技术领域
本公开属于生物技术领域,具体涉及一种mRNA,及其制备方法和应用。
背景技术
严重急性呼吸系统综合症冠状病毒(SARS-CoV-2,新冠病毒)是一种有包膜的正义单链RNA病毒,可导致新型冠状病毒病(COVID-19),引发机体产生发热、乏力、干咳等症状,严重的还容易引起呼吸困难,最终导致死亡。
新冠病毒颗粒的结构包括入侵宿主细胞所需的遗传物质RNA和结构蛋白。新冠病毒侵染细胞后,遗传物质RNA被用来编码构成病毒颗粒的结构蛋白,指导病毒组装、转录、复制和宿主控制的非结构蛋白,以及功能尚未确定的辅助蛋白。SARS-CoV-2的结构蛋白包括包膜(E)蛋白、表面刺突(S)蛋白、膜(M)蛋白和核衣壳(N)蛋白。S蛋白位于病毒颗粒的外部,介导新冠病毒颗粒的附着和进入宿主细胞,是新冠疫苗开发、抗体疗法和基于抗原的诊断测试的主要靶标。
S蛋白是新冠病毒的主要免疫原区域,绝大多数疫苗免疫后产生的中和抗体及恢复期患者血清中的中和抗体均由S蛋白的受体结合结构域(RBD)产生。最近有报道发现,S蛋白的S1/S2连接处的弗林酶切位点缺失或突变会请确认引起新冠病毒在Vero细胞中呈现复制增强,在动物中呈现毒力减弱的状态,且这种含有弗林酶切位点缺失的病毒能够诱导产生很强的体液免疫和细胞免疫反应。
截止到2021年8月9日,由COVID-19引发的疫情在全世界范围内累计确诊病例达到202935805例,累计死亡病例4298483例。目前,已有灭活疫苗、重组蛋白疫苗、mRNA疫苗、腺病毒载体疫苗、减毒流感病毒载体疫苗等新冠疫苗陆续上市或进入临床试验阶段。在保证安全性的前提下,如何获得一种免疫原性更好的新冠疫苗,是新冠疫苗开发设计的重要目标。
发明内容
为了解决现有技术中存在的上述技术问题之一,本公开对新冠病毒SARS-CoV-2中表达S蛋白的mRNA核酸序列进行了改造,提供了一种免疫原性得到显著提高的mRNA序列,可用于新冠疫苗的开发。
根据本公开的一个方面,提供了一种分离的mRNA,所述mRNA编码重组表面刺突(S)蛋白,所述重组S蛋白来源于严重急性呼吸系统综合症冠状病毒(SARS-CoV-2),且包含在N末端结构域中的缺失和弗林酶切位点结构域的缺失。本公开的S蛋白mRNA具有与新冠病毒野生型毒株的S蛋白mRNA相比显著提高的免疫原性。
根据本公开的一些实施方式,所述SARS-CoV-2可以选自原始病毒株、P.1病毒株、P.2病毒株、B.1.351病毒株、B.1.1.7病毒株、C.37病毒株或B.1.617.2病毒株。根据本公开的一些实施方式,所述mRNA表达的S蛋白可以具有如SEQ ID NO:1~7中任一项所示的氨基酸序列。根据本公开的一些实施方式,所述mRNA表达的S蛋白可以具有与SEQ ID NO:1~7中任一项所示的氨基酸序列具有至少85%同一性的氨基酸序列,例如,具有与SEQ ID NO:1~7中任一项所示的氨基酸序列具有至少85%,86%,87%,88%,89%,90%,91%,92%,93%,94%,95%,96%,97%,98%或99%同一性的序列。
根据本公开的一些实施方式,所述N末端结构域中可以包括如IX1X2SGTNX3X4所示的序列的缺失,其中X1不存在或者选自组氨酸,X2不存在或者选自缬氨酸,X3选自甘氨酸或缬氨酸,和/或X4选自苏氨酸或异亮氨酸。根据本公开的一些实施方式,所述N末端结构域中可以包括如SEQ ID NO:8~10中任一项所示的氨基酸序列的缺失。
根据本公开的一些实施方式,所述弗林酶切位点结构域的缺失包括如X5RRAR所示的序列的缺失,其中X5选自组氨酸、精氨酸或脯氨酸。根据本公开的一些实施方式,所述弗林酶切位点结构域的缺失包括如SEQ ID NO:11~13中任一项所示的氨基酸序列的缺失。
根据本公开的一些实施方式,所述mRNA表达的S蛋白可以具有如SEQ ID NO:14~20中任一项所示的氨基酸序列。根据本公开的一些实施方式,所述mRNA表达的S蛋白可以具有与SEQ ID NO:14~20中任一项所示的氨基酸序列具有至少85%同一性的氨基酸序列,例如,具有与SEQ ID NO:14~20中任一项所示的氨基酸序列具有至少85%,86%,87%,88%,89%,90%,91%,92%,93%,94%,95%,96%,97%,98%或99%同一性的序列。
根据本公开的一些实施方式,所述mRNA还可以包括:5’UTR序列、3’UTR序列和多聚腺苷酸(PolyA)序列。根据本公开的一些实施方式,所述mRNA按照从5’到3’的方向可以依次包括:5’UTR序列、编码的所述重组S蛋白的序列、3’UTR序列和多聚腺苷酸序列。根据本公开的一些实施方式,所述mRNA的5’端还可以包括在5’帽结构。

