本发明公开了一种药物载体及其制备方法与应用。所述药物载体自内由外包括镁基微马达、水凝胶涂层和酯化淀粉层;所述酯化淀粉层采用M细胞靶向性配体进行改性。所述药物载体可以抵抗胃酸、胃蛋白酶以及胰酶等消化道苛刻环境对药物的破坏,提高M细胞对疫苗物质的摄入量,提高肠道黏膜免疫应答水平具有结肠靶向特性,对药物尤其是蛋白质多肽等大分子药物的递送效率高。

技术领域
本发明属于医药领域,尤其涉及一种药物载体及其制备方法与应用。
背景技术
口服疫苗具有较好的依从性且简易便捷,是一种较为理想的疫苗给药方式。然而,以蛋白质和多肽类生物大分子为代表的口服疫苗在经过人体消化道的过程中,胃肠中苛刻的消化环境易于导致蛋白质和多肽生物大分子结构的破坏,致使生物利用度的下降。而传统口服疫苗药剂主要是通过过量的给药以达到免疫治疗的效果,存在免疫响应性低的不足,且易于导致不良风险。因而,如何提高口服疫苗在肠道中的递呈能力,并产生有效的黏膜免疫反应是口服疫苗成功的关键。
Peyer区作为肠腔和肠下淋巴滤泡组织的界面,其上所具有的特殊上皮细胞—M细胞,能够转运肠腔内的抗原性物质至肠下淋巴组织,从而激发人体肠道黏膜免疫反应。而传统微纳米药物制剂主要通过被动扩散的方式进行运动,只能在体液中进行布朗运动,其跨越细胞及组织屏障的能力相对较弱。
发明内容
本发明旨在至少解决上述现有技术中存在的技术问题之一。为此,本发明提出一种药物载体,所述药物载体能够提高M细胞靶向性,增加疫苗在Peyer区的主动富集,即可进而提高肠道黏膜免疫水平。
本发明还提出一种具有上述药物载体的制备方法。
本发明还提出一种具有上述药物载体的应用。
根据本发明的一个方面,提出一种药物载体,所述药物载体自内由外包括镁基微马达、水凝胶涂层和酯化淀粉层。
在本发明的一些实施方式中,所述酯化淀粉层采用M细胞靶向性配体进行改性。

一种口服疫苗,所述疫苗包括上述药物载体,优选地,所述疫苗药物分散在水凝胶层中。
根据本发明的实施方式,至少具有以下有益效果:与传统的基于被动扩散的微纳米药物载体相比,自驱动微纳米马达能够将外界能量转化为机械动能,从而实现自主地运动,具有更强的主动性和选择性。本发明方案通过在镁微粉表面修饰Au制备得到非对称微马达。随后利用水凝胶材料对马达进行药物包载,并利用兼具抵抗胃酸和胰淀粉酶响应性降解特性的酯化淀粉材料对微马达进行包衣处理,制备得到具有自主运动能力的药物载体。所述药物载体降低了胃酸、胃蛋白酶以及胰酶等消化道苛刻环境对药物的破坏,同时增强药物在肠道的吸收,进一步地,所述酯化淀粉材料采用M细胞靶向性配体进行改性,有效的提高M细胞对疫苗物质的摄入量,增加了肠道黏膜免疫应答水平,对药物尤其是蛋白质多肽类大分子药物的递送效率高,降低了疫苗药物的添加量。

具体实施方式
以下将结合实施例对本发明的构思及产生的技术效果进行清楚、完整地描述,以充分地理解本发明的目的、特征和效果。显然,所描述的实施例只是本发明的一部分实施例,而不是全部实施例,基于本发明的实施例,本领域的技术人员在不付出创造性劳动的前提下所获得的其他实施例,均属于本发明保护的范围。
M细胞(M cell):是散布于肠道黏膜上皮细胞间的一种特化的抗原转运扁平细胞,能将大部分抗原由肠腔转运到上皮下的淋巴组织,从而诱导机体产生免疫黏膜免疫应答或免疫耐受。
实施例1
本实施例提供一种药物载体,制备方法包括以下步骤:
1、酯化淀粉膜的制备
(1)以玉米淀粉为原料,利用酸酐对其进行高取代酯化修饰,获得酯化淀粉并对其酯化侧链的取代度进行调控(DS值1.5~2.5)。将获得的酯化淀粉经过105℃干燥处理,10g干基淀粉加入到100mL丙酮中加热溶解,室温下加入相应摩尔比例的N,N-碳酰二咪唑(CDI)活化2h,加入相应比例的M细胞靶向肽30℃下进行接枝反应24h。反应结束后用500Da分子量的透析袋对样品进行分离,以除去反应中间产物,醇沉处理,45℃下干燥,粉碎,得到M细胞靶向性肽修饰的高取代功能化酯化淀粉。
