技术领域
本发明涉及胰酶(pancreatin)和在生产胰酶过程中减少病毒感染性或灭活病毒和微生物内容物的方法。
背景技术
胰酶由在兽医检查后发现适合人类消费的动物的猪胰腺组织生产。胰酶是消化酶的混合物,主要是从猪胰中提取的淀粉酶、蛋白酶和脂肪酶。胰酶由于其重要的治疗特性和高度的安全性,长期以来被用作酶替代疗法的药物制剂。多种胰酶可作为消化酶补充剂商购获得,以帮助消化并增强营养物质的吸收。临床上胰腺酶(pancreatic enzyme)替代疗法是治疗胰腺外分泌功能不全的主要手段,所述胰腺外分泌功能不全与囊性纤维化、慢性胰腺炎、胃肠道旁路手术术后(post-gastrointestinal bypass surgery)、胰腺切除术术后等有关。
得自任何动物或人源材料的所有生物制品的共同特征是病毒污染的风险。生物污染可能来自原料或在生产过程中引入的外来物质。
病毒是仅在其他生物体的活细胞内复制的小的传染原。病毒由被蛋白质壳和脂质层(某些情况下)包围的核酸(RNA或DNA)组成。仅产生蛋白质壳的病毒通常称为无包膜病毒,壳中含有蛋白质和脂质成分的病毒通常称为包膜病毒。它们的尺寸范围从约15nm到约450nm,并且不能用光学显微镜看到。已经通过电子显微镜、NMR波谱和X射线晶体学研究了病毒的形状和结构。
病毒可感染生命形式的所有类型,从动物和植物到细菌和古细菌。由于病毒不能独立复制,它们依赖于宿主。因此,它们几乎存在于世界上所有的生物体中。已知的病毒很少对人有致病性,因为它们具有很高的宿主特异性。几个国家主管部门已经敦促胰酶生产商改善生产工艺的病毒灭活/去除能力;然而,由于大部分可去除或灭活病毒的条件也会导致失活产物(被破坏的酶活性),所以改善的尝试取得有限的成功。规定和安全问题要求生物制药如胰酶活性药物成分(API)中的病毒清除(病毒去除或灭活)。因此,生产来自生物组织的药物产品的公司正面临来自监管机构的额外压力,以通过将所有污染物降至可能的最低水平来提高其产品的安全水平。

迄今为止,还没有研发出可靠的方法去除或灭活胰酶样品中的所有病毒污染物。这是由于胰酶中的活性酶与许多已知的失活条件(包括加热、低pH、氧化和电离辐射)不相容。尽管如此,已经公布了几种灭活或减少病毒和微生物的方法。
Tijsensen等人(US2014/0017223 A1)公开了制备胰腺酶制剂(pancreaticenzyme preparation,PEP)的方法,包括使β方丙内酯(BPL)与含有一种或多种胰腺酶的制剂反应足够的时间以降低制剂中的病毒感染性。

