Abstract
本发明公开了一种白刺果多糖干燥方法,包括以下步骤:取白刺果,在其中加入水,然后水浴加热后过滤,得到过滤浆液;在所述过滤浆液中加入辅料,加热并搅拌均匀,得到混合物料;采用喷雾干燥法处理所述混合物料,得到纯化的白刺多糖干粉。本发明的白刺果多糖干燥方法操作简单,能耗小,时间短,不破坏多糖结构与活性,产品品质好,成本低,环境污染小,产物总收率>85%,产物中的多糖含量>80%。

技术领域
本发明涉及一种干燥方法,尤其涉及一种白刺果多糖干燥方法。
背景技术
白刺为蒺藜科白刺属灌木,由林奈1759年命名,在青海柴达木盆地有大量的分布,其抗逆性强,耐干旱、耐盐碱特点,能适应严酷恶劣的环境,是典型的沙漠植物。白刺除了固沙等生态作用外,还具有良好的药用价值,如滋阴润肺、解表安神,是蒙、藏、回等少数民族的传统地道药材。由于青海省柴达木盆地受现代工业化污染较少,人口稀疏,环境十分洁净,此处白刺具有纯天然、绿色、无污染等特点,是现代食品、药品和保健品的原辅料来源,白刺果即可生食,也可熟食,经检测白刺营养成分相当丰富,具有很高的开发价值。
从白刺果中提取水溶性多糖,使其资源得到充分利用。多糖为天然有机化合物中的大分子,由单糖通过糖苷键连接而成。越来越多的国内外研究证明活性多糖具有抗肿瘤、增强免疫力、抗氧化、降血糖等作用。本研究对白刺果多糖进行纯化精制,获得高纯度白刺果多糖,对白刺果资源的开发利用具有重要意义。
干燥是白刺果多糖产品工业化重要工序之一,它直接关系到最终产品的质量。干燥的方式有很多,如喷雾干燥、冷冻干燥、真空干燥、微波干燥、热泵干燥等,其中喷雾干燥由于成本较低,工艺简单,方便快捷,可直接干燥制粉等优点,在食品、医药、化工等行业得到广泛的应用。
目前,多糖类干燥主要采取真空冷冻干燥,例如专利CN103772518 A中采用冷冻干燥法干燥山药多糖;专利CN101899120 A中采用冷冻干燥法干燥红枣多糖。冷冻干燥可以较好的保护多糖的活性,但是其干燥时间长,能耗高,且干燥后产品呈块状或棒针状,因此制约着多糖工业化生产。
发明内容
发明目的:本发明旨在提供一种品质好、效率高、能耗小的白刺果多糖干燥方法。
技术方案:本发明的白刺果多糖干燥方法,包括以下步骤:
(1)取白刺果,在其中加入水,然后水浴加热后过滤,得到过滤浆液;
(2)在所述过滤浆液中加入辅料,加热并搅拌均匀,得到混合物料;
(3)采用喷雾干燥法处理所述混合物料,得到纯化的白刺多糖干粉。
进一步地,步骤(1)中,水浴加热温度为50~100℃,优选80℃~100℃。
进一步地,步骤(1)中,过滤温度为20~80℃,优选40℃~60℃。
进一步地,步骤(2)中,所述辅料为麦芽糊精和木薯淀粉,其中麦芽糊精和木薯淀粉的质量比例为1~5:1,优选2~3:1。
进一步地,步骤(2)中,所述过滤浆液与辅料的质量比例为50~200:1,优选100:1。
进一步地,步骤(2)中,搅拌时间为10~30min。
进一步地,步骤(3)中,所述喷雾干燥法的参数为:风温度为115~155℃,进料量为10~30ml/min,进气压力为0~0.2MPa,出风温度为80~100℃。
有益效果:与现有技术相比,本发明具有如下显著优点:白刺果多糖干燥方法操作简单,能耗小,时间短,不破坏多糖结构与活性,产品品质好,成本低,环境污染小,产物总收率>85%,产物中的多糖含量>80%。
具体实施方式
实施例1
白刺果多糖干燥方法包括:
(1)取白刺果2kg,加入去离子水10升,充分搅拌均匀,水浴煮沸2小时,其中水浴温度为100℃。趁热过滤去除不溶物,过滤温度80℃,得白刺果多糖原浆液11升;
(2)将白刺果多糖原浆液11升,加入55g辅料,辅料比例为麦芽糊精:木薯淀粉=1:1,加热搅拌10min;
(3)将11L加入辅料的多糖原浆液直接进料,喷雾干燥工艺为进风温度130℃,进料量12ml/min,进气压力0.05MPa,出风温度90℃。获得高含量的白刺果多糖成品271g,呈紫红色粉末状,不粘壁,检测后得多糖含量为87.0%,含水量4.56%,平均粒径9.56μm。
实施例2
白刺果多糖干燥方法包括:
