背景技术
阿尔茨海默病(AD)是一种神经退行性疾病,其主要病理特征是神经元功能的进行性丧失。β-淀粉样蛋白(Aβ)是淀粉样前体蛋白 (APP)的蛋白水解产物,是AD病理过程中的致病因素之一。Aβ包括许多亚型,人体内最常见的Aβ亚型是Aβ1-40和Aβ1-42。Aβ1-40和Aβ1-42都经历从单体、寡聚体到聚集体的转变。Aβ1-42比Aβ1-40具有更大的毒性并且更容易形成聚集体,这种聚集体会造成Aβ在脑部的沉积,进而导致产生神经毒性,AD的进展与导致的神经毒性密切相关。因此,Aβ1-42是AD早期诊断的重要生物标志物。
目前,AD的临床诊断方法主要基于正电子发射断层扫描(PET) 和单光子发射计算机断层扫描(SPECT)两种。这两种方法均需要含有放射性核素的探针作为造影剂,且由于放射性核素的存在,可能不利于人体健康。此外,这两种方法的数据采集时间过长,花费较高。与这两种方法相比,光学成像不使用放射性核素,可以对大脑进行实时成像。然而,大脑有一个先天屏障来维持大脑内部微环境的稳态,称为血脑屏障(BBB)。BBB由脑毛细血管内皮细胞(BCECS)、周细胞、星形胶质细胞和神经元细胞的紧密连接形成。BCECS之间的紧密连接阻止了化合物从血液到大脑的细胞旁转运。因此,几乎100%的大分子和98%以上的有机小分子都被BBB阻断。Aβ1-42通常积聚在大脑海马和皮层,由于血脑屏障的存在,体内Aβ1-42的成像和诊断变得十分困难。
商业探针硫磺素T(ThT)是一种用于体外海马和皮层染色的标准探针之一,用于确认Aβ的存在。因此,ThT已成为AD成像和诊断的金标准探针之一。虽然ThT在检测Aβ方面具有一些优势,但它也有几个内在的缺陷,例如BBB渗透性差(脂水分配系数Log P= -0.14)、发射波长短(PBS中的λem=490nm)。血脑屏障通透性受多种因素影响,这主要包括物质的亲酯性和亲水性、有机小分子的分子量,物质与血浆蛋白的结合程度等。然而,物质的血脑屏障通透性的实际情况很复杂,上述因素也是仅供参考的可能的影响因素,并不能仅仅根据这些因素来判断物质能否穿过血脑屏障。通常,Log P值介于2和 5之间且分子量小于500Da的探针,具有近红外区域的发射波长,对于穿透BBB和促进体内脑成像至关重要。由于ThT具有较低的Log P 值(Log P<1),其无法穿过BBB,因此只能用于对Aβ的体外检测。
在开发用于Aβ检测的荧光探针的过程中,不免会遇到有一些检测性能良好的探针,但是由于同样无法穿透BBB,尚无法用于体内Aβ的实时检测。
因此,亟需开发一种递送这类小分子探针穿过血脑屏障的通用策略,以解决探针无法在体内实时成像Aβ这一问题。
发明内容
为了解决现有技术中存在的问题,本发明的目的是通过改变小分子探针的发射波长,在保证小分子探针对Aβ的检测能力的同时,解决其不能穿过血脑屏障这一关键问题。
为了实现本发明目的,本发明的技术方案如下:
第一方面,本发明提供一种用于β-淀粉样蛋白体内检测的复合物,所述复合物由经乳铁蛋白、转铁蛋白和其他类型转运受体中至少一种修饰的介孔硅与小分子探针掺杂而成;
所述小分子探针选自硫磺素T和SZIs中的至少一种。
所述小分子探针的LogP值为-0.14~1.26,分子量为318.86~448.37。 Log P值由以下网站计算(http://www.vcclab.org/web/alogps/)。