Kurfurst等人(US2009/0233344 A1)公开了一种通过处理具有0.5重量%或更少残留水分的样品来减少样品的病毒和微生物污染的方法,将胰酶在84℃的温度(优选80℃及以下)热处理48小时或30小时,其中所得的胰酶的活性为至少50%的生物活性。所公开的胰酶病毒感染性降低了大于1log10的log10折减系数(reduction factor)。
Mann(US2009/6749851)公开了用于灭菌消化酶制剂的方法,以降低活性生物污染物如病毒、细菌、酵母、霉菌和真菌的水平。包含消化酶的组合物的处理包含通过(a)降低温度、(b)还原溶剂、或(c)向组合物中加入稳定剂,然后照射组合物来稳定组合物。
Becher等人(US2009/0130063 A1)公开了一种从胰酶样品中分离感染性病毒载量、通过使用离心和超离心来定量测定胰酶样品中的病毒载量的方法。该方法仅限于适于离心的液体样品。
Braeuniger等人(Int.J.Hyg.Environ.Health(2000)203:71-75)公开了热灭活牛细小病毒(BPV)的用途。已经证明BPV可以由暴露于热和残留水分而被失活。然而Braeuniger等人没有公开加热对酶活性和组成变化的任何影响。
Lewis(US1971/3,956,483)公开了胰酶加工和细菌减少的同时保持淀粉分解、蛋白水解和脂肪分解活性的方法。该方法包括将胰酶加热至49-82℃之间的足够高的温度。然而,Lewis没有提供灭活或减少病毒数量的方法。
特别的挑战是灭活或减少来自活性物质是酶混合物的生物提取物基质的病毒,而在该过程中不破坏或改变蛋白质的酶活性或比例。需要一种方法,其中含有固体的生物提取物中的病毒和细菌内容物被最大可能程度地降低或最小化,并与保持所需的药物活性成分的纯度、组成和效价一致。该方法必须同样适用于固体和悬浮液。
根据ICH Q5A:“Viral Safety Evaluation of Biotechnology ProductsDerived from Cell Lines of Human or Animal Origin”,本发明研发的方法需要减少或灭活源于生物产品中的病毒,同时需要维持酶活性和组成比例(例如脂肪酶、蛋白酶和淀粉酶)处于可接受的水平。原则上,药物活性剂不应含有感染性病毒。目前的生产过程不能以足够的安全限度(safety margin)减少或灭活潜在存在的无包膜病毒,并且必须实施额外的病毒减少步骤。
PAA被认为是能够失活多种细菌(Cronmiller,J.R,等人(1999),Efficacy ofConventional Endoscopic Disinfection and Sterilization Methods AgainstHelicobacter pylori Contamination.Helicobacter 4:198-203.)、真菌(Werner andWewalka(1973),Oxidation of vitamin A alcohol with peraceticacid.Tetrahedron29:47-50)和病毒(Kline and Hull(1960),The Virucidal Propertiesof Peracetic Acid.American Journal of Clinical Pathology 33:30-33)的有效消毒剂。
过乙酸(PAA)也被用作喷施水果和蔬菜的杀菌剂(Greenspan and MacKeller(1951),"The Application of Peracetic Acid Germicidal Washes to Control Moldof Tomatoes(Food Technology 5:95-97)。
Baldry(J.Applied Bacteriology.(1983)54:417-423)报道了使用含有1.3mmol/L的PAA的溶液在25℃在1分钟内使蔬菜细菌的数量减少了106倍。

发明概述
本发明提供了减少或灭活生物提取物的微生物和病毒污染的方法,而不影响生物提取物中所含酶的活性并且不改变生物提取物中所含酶的组成。此外,该方法在最终产品中不产生任何有毒化合物或残留物。
在一个实施方案中,提供了胰酶制剂(preparation)。该制剂具有降低的病毒感染性,并且包含一种或多种胰酶中的酶和PAA。在一些实施方案中,该制剂包含一个或多个猪胰腺。
在一些实施方案中,胰酶制剂的脑心肌炎病毒(EMC)的病毒感染性比未用PAA处理的胰酶对照样品的病毒感染性低至少1log10;鼠细小病毒(MMV)的病毒感染性比未用PAA处理的胰酶对照样品的病毒感染性低至少1log10;猪细小病毒(PPV)的病毒感染性比未用PAA处理的胰酶对照样品的病毒感染性低至少1log10,和/或无包膜病毒的病毒感染性比未用PAA处理的胰酶对照样品的病毒感染性低至少1log10。
在一些实施方案中,至少一种胰酶中的酶衍生自动物来源。

在另一个实施方案中,提供了一种生产胰酶制剂的方法。该方法包括提供胰腺和使胰腺与PAA在反应中反应的步骤。与PAA的反应降低了病毒感染性并减少了胰酶制剂中的细菌计数。
在一些实施方案中,反应后胰酶制剂中的无包膜病毒的病毒感染性与PAA反应之前的胰腺的无包膜病毒的病毒感染性相比低至少1log10。