(2)将制备的高取代功能化酯化淀粉溶于乙酸乙酯中形成1w/v%的成膜溶液。
(3)将该溶液铺于平板之上,通过流延法制备酯化淀粉膜。通过调节淀粉材料的用量对膜的厚度进行调控,通过调控酯化淀粉取代度对膜的机械性能进行调控。
2、非均相反应羧甲基淀粉基崩解剂的制备
(1)利用甲醇对淀粉进行处理,以除去淀粉中存在的脂质。
(2)将9.45g的氯乙酸溶解于62.5mL的95%乙醇在中,加入11mL 30w/w%的氢氧化钠溶液,反应5分钟。
(3)加入40.5g干基淀粉。待搅拌均匀后再加入11mL 30%的氢氧化钠,同时加入62.5mL 95%乙醇。
(4)将烧瓶移入55℃水浴反应4h(冰醋酸调pH至6.5~7.0)。
(5)反应结束后取出反应物用95%乙醇洗涤、抽滤3次(以抽滤液中没有氯离子存在时为标准,用滴加硝酸银不产生沉淀为准)。
(6)将反应的淀粉在室温下晾干2~3小时,随后40℃恒温箱3小时,过30目筛去除糊化的颗粒。
3、智能响应型M细胞靶向微马达(药物载体)的制备
(1)对镁微粒进行筛选,获得粒径为20-40μm的镁粉微粒;以镁微粒为核心,采用离子溅射法(电流25-35mA,处理时间2-5min),制备得到包覆Au的微马达。
(2)采用旋涂技术,将壳聚糖溶液包覆在微马达表面,壳聚糖涂层厚度在1-3μm;将优化的功能化酯化淀粉材料溶于乙酸乙酯中形成1w/v%的成膜溶液,并利用其对微马达表面进行薄膜包衣处理,酯化淀粉膜厚度约1~3μm,制备得到药物载体(如图2所示)。将羧甲基淀粉、微晶纤维素以1:3的质量比混合,以一定的比例加入所制备的微马达,混合均匀。
(3)采用压片法制备粒径大小约为3-5毫米的含镁基微马达的片剂。
(4)将一定量的Eudragit L100-55溶于异丙醇中,配制成浓度为6.5w/v%的溶液,待完全溶解后,过200目筛网制备肠溶性包衣液。
(5)随后调流化床真空悬浮包衣机工作参数(风机转速1500~1800rpm、物料温度25~29℃、进风温度40~43℃、流化压力和雾化压力0.01~0.1MPa),包衣液进样流速为5~7rpm,利用包衣液对载药片进行肠溶包衣。
(6)通过调节包衣时间,对包衣膜的厚度进行调节,包衣增重4%~7%的微片,获得智能响应型微马达疫苗递送系统。
未载药的智能响应型微马达疫苗口服递送系统的制备流程图如图1所示,表征图如图2所示。
实施例2
本实施例提供一种载有口服疫苗的药物(微片剂形式),其制备方法包括以下步骤:
(1)分别采用450目与800目筛网对镁微粒进行分筛,分别获得粒径20~28μm的镁粉微粒。采用离子溅射法(电流25-35mA,处理时间2-4min),制备得到包覆Au的微马达。
(2)采用旋涂技术,将含口服疫苗药物的壳聚糖溶液(口服疫苗药物在壳聚糖溶液中的浓度为0.05w/w%)包覆在微马达表面,壳聚糖涂层厚度在1-3μm;将优化的功能化酯化淀粉材料(DS值1.5~2.5,M细胞靶向性配体接枝量为0.5~1.5%)溶于乙酸乙酯中形成1w/v%的成膜溶液,并利用其对微马达表面进行薄膜包衣处理,从而形成较好的非对称结构,制备得到药物载体,酯化淀粉膜厚度约1~3μm,所制备的药物载体粒径大小在35~40μm。将羧甲基淀粉(羧甲基取代度在0.015~0.10之间)、微晶纤维素以1:3的质量比混合,以一定的比例加入所制备的药物载体,混合均匀。
(3)采用压片法将质量比为10%~25%药物载体封装到直径大小为5mm的片剂中,。