其中,所述SZIs为已经报道的小分子探针,具体可为参考文献 (MarekAntonIvica Sigmundová,Pavol Zahradník, Roman Dědic,MagdalénaHromadová.Photostability of D–π–A nonlinear optical chromophores containing abenzothiazolium acceptor.Journal of Physical Organic Chemistry,2011,24,450-459.)中记载的“1”物质或“2”物质。
作为优选,所述复合物的粒径为40~70nm。
进一步地优选,所述复合物中,介孔硅材料与修饰物的摩尔比为 83:0.071~75:0.14。
第二方面,本发明提供了所述复合物的制备方法,具体为:将经乳铁蛋白、转铁蛋白和其他类型转运受体中至少一种修饰的介孔硅与小分子探针掺杂,静置,离心后得到所述复合物。
作为优选,将经修饰的介孔硅与小分子探针按25:1~100:1的质量比掺杂。
其中,经修饰的介孔硅粒径为40~70nm,由于掺杂后小分子探针将进入介孔硅的孔隙中,因此所述复合物的粒径并不会有明显变化。
作为一种示例性说明,将介孔硅经乳铁蛋白修饰,修饰后,乳铁蛋白与介孔硅的摩尔比为0.071:83~0.088:83,其中,介孔硅的摩尔量以SiO2的摩尔量计。
针对以上示例所制备的复合物,其制备方法包括如下步骤(参见图1):
1)以十六烷基三甲基氯化铵(CTAC)为模板,三乙醇胺(TEA) 作为碱,正硅酸乙酯(TEOS)为原料制备介孔硅纳米材料,以摩尔比计,三者摩尔比为1.25:5.36:6.69;
2)以3-氨丙基三乙氧基硅烷(APTES)和步骤1)制得的介孔硅纳米材料为原料制备氨基-介孔硅(MSN-NH2),以摩尔比计,二者的质量比为0.83:0.43,其中介孔硅材料的摩尔量以SiO2对应的摩尔量计;
3)以双羧基聚乙二醇(HOOC-PEG-COOH)、1-乙基-(3-二甲基氨基丙基)碳酰二亚胺(EDC)、N-羟基琥珀酰亚胺(NHS)和步骤2) 制得的氨基-介孔硅(MSN-NH2)为原料合成PEG修饰的介孔硅 (MSN-PEG),以摩尔比计,四者的摩尔比为0.167:0.156:0.1:0.083,其中MSN-NH2的摩尔量以SiO2对应的摩尔量计;
4)以1-乙基-(3-二甲基氨基丙基)碳酰二亚胺(EDC)、N-羟基琥珀酰亚胺(NHS)、乳铁蛋白(Lf)和步骤3)制得的PEG修饰的介孔硅(MSN-PEG)为原料,制备乳铁蛋白修饰的介孔硅(MSN-Lf),以摩尔比计,其摩尔比为0.156:0.1:0.177:0.083,其中MSN-PEG的摩尔量以SiO2对应的摩尔量计;
5)将步骤4)制得的乳铁蛋白修饰的介孔硅(MSN-Lf)和小分子探针掺杂静置2小时,离心后得到所述复合物。
第三方面,本发明提供了所述复合物在制备用于β-淀粉样蛋白体内检测试剂方面的应用。
进一步地,由于对β-淀粉样蛋白的检测可用于早起诊断阿尔茨海默症,故本发明还提供了所述复合物在制备阿尔茨海默病诊断试剂方面的应用。
本发明涉及到的原料或试剂均为普通市售产品,涉及到的操作如无特殊说明均为本领域常规操作。
在符合本领域常识的基础上,上述各优选条件,可以相互组合,得到具体实施方式。
本发明的有益效果在于:
本发明开创性的将SZIs小分子探针用于β-淀粉样蛋白的检测,并提出一种经修饰介孔硅/小分子探针的纳米复合物的通用策略,辅助 SZIs、ThT等小分子探针高效率穿过血脑屏障,使得利用这些小分子探针对脑部的β-淀粉样蛋白进行体内实时成像成为可能。

为了能够更清楚地理解本发明的上述目的、特征和优点,下面将对本发明的方案进行进一步描述。需要说明的是,在不冲突的情况下,本发明的实施例及实施例中的特征可以相互组合。