本发明任何实施方案中的胰酶和/或胰腺酶可以衍生自动物来源(阉猪、公牛、羊(sheep)、猪、牛等)。胰酶中的酶包括但不限于脂肪酶、蛋白酶和淀粉酶。

具体实施方式
本文所述的方法包括用一定浓度的过乙酸处理动物来源的所需提取物(例如胰腺)。没有限制,动物胰腺可以是牛或猪胰腺。有利地,通过本文所述的方法产生的所得胰酶以及包含所述胰酶的药物组合物可以通过实验室规模、中试规模和生产规模操作获得。
胰腺的过乙酸处理(PAA):本发明涉及一种在胰酶生产过程中减少或灭活病毒和微生物内容物的方法,包括将动物胰腺例如猪胰腺用一定浓度的可用于冷灭菌或去污染的试剂(如过氧化氢、过碳酸盐、过苯甲酸、过氧酸、过乙酸等)处理。所述方法可以进一步包括提供胰酶产品,例如来自经处理的腺的胰酶API。
PAA的灭菌效力与其迅速渗透到微生物中以及释放对微生物酶的氧化和破坏至关重要的氧和自由基有关。Pruss等人(1999;"Virus Safety of Avital Bone TissueTransplants:Evaluation of Sterilization Steps of Spongiosa Cuboids Using aPeracetic Acid-Methanol Mixture".Biologicals 27:195-201)。当在室温(RT)向掺加病毒的海绵状立方体加入1%PAA/24%乙醇(PES)4小时,报道的滴度减少了超过4log10,有效地灭活了所测试的(人免疫缺陷)病毒2型(HIV-2)、甲型肝炎病毒(HAV)、脊髓灰质炎病毒(PV-1)、伪狂犬病病毒(PRV)、猪细小病毒(PPV)和牛病毒性腹泻病毒(BVDV)的大多数病毒,除了甲型肝炎病毒(HAV)。PAA浓度在<100ppm时在不到5分钟的时间内使革兰氏阳性和革兰氏阴性细菌、真菌和酵母失活。对于病毒,剂量范围很宽(100-40000ppm)。
过乙酸(PAA)是式为CH3CO3H的有机化合物。这种有机过氧化物是一种无色的液体,具有令人联想到醋酸的辛辣气味的特点。它是一种强氧化剂,具有很强的腐蚀性,由乙醛自氧化工业生产。在有或没有共溶剂如醇的情况下,PAA可用于本文所述的反应中。
根据本发明的实施方案,根据本领域技术人员已知的方法,在提取胰酶API之前,用PAA预处理动物腺体。预处理由将整个动物腺体浸入含有PAA的溶液中组成,从而降低病毒活性和细菌计数。在其他实施方案中,预处理可由将切片的、切碎的或磨碎的动物腺体浸入含有PAA的溶液中组成,从而降低病毒活性和细菌计数。动物腺体的预处理可在不同浓度的PAA、温度、pH和时间长度下进行。
在优选的实施方案中,使用PAA浓度在约100ppm至40000ppm的水溶液进行预处理。在更优选的实施方案中,使用约500ppm至约25000ppm的PAA浓度。在一些实施方案中,可使用100-5000ppm、5000-10000ppm、10000-20000ppm、20000-30000ppm、30000-40000ppm或大于40000ppm的PAA浓度。
在优选的实施方案中,PAA预处理在约1.0至约5.5的pH进行。在更优选的实施方案中,PAA预处理在约2.0至约5.0的pH进行。
在优选的实施方案中,PAA预处理在约室温进行。在一些实施方案中,PAA预处理在约15℃和22℃的温度进行。在其他实施方案中,PAA预处理可以用含有PAA的冷藏溶液进行。在这类实施方案中,PAA预处理可以在刚好高于水溶液的冰点至室温的温度范围内进行,例如约4℃。
在优选的实施方案中,PAA预处理进行约0.5分钟至约1小时的时间。在更优选的实施方案中,PAA预处理进行约5分钟至约30分钟的时间。在一些实施方案中,可以进行0.5-5分钟、5-10分钟、10-20分钟、30-60分钟或超过60分钟的PAA预处理。
在PAA预处理之后,经处理的动物腺体可以用作提取所需酶产品的来源材料,如本领域通常已知的。
在优选的实施方案中,动物腺体是胰腺,特别是猪胰腺。其他动物的腺体、器官和组织也可用PAA预处理,包括但不限于肺、脑、胆囊、垂体、肠和肠粘膜、肾上腺、胆汁和胸腺,用于生产其他动物衍生产品,包括酶产品。
用PAA对动物腺体进行预处理是特别有利的,因为预处理显著降低了经处理的腺体材料上的病毒感染性和细菌计数。出乎意料地,与由未经PAA预处理的动物腺体用相同方法产生的提取物的酶谱相比,由PAA处理的动物腺体产生的酶产品没有显著改变提取物的酶谱。
术语“USP单位”是指用于测量胰酶中存在的活性酶的效价的单位。
淀粉酶活性的1USP单位是包含在以初始速率分解淀粉的胰酶的量,使得在USP淀粉酶活性测定的条件下每分钟0.16μEq的糖苷键被水解。
脂肪酶活性的1USP单位是包含在胰酶中的量,使得在脂肪酶活性测定的条件下于37℃在pH9.0下每分钟释放1.0μEq的酸。
蛋白酶活性的1USP单位是包含在胰酶中的量,其在蛋白酶活性测定的条件下以初始速率水解酪蛋白,使得在每分钟释放的不被三氯乙酸沉淀的肽的量在280nm处的吸光度与15nmol的酪氨酸相同。