根据本公开的又一方面,提供了如上所述的mRNA、如上所述的DNA序列、如上所述的递送系统,在制备用于对象的SARS-CoV-2疫苗中的用途。根据本公开的一些实施方式,所述对象可以包括哺乳动物,优选为人、猴、骆驼、牛、马、山羊、绵羊、猪、猫、狗、兔、小鼠或大鼠。本公开的mRNA或DNA可以根据对象的不同,而对RNA或DNA序列进行优化。
根据本公开的又一方面,提供了一种用于制备如上所述的mRNA的方法。根据本公开的一些实施方式,所述方法包括:对来源于严重急性呼吸系统综合症冠状病毒(SARS-CoV-2)的S蛋白mRNA进行改造,其表达的S蛋白在N末端结构域和弗林酶切位点结构域存在缺失。
根据本公开的一些方式,所示方法包括以下步骤:在编码上述S蛋白的核苷酸序列的两端加上5’UTR和3’UTR,以及多聚腺苷酸(PolyA)尾。筛选正确的重组子,用重组子转化大肠杆菌,获得大量纯净的质粒后,进行体外mRNA转录及mRNA的纯化。纯化的mRNA通过脂质体包裹,形成纳米脂质颗粒,可以用作新冠候选mRNA疫苗。
本公开通过实验证明,由于弗林酶切位点的缺失会导致S1和S2处的连接无法断开,因而更好的保证S蛋白的完整性,从而能够诱导产生很强的体液免疫和细胞免疫反应。本公开获得的mRNA变体相比于新冠病毒原型株S蛋白mRNA的免疫原性显著提高,因此是作为免疫原或抗原用于免疫对象的理想疫苗候选蛋白。

SARS-CoV-2病毒颗粒的S蛋白与细胞表面受体血管紧张素转化酶2(ACE2)结合,以促进其进入细胞。由于S蛋白对于病毒进入至关重要,因此它已成为疫苗开发和治疗性抗体干预的目标。SARS-CoV-2的S蛋白被弗林酶(Furin)切割,产生S1和S2片段。S1蛋白包括N末端结构域(NTD)和受体结合结构域域(RBD),而S2蛋白促进膜融合。
目前发现了七株SARS-CoV-2病毒,即原始病毒株、P.1(Gamma)病毒株、P.2(Zeta)病毒株、B.1.351(Beta)病毒株、B.1.1.7(Alpha)病毒株、C.37(Lambda)病毒株或B.1.617.2(Delta)病毒株。原始病毒株的S蛋白具有如SEQ ID NO:1所示的氨基酸序列。P.1(Gamma)病毒株的S蛋白具有如SEQ ID NO:2所示的氨基酸序列。P.2(Zeta)病毒株的S蛋白具有如SEQID NO:3所示的氨基酸序列。B.1.351(Beta)病毒株的S蛋白具有如SEQ ID NO:4所示的氨基酸序列。B.1.1.7(Alpha)病毒株的S蛋白具有如SEQ ID NO:5所示的氨基酸序列。C.37(Lambda)病毒株的S蛋白具有如SEQ ID NO:6所示的氨基酸序列。B.1.617.2(Delta)病毒株的S蛋白具有如SEQ ID NO:7所示的氨基酸序列。图1中示出了这七株毒株的S蛋白的序列比对结果。