在下面的描述中阐述了很多具体细节以便于充分理解本发明,但本发明还可以采用其他不同于在此描述的方式来实施;显然,说明书中的实施例只是本发明的一部分实施例,而不是全部的实施例。
下面将结合实施例对本发明的优选实施方式进行详细说明。需要理解的是以下实施例的给出仅是为了起到说明的目的,并不是用于对本发明的范围进行限制。本领域的技术人员在不背离本发明的宗旨和精神的情况下,可以对本发明进行各种修改和替换。
下述实施例中所使用的实验方法如无特殊说明,均为常规方法。
下述实施例中所用的材料、试剂等,如无特殊说明,均可从商业途径得到。
其中,下述实施例中所述的SZI-1和SZI-2为参考文献(Marek Cigáň, AntonIvica Sigmundová,Pavol Zahradník,Roman Dědic, Magdaléna Hromadová.Photostability of D–π–A nonlinear optical chromophores containing abenzothiazolium acceptor.Journal of Physical Organic Chemistry,2011,24,450-459.)中记载的“1”物质和“2”物质,可参考文献中的合成方法合成。
实施例1制备乳铁蛋白修饰的介孔硅
1、将2g十六烷基三甲基氯化铵(CTAC),0.08g三乙醇胺(TEA) 作为碱,于开水浴下加热搅拌1h,加入1.5mL正硅酸乙酯(TEOS)反应1h制备介孔硅纳米材料,以摩尔比计,三者摩尔比为1.25:5.36:6.69。透射电镜结果如图2(I)所示,粒径范围40-50nm。MSN的zeta电势为 -20.08mV(图3)。
2、将100μL 3-氨丙基三乙氧基硅烷(APTES)和5mg介孔硅为原料制备氨基-介孔硅(MSN-NH2),以摩尔比计,二者的质量比为0.83: 0.43,其中介孔硅材料的摩尔量以SiO2对应的摩尔量计。透射电镜结果如图2(II)所示,粒径范围40-50nm。MSN-NH2的zeta电势为17.64mV (图3)。
3、将0.1g双羧基聚乙二醇(HOOC-PEG-COOH)、0.032g 1-乙基- (3-二甲基氨基丙基)碳酰二亚胺(EDC)、0.024g N-羟基琥珀酰亚胺的摩尔比为0.167:0.156:0.1:0.083,其中MSN-NH2的摩尔量以SiO2对应的摩尔量计。粒径范围40-50nm。MSN-PEG的zeta电势为19.91mV (图3)。
4、将5mg MSN-PEG、0.03g EDC、0.01g NHS、0.1g乳铁蛋白(Lf)常温搅拌过夜,制备乳铁蛋白修饰的介孔硅(MSN-Lf),以摩尔比计,其(NHS)、5mg MSN-NH2搅拌过夜制备MSN-PEG,以摩尔比计,四者摩尔比为0.156:0.1:0.177:0.083,其中MSN-PEG的摩尔量以SiO2对应的摩尔量计。透射电镜结果如图2(III)所示粒径范围40-50 nm。MSN-Lf的zeta电势为8.80mV(图3)。
实施例2制备乳铁蛋白修饰的介孔硅/小分子探针(SZIs)复合物
将5mg实施例1制备的MSN-Lf和50μg小分子探针SZIs掺杂静置2 小时,离心后得到介孔硅/小分子探针复合物,其粒径约为50nm。透射电镜图如图2(IV)所示,粒径范围为40-50nm,复合物的zeta电势如图3所示,结果为5.67mV。
实施例3乳铁蛋白修饰的介孔硅/小分子探针复合物的性能验证
本实施例用于验证实施例2制备的乳铁蛋白修饰的介孔硅/小分子探针复合物(简称复合物)在穿透血脑屏障方面的能力与程度。