进行病毒清除发明是为了建立用于减少和/或灭活可能以已知的或未知的污染物存在的病毒的方法的能力和效力。这些实验是通过掺加病毒并在随后的过程步骤中测量其减少或灭活来进行的。病毒清除实验在提供病毒学工作的实验室中进行。
实施例1:
为了制备胰酶,将3kg解冻的全猪胰腺和3kg猪胰腺切成约2英寸的条,在室温(18-22℃)将其浸入两种不同浓度的PAA中,分别为500ppm和2000ppm达10分钟和15分钟。用工艺用水冲洗经处理的腺体,并将其置于无菌样品袋中并在-20℃保存冷冻不少于12小时。每次测试总共6个样品。制备进行2轮。第二轮不包括条状腺体,因为在第一轮中观察到条状腺体变得难以进一步处理。

处理后,用本发明的方法的额外步骤加工腺体以获得胰酶粉末,其可包括:1)冷冻的全胰腺和2英寸的条状腺体从冷冻材料研磨;2)磨碎的材料被活化,然后活化淬灭;3)与不溶物分离;4)胰酶沉淀;5)洗涤和分离;6)干燥固体残留物;7)粉磨;和8)获得干燥的非无菌的活性药物成分(API)胰酶终产物。
本文提供的实施例是测定PAA对全猪胰腺和2英寸条状猪胰腺的作用。测试两组腺体的好氧微生物总数(TAMC)、(减少生物负荷)以及在PAA处理之前和之后的病毒灭活。测试成品样品的酶活性。对于经处理的全腺体,PAA处理显著减少了TAMC(表1)。未经处理的腺体具有24000和20000CFU/g的计数,并且在PAA处理之后,TAMC结果减少到10和90CFU/g。对于条状腺体也是如此。未经处理的条状腺体具有9700CFU/g的TAMC结果,经处理的腺体的计数减少到130和45CFU/g。全腺体成品样品的TAMC结果略有下降。对照样品为50和290CFU/g。用500ppm和2000ppm的PAA处理后,结果为10-25CFU/g。条状腺体保持一致。对照样品<10CFU/g,经处理的腺体为30和10CFU/g(表2)。

评估潜在的过程变化的相关分析是对潜在的酶降解的评估。表1和表2中给出的数据表明,在选定浓度的PPA下,缺乏显著的酶降解。图1显示用PAA处理之前和之后样品中TAMC减少1至2log10。

实施例2:
为了制备胰酶,将3kg解冻的全腺体和3kg磨碎的腺体在室温(18-22℃)浸入两种不同浓度的PAA中,分别为500ppm和2000ppm达10分钟和15分钟。用纯化水冲洗经处理的腺体,并将其置于无菌样品袋中并在-20℃保存冷冻不少于12小时。每次测试总共6个样品。制备进行2轮。由于磨碎的腺体在PAA处理后形成浆液而变得难以处理,第二轮不包括磨碎的腺体,。

处理后,如实施例1中所述用本发明方法的额外步骤处理腺体以获得胰酶粉末。在PAA处理之前,全腺体和磨碎的腺体具有相似的TAMC结果,为1400和1100CFU/g。经过500ppm和2000ppm的PAA处理后,经处理的全腺体的TAMC结果完全相同,为10CFU/g。PAA处理后没有测试磨碎的腺体。见表3的结果。