(1)取白刺果200克,加入去离子水1000毫升,充分搅拌均匀,水浴煮2小时,其中水浴温度为80℃。趁热过滤去除不溶物,过滤温度为60℃,得白刺果多糖原浆液1100毫升;
(2)将白刺果多糖原浆液1100毫升,加入11g辅料,辅料比例为麦芽糊精:木薯淀粉=3:1,加热搅拌10min;
(3)取500ml加入辅料的多糖原浆液直接进料,喷雾干燥工艺为进风温度115℃,进料量10ml/min,进气压力0.1MPa,出风温度80℃。获得高纯度的白刺果多糖成品29g,呈紫红色粉末状,不粘壁,检测后得多糖含量为87.2%,含水量4.35%,平均粒径9.16μm。
实施例3
白刺果多糖干燥方法包括:
(1)取风干的白刺果10kg,加入去离子水50升,充分搅拌均匀,水浴煮2小时,其中水浴温度为90℃。趁热过滤去除不溶物,过滤温度为40℃,得白刺果多糖原浆液55升;
(2)将白刺果多糖水提取液55升,加入1.1Kg辅料,辅料比例为麦芽糊精:木薯淀粉=2:1,加热搅拌20min;
(3)取20L含辅料的多糖原浆液直接进料,喷雾干燥工艺为进风温度120℃,进料量20ml/min,进气压力0.15MPa,出风温度87℃。获得高纯度的白刺果多糖成品780g,呈紫红色粉末状,不粘壁,检测后得多糖含量为88.1%,含水量为4.18%,平均粒径9.36μm。
实施例4
白刺果多糖干燥方法包括:
(1)取风干的白刺果100kg,加入去离子水500升,充分搅拌均匀,水浴煮2小时,其中水浴温度为50℃。趁热过滤去除不溶物,过滤温度为20℃,得白刺果多糖原浆液570升;
(2)将白刺果多糖水提取液570升,加入5Kg辅料,辅料比例为麦芽糊精:木薯淀粉=5:1,加热搅拌30min;
(3)取100L含辅料的多糖原浆液直接进料,喷雾干燥工艺为进风温度155℃,进料量30ml/min,进气压力0.2MPa,出风温度100℃。获得高纯度的白刺果多糖成品3.61kg,呈紫红色粉末状,不粘壁,检测后得多糖含量为81.0%,含水量为4.75%,平均粒径12.1μm。
实施例5
白刺果多糖干燥方法包括:
(1)取风干的白刺果10kg,加入去离子水50升,充分搅拌均匀,水浴煮2小时,其中水浴温度为70℃。趁热过滤去除不溶物,过滤温度为40℃,得白刺果多糖原浆液52升;
(2)将白刺果多糖水提取液52升,加入500g辅料,辅料比例为麦芽糊精:木薯淀粉=4:1,加热搅拌30min;
(3)取50L含辅料的多糖原浆液直接进料,喷雾干燥工艺为进风温度135℃,进料量10ml/min,进气压力0.01MPa,出风温度95℃。获得高纯度的白刺果多糖成品1.77kg,呈紫红色粉末状,不粘壁,检测后得多糖含量为82.4%,含水量为5.18%,平均粒径11.3μm。
表1实施例1-5的白刺果多糖成品
由上表可知,通过本方干燥的白刺果多糖,含水率最低可达4.18%,说明干燥效果好,产品成粉末状且颗粒细小,未出现块状或棒针状,成品多糖含量高,多糖的结构与活性未遭到破坏。
Claims (7)
1.一种白刺果多糖干燥方法,其特征在于,包括以下步骤:
(1)取白刺果,在其中加入水,然后水浴加热后过滤,得到过滤浆液;
(2)在所述过滤浆液中加入辅料,加热并搅拌均匀,得到混合物料;
(3)采用喷雾干燥法处理所述混合物料,得到纯化的白刺多糖干粉。
2.根据权利要求1所述的白刺果多糖干燥方法,其特征在于,步骤(1)中,水浴加热温度为50~100℃。
3.根据权利要求1所述的白刺果多糖干燥方法,其特征在于,步骤(1)中,过滤温度为20~80℃。
4.根据权利要求1所述的白刺果多糖干燥方法,其特征在于,步骤(2)中,所述辅料为麦芽糊精和木薯淀粉混合物,其中麦芽糊精和木薯淀粉的质量比例为1~5:1。
5.根据权利要求1所述的白刺果多糖干燥方法,其特征在于,步骤(2)中,所述过滤浆液与辅料的质量比例为50~200:1。
6.根据权利要求1所述的白刺果多糖干燥方法,其特征在于,步骤(2)中,搅拌时间为10~30min。
7.根据权利要求1所述的白刺果多糖干燥方法,其特征在于,步骤(3)中,所述喷雾干燥法的参数为:风温度为115~155℃,进料量为10~30ml/min,进气压力为0.05~0.2MPa,出风温度为80~100℃。