1、定性实验
分别配制47.5μg/mL(MSN-Lf@SZI-1)和40μg/mL(MSN-Lf@SZI-2) 的复合物PBS溶液和小分子探针(两个不同的小分子探针SZIs,分别为 SZI-1和SZI-2)PBS溶液,经小鼠尾静脉注射2mg/kg到小鼠体内,在不同时间点下于IVIS活体成像仪下进行荧光成像,结果如图4所示。
由图4可知,将单独的小分子探针SZIs对经小鼠尾静脉注射后,在小鼠脑部未发现荧光信号,说明单独的小分子探针具有差的BBB穿透率,无法穿过BBB对脑部进行成像,如图4(a)所示。与之形成强烈对比的是,将乳铁蛋白修饰的介孔硅材料/小分子探针复合物经小鼠尾静脉注射后,在小鼠脑部观察到来自小分子探针的荧光信号,说明乳铁蛋白修饰的介孔硅材料/小分子探针复合物可以穿过血脑屏障,对小鼠脑部进行实时成像,如图4(b)所示。
2、定量实验
分别配制47.5μg/mL(MSN-Lf@SZI-1)、40μg/mL(MSN-Lf@SZI-1) 的复合物PBS溶液,以0.13mg/kg注射剂量经小鼠尾静脉注射后,5分钟后给予小鼠安乐死。解剖出小鼠脑部,匀浆处理后,使用乙腈萃取,萃取液经LC-MS测定后,计算探针的血脑屏障穿透率,结果如图5、图 6所示。
其中,图5(a)为小分子探针SZI-1的LC-MS图,图5(b)为小分子探针的标准曲线,图5(c)为小鼠脑部小分子探针的LC-MS图,图5 (d)为小分子探针的质谱,图5(e)为小鼠脑部小分子探针的质谱。
由图5(a)和(b)可知,小分子探针的标准曲线方程为y= 90.661x+11.623。在图5(c)中,我们观察到来自脑部的小分子探针的 LC-MS信号,这说明探针确实到达了脑部。此外,如图5(d)和(e) 所示,我们也观察到来自小分子探针对应的质谱分子量,这证实我们的小分子探针确实被递送到了脑部。
其中,图6(a)为小分子探针SZI-2的LC-MS图,图6(b)为小分子探针的标准曲线,图6(c)为小鼠脑部小分子探针的LC-MS图,图6 (d)为小分子探针的质谱,图6(e)为小鼠脑部小分子探针的质谱。
由图6(a)和(b)可知,小分子探针的标准曲线方程为y=566.05 x-20.01。在图6(c)中,我们观察到来自脑部的小分子探针的LC-MS 信号,这说明探针确实到达了脑部。此外,如图6(d)和(e)所示,我们也观察到来自小分子探针对应的质谱分子量,这进一步证实我们的小分子探针确实被递送到了脑部。
实施例4制备乳铁蛋白修饰的介孔硅/小分子探针(硫磺素-T)复合物
本实施例与实施例2的区别在于:将5mg实施例1制备的MSN-Lf和 50μg硫磺素-T掺杂静置2小时,离心后得到介孔硅/小分子探针复合物,其粒径约为50nm。
实施例5通用策略验证
为了证实本发明的策略是一个通用的策略,我们使用乳铁蛋白修饰的介孔硅材料对市售硫磺素-T进行负载得到实施例4所述复合物,其浓度为37μg/mL(MSN-Lf@ThT),采用与实施例3相同的方法测试硫磺素-T是否可以到达脑部,并使用HPLC对小鼠脑中的硫磺素-T进行测定。
测定结果如图7所示,我们同样从小鼠脑萃取液中观察到来自硫磺素-T的HPLC信号。此外,脑萃取液中的硫磺素-T的存在由其对应的紫外吸收曲线所证实,响应的紫外吸收曲线如图7(e)和图7(f)所示。这些实验结果证实,我们的材料可以有效递送水溶性小分子探针到达脑部,并对脑部的β-淀粉样蛋白进行成像。
实施例6小分子探针在溶液中对Aβ聚集体的荧光响应及解离常数
1、荧光响应