实施例3:
测量病毒感染性活性:可以通过本领域已知的任何方法测量初始的和经处理的胰酶中的病毒活性。优选地,在经处理的胰酶中测量一种或多种病毒的病毒感染性。在一个实施方案中,测量PCV2、PPV和/或其他病毒的病毒感染性。
在一个实施方案中,感染性测定使用在组织培养孔中生长的细胞培养物,在孔中加入样品并测量细胞的感染。
根据所述生产方法用500ppm和2000ppm PAA溶液处理胰酶全腺体生产胰酶API样品。表5中列出的测试样品是溶解在含有Mg2+和Ca2+的Dulbecco磷酸盐缓冲盐水(PBS)中的胰酶API样品,以产生10%悬浮液用于测定感染性猪圆环病毒2型(PCV 2)的滴度,所述测定使用PK-13细胞(猪肾细胞)通过基于细胞的体外测定进行。在干扰测定中分析的测试项目掺加3000TCID50/mL的PCV-2。使用最终以1:800稀释的每种溶液进行分析。滴度由大容量铺板(Large Volume Plating)测定。此外,在比较终点滴定测定中评估测试项目对PCV-2感染的干扰作用。测试项目没有显示对PCV-2感染的负面干扰作用。结果总结于表5。

实施例4:
发现500ppm和2000ppm的PAA表面处理减少好氧微生物总数(TAMC)。然而,如通过PCV-2感染性测试(对内源性病毒)所测量的,500和2000ppm的PAA表面处理没有可辨别的效果。因此测试了用于表面处理的PAA的增加的浓度。在图2的流程图中详细描述了包括该过程的表现、掺加(spiking)病毒和用PAA处理腺体的实验。
对于具有较高浓度的PAA的病毒灭活或减少实验,选择模式病毒EMC、MMV和PPV。
EMC是小的无包膜病毒(20-30nm),具有属于小RNA病毒科(Picornaviridae)家族的RNA基因组,并且是二十面体结构。它具有相对的热稳定性、耐有机溶剂、非离子型洗涤剂、且在酸性pH下稳定。这种病毒是人类小RNA病毒如脊髓灰质炎病毒、甲型肝炎病毒和鼻病毒(rhino virus)的代表。
MMV是小的无包膜病毒(18-24nm),具有属于细小病毒科(Parvoviridae)家族的DNA基因组,并且是二十面体结构。它耐高温、脂质溶剂,且在酸性pH稳定。这种病毒被认为是对多种物理和化学疗法的严格测试,并且是在世界各地人群中以高血清阳性率发现的人类细小病毒和圆环病毒的代表模式。
猪细小病毒(PPV)是细小病毒科家族成员,具有单链DNA作为基因组。PPV与生殖问题有关,包括流产、死胎、小窝、新生儿死亡和弱仔猪。PPV在猪的肠内正常繁殖而不会引起临床症状。血清阴性的母畜在妊娠前半期被感染且病毒穿过胎盘时发生疾病。它对大多数消毒剂有抗性。
所进行的实验是测定纯化过程是否使病毒失活,如在适当细胞系中50%组织培养感染剂量(TDID50)终点测定法的变化所测量的。测量每个样品的pH值以确保pH在6-8之间。使用测定稀释剂(含有2%FBS的EMEM)根据细胞毒性/病毒干扰结果稀释样品。表6中描述测定方法。在对收集的样品进行TDID50测定之前,将合适的细胞接种到板中。在接种前观察板的细胞毒性。所有对照稀释每孔均接种50μl,8个重复孔。所有测试物品稀释都接种在8个重复孔中。