实施例1
选取SEQ ID NO:23和SEQ ID NO:24所示的RNA序列,两端分别添加5’UTR和3’UTR,构建到pUC57质粒中,然后转化大肠杆菌。通过放大培养阳性菌株,提取大量质粒。质粒经过纯化,进行体外转录成mRNA。使用牛痘病毒加帽酶将mRNA 5’端加帽为Cap0结构,使用2’-O-甲基转移酶进一步加帽为Cap1结构。后经过纯化,得到纯净的mRNA,用于脂质纳米颗粒(LNP)的制备。
实施例2
将一定比例的可电离阳离子脂质、中性脂质、胆固醇和PEG脂质溶解于乙醇,充分振荡混匀后,作为油相。其次,将mRNA溶解于pH=4~5的柠檬酸缓冲溶液中,作为水相。再次,在微流控设备中,插入微流控芯片,并将油相、水相装入注射器中后,分别拆入芯片的注入口中;微流控系统中,设置温度为室温,注射总流速为6-24mL/min,流速比(油相:水相)为1:3,启动微流控设备,收集LNP初乳。采用超滤管将LNP初乳的柠檬酸缓冲液和乙醇置换为PBS(pH=7.4)。测定LNP粒子的粒径、Zeta电位以及各组分含量。
实施例3
按照实施例2的方法进行脂质体的包裹,得到相应的S-LNP(含SEQ ID NO:23)和S-del-LNP(SEQ ID NO:24),每组10只,免疫剂量为20μg/只,0、21天免疫小鼠后,第35天采血进行S蛋白特异的IgG抗体效价和中和抗体效价检测,结果分别示于图2和图3所示。
从图2可以看出,S蛋白特异的IgG抗体效价,S-LNP约为2580318,S-del-LNP免疫后约为4597605。与S-LNP相比,S-del-LNP免疫后特异的IgG抗体效价提高约1.8倍。从图3可以看出,血清中和抗体效价GMT,S-LNP约为1:3000,S-del-LNP免疫后的GMT为1:7094。与S-LNP相比,S-del-LNP血清中和抗体效价提高约2.4倍且有统计学差异。
实施例4
本实施例中,分别构建了含有编码重组S蛋白(SEQ ID NO:14)的mRNA。根据实施例1表达纯化后,按照实施例2的方法进行脂质体的包裹得到S-del-LNP。每组10只,免疫剂量为20μg/只,0、21天免疫小鼠后,进行细胞免疫检测,结果如图4和图5所示。
细胞免疫指标为:(1)加强免疫后7天用ELISPOT检测诱导细胞产生细胞因子情况:IFN-γ,IL2,IL4,IL5;(2)加强免疫后9天进行CD4+T细胞、CD8+T细胞、效应记忆T细胞亚群检测。
从图4可以看出,含编码重组S蛋白的mRNA能够显著提高IFN-γ、IL2和IL4的产生。从图5可以看出,含编码重组S蛋白的mRNA能够显著提高效应记忆T细胞亚群的数量。

Claims (8)

1.一种分离的mRNA,其特征在于,所述mRNA编码重组表面刺突(S)蛋白,所述重组S蛋白来源于严重急性呼吸系统综合症冠状病毒(SARS-CoV-2),且包含在N末端结构域中的缺失和弗林酶切位点结构域的缺失,所述重组S蛋白的氨基酸序列为SEQ ID NO: 14~20中任一项所示的氨基酸序列。
2.根据权利要求1所述的mRNA,其特征在于,所述SARS-CoV-2选自原始病毒株、P.1病毒株、P.2病毒株、B.1.351病毒株、B.1.1.7病毒株、C.37病毒株或B.1.617.2病毒株。
3. 根据权利要求1所述的mRNA,其特征在于,所述mRNA还包括:5’ UTR序列、3’ UTR序列和多聚腺苷酸序列,
其中,所述mRNA按照从5’到3’的方向依次包括:5’ UTR序列、编码的所述重组S蛋白的序列、3’ UTR序列和多聚腺苷酸序列。
4.一种编码如权利要求1至3中任一项所述的mRNA的DNA。
5.一种递送系统,其特征在于,所述递送系统包括如权利要求1至3中任一项所述的mRNA。
6.如权利要求1至3中任一项所述的mRNA、如权利要求4所述的DNA或如权利要求5所述的递送系统,在制备用于对象的SARS-CoV-2疫苗中的用途。
7.如权利要求6所述的用途,其特征在于,所述对象为哺乳动物。
8.如权利要求7所述的用途,其特征在于,所述对象为人、猴、骆驼、牛、马、猫、狗、兔、小鼠或大鼠。
