Abstract
本发明公开了一种樱桃番茄采前与采后技术结合应用的绿色综合保鲜方法,包括以下步骤:(1)在樱桃番茄育苗播种前,将干种子用清水浸种后,再用混合药剂A浸泡;混合药剂A包括γ‑氨基丁酸、水杨酸和超敏蛋白;(2)于樱桃番茄小果期和转色期,分别用混合药剂B喷雾处理;混合药剂B包括γ‑氨基丁酸、水杨酸、复硝酚钠和乳化剂;(3)采摘樱桃番茄果实,用混合药剂C浸果。该综合保鲜方法处理可显著降低樱桃番茄果实在常温贮藏中的腐烂率,且较好维持可溶性固形物、维生素C、柠檬酸及番茄红素等营养品质,具有良好的防腐保鲜效果。该保鲜方法实施方便,操作简单,同时绿色安全高效,对生态环境友好,具有良好的经济效益和社会效益。

技术领域
本发明涉及樱桃番茄保鲜技术领域,具体涉及一种樱桃番茄采前与采后技术结合应用的绿色综合保鲜方法。
背景技术
樱桃番茄(Lycopersicon esculentum var.Cerasiforme)又称圣女果、小西红柿、小番茄果等,既是蔬菜又是水果,营养丰富。因其外观玲珑可爱,含糖度很高,口味香甜鲜美,风味独特而广受消费者喜爱。樱桃番茄主要分布在我国海南、广东、广西、云南、四川与福建等地。近年来樱桃番茄产业取得了迅速发展,品种越来越丰富,产量不断增加,但同时也面临着各种问题带来的挑战。首先,樱桃番茄在田间生长过程中的病害发生情况日益增多,防治难度也不断加大,一些病原菌容易潜伏浸染于果实表面,导致果实采后贮运中极易腐烂霉变,造成巨大的经济损失;其次,樱桃番茄属于呼吸跃变型果实,采后呼吸代谢旺盛,不耐贮藏保鲜,其皮薄多汁的特性使其在贮运过程中若不采取有效的保鲜措施极易失水萎蔫,继而出现霉变腐烂,直到完全失去商品价值。目前,销售企业的樱桃番茄保鲜多集中在采后,但繁杂的采后处理会增加劳动力成本和损耗;而种植者在田间生产中使用化学农药不当或滥用,往往会造成樱桃番茄品质下降,也不利于食品安全和生态环境的保护。因此,从采前入手,研发绿色安全的采前与采后技术结合应用的综合保鲜技术具有十分重要的现实意义。
目前,抗病性诱导技术已经成为果蔬采后防腐保鲜研究的热点之一。诱抗剂对病原物没有直接的杀菌作用,但能够诱导植物产生免疫活性,起到抗病、防病的作用,广泛应用于农业生产。水杨酸(salicylic acid,SA)是一种类似植物激素的内源信号分子,能够激活植物产生系统获得性抗性,在植物成熟、衰老等生理过程的调控和抗逆反应的诱导过程中起重要作用。研究表明,采前或采后外源SA处理均能提高芒果、香蕉、番茄、梨等果蔬对采后病害的抗性,达到良好的防腐保鲜作用。γ-氨基丁酸(γ-aminobutyric acid,GABA)是一种四碳氨基酸,广泛存在于动物、植物和微生物中,在植物生长发育和抗性反应中发挥着关键性的作用。已有研究表明,GABA处理能够提高桃和西葫芦采后冷藏期间的抗冷性,减轻果实冷害。也有研究报道,GABA处理能诱导采后梨果实对青霉病的抗性,抑制甜樱桃果实的腐烂。目前尚未发现水杨酸和γ-氨基丁酸在樱桃番茄上通过育苗期浸种处理提高果实采后防腐保鲜效果的应用研究报道。