阳性对照样品在测试的每一天给出每种病毒的病毒滴度±1.0log10的检定滴度,证实测定中的病毒在预定的规范内进行。阴性对照显示正常的形态和汇合,以及每种病毒没有病毒诱导的细胞病变效应(CPE),证实检测细胞系在预定的规范内进行。
当分析PPV病毒的干扰数据时,确定在所有测试的非细胞毒性稀释中都存在显著的干扰。未稀释的和第一次稀释(1:5)回收样品具有细胞毒性,而第二次和第三次连续稀释(分别为1:25和1:125)回收样品的病毒滴度大约低于预期值1.8log10。测试品来源于猪,PPV是猪病毒。病毒干扰很可能是由于宿主中存在的中和抗体。这种干扰不太可能通过进一步稀释而得到缓解。病毒干扰的存在妨碍了滴定的精确测量,从而妨碍了清除的评估。用PPV进行的实验部分被终止。
评估纯化过程减少或失活掺入各个负荷材料中的脑脊髓炎病毒(EMC)和鼠细小病毒(MMV)的能力。使用回收对照、0分钟样品和10000ppm、30分钟样品计算清除。EMC的测试结果显示在15分钟处约1.75log10的折减系数,在使用30分钟样品计算时,约0.8log10。MMV测试结果显示显著的3.0±0.9log10折减系数,如图3中的动力学图所示。使用回收对照、0分钟样品和10000ppm 30分钟样品计算减少。
实施例5:
电子显微镜研究:参照图4A-4D,进行透射电子显微术(TEM)和扫描电子显微术(SEM)以显示用不同浓度PAA处理的胰腺组织和胰酶粉末表面上的细菌和病毒的存在,并与未处理的(对照)样品进行比较。
用TEM和SEM分析用PAA处理的胰腺组织和未处理的样品以及用PAA处理产生的所得胰酶粉末。对于SEM,用导电材料的超薄镀层涂覆样品,通过低真空蒸汽溅射镀层(low-vacuum sputter coating evaporation)沉积在样品上。目前使用的镀层中的导电材料是金,以观察细菌和病毒。使用ZEISS LEO-1530DSM扫描电子显微镜来产生图4A和4B的图像。图4A显示用20000ppm PAA处理30分钟的胰腺组织,并且没有显示细菌活性的迹象(比例尺为2μm)。图4B显示未处理的胰腺组织表面存在丰富的细菌菌落(比例尺为10μm)。病毒尺寸非常小(15-100nm),在SEM中不可能观察到。对于TEM,用20000ppm PAA处理胰腺组织和胰酶粉末样品30分钟,对照样品用2%乙酸双氧铀(aq)染色。使用型号H-7600HITACHI透射电子显微镜(version 3.01)来产生图4C和4D的图像。观察每个处理的三张TEM载玻片。每个TEM载玻片有400个网格来观察。每张载玻片最少有10-15个网格被观察。胰酶对照样品(169)在胰酶粉末和胰酶表面未处理的腺体组织中确实显示存在病毒。图4C是未经处理的胰酶粉末样品(样品169)的TEM图像,显示病毒的存在并由箭头指示(比例尺为20nm)。图4D是未经处理的胰腺表面组织样品的TEM切片,显示病毒的存在并由箭头指示(比例尺为200nm)。在胰酶粉末样品ID#166、167和168中未发现病毒。在用20000ppm PAA处理30分钟的胰腺腺组织中没有观察到病毒。
实施例6:
研发了在加工之前用PAA处理猪胰腺的方法,以在生产过程中减少外源材料和灭活病毒而不影响胰酶的酶性能。该实验的目标是证明等效的酶活性,不管是否存在腺体的预处理。此外,经过病毒灭活处理的腺体通过加工减少病毒载量,并且在最终产品中减少病毒载量而不影响酶活性(组成和效价)。通过不同的研究测量不同腺体处理的病毒载量。该研究只测量了整个过程中腺体的酶活性。该过程重复了八次以完全限定所述过程。
在该研究中测试了三种处理条件。第一种处理条件将腺体以1:1(腺体重量:PAA溶液重量)的比例浸入静态的10000ppm过乙酸溶液中5分钟和30分钟的处理时间(样品163和164)。第二种处理条件将腺体浸入搅拌的20000ppm过乙酸溶液中,其比例为1:10(腺体重量:PAA重量),腺体固定在处理笼中并每5分钟倒置。第三种处理条件将腺体浸入搅拌的40000ppm过乙酸溶液中30秒,其比例为1:10(腺体重量:PAA重量)。然后将腺体从过乙酸溶液中取出,使过乙酸在腺体表面上反应30分钟。在所有三种情况下,将腺体用水冲洗、置于容器中并冷冻,然后进行胰酶提取过程。
然后将腺体加工并冷冻干燥。粉磨所得材料以产生平均粒径为180μm的胰酶粉末。分析胰酶粉末的胰酶活性。在表7中总结了每个运行的过程参数。

