸89Zr标记最终C'点放射性免疫偶联物(89Zr-DFO-C’点-PEG-scFv,6)。图1中所图解说明的示意图并不限于scFv且可包括不同类型的抗体片段,例如sdAb。
图2A和2B显示在健康裸小鼠中在不同注射后时间点(10min、1h、第1天、第3天和第6天)89Zr-DFO-C’点-PEG的体内(图2A)冠状和(图2B)矢状PET图像。C’点-PEG-Mal与GSH之间的反应比保持在1:20。PET图像是通过使用Focus 120MicroPET扫描仪获取。
图3显示在第6天时在健康裸小鼠中89Zr-DFO-C’点-PEG的生物分布数据。观察到小于2%ID/g的骨(和关节)摄取。

图7显示在注射后48h时在健康裸小鼠中89Zr-DFO-C’点、89Zr-DFO-C’点-DBCO和89Zr-DFO-C’点-PEG-sdAb的生物分布数据。通过最佳化来自每一C’点的DFO、DBCO和sdAb的数量可实现经改良的药代动力学特征(具有延长的血液循环半衰期和较低肝摄取)。
Claims (20)
1.一种免疫偶联物,其包含:
经有机聚合物涂布的纳米粒子;
偶联到所述有机聚合物涂布的纳米粒子的抗体片段;和
通过连接体偶联到所述有机聚合物涂布的纳米粒子的治疗剂;
其中所述纳米粒子具有不大于20纳米的直径,
其中所述纳米粒子包含基于二氧化硅的核心和包围所述核心的至少一部分的二氧化硅壳;
其中所述抗体片段是单链可变片段(scFv);并且
其中所述有机聚合物是聚乙二醇(PEG)。
2.根据权利要求1所述的免疫偶联物,其中所述连接体是可裂解连接体。
3.根据权利要求2所述的免疫偶联物,其中所述可裂解连接体选自由以下组成的群组:肽、肼和二硫化物连接体。
4.根据权利要求1所述的免疫偶联物,其中所述连接体是肽连接体。
5.根据权利要求1所述的免疫偶联物,其中所述连接体是被溶酶体蛋白酶裂解的肽连接体。
6.根据权利要求5所述的免疫偶联物,其中所述溶酶体蛋白酶是组织蛋白酶-B。
7.根据权利要求1所述的免疫偶联物,其中所述连接体是二肽连接体。
8.根据权利要求7所述的免疫偶联物,其中所述二肽连接体是缬氨酸-瓜氨酸连接体。
9.根据权利要求1所述的免疫偶联物,其中所述抗体片段为25kDa到30kDa。
10.根据权利要求1所述的免疫偶联物,其中所述纳米粒子包含在所述核心内的荧光化合物。
11.根据权利要求1所述的免疫偶联物,其中所述纳米粒子附接有1到10个抗体片段。
12.根据权利要求1所述的免疫偶联物,其中所述纳米粒子具有不大于15纳米的直径。
13.根据权利要求1所述的免疫偶联物,其中所述纳米粒子具有介于1nm到20nm范围内的直径。
14.根据权利要求1所述的免疫偶联物,其中所述抗体片段包含抗VEGF-A。
15.根据权利要求1所述的免疫偶联物,其中所述免疫偶联物包含一或多种成像剂。
16.根据权利要求15所述的免疫偶联物,其中所述一或多种成像剂包含PET示踪剂。
17.根据权利要求15所述的免疫偶联物,其中所述一或多种成像剂包含荧光团。
18.根据权利要求1所述的免疫偶联物,其中所述治疗剂包含化学治疗药物。
19.根据权利要求1所述的免疫偶联物,其中所述治疗剂包含放射性同位素。
20.根据权利要求19所述的免疫偶联物,其中所述放射性同位素是选自由以下组成的群组的成员:99mTc、111In、64Cu、67Ga、186Re、188Re、153Sm、177Lu、67Cu、123I、124I、125I、11C、13N、15O、18F、186Re、188Re、153Sm、166Ho、177Lu、149Pm、90Y、213Bi、103Pd、109Pd、159Gd、140La、198Au、199Au、169Yb、175Yb、165Dy、166Dy、67Cu、105Rh、111Ag、89Zr、225Ac和192Ir。