超敏蛋白(Harpin蛋白,商品名:康壮素)是多肽类化合物,是具有诱导植物产生过敏反应(HR)的激活蛋白,能够激发植物产生系统获得抗性(SAR)。Harpin蛋白是由采用含有天然基因的工程菌发酵获得,易被植物表面受体识别,可以激活植物体内内源信号传导,是一类广谱的植物机能激活剂。复硝酚钠(compound sodium nitrophenolate,CSN)又名爱多收(atonik)、丰产素、特丰收,化学成分为5-硝基愈创木酚钠、邻硝基苯酚钠、对硝基苯酚钠;它在与作物接触后能迅速渗透到植物体内,促进细胞的原生质流动,提高细胞活力;同时还能促进生长发育、防止落花落果、改善产品品质、提高产量及作物的抗逆能力。有研究报道,复硝酚钠在荔枝、芒果上应用具有防腐保鲜效果。目前尚未发现超敏蛋白和复硝酚钠在樱桃番茄防腐保鲜上有应用报道。
樱桃番茄保鲜是一项综合的系统工程,采前和采后都不能被忽视。将育苗浸种技术、采前诱抗技术、采后绿色保鲜处理和贮藏有效结合起来,才能达到理想效果。但在实际生产中,采前与采后处理往往出现严重脱节,不能有机整合起来。
基于上述理由,提出本申请。
发明内容
针对上述现有技术存在的问题或缺陷,本发明的目的在于提供一种樱桃番茄采前与采后技术结合应用的绿色综合保鲜方法。本发明采前和采后并重,突出绿色安全高效,将育苗浸种技术、生长期诱抗技术与采后绿色保鲜处理有机结合,提出一套樱桃番茄采前与采后技术结合应用的绿色综合保鲜方法,该保鲜方法既安全环保高效又操作便捷,具有一定的应用前景。
本发明技术方案主要包括以下内容:
一种樱桃番茄采前与采后技术结合应用的绿色综合保鲜方法,包括以下步骤:
(1)在樱桃番茄育苗播种前,将干种子浸种后,用混合药剂A浸泡;
混合药剂A包括γ-氨基丁酸、水杨酸和超敏蛋白;
(2)于樱桃番茄小果期和转色期,分别用混合药剂B喷雾处理;
混合药剂B包括γ-氨基丁酸、水杨酸、复硝酚钠和乳化剂;
(3)采摘樱桃番茄果实,用混合药剂C浸果;
混合药剂C包括γ-氨基丁酸和水杨酸。
优选的,混合药剂A包括100~2000mg/Lγ-氨基丁酸、10~100mg/L水杨酸和100~500mg/L超敏蛋白。
优选的,混合药剂B包括100~1000mg/Lγ-氨基丁酸、10~100mg/L水杨酸、200~1000mg/L复硝酚钠和体积百分比浓度为0.05~0.2%的乳化剂。
优选的,所述乳化剂为阿维菌素4209-A辛硫磷。
优选的,混合药剂C包括100~2000mg/Lγ-氨基丁酸和10~100mg/L水杨酸。

本发明所取得的效果:
本发明采用的绿色综合保鲜方法,以育苗期浸种处理、田间生长期诱抗处理、采后诱抗处理与常温贮藏相结合,达到减轻樱桃番茄采后腐烂的发生,提高果实防腐保鲜效果的目的。经该方法处理的樱桃番茄果实,在常温(25±1)℃、相对湿度80~90%的库中贮藏15d时,果实腐烂明显减轻,腐烂率为1.33~5.03%,与对照(16.38%)相比降低了11.35~15.05%,防效达到69.29~91.88%。本发明采前与采后技术结合应用的绿色综合保鲜方法可有效降低樱桃番茄常温贮藏中的腐烂率,并较好维持营养品质,提高防腐保鲜效果,该方法安全环保高效,操作便捷,容易推广应用。
具体实施方式
为了更好理解本发明技术内容,下面提供具体实施例,对本发明做进一步的说明。
本发明下述各例中涉及的樱桃番茄果实腐烂果的定义以及腐烂率的计算方法如下:
调查常温贮藏期樱桃番茄果实的腐烂情况,判定标准以果实全部或局部霉变、腐烂或流汁均计入腐烂果。
腐烂率(%)=腐烂果数×100/调查总果数。
防效(%)=(对照果腐烂率-处理果腐烂率)×100/对照果腐烂率。
本发明下述各例中涉及的樱桃番茄果实营养品质指标测定方法如下:
可溶性固形物含量测定:取15个果实,将果肉用两层纱布包裹,挤出果汁,用手持折光仪测定可溶性固形物含量,共测定15个数据取平均值,结果表示为%。