实施例:7
用PAA处理的所有样品产生可接受的酶活性。用于鉴定这些样品的RP-HPLC色谱图指示了胰酶的适当组成(参见图5,用于比较可接受的胰酶的色谱图)。
使用总过氧化物残余量的极限测试来检查胰酶API中任何剩余的PAA的存在。该方法的检测极限为溶液中2ppm、样品中200ppm H2O2和样品中100ppm总O·(氧自由基)。PAA处理和冲洗样品显示未检测到,指示PAA可从胰酶中去除至低于检测水平。
基于剩余批次的这些分析结果,样品的过乙酸处理条件不影响最终产品的性能或组成、纯度、鉴定和效价。
实施例8:
然后测试胰酶样品用于病毒评估。表8总结了这些评估的结果。所有用于特定病毒测定的样品测试均为阴性,并且低于PPV测定的检测极限。显示处理间差异的唯一样品是PCV2测定,其中用20000ppm过乙酸处理的样品显示出比其他样品更低的值,虽然解冻的对照样品显示出与用20000ppm过乙酸处理的样品之一相当的值。尚不清楚初始腺体的细菌和病毒载量是否相等。然而,用20000ppm过乙酸处理30分钟的腺体产生的样品显示了等于或低于任何其他样品的PCV2测定结果。

实施例9:
然后使用两种不同的专利胰酶提取过程(指定为“方法1”和“方法2”)将所得腺体转化成胰酶,产物产率为9-15%。无论是否用过乙酸处理腺体,胰酶样品的活性都相似(参见表7)。处理条件高达40000ppm的过乙酸、浸泡时间长达30分钟也不影响产生的胰酶的活性或效价。得自使用方法1和方法2从胰酶提取过程的结果被一并呈现在图6中。此外,所有样品的反相HPLC鉴定测试结果对胰酶都是阳性的(见图5)。
病毒结果表明,所有样品不论是用过乙酸处理还是未处理,对于所有测试的病毒组(viral panel)均检测为阴性,并且低于PPV测定的检测极限。用20000ppm过乙酸处理的样品显示与PCV2测定相等或较低的测定值。
注意PPV和PCV2不是病毒灭活效力的良好指标,因为病毒的研究滴度是未知的。PCR结果不是病毒感染性的指示。

1.过乙酸用于通过包括以下步骤的方法制备胰酶制剂的用途:
(a)提供胰腺或组织;
(b)在酶提取之前使胰腺或组织与过乙酸在反应中反应;以及
(c)提取胰酶中的酶;

6.权利要求1或2所述的用途,其中所述胰酶制剂具有的鼠细小病毒(MMV)的病毒感染性比未用过乙酸处理的胰酶对照样品的病毒感染性低至少1log10。
7.权利要求1或2所述的用途,其中所述胰酶制剂具有的猪细小病毒(PPV)的病毒感染性比未用过乙酸处理的胰酶对照样品的病毒感染性低至少1log10。
8.权利要求1或2所述的用途,其中所述胰酶制剂具有的无包膜病毒的病毒感染性比未用过乙酸处理的胰酶对照样品的病毒感染性低至少1log10。
9.权利要求1或2所述的用途,其中所述至少一种胰酶中的酶选自脂肪酶、蛋白酶和淀粉酶。
10.权利要求1或2所述的用途,其中所述制剂包括胰酶活性药物成分(API)。
11.权利要求10所述的用途,其中所述胰酶API为粉末形式。
12.权利要求10所述的用途,其中所述胰酶API具有降低的病毒感染性和减少的细菌计数。
13.权利要求10所述的用途,其中所述胰酶API包括至少2.0USP单位脂肪酶、至少25USP单位蛋白酶和至少25USP单位淀粉酶。
14.权利要求10所述的用途,其中所述胰酶API具有的PPV、EMC和MMV的病毒感染性比未用过乙酸处理的胰酶API对照样品的病毒感染性低至少1log10。
15.权利要求10所述的用途,其中所述胰酶API具有的MMV病毒感染性比未用过乙酸处理的胰酶API对照样品的病毒感染性低至少2log10。
16.权利要求10所述的用途,其中所述胰酶API具有的MMV病毒感染性比未用过乙酸处理的胰酶API对照样品的病毒感染性低2log10至4log10。