(4)将一定量的Eudragit L100-55溶于异丙醇中,配制成浓度为6.5w/v%的溶液,待完全溶解后,过200目筛网制备肠溶性包衣液。
(5)随后调流化床真空悬浮包衣机工作参数(风机转速1500~1800rpm、物料温度25~29℃、进风温度40~43℃、流化压力和雾化压力0.01~0.1MPa),包衣液进样流速为5~7rpm,利用包衣液对载药片进行肠溶包衣,肠溶包衣厚度约为50~80μm,回肠响应时间约为15~20min,从而获得可用于口服疫苗递送的药物载体系统,微片质量为600~800mg。所制备的微马达的运动速度在65μm/s,运动时间2.5min~3.5min,有效黏膜穿透厚度为3mm~4mm,M细胞靶向效率为55%~65%。
制备得到的药物表征图如图3所示,从图中可以看出,本发明方案成功制备得到了20~28μm的药物。
实施例3
本实施例提供一种载有口服疫苗的药物(微片剂形式),其制备方法包括以下步骤:
(1)采用450目筛网对镁微粒进行分筛,获得粒径30~35μm的镁粉微粒。采用离子溅射法(电流25-35mA,处理时间2-4min),制备得到包覆Au的微马达;
(2)采用旋涂技术,将含口服疫苗药物的壳聚糖溶液(口服疫苗药物在壳聚糖溶液中的浓度为0.05w/w%)包覆在微马达表面,壳聚糖涂层厚度在1-3μm;将优化的功能化酯化淀粉材料(DS值1.5~2.5,M细胞靶向性配体接枝量为0.5~1.5%)溶于乙酸乙酯中形成1w/v%的成膜溶液,并利用其对微马达表面进行薄膜包衣处理,酯化淀粉膜厚度约1~3μm,制备得到药物载体,粒径大小为35~40μm。将羧甲基淀粉(羧甲基取代度在0.015~0.10之间)、微晶纤维素以1:3的质量比混合,以一定的比例加入所制备的药物载体,混合均匀。
(3)采用压片法将质量比为10%~25%药物载体封装到直径大小为5mm的片剂中,。
(4)将一定量的Eudragit L100-55溶于异丙醇中,配制成浓度为6.5w/v%的溶液,待完全溶解后,过200目筛网制备肠溶性包衣液。
(5)随后调流化床真空悬浮包衣机工作参数(风机转速1500~1800rpm、物料温度25~29℃、进风温度40~43℃、流化压力和雾化压力0.01~0.1MPa),包衣液进样流速为5~7rpm,利用包衣液对载药片进行肠溶包衣,肠溶包衣厚度约为50~80微米,回肠响应时间约为15~20min,从而获得可用于口服疫苗递送的药物载体系统,微片质量为600~800mg。所制备的微马达运动速度在55μm/s,运动时间3.5min~5min,有效黏膜穿透厚度为2mm~3mm,M细胞靶向效率为45%~55%。
制备得到的药物的表征图如图4所示,从图中可以看出,本发明方案成功制备得到了30~35μm的药物。
测试例
1、药物(微片剂)的表征
以实施例3所制得的口服疫苗药物为材料,使用扫描电镜和倒置荧光显微镜对药物载体的载药特性进行表征,测试结果如图4和图5所示,从图中可以看出本发明方案成功制备了口服疫苗药物,制备得到的片状药物的形貌图如图6所示。
无马达口服疫苗药物的制备:与实施例3的区别仅在于不包括步骤(1)制备得到微马达的步骤。继续测定有马达的实施例3制备得到的含有微马达的口服疫苗药物和无马达口服疫苗药物进行扫描电镜观察表面形貌,测试结果如图7所示。图8为实施例3制备得到的药物载体片剂的表面扫描电镜及EDX形貌图。
2、细胞毒性分析
将市购正常结肠细胞(购自上海楚天生物科技有限公司)接种在96孔板中,并在37℃、含5%CO2的细胞培养箱中培养过夜。用不同浓度(50μg/mL、100μg/mL、200μg/mL)的实施例1制备的药物载体处理细胞,并在37℃、含5%CO2的细胞培养箱中继续培养48h。采用商业的MTT检测试剂盒测定细胞的细胞活力。实验重复三次,数据用GraphPad Prism 6.0软件分析。