维生素C和柠檬酸含量测定:称取果肉样品0.5g于10mL离心管中,加5mL超纯水提取,在10 000g下离心5min,取上清液,经0.22μm水相滤膜过滤后待测。使用仪器为waters2695高效液相色谱仪,配有2998紫外检测器,色谱柱为Agilent-C18(250mm×4.6mm,粒径5μm),流动相为0.5%NH4H2PO4-H3PO4缓冲溶液;流速为1mL/min;柱温为35℃;进样量为10.0μL;检测波长分别为254nm和210nm,分别用于检测维生素C和柠檬酸含量。每个样品重复测定3次数据,取平均值,维生素C结果以mg/100g表示,柠檬酸结果以g/kg表示。
番茄红素含量测定:称取0.5g果肉样品于10mL的离心管中,加5mL的乙醇与三氯甲烷等体积配制的提取液,在50℃水浴锅中水浴60min后,10 000g下高速离5min,经0.22μm水相滤膜过滤后待测。使用仪器为waters 2695高效液相色谱仪,配有2998紫外检测器,色谱柱为Agilent-C18(250mm×4.6mm,粒径5μm),流动相A为乙腈,流动相B为异丙醇;流速为1mL/min;柱温为35℃;进样量为10.0μL;检测波长为450nm。每个样品重复测定3次数据,取平均值,番茄红素结果均以mg/kg表示。
本发明采用的诱导抗病保鲜剂的选择及处理方法是在下述理论和实验结果基础上做出的:
发明人的研究表明,采后SA、GABA及其结合处理能够延缓樱桃番茄果实的腐烂发生,并且GABA(1000mg/L)与SA(40mg/L)的结合处理效果优于GABA(1000mg/L)和SA(40mg/L)单独处理效果(见表1);研究发现,诱抗剂SA(40mg/L)和GABA(1000mg/L)于采前小果期(苗移栽后60d)和转色期(苗移栽后100d)处理2次的防腐保鲜效果要好于仅在小果期(苗移栽后60d)喷施1次的效果(见表2);又研究发现,于采前生长期小果期(苗移栽后50d)和开始转色期(苗移栽后100d)2次喷施SA、GABA、CSN及其结合处理能够延缓樱桃番茄果实的腐烂发生,并且GABA(1000mg/L)、SA(40mg/L)与CSN(500mg/L)的结合处理效果优于GABA、SA和CSN单独处理效果(见表3);发明人研究发现,育苗期播前SA(40mg/L)、GABA(1000mg/L)浸种处理能够提高樱桃番茄果实采后常温贮藏中防腐保鲜效果,并且(SA+GABA)复合浸种处理效果显著优于SA、GABA单独浸种处理,而(SA+GABA+超敏蛋白)复合浸种处理效果优于(SA+GABA)复合浸种处理(见表4);发明人进一步将育苗期浸种处理A、田间生长期诱抗处理B与采后诱抗处理C相结合,研究发现该综合处理方法,可以显著降低樱桃番茄果实常温贮藏期的腐烂率,提高果实防腐保鲜的效果,并且效果显著优于采后处理C、(生长期处理B+采后处理C)(见表5)。因此,该综合处理方法具有良好的防腐保鲜效果。

(1)在樱桃番茄育苗播种前,将干种子先用清水浸泡3h,然后用混合药剂A浸泡3h;混合药剂A包括100mg/Lγ-氨基丁酸、10mg/L水杨酸和100mg/L超敏蛋白;
(2)将步骤(1)浸种处理后的种子,用湿润双层纱布或棉花团包裹,在30~35℃恒温培养箱中培养18h进行催芽;
(3)将步骤(2)催芽后的种子,拌入适量的杀虫粉剂(氯氰菊酯含量0.25%)和种子处理干粉剂(甲霜灵含量35%),在育苗盘上播种后放置于育苗棚中育苗约40d;