配置50nM的SZI-1和SZI-2的PBS溶液,分别测定SZIs对Aβ聚集体 (20μg/mL),牛血清白蛋白(BSA,10μg/mL)的荧光响应。结果如图8(a)和(c)所示。由图8(a)可知,PBS溶液本身没有荧光,SZI-1 的荧光强度约为50,与BSA结合后的荧光强度与SZI-1本身相差不大。当SZI-1与Aβ聚集体作用后,其荧光发生了显著增强,强度值约为150,大约增强了3倍。由图8(c)可知,PBS溶液本身没有荧光,SZI-2的荧光强度约为200,与BSA结合后的荧光强度相比SZI-2本身略有降低。当 SZI-1与Aβ聚集体作用后,其荧光发生了增强,强度值约为340,大约增强了1.7倍。
2、解离常数
配置一定浓度梯度的SZIs+Aβ聚集体的溶液,浓度梯度分别为20, 30,50,100,200,500,1000,1500nM。然后测定其荧光强度值,使用GraphPad Prism软件计算解离常数Kd,Kd结果可由该软件直接给出。由图8(b)可知,SZI-1与Aβ聚集体的结合曲线的Kd值为507.6±94.75nM。由图8(b)可知,SZI-2与Aβ聚集体的结合曲线的Kd值为 600.6±94.82nM。
实施例7介孔硅/小分子探针复合物对转基因小鼠的脑部Aβ染色确认
以实施例3的定量方法,给予小鼠尾静脉注射,于不同时间点拍摄脑部荧光后,给予小鼠安乐死,解剖出小鼠脑部,制备切片。使用小分子探针染色,确认对Aβ的染色效果。染色结果如图9所示。图9(a) 和(b)分别为两个不同的小分子探针SZIs对转基因小鼠APP/PS1的脑切片染色的结果。使用标准商用试剂硫磺素-T进行对照,并将来自硫磺素-T的荧光信号和小分子探针的信号进行merge处理,merge效果如图9(i)和(j)所示。由该结果可以看出,小分子探针达到了与硫磺素-T相似的染色结果,证实小分子探针确实可以对β-淀粉样蛋白进行染色。

1.一种用于β-淀粉样蛋白体内检测的复合物,其特征在于,所述复合物由经乳铁蛋白、转铁蛋白和其他类型转运受体中至少一种修饰的介孔硅与小分子探针掺杂而成;
所述小分子探针选自硫磺素T和SZIs中的至少一种。
2.根据权利要求1所述的复合物,其特征在于,所述复合物的粒径为40~70nm。
3.根据权利要求2所述的复合物,其特征在于,所述介孔硅与修饰物的摩尔比为83:0.071~75:0.14。
4.权利要求1~3任一项所述复合物的制备方法,其特征在于,将经乳铁蛋白、转铁蛋白和其他类型转运受体中至少一种修饰的介孔硅与小分子探针掺杂,静置,离心后得到所述复合物。
5.根据权利要求4所述的制备方法,其特征在于,将经修饰的介孔硅与小分子探针按25:1~100:1的质量比掺杂。
6.根据权利要求5所述的制备方法,其特征在于,经修饰的介孔硅粒径为40~70nm。
7.根据权利要求6所述的制备方法,其特征在于,介孔硅经乳铁蛋白修饰,修饰后,乳铁蛋白与介孔硅的摩尔比为0.071:83~0.088:83。
8.根据权利要求7所述的制备方法,其特征在于,包括如下步骤:
1)以十六烷基三甲基氯化铵(CTAC)为模板,三乙醇胺(TEA)作为碱,正硅酸乙酯(TEOS)为原料制备介孔硅纳米材料,以摩尔比计,三者摩尔比为1.25:5.36:6.69;
2)以3-氨丙基三乙氧基硅烷(APTES)和步骤1)制得的介孔硅纳米材料为原料制备氨基-介孔硅(MSN-NH2),以摩尔比计,二者的质量比为0.83:0.43,其中介孔硅材料的摩尔量以SiO2对应的摩尔量计;

5)将步骤4)制得的乳铁蛋白修饰的介孔硅(MSN-Lf)和小分子探针掺杂静置2小时,离心后得到所述复合物。