实验结果如图9所示,从图中可以看出,本发明方案制备的药物载体基本不影响正常细胞的活性。
3、药物载体系统对M细胞靶向性分析
实验材料:
实验组:按照实施例2所述的药物载体的制备方法制备得到的药物载体,与实施例2的区别仅在于M细胞靶向性肽接枝量为1.5%。
对照组:按照实施例2所述的药物载体的制备方法制备得到的药物载体,与实施例2的区别仅在于M细胞靶向性配体接枝量为0.5%。
实验步骤:用DMEM培养基溶液将Caco-2细胞(购自中国典型培养物保藏中心(武汉大学))重悬形成细胞悬浮液,首先将0.5mL Caco-2细胞(1×106/mL)悬浮液置于12孔聚碳酸酯Transwell滤膜上(3μm,1.12cm2膜表面积)37℃,5%CO2下培养14天,每两天更换培养基。培养结束后,将5×105/mL的Raji细胞悬浮液1mL加入到内嵌滤膜的底部进行与Caco-2细胞的共培养,以触发形成M细胞。共培养持续4-5天。M细胞模型后,将M细胞靶向肽修饰的药物载体与细胞相互接触2h。随后,在培养皿中加入足量PBS,避光条件下振动清洗细胞,使用单克隆抗体及Hoechst 33342等试剂对细胞进行免疫荧光染色处理,染色结束后采用共聚焦显微镜观察M细胞靶向性微马达对M细胞的靶向性及靶向效率。实验结果如图10所示,其中,图A为对照组M细胞靶向肽修饰的微马达对M细胞的靶向特性,图B为实验组M细胞靶向肽修饰的微马达对M细胞的靶向特性,从图中可以看出,本发明方案制备的药物载体可以高效的用于制备M细胞靶向药物。
4、药物载体系统黏膜穿透厚度分析
实验步骤:利用肠黏膜主要成分黏蛋白构建肠粘液层模拟体系(8mg/mL),并将配置好的黏蛋白溶液置于水平放置的细胞皿中,并将不同类型的马达置于同一深度位置,经随后利用倒置显微镜对微马达的黏膜穿透厚度分析。结果表明,由20~28μm的镁微粒构筑的微马达的有效黏膜穿透厚度为3mm~4mm。由粒径30~35μm的镁粉微粒的有效黏膜穿透厚度为2mm~3mm。
与传统的基于被动扩散的微纳米药物载体相比,自驱动微纳米马达能够将外界能量转化为机械动能,从而实现自主地运动,因此具有更强的主动性和选择性。因此,本发明方案提供了一种新型药物载体,提高M细胞靶向性,增加疫苗在Peyer区的主动富集,即可进而提高肠道黏膜免疫水平。
上面结合附图对本发明实施例作了详细说明,但是本发明不限于上述实施例,在所属技术领域普通技术人员所具备的知识范围内,还可以在不脱离本发明宗旨的前提下作出各种变化。此外,在不冲突的情况下,本发明的实施例及实施例中的特征可以相互组合。
Claims (7)
1.一种药物载体,其特征在于,所述药物载体自内由外包括镁基微马达、水凝胶涂层和酯化淀粉层;所述酯化淀粉层采用M细胞靶向性配体进行改性;所述酯化淀粉层中M细胞靶向性配体的接枝量为0.1-3%;所述水凝胶涂层为壳聚糖涂层或赖氨酸涂层;所述镁微粒的粒径为5-40μm;所述水凝胶涂层的厚度为1-3μm;所述酯化淀粉层厚度为0.5-5μm。
2.根据权利要求1所述的药物载体,其特征在于,接枝量为0.5-1.5%。
3.根据权利要求1所述的药物载体,其特征在于,所述镁基微马达为Au包覆的镁微粒。
4.根据权利要求1所述的药物载体,其特征在于,所述酯化淀粉层的厚度为1-3μm。
5.权利要求1-4中任一项所述的药物载体的制备方法,其特征在于,包括以下步骤:将水凝胶包覆在镁基微马达上,再采用酯化淀粉进行包衣,制得所述药物载体。
6.权利要求1-4中任一项所述的药物载体制备口服免疫药物中的应用。
7.根据权利要求6所述的应用,其特征在于,所述药物为多肽类药物和/或蛋白质类药物。