(4)将步骤(3)育好的樱桃番茄苗移栽于大田,进行常规生产栽培管理;
(5)在步骤(4)移栽于大田的樱桃番茄小果期(苗移栽后50d)和转色期(苗移栽后100d),分别用混合药剂B喷雾处理,直至将果面喷施均匀;混合药剂B包括100mg/Lγ-氨基丁酸、10mg/L水杨酸、200mg/L复硝酚钠和体积百分比浓度为0.05%的乳化剂阿维菌素4209-A辛硫磷;
(6)采摘经步骤(5)田间处理的果面全部转红的樱桃番茄果实,用混合药剂C浸果2min;混合药剂C包括100mg/Lγ-氨基丁酸和10mg/L水杨酸;
(7)经步骤(6)浸果处理的樱桃番茄果实,风干后装入泡沫箱中,置于(25±1)℃常温库中贮藏。
实施例2一种樱桃番茄采前与采后技术结合应用的绿色综合保鲜方法
(1)在樱桃番茄育苗播种前,将干种子先用清水浸泡4h,然后用混合药剂A浸泡2h,混合药剂A包括200mg/Lγ-氨基丁酸、20mg/L水杨酸和200mg/L超敏蛋白;
(2)将步骤(1)浸种处理后的种子,用湿润双层纱布或棉花团包裹,在30~35℃恒温培养箱中培养18h进行催芽;
(3)将步骤(2)催芽后的种子,拌入适量的杀虫粉剂(氯氰菊酯含量0.25%)和种子处理干粉剂(甲霜灵含量35%),在育苗盘上播种后放置于育苗棚中育苗约40d;
(4)将步骤(3)育好的樱桃番茄苗移栽于大田,进行常规生产栽培管理;
(5)在步骤(4)移栽于大田的樱桃番茄小果期(苗移栽后50d)和转色期(苗移栽后100d),分别用混合药剂B喷雾处理,直至将果面喷施均匀;混合药剂B包括200mg/Lγ-氨基丁酸、20mg/L水杨酸、500mg/L复硝酚钠和体积百分比浓度0.1%乳化剂阿维菌素4209-A辛硫磷;

(5)在步骤(4)移栽于大田的樱桃番茄小果期(苗移栽后50d)和转色期(苗移栽后100d),分别用混合药剂B喷雾处理,直至将果面喷施均匀,混合药剂B包括2000mg/Lγ-氨基丁酸、100mg/L水杨酸、1000mg/L复硝酚钠和体积百分比浓度为0.2%的乳化剂阿维菌素4209-A辛硫磷;
(6)采摘经步骤(5)田间处理的果面全部转红的樱桃番茄果实,用混合药剂C浸果8min;混合药剂C包括2000mg/Lγ-氨基丁酸和100mg/L水杨酸;
(7)经步骤(6)浸果处理的樱桃番茄果实,风干后装入泡沫箱中,置于(25±1)℃常温库中贮藏。
对比例1一种樱桃番茄保鲜方法
(1)在樱桃番茄育苗播种前,将干种子先用清水浸泡6h;
(2)将步骤(1)浸种处理后的种子,用湿润双层纱布或棉花团包裹,在30~35℃恒温培养箱中培养18h进行催芽;
(3)将步骤(2)催芽后的种子,拌入适量的杀虫粉剂(氯氰菊酯含量0.25%)和种子处理干粉剂(甲霜灵含量35%),在育苗盘上播种后放置于育苗棚中育苗约40d;
(4)将步骤(3)育好的樱桃番茄苗移栽于大田,进行常规生产栽培管理;
(5)在步骤(4)移栽于大田的樱桃番茄小果期(苗移栽后50d)和转色期(苗移栽后100d),用体积百分比浓度为0.1%的乳化剂(阿维菌素4209-A辛硫磷)将果面喷施均匀。
(6)采摘经步骤(5)田间处理的果面全部转红的樱桃番茄果实,用清水浸果5min;
(7)经步骤(6)浸果处理的樱桃番茄果实,风干后装入泡沫箱中,置于(25±1)℃常温库中贮藏。
对比例2一种樱桃番茄采前与采后技术结合应用的绿色综合保鲜方法
(1)在樱桃番茄育苗播种前,将干种子先用清水浸泡6h;
(2)将步骤(1)浸种处理后的种子,用湿润双层纱布或棉花团包裹,在30~35℃恒温培养箱中培养18h进行催芽;

(5)在步骤(4)移栽于大田的樱桃番茄小果期(苗移栽后50d)和转色期(苗移栽后100d),用体积百分比浓度为0.1%的乳化剂(阿维菌素4209-A辛硫磷)将果面喷施均匀。
(6)采摘经步骤(5)田间处理的果面全部转红的樱桃番茄果实,用混合药剂C浸果5min;混合药剂C包括1000mg/Lγ-氨基丁酸和40mg/L水杨酸;
(7)经步骤(6)浸果处理的樱桃番茄果实,风干后装入泡沫箱中,置于(25±1)℃常温库中贮藏。
表6实施例及对比例采前与采后结合处理对樱桃番茄腐烂和品质的影响(贮藏15d)
注:表中同列数据后不同小写英文字母表示在5%水平上差异显著。
结果显示:实施例1~实施例4樱桃番茄贮藏15d后,果实腐烂率为1.33%~5.03%,比对照(16.38%)降低了11.35%~15.05%,防效达到69.29%~91.88%,且较好维持可溶性固形物、维生素C、柠檬酸及番茄红素等营养品质。说明实施例1~实施例4处理方法具有良好的樱桃番茄果实的防腐保鲜效果。

Claims (10)
1.一种樱桃番茄采前与采后技术结合应用的绿色综合保鲜方法,其特征在于,包括以下步骤:
(1)在樱桃番茄育苗播种前,将干种子用水浸种后,用混合药剂A浸泡;
混合药剂A包括γ-氨基丁酸、水杨酸和超敏蛋白;
(2)于樱桃番茄小果期和转色期,分别用混合药剂B喷雾处理;
混合药剂B包括γ-氨基丁酸、水杨酸、复硝酚钠和乳化剂;
(3)采摘樱桃番茄果实,用混合药剂C浸果;
混合药剂C包括γ-氨基丁酸和水杨酸。
2.根据权利要求1所述樱桃番茄采前与采后技术结合应用的绿色综合保鲜方法,其特征在于,混合药剂A包括100~2000mg/Lγ-氨基丁酸、10~100mg/L水杨酸和100~500mg/L超敏蛋白。
3.根据权利要求1所述樱桃番茄采前与采后技术结合应用的绿色综合保鲜方法,其特征在于,混合药剂B包括100~1000mg/Lγ-氨基丁酸、10~100mg/L水杨酸、200~1000mg/L复硝酚钠和体积百分比浓度为0.05~0.2%的乳化剂。
4.根据权利要求1所述樱桃番茄采前与采后技术结合应用的绿色综合保鲜方法,其特征在于,所述乳化剂为阿维菌素4209-A辛硫磷。
5.根据权利要求1所述樱桃番茄采前与采后技术结合应用的绿色综合保鲜方法,其特征在于,混合药剂C包括100~2000mg/Lγ-氨基丁酸和10~100mg/L水杨酸。
6.根据权利要求1所述樱桃番茄采前与采后技术结合应用的绿色综合保鲜方法,其特征在于,混合药剂A包括1000mg/Lγ-氨基丁酸、40mg/L水杨酸和500mg/L超敏蛋白;混合药剂B包括1000mg/Lγ-氨基丁酸、40mg/L水杨酸、500mg/L复硝酚钠和体积百分比浓度为0.1%的乳化剂;混合药剂C包括1000mg/Lγ-氨基丁酸和40mg/L水杨酸。
7.根据权利要求1所述樱桃番茄采前与采后技术结合应用的绿色综合保鲜方法,其特征在于,用水浸种时间为3~5h。
8.根据权利要求1所述樱桃番茄采前与采后技术结合应用的绿色综合保鲜方法,其特征在于,混合药剂A浸泡1~3h。
9.根据权利要求1所述樱桃番茄采前与采后技术结合应用的绿色综合保鲜方法,其特征在于,混合药剂C浸果2~8min。
10.根据权利要求1所述樱桃番茄采前与采后技术结合应用的绿色综合保鲜方法,其特征在于,所述小果期为苗移栽后50~60d,所述转色期为苗移栽后100~110d。
